무 쥬스 제조를 위한 starter로써 동치미에서 분리한 유산균의 동정 및 발효 특성 (Identification and Fermentation Characteristics of Lactic Acid Bacteria Isolated from Dongchimi as Starter for Radish Juice)

  • 본 연구는 동치미에서 유산균을 분리하여 동치미의 가능성이 강화된 무 주스를 제조하기 위한 starter로써의 가능성을 검토하기 위하여 유산균의 발효특성을 조사하였다. 동치미에서 분리 동정된 40 균주는 당 발효능과 양상에 따라 5개의 group으로 분류되었다. Species별 분포를 살펴본 결과는 Leuconostoc mesenteroides, Lactobacillus brevis, Lactobacillus sake, Lactobacillus plantarum, Lactobacillus fermentum의 5개 균종이었으며, 유산균의 분포는 Leuconostoc mesenteroides가 52.5%로 최 우점종으로 나타났다. 분리된 5개의 group에서 가장 생육능이 왕성한 1개 균주씩 Leuconostoc mesenteroides J-9, Lactobacillus brevis J-12, Lactobacillus plantarum J-39, Lactobacillus fermentum J-7 등 5균주를 선정하여 한외 여과한 무즙에 각각 0.3($10^6$ cfu/㎖)%씩 접종하여 25${\circ}C$에서 9일간 숙성하면서 pH 및 총 산도, 비휘발성유기산함량의 변화 등을 조사하였다. 각 접종 균주 중 Lactobacilus plantarum의 생육능이 가장 빠르게 나타났으며 Lactobacillus sake가 가장 느리게 성장하였다. 발효 개시 전의 한외 여과한 무즙의 pH 및 총 산도는 6.30~6.36, 0.09~1.0%이었으나 9일째 pH 3.2~4.3, 0.85~1.2%로 나타났다. 각 접종 균주의 pH, 산도의 변화를 비교해보면 미생물의 생육능과 상관성이 높아 유산균수의 증가속도가 빠를수록 pH는 빠르게 감소하고, 산도는 빠르게 증가하였다. 비휘발성 유기산의 변화를 보면 모든 접종구에서 시트르산, 말산, 말론산, 숙신산은 감소하는 경향으로 나타났으며 락트산은 발효가 진행됨에 따라 증가하였다. 관능적으로 맛에 좋은 영향을 주는 유기산으로 알려진 숙신산과 락트산의 함량을 보면 L. mesenteroides J-9가 가장 높게 나타났다. 락트산 단독의 경우는 L. plantarum J-39m, L. brevis J-12 접종구의 순으로 높게 나타났다. 이 ${\cdot}$ 화학적 검사와 향미에 대한 관능검사결과 L. mesenteroides J-9, L. brevis J-12, L. fermentum J-7이 무쥬스의 젖산발효를 위한 starter로써 적합한 것으로 나타났다.

토마토 주스의 미생물 발효 산도 특성에 관한 연구 (A Study on the pH Characterization for Microbial Fermentation in Tomato Juice)

  • This study was conducted to know the behavior of pH behavior in the tomato juices to find out an effective medium for microbial cultivation. Bacterial culture media is a material consist a mixture of nutrients used to grow microorganisms on or in it. In addition, microbial culture media can also be used for isolation, propagation, testing the nature physiological, and calculation of the number of microorganisms. Fresh tomato juice is used for basic ingredient, therein added salt, sugar and EM (Effective Microbial). The fermented solution placed in a room with a temperature of 40oC. Data retrieval before the pH value reached a constant value is done every 12 hours, after constant rate data collection was done every 24 hours. The pH value has been steady after 372 hours of fermentation process (15.5 days). From the results obtained that the amount of additional ingredient which added into tomato juice does not affect final pH value of solution. Thereby the most effective treatment for microbial cultivation media is treatment number four.

Effect of Triticale Dried Distillers Grains with Solubles on Ruminal Bacterial Populations as Revealed by Real Time Polymerase Chain Reaction

  • Real time PCR was used in this study to determine the effect of triticale dried distillers grains with solubles (TDDGS) as a replacement for grain or barley silage in finishing diets on the presence of six classical ruminal bacterial species (Succinivibrio dextrinosolvens, Selenomonas ruminantium, Streptococcus bovis, Megasphaera elsdenii, Prevotella ruminicola and Fibrobacter succinogenes) within the rumen contents of feedlot cattle. This study was divided into a step-wise adaptation experiment (112 days) that examined the effects of adaptation to diets containing increasing levels of TDDGS up to 30% (n = 4), a short-term experiment comparing animals (n = 16) fed control, 20%, 25% or 30% TDDGS diets over 28 days, and a rapid transition experiment (56 days) where animals (n = 4) were rapidly switched from a diet containing 30% TDDGS to a barley-based diet with no TDDGS. It was found that feeding TDDGS as replacement for barley grain (control vs. 20% TDDGS) decreased 16S rRNA copy numbers of starch-fermenting S. ruminantium and S. bovis (p<0.001 and p = 0.04, respectively), but did not alter 16S rRNA copy numbers of the other rumen bacteria. Furthermore, feeding TDDGS as a replacement barley silage (20% vs. 25% and 30% TDDGS) increased 16S rRNA copy numbers of S. ruminantium, M. elsdenii and F. succinogenes (p<0.001; p = 0.03 and p<0.001, respectively), but decreased (p<0.001) the 16S rRNA copy number of P. ruminicola. Upon removal of 30% TDDGS and return to the control diet, 16S rRNA copy numbers of S. ruminantium, M. elsdenii and F. succinogenes decreased (p = 0.01; p = 0.03 and p = 0.01, respectively), but S. dextrinosolvens and S. bovis increased (p = 0.04 and p = 0.009, respectively). The results suggest that replacement of TDDGS for grain reduces 16S rRNA copy numbers of starch-fermenting bacteria, whereas substitution for barley silage increases 16S rRNA copy numbers of bacteria involved in fibre digestion and the metabolism of lactic acid. This outcome supports the contention that the fibre in TDDGS is highly fermentable.

