Analysis on the International Trends in Safe Management of Very Low Level Waste Based upon Graded Approach and Their Implications (차등접근법에 근거한 극저준위폐기물의 안전관리 국제동향 및 시사점에 대한 고찰)

  • Recently, International Atomic Energy Agency and major leading countries in radioactive waste management tend to subdivide the categories of radioactive waste based upon risk-graded approach. In this context, the category of very low level waste has been newly introduced, or optimized management options for this kind of waste have been pursued in many countries. The application of engineered surface landfill type facilities dedicated to dispose of very low level waste has been gradually expanded, and it was analyzed that their design concept of isolation has been much advanced than those of the old fashioned surface trench-type disposal facilities for low and intermediate level waste, which were usually constructed in 1960's. In addition, the management options for very low level waste in major leading countries are varied depending upon and interfaced with the affecting factors such as: national framework for clearance, legal and practical availability of low and intermediate level waste repository and/or non-nuclear waste landfill, public acceptance toward alternative waste management options, and so forth. In this regard, it was concluded that optimized long-term management options for very low level waste in Korea should be also established in a timely manner through comprehensive review and discussions, in preparation of decommissioning of large nuclear facilities in the future, and be implemented in a systematic manner under the framework of national policy and management plan for radioactive waste management.

The Isolation and Culture Characterization of a Lipolytic Enzyme Producing Strain from Meju (메주로부터 지질분해 효소 생산 균주의 분리 및 배양학적 특성)

  • For screening of useful enzymes producing microorganisms from Meju, we isolated high lipase producing strains and their lipolytic enzyme activities were then tested. The lipolytic enzyme activities of isolated microorganisms were therefore tested on the Y124 strain. The gene sequence analysis of ITS from Y124 strain revealed Yarrowia lipolytica. Lipase production by the Y124 strain was studied in media containing various carbon sources. The Y124 strain drastically increased lipolytic enzyme activity in YPO media containing olive oil, as well as in YPDO media containing both olive oil and glucose. Maximal lipase production was achieved in YPD (yeast extract-peptone-D-glucose) media containing 0.7% olive oil when cultured at $30^{\circ}C$ for 8 hrs. The lipase produced from the Y124 strain showed the highest activity in p-NPO (p-nitrophenyl octanoate ($C_8$)), amongst the various p-nitrophenyl esters.

  • A bacterium which utilizes ${\varepsilon}$-caprolactam as a sole source of carbon and nitrogen was isolated from sludge of Shinchun river in Taegu and identified as Arthrobacter globiformis N-2-l. The growth medium for the optimum culture condition was composed of 0.4% ${\varepsilon}$-caprolactam, 0.02% $K_2HPO_4$, 0.05% $KH_2PO_4$, 0.02% $MgSO_4{\cdot}7H_2O$, 0.01% $FeCl_3{\cdot}6H_2O$ and 0.05% yeast extract. The optimum pH and temperature for growth were 7.0 and $30^{\circ}C$ respectively. The bacterial growth on the ${\varepsilon}$-caprolactam medium did not require any other organic nitrogen source such as yeast extract, although it was remarkably stimulated by the yeast extract. The bacteria utilized wide range of sugars and organic acids such as ${\alpha}$-ketoglutarate, adipate and P-hydroxybenzoate. The bacteria could use all kind of amino acids, ${\varepsilon}$-Caprolactam in the medium was consumed completely in the timecourse culture at $30^{\circ}C$ for 60 hr on the shaker by the bacteria. Decomposition product of ${\varepsilon}$-caprolactam by Arthrobacter globiformis N-2-1 was ${\varepsilon}$-aminocaproic acid.

Polyphenol Compounds and Biochemical Characteristics of Polyphenol Oxidase in Chinese Chestnut (밤의 Polyphenol물질과 Polyphenol Oxidase의 생화학적 특성)

  • This study was conducted to understand browning characteristics of Chinese Chestnut during processing and storage. For this, the isolation and identification of polyphenolic compounds and the biochemical characteristics of polyphenol oxidase(PPO) were investigated. The content of total phenol was $6.5{\mu}g/g$ and it was consisted of ferulic acid, caffeic acid, synapic acid, p-coumaric acid, gallic acid and salicylic acid in order. PPO was purified 11.7 fold through ammonium sulfate fractionation, DEAE-cellulose column chromatography and Sephadex G-200 column chromatography. Purified enzyme showed single protein and activity band by polyacrylamide gel electrophoresis. The optimum pH and temperature of PPO were 5.9 and $45^{\circ}C$, respectively. The activity of PPO was lost 93% by exposing at $80^{\circ}C$ for 15minutes. $Mg^{{+}{+}}$, $Ca^{{+}{+}}$, $Zn^{{+}{+}}$ increased the activity of PPO, but $Fe^{{+}{+}}$, $K^+$, $Hg^{{+}{+}}$ inhibited PPO at 10mM concentration. L-ascorbic acid, thiourea, sodium chloride and L-cysteine were effective inhibitors of PPO. The activity of PPO was higher for o-diphenols than other polyphenols. The Km value of PPO for catechol was 5mM.

