  • This paper discusses an on-line fuzzy dynamic model(FDM) identification of nonlinear processes for the design of fuzzy model based fault detection and isolation(FDI). The dynamic behavior of a nonlinear process is represented by a fuzzy aggregation of a set of local linear models. The identification is divided into two procedures. The first is the off-line identification of membership function. The second is the on-line identification of the local linear models. Then, we propose a residual generation scheme based on the parameters of local linear models and show that the scheme can be used for the design of FDI

Isolation and identification of mammalian orthoreovirus type 3 from a Korean roe deer (Capreolus pygargus)

  • Mammalian reovirus (MRV) causes respiratory and intestinal disease in mammals. Although MRV isolates have been reported to circulate in several animals, there are no reports on Korean MRV isolates from wildlife. We investigated the biological and molecular characteristics of Korean MRV isolates based on the nucleotide sequence of the segment 1 gene. In total, 144 swabs from wild animals were prepared for virus isolation. Based on virus isolation with specific cytopathic effects, indirect fluorescence assays, electron microscopy, and reverse transcription-polymerase chain reaction, only one isolate was confirmed to be MRV from a Korean roe deer (Capreolus pygargus). The isolate exhibited a hemagglutination activity level of 16 units with pig erythrocytes and had a maximum viral titer of 105.7 50% tissue culture infectious dose (TCID50)/mL in Vero cells at 5 days after inoculation. The nucleotide and amino-acid sequences of the partial segment S1 of the MReo2045 isolate were determined and compared with those of other MRV strains. The MReo2045 isolate had nucleotide sequences similar to MRV-3 and was most similar (96.1%) to the T3/Bat/Germany/342/08 strain, which was isolated in Germany in 2008. The MReo2045 isolate will be useful as an antigen for sero-epidemiological studies and developing diagnostic tools.

  • For the purpose of the production of yeast cell protein from rice bran oil, it's assimillating yeast(E222) was isolating from soil and the resulting of identification was shown that it was belonging to Candida aibican species Total free amino acids from yeast cells were shown 0.05% per gram. Nine other species of amino acids as well as glycine, glutamic acid, alanine, leucine and aspartic acid were produced from yeast cells.

Isolation and Antimicrobial Susceptibility of Nontuberculous Mycobacteria in a Tertiary Hospital in Korea, 2016 to 2020

  • Background: There is a global increase in isolation of nontuberculous mycobacteria (NTM). The aim of the study was to analyze longitudinal trends of NTM identification and pattern of antimicrobial susceptibility testing. Methods: NTM recovery rates, distribution of NTM species identification, and antimicrobial susceptibility pattern of NTM at Pusan National University Yangsan Hospital between January 2016 and December 2020 were retrospectively analyzed. Results: A total of 52,456 specimens from 21,264 patients were submitted for mycobacterial culture, of which 2,521 from 1,410 patients were NTM positive over five years (January 2016 to December 2020). NTM isolation showed an increasing trend from 2016 to 2020 (p<0.001, test for trend) mainly caused by Mycobacterium avium complex. The vast majority of M. avium complex were susceptible to key agents clarithromycin and amikacin. For Mycobacterium kansasii, resistance to rifampin and clarithromycin is rare. Amikacin was the most effective drug against Mycobacterium abscessus subspecies abscessus and Mycobacterium subspecies massiliense. Most of M. subspecies massiliense were susceptible to clarithromycin, while the majority of M. abscessus subspecies abscessus were resistant to clarithromycin (p<0.001). Conclusion: There was an increasing trend of NTM isolation in our hospital. Resistance to key drugs was uncommon for most NTM species except for M. abscessus subspecies abscessus against clarithromycin.