THE EFFICIENCY OF UTILIZATION OF METABOLIZABLE ENERGY OF MILK-REPLACER-FED CALVES AT WEANING PERIOD

  • A total of 22 energy balance trials were done for calves fed a liquid milk replacer, calf starter and second cut mixed hay during weaning period. Milk replacer supplied 50% of the total dietary energy, calf starter 42% and hay 8% Live weight of calves averaged 64.6 ($S.D.{\pm}7.8$) kg and daily gain 0.54 (${\pm}0.22$) kg. The metabolizability of gross energy averaged 0.751. A regression was calculated relating energy retention (ER, $kJ/kg^{0.75}$) to the intake of metabolizable energy (IME, $kJ/kg^{0.75}$): $$ER=0.69({\pm}0.09)IME-395,\;r=0.888,\;P<0.01,\;S.E.{\pm}7.1$$. Metabolizable energy for maintenance (MEm) was calculated to be $572kJ/kg^{0.75}$ when ER = 0. The amount of IME over MEm for an individual animal (MEg, $kJ/kg^{0.75}$) was regressed on average daily gain (ADG, kg) by the method of regression through the origin: $$MEg=364({\pm}55)ADG,\;r=0.634,\;P<0.01,\;S.E.{\pm}12$$. The amount of ME required for maintenance and growth was estimated to be $936kJ/kg^{0.75}$.

  • 한우의 mt DNA cytochrome oxidase subunit I, II, 및 III complex지역의 유전적 다형현상을 제한효소를 이용하여 검출하였다. PCR primer 6종에 대하여 20가지 제한효소를 처리하였으며, Pst I, Pvu II, Rsa I, Eco RI, Bgl II, and Msp I 제한효소를 사용하여 유전적 변이를 검출하였다. 검출된 변이체와 한우의 성장과의 관련성을 조사한 결과 cytochrome oxidase subunit III complex 지역의 유전염기서열을 근거로 제작한 primer Mt9 좌위에서 제한효소 PvuII를 이용한 절단형과 체중형질 인 WT15(P<0.05) 및 WT18(P<0.01)에서 고도의 유의성이 관찰되었다. 아울러 , Mt9-Pvu II(P=0.07), Mt6-Bgl II(P=0.05), and Mt8-Rsa I(P=0.05) 좌위 또한 WT9, WTl5, and WT15에서 각각 통계적 유의성이 관찰되었다. 따라서 본 결과는 cytochrome oxidase subunit III complex segments가 candidate gene으로서 기초적 유전정 보 제공은 물론 유전적 개량을 위해 사용될 수 있을 것으로 사료된다.

  • Let p be an analytic function defined on the open unit disk 𝔻. We obtain certain differential subordination implications such as ψ(p) := pλ(z)(α+βp(z)+γ/p(z)+δzp'(z)/pj(z)) ≺ h(z) (j = 1, 2) implies p ≺ q, where h is given by ψ(q) and q belongs to 𝒫, by finding the conditions on α, β, γ, δ and λ. Further as an application of our derived results, we obtain sufficient conditions for normalized analytic function f to belong to various subclasses of starlike functions, or to satisfy |log(zf'(z)/f(z))| < 1, |(zf'(z)/f(z))2 - 1| < 1 and zf'(z)/f(z) lying in the parabolic region v2 < 2u - 1.

Effects of Fermented Soy Protein on Growth Performance and Blood Protein Contents in Nursery Pigs

  • Fifty-four cross-bred ((Landrace${\times}$Yorkshire)${\times}$Duroc) pigs (13.47${\pm}$0.03 kg average initial BW) were evaluated in a 42 d growth assay to determine the effects of the fermented soy product (FSP). The dietary treatments were: FSP 0 (corn-soybean basal diet), FSP 2.5 (FSP 0 amended with 2.5% FSP), and FSP 5 (FSP 0 amended with 5% FSP). The body weight at the end of the experiment increased linearly (p = 0.05) as the FSP levels in the diets increased. In addition, the ADG and G/F ratio also increased (linear effect, p = 0.06) as the levels of FSP increased. However, there was no effect of FSP on ADFI or DM digestibility (p>0.05). Furthermore, the N digestibility increased as the FSP levels increased (linear effect, p = 0.003), although the total protein concentration in the blood was not affected by FSP (p>0.05). Additionally, the albumin concentration was higher in pigs fed diets that contained 2.5% FSP than in pigs in the control group or the FSP 5 group (quadratic effect, p = 0.07). The creatinine concentrations were also evaluated at d 42 and found to be greater in pigs that received the FSP 2.5 diet (quadratic effect, p = 0.09). Moreover, the creatinine concentration increased linearly in response to FSP treatment (p = 0.09). Finally, although the BUN concentration on the final day of the experiment was greater in pigs that received the FSP 2.5 diet (quadratic effect, p = 0.10), there were no incremental differences in BUN concentrations among groups (p>0.05). Taken together, the results of this study indicate that feeding FSP to pigs during the late nursery phase improves growth performance and N digestibility.
