Analysis of adequacy of oil emulsion for hazardous materials storage tank due to isooctane leak (아이소옥테인 누출에 따른 위험물저장탱크의 방유제 적정성 분석)

  • The ALOHA program was used to propose an improvement plan for antifouling agents by reflecting the effects of toxicity, overpressure, and radiant heat around the storage tank of dangerous goods manufacturers. The ALOHA program was used to analyze the damage effects of isooctane on the toxicity, overpressure, and radiant heat of isooctane for the surrounding effects of the isooctane leakage accident, which is the first class of petroleum. As a result, it was analyzed that the toxicity was 117m in the area above the PAC-2 concentration, the overpressure was 114m in the range of 1 psi or more, and the radiant heat was 1,900m in the range of 2kW/m2 or more. Therefore, it is necessary to implement a performance-oriented dike design by applying a risk assessment reflecting the physical and chemical characteristics of dangerous substances that reduce and store the amount of octane in the antifouling agent by installing an dike for each storage tank of isooctane at a minimum distance of 38m from the center of the tank.

Protein Separation with CTAB/Hexanol/Isooctane Reverse Micellar System (CTAB/Hexanol/Isooctane 역미셀계를 이용한 단백질 분리)

  • The solubilization and desolubilization of proteins in CTAB/hexanol/isooctane reverse micellar system were investigated for the selective separation of proteins. Several proteins were used, including bovine serum albumin (BSA), pepsin, trysin and ribonuclease-a. Most proteins could be solubilized into reverse micelles in the pH range above the isoelectric point of each protein, where the net charge of protein was opposite to that of surfactant. However BSA was solubilized above pH 10, which is serveral pH units above the pI 4.9. The kinds of anions in aqueous phase influenced on protein solubilization while no significant trend was observed with different cations, Protein solubilization decreased with increase of the ion size in the order of F -, C1-, Br- and I -. The size of CTAB micelles did not change significantly with increasing ionic strength, but the solubilization decreased. Protein desolubilization showedropposite behaviors to the solubilization. Several model mixtures such as pepsin/ trypsin, pepsin/ribonuclease-a and BSAlribonucleaee-a were successfully separated from each other without changing enzymatic activities.

  • The effect of organic solvents on the stability and catalytic activity of the microbial lipase from Rhizopus arrhizus for interesterification of fats and oils has been examined. The reaction system used was nonaqueous solvent system (two phase system). The solvents examined were 5 hydrocarbons (n-hexane, n-heptane, n-octane, isooctane, and cyclohexane) and 3 ethers (diethylether, diisopropylether, di-n-butylether). The results revealed that diisopropylether and isooctane are superior to the other solvents examined for interesterification of fats and oils in two phase systems.

  • The removal of cadmium, copper and chromium ions was carried in a phase transfer reactor using W/O(water in oil) microemulsion containing sodium di[2-ethylhexyl] sulfosuccinate(AOT) and isooctane system. Removal efficiencies and mass transfer rate of $Cd^{2+}$, $Cu^{2+}$ and $Cr^{3+}$ were increasing with increasing pH of aqueous solution. However, $Cr^{6+}$ was not extracted by W/O microemulsion with AOT/isooctane system. It was found that removal of heavy metal ions were required an attractive electrostatic interaction between the metal ions and W/O microemulsion. The relationship between mass transfer rate. Jo of $Cd^{2+}$, $Cu^{2+}$ and $Cr^{3+}$ and pH of aqueous solution by W/O microemulsion suggested.

Characterization of Lipoxygenase in AOT/Isooctane Reversed Micelles (AOT/isooctane 역미셀계내에서의 lipoxygenase 반응 특성)