  • 돼지 호흡기 생식기증 바이러스(porcine reproductive and respiratory syndrome virus: PRRSV) 감염이 의심되는 농장으로부터 수집된 혈청가검물 646개로부터 MARC-145 cell을 이용하여 PRRSV 분리를 시도한 바 MARC-145 세포단층상에 세포변성효과(cytopathic effects : CPE)를 나타내는 바이러스 36주를 분리하였다. 분리된 36주가 PRRSV인지 여부를 확인하기 위하여 PRRSV를 실험적으로 접종한 혈청을 이용하여 간접형광항체시험과 혈청중화시험을 실시한 결과 36주 모두가 PRRSV로 동정되었다. 혈청학적인 동정법과 더불어 reverse-transcription polymerase chain reaction을 이용하여 PRRSV open reading frame 5(ORF5)의 유전자를 증폭한 결과 선발된 6주 모두에서 80bp의 flanking sequencing를 포함하여 약 680bp의 ORF5의 유전자를 증폭할 수 있었다.

  • 철저한 혐기적 방법으로 배양분리한 결과 편성 혐기성세균의 분리율이 높아졌고, 검체당 평균 3.3주가 분리되었다. 혈액배지인 NATB배지와 BA배지에서 비혈액배지인 NAT배지와 GAM배지에서 보다 세균 분리효과가 우수했다. NATB배지에는 혐기성 그람 양성 구균에 좋은 분리효과를 나타내었고, 그람 음성 간균은 15주 분리되었다. 이 중 Bacteroides가 12주로서 가장 많이 존재하는 균종이었다.

  • Demand for reliability and safety in modem systems has been increased in the research on fault detection and isolation. At traditional approaches to fault detection, redundant sensors have been used. More advanced methods are the residual analysis of signals which are created by the comparison between the actual plant behavior and the output response of a mathematical model. However, mathematical system models are difficult to obtain by using physical laws. These problems can be solved by system identification. In this paper, the transfer function of a direct current motor is estimated by using the system identification. And, the efficiency of the fault detector design is verified by using experiments.

메주발효에 관여하는 우량균주의 분리, 동정 및 생육특성 (Isolation, Identification and Growth Characteristics of Main Strain Related to Meju Fermentation)

  • 전국에 분포되어 있는 우수메주를 일반분석 및 관능평가를 통해 선발하고 선발된 메주의 우수 균주를 분리 동정하여 형태학적 생육특성을 조사하였다. 지역별 메주의 성분분석 결과, 수분함량은 7.2∼28.8%, 조단백질 함량은 32.7∼42.3%의 범위를 나타내었다. 아미노태질소 함량은 경북지역의 메주가 770.8 mg%이었고, 전북지역 메주가 239.9 mg%로 가장 낮았다. 각 지역에서 생산된 메주로 만든 된장의 유리아미노산 함량은 전북 된장이 4,169.6 mg%, glutamic acid는 경북이 499.4 mg%로 가장 높은 함량을 나타내었다. 그리고 메주의 관능검사 결과 경북지역에서 수거한 메주가 색상, 외관, 종합적 품질평가에서 타지역과 유의성(p<0.05) 있게 좋은 것으로 나타났다. 경북지역 메주에서 분리한 균의 형태학적 특성은gram 양성, 호기성 간균이며 운동성이 있었다. 세포의 지방산 조성중 주요 성분의 함량은 $C_{15:0}$ anteiso 지방산이 30.7%, $C_{15:1}$ iso 지방산이 28.9%, $C_{17:0}$ anteiso 지방산이 13.3%,그리고 $C_{17:0}$ iso지방산이 11.2%를 나타내었다. 이 균주의 identification coefficient는 표준균주와 같은 0.653을 나타내었으며, Bergey의 매뉴얼과 지방산 profile에 따라서 분석한 결과 B. licheniformis NH20으로 동정되었다. B. licheniformis NH20의 최적 생육 온도는 32$^{\circ}C$, 최적 초기 pH는 7.0, 최적 염도는 2%, 그리고 배양시간은 9시간이었다. 9시간이었다.

Isolation and Identification of Inhibitory Compounds on TNF-$\alpha$ Production from Magnolia fargesii

  • Three TNF-$\alpha$inhibitory lignans were isolated from the flower buds of Magnolia fargesii through bioassay-guided isolation. They were identified as eudesmin, magnolin and lirioresinol-B dimethylether on the basis of their spectroscopic data. All three lignans showed inhibitory effects on TNF-$\alpha$ production in LPS-stimulated murine macrophage cell line, RAW264.7 and eudesmin showed the strongest activity ($IC_{50}=51{\mu}M)$.
