  • Contents of free amino acids and organic acids in the leaves of Ailanthus altissima were surveyed through the course of this study. The results were as follows; 1. Lysine, histidine, arginine, tryptophan, aspartic acid, glutamic acid, threonine, serine, proline, glycine, alanine, valine, leucine, isoleucine, methionine, tyrosine and phenylalanine were presented in the leaves of A. altissima, and glutamic acid showed the highest amount and was more than about 48% of total free amino acids. 2. Fumaric acid was the major organic acid in the leaves of A. altissima, and also formic acid, acetic acid, maleic acid, succinic acid, oxalic acid, citric acid, tartaric acid and sorbic acid were determined, and two unknown were found.

Studies on the Nutrition of Amino Acids and Vitamins in Undaria pinnatifida (Harvey) Suringar (미역의 아미노酸 및 비타민에 對한 營養學的 硏究)

  • Leuconostoc mesenteroides P-60, Lactobacillus arabinosus 17-5, Streptococcus faecalis R have been successfully used for the quantitative determination of sixteen amino acids in Undaria pinnatifida (Harvey) Suringar hydrolysate by alkaline and hydrolysis for succesive two hours from two to twelve hours, by means of microbiological assay. And thiamine and riboflavin were fluorometrically determinated by thiochrome and lumiflavin in powder (80mesh) of Undaria pinnatifida (Harvey) Suringar. The results were as follows: 1) Arginine contents was the highest in hydrolysate for two hours, but longer the hydrolysis, the more content Undaria pinnatifida was decreased. 2) The adequate contents of other amino acids were obtained by hydrolysis for six hours. 3) Growth check and improve of Lactobacillus were not identified in determination by microbiological assay for Undaria pinnatifida. 4) The following values were obtained in Undaria pinnatifida hydrolysate six hours: asparatic acid 466, arginine 230, lysine 317, histidine 74, isoleucine 242, methionine 202, phenylalanine 256, proline 231, threonine 231, tyrosine 161, valine 415, glycine 302, leucine 414, glutamic acid 625, cystine (5 hrs.) 53 and tryptophan (8 hrs.) 90mg per nitrogen one gram. 5) Protein score was 81 (limiting factor was isoleucine) and essential amino acids pattern was of satisfactory results. And methionine contained was higher than FAO value or milk value. 6) Sulphur contained amino acids (methionine plus cystine) contained in Undaria pinnatifida were 225mg/N-g. That was satisfactory results. 7) Absorption spectrum of wave length were not different 1% HAc from buffer-sol. (pH 6.8) in dilution for determination of riboflavin. Both methods might be suitable. 8) Thiamine and riboflavin contained in Undaria pinnatifida were ($B_1,\;82.51{\pm}1.1){\gamma}/N-g\;and\;(B_2,\;115.29{\pm}1.5){\gamma}/N-g.$.

Preparation of Fish Sauce from Mackerel Scrap (고등어 가공잔사를 이용한 어간장의 제조)

  • The purpose of this study is to prepare the fish sauce front mackerel scrap which usually comprises $40{\sim}50%$ of raw fish in processing. Mackerel scrap was chopped, mired with equal weight of water, and then hydrolyzed by autolysis. The optimal conditions for hydrolysis of .mackerel scrap were at $55^{\circ}C$ for 4 hours. The maximum hydrolyzed rate of protein was 65% by autolysis. Crude protein content (6.5%) and color of mackerel sauce were similar to those of traditional soybean sauce. The abundant amino acids in mackerel sauce were leucine (22.8%), isoleucine (15.0%), phenylalanine (12.6%) and valine (12.5%). In sensory evaluation, mackerel sauce was at least equal to the traditional soybean sauce in product quality.

Optimization of the Taste Components Composition in Traditional Korean Soybean Paste (한국 재래식 된장의 맛성분 조성의 최적화)

  • We investigated main effective taste components and optimization of taste component composition in traditional Korean soybean paste. In optimization of taste components the original data with 19 kinds of taste components which is consisted of bitter taste transformed with square root could explain the taste up to 78% (contributing proportion of leucine was 14.7%, isoleucine 12.7%, methionine 5.0%, higtidine 4.7% and arginine 1.1%), palatable taste (cysteine 8.4%, aspartic acid 2.0% and glutamic acid 0.7%), sweet taste (threonine 6.3%, serine 5.6%, sucrose 4.7%, glycine 1.6%, lysine 1.2%, fructose 0.6%, alanine 0.4% and glucose 0.3%), sour taste (oxalic acid 3.9% and succinic-fumaric-citric acid 3.6%) and saline taste (ash 0. 3%). In order to optimize the taste of traditional soybean paste, the constitution of taste components was analysed by multiple regression between the original data transformed with square root sensory scores of the soybean paste. This way explained the sensory evaluation best score.

  • Sex pheromone blends of large black chafer, Holotrichia parallela (Motschulasky), which consist of a major component L-isoleucine methyl ester (LIME) and a minor component (R)-(-)-linalool, showed different attractiveness depending on the mixing ratios of the two components in potato fields in South Korea. The best ratio for H. parallela attraction was 2.5:1 of LIME and (R)-(-)-linalool. The attraction ability of pheromone traps bated with 2.5:1 ratio did not drop up to 14m, but significantly decreased at 21m from H parallela release point. Korean population of H. parallela showed almost same periodical activity to sex pheromone with that reported in Japan, which showing a circabidian periodicity of 48h cycle.