  • An investigation was carried out to study the characteristics of lipoxygenase in dioctyl sulfosuccinate (aerosol-OT, AOT)/isooctane revered micelles of microaqueous system containing infinitesimal water. ${\alpha}-Linoleic$ acid as a substrate could be analyzed by the colorimetric methodology using 5%(w/v) cupric acetate-pyridine solution and the activity of lipoxygenase was able to be assayed by the degree of ${\alpha}-linoleic$ acid consumption per minute. Optimal pH, temperature, and R-value ([water]/[AOT]) were determined as the value of 5.0, $25^{\circ}C$, and 10.0, respectively. Kinetic analysis of the enzyme reaction under the optimal conditions showed that the values of $K_m$ and $V_{max}$ were 0.31 mM of ${\alpha}-linoleic$ acid and $384.16{\mu}mol$ of ${\alpha}-linoleic$ acid decomposed/min, respectively. The results indicate the reaction to be lipoxygenase-catalyzed oxidation of ${\alpha}-linoleic$ acid in AOT/isooctane reversed micellar system. The inhibitory effect of natural antioxidants on lipoxygenase showed little inhibitory effect of L-ascrobic acid while ${\alpha}-tocopherol$ showed 72% of inhibitory effect.

유기용매 내에서 중쇄지방질의 합성

  • Using 20 lipases from various microbial origins medium chain glycerides, namely, mono-, di-, and tri-carproyl glycerols from glycerol and acid were synthesized in isooctane. Enzyme reaction was performed at 0.35 M of capric acid, 0.025M of glycerol and the same mass of silica gel to remove water in 5ml of isooctane with 30mg of lyophilized lipase. Among 20 lipases, eleven lipases showed good synthetic activities, especially lipase from Pseudomonas aeruginosa (Lipase PS), Rhizomucor miehei origined lipase and Chromobacterium viscosum lipase (Lipase CV) showed good activities for production of tricaproylglycerol, while Lipase OF-360 (origined from Candida rugosa) and Lipase D (Rhizopus delemar) were good for production of dicaprolyglycerol. The lipases, especially Lipase PS, have high thermal stability at $ 60^{circ}C$, and optimum pH of lyophilization for dehydrating the lipase was pH 6.5.

Reversed Micellar Protein Extraction in a Hollow Fiber Membrane Extractor (단백질 추출용 역미셀 실관막장치에 관한 연구)

  • Solubilization phenomena of a protein in a reversed micellar solution were investigated and a hollow fiber membrane extractor was tested for reversed micellar protein extraction equipment. Alkaline protease was used as a model protein compound and the reversed micellar solution was consisted of AOT and isooctane. It was found that protein solubilization was strongly influenced by ionic strength and pH. The distribution coefficient of the protease between the aqueous solution and the AOT/isooctane solution was also observed to be as high as 4.0 within the scope of this experiment. A hollow fiber membrane extractor was constructed and tested for the protein extraction. The overall mass transfer coefficient at a typical experimental condition of this study was observed to be $6.7{\times}10^{-5}cm/s$. It was also found that the mass transfer resistance on reversed micellar solution was the dominant resistance for the protein transfer.

  • The enantioselective hydrolysis of racemic styrene oxide in organic solvents was conducted using a recombinant E. coli expressing protein-engineered Mugil cephalus epoxide hydrolase (McEH). The volumetric total activity of the recombinant E. coli was enhanced 2.2-fold by IPTG induction at a mid-exponential growth phase. Among organic solvents with different log P values, isooctane was chosen based on the high activity and the enantioselectivity of McEH. Some lyoprotectants such as skim milk or sucrose enhanced the McEH activity. Enantiopure (S)-Styrene oxide with a 98% ee was obtained from the racemic styrene oxide with a 53.6% yield based on its theoretical yield in isooctane.

  • It is difficult to accurately evaluate the effect of lipophilic compounds in aqueous reaction system of enzymes because they are immiscible with water. To screen lipophilic inhibitors of tyrosinase which catalyzes the synthesis of melanin in vivo, an optically clear organic system composed of organic solvent, surfactant, and water, often called reverse micelles(RM), was introduced. Optimal RM to let tyrosinase act normally was composed of isooctane as an organic solvent and dioctyl sulfosuccinate(AOT) of 100 mM as a surfactant. When a molar ratio of water to surfactant was 15, tyrosinase(105.3 units) in RM showed a similar reactivity toward 3,4-dihydroxyphenylalanine(0.18 mM) as in the aqueous assay system. In the presence of cinnamic acid, the product formation of tyrosinase reaction was proportional to the reaction time. This indicates that the inhibitory effect of lipophilic compounds could be analyzed in RM.