Isoleucine at position 150 of Cyt2Aa toxin from Bacillus thuringiensis plays an important role during membrane binding and oligomerization

  • Cyt2Aa2 is a mosquito larvicidal and cytolytic toxin produced by Bacillus thuringiensis subsp. darmstadiensis. The toxin becomes inactive when isoleucine at position 150 was replaced by alanine. To investigate the functional role of this position, Ile150 was substituted with Leu, Phe, Glu and Lys. All mutant proteins were produced at high level, solubilized in carbonate buffer and yielded protease activated product similar to those of the wild type. Intrinsic fluorescence spectra analysis suggested that these mutants retain similar folding to the wild type. However, mosquito larvicidal and hemolytic activities dramatically decreased for the I150K and were completely abolished for I150A and I150F mutants. Membrane binding and oligomerization assays demonstrated that only I150E and I150L could bind and form oligomers on lipid membrane similar to that of the wild type. Our results suggest that amino acid at position 150 plays an important role during membrane binding and oligomerization of Cyt2Aa2 toxin.

Effect of Bleaching and Permanent Wave Manipulation on the Amino Acid Composition of Hair (모발 탈색 및 퍼머넌트 웨이브가 모발의 아미노산 조성에 미치는 영향)

  • Variations of the seventeen amino acids(aspartic acid, threonine, serine, glutamic acid, proline, glycine, alanine, cystein, valine, methionine, isoleucine, leucine, tyrosine, phenylalanine, histidine, lysine, arginine) were analyzed in human hair sample by amino acid auto analyzer(AAA). The effect of bleaching and permanent wave manipulation on the amino acid composition of hair were investigated. Hair samples were collected from 10 males in their thirties. Hair samples were treated with 10 mL of 6 N hydrochloric acid at $110^{\circ}C$ for 24 h and analysed by AAA. The results showed that the amino acid content of normal hair(73.9%) decreased to 71.5% and 69.3% after bleaching and permanent wave treatment, respectively. Furthermore, mean contents of lysine and tyrosine in amino acids showed obviously decreased about 25% by permanent wave and bleaching treatment. On the other hand, serine, cystein, leucine and histidine were not changed.

Quality Changes of Citrus kiyomi by Chitosan and Calcium Treatment and Storage Warehouse (키토산 및 칼슘 처리와 저장고 형태에 따른 청견의 저장 중 품질변화)

  • Quality changes of citrus tangor(Citrus kiyomi) during storage by chitosan and calcium treatment and storage warehouse were investigated. Citrus fruits were treated with 2000-folds diluted iminoctadime-triacetate solution, and 1.5% chitosan with 0.5% CaC1$_2$solution, and were at 30$^{\circ}C$ for 24 hr before storage. The citrus fruits of about 12 kg/26 L plastic container were stored at 4$^{\circ}C$ with 87% relative humidity. Decay ratio of citrus with precise temperature and humidity control were lower than the others during storage. Weight loss, moisture content of peel and flesh were decreased slowly during storage. 15 ∼ 18% of acid content were decreased on 120 days' storage. Vitamin C content were decreased rapidly during storage. 23 free amino acids were detected. Isoleucine and leucine were not detected, but alanine was detected only on stored citrus. Citrus fruits was kept well on its commodity quality for 3 month at 4$^{\circ}C$ with 87% relative humidity.

Analysis of Amino Acids, Vitamins and Minerals of Fruiting Body of Fomitopsis pinicola (소나무 잔나비버섯의 아미노산, 비타민 및 무기물 분석)

  • The nutritional composition of Fomitopsis pinicola (F. pinicola) fruiting body has been analyzed for medicinal and edible uses. The contents of crude fibers, carbohydrates, crude protein, moisture, crude fats and ashes were 43.3%, 26.3%, 12.8%, 12.6%, 3.3% and 1.7%, respectively. Eighteen amino acids were found in F. pinicola. Among total amino acids, glutamate content was the highest (457 mg/100 g dry mushroom) and arginine, glycine, valine, aspartate and isoleucine were followed. Concerning free amino acids, glutamine, arginine, trytophan, and glutamate were dominant. The vitamin E content was the highest (276 mg/100 g dry mushroom), then vitamin H and vitamin B, were followed. The mineral contents were as follows: K 165.06 mg, P77.57 mg, Mg 46.11 mg, Fe 21.56 mg, and Ca 16.90 mg based on 100 g dry mushroom.

Studies on Lipid Oxidative Browning - Effects of Water Activities and Temperatures on Maillard Reaciton of Amino Acids-Oil - (지질산화에 의한 갈변에 관한 연구 - 지질과 아미노산의 Maillard 반응에 있어서 Aw와 온도의 영향 -)

  • This study was carried out in order to investigate the browning reaction of fish oil-amino acid model system at different temperatures and watger activities. The 23 amino acids, induced during dehydration in the presence of oil and avicel(5 to $45^{\circ}C$), Aw 0.33 to 0.95, were resulted in three types of browning patterns : Type I showed high browning rates at Aw 0.33, 0.95 than at Aw 0.52, 0.75(phenylalanine, trans-4-hydroxy-L-proline, methionine, valine). Type II showed high browning rates decreased with increasing water activity(poline, leucine, isoleucine, arginine). Type III showed high browning rates at Aw 0.52, 0.75 than at Aw 0.33, 0.95(tryptophan, cystein, threonine, lysine). The temperature effect on the browning development of the four most active amino acids : phenylalanine, valine, trnas-4-hydroxy-proline and methionine are shown to represent the 23 amino acids. Above $25^{\circ}C$ the browning rate began to increase. Activation energy of the amino acids-fish oil was 8 to 40kcal/mole, and $Q_{10}$ were 2 to 10.