Isolation of Plasmid DNA and Physiological Characteristics of Rhizobium japonicum (Rhizobium japonicum의 생리적(生理的) 특성(特性) 및 Plasmid DNA의 분리(分離))

  • This study was conducted to investigate the physiological characteristics and to isolate plasmid DNA of R. japonicum strains. The results obtained were as follows; Strains S117, S118, 005, 011 and DY-1 were slow-growers and showed alkaline reaction, whereas strains S110, S111, S114, S116, S120 and 010 were fast-growers and produced acid reaction in YEM broth. All the fast- and slow-growing R. japonicum showed gram negative and formed mucous white colony on agar plate. After 7 days, the colonies of the fast-growers were between 2.0 and 4.0mm in diameter, whereas those of slow-growers were approximately between 0.5 and 1.5mm. The fast-growers were uniformly sensitive to the pH of 4.5 and tolerant of the pH of 9.5, whereas the reverse was found for the slow-growers. All the fast-growers were able to grow in the presence of 2% NaCl however the slow-growers were not grown. All the microorganisms grew rapidly in simple mineral salt medium containing as the sole source of carbon. Starch was rarely utilized. All the fast-growers utilized sucrose. The slow-growing R. japonicum strains examined usually contained 1 to 3 plasmid DNA ranging between 15Kb and 250 Kb, whereas the fast-growing R. japonicum strains contained 1 to 3 plasmid DNA ranging from 20 Kb to 250Kb.

Structural and Chemical Characterization of Aquatic Humic Substances in Advanced Water Treatment Processes (고도정수처리 공정에서 수질계 휴믹물질의 구조 및 화학적 특성분석)

  • Humic substances HS) from process waters at advanced water treatment plant consisted of GAC and Ozone/GAC processes were isolated and extracted by physicochemical fractionation methods to investigate their characteristics. They are characterized for their functionality, chemical composition, spectroscopic characteristics using FT-IR and $^1H$-NMR spectroscopy. Humic fraction gradually decreased from 36.3% to 24.2% from 0.45 to 0.30 mgC/L) through ozonation and carbon adsorption. The humic fraction was isolated into the phenolic and carboxylic groups using A-21 resin, and the concentration of phenolic groups gradually decreased from 38.4% to 23.5% (from 4.9 to $3.2\;{\mu}M/L$ as phenolic-OH) through ozonation and carbon adsorption. In the case of carboxylic groups, the concentration decreased from 61.6% to 43.3% (from 7.8 to $5.8\;{\mu}M/L$ as COOH) through the water treatment processes. On the other hand, concentrations of those roups decreased from 38.4% to 24.0% and 61.6% to 44.9% through carbon adsorption without ozonation, respectively. The structural changes of HS identified from FT-IR and $^1H$-NMR were consistent with the results from the isolation of functional groups in HS.

Isolation, Identification and Characterization of Bacteria Degrading Crude Oil (원유 분해 미생물의 분리, 동정 및 특성)

  • Crude oil-degrading bacteria were isolated from the sites contaminated by oil products. The isolates were identified as Acinetobacter sp. A132, Pseudomonas putida A422, Pseudomonas aeruginosa F721, F722, and Xanthomonas maltophilia B823. The results of investigation on the degradability of crude oil indicated that the strain A132 had the highest rate of $6.04g/L{\cdot}day$. Also, the strain A132 and F722 almost degraded each of n-alkane compounds between $nC_{10}$ and $nC_{32}$. The strain A422 degraded benzene and xylene but not n-alkane. The strain B823 grew somewhat in crude oil but did not entirely degrade other substrates used in this study. The results of the GC/FID analysis on the degradability of the mixed n-alkane compounds showed that the strain F722 could degrade 100% of the compounds with $nC_7{\sim}nC_{10}$ and more than 80% of those with $nC_{11}{\sim}nC_{24}$.

  • Procedures to separate motile. normal & motile and acrosome-reacted sperm with high efficiency have clinical application in Assisted Reproductive Technology in terms of increasing the probability of fertilization by a normal sperm and subsequent normal embryonic development. This study evaluated the effects of 10 sperm preparation techniques [Swim-up from a washed pellet (SU). Swim-up from semen (SO). Continuous Percoll Gradients I (PIC). Discontinuous Percoll Gradients I (PID). Continuous Percoll Gradients II(P II C). Discontinuous Percoll Gradients II(P II D), SpermPrep (SFC). Wang's tube (WT). Albumin Gradients (AG), Low temperature capacitation (LTC)] on motility (%), normal morphology (%), motile sperm recovery rate(%). morphologically normal & motile sperm recovery rate (%), true acrosome reaction (%) and fertilizing ability. A P II D proved to be an effective means of separating morphologically normal & motile sperm. Our results indicated the P II D has advantages as compared with other methods in terms of recovery rate. enhancement of motility and normal morphology. And a LTC seems to be an effective means of enhancing the true acrosome reaction and fertilizing ability. These results suggest that the combined method of LTC and P II D for separation of morphologically normal & motile sperm and acrosome reacted sperm may be a useful procedure for intrauterine insemination and in vitro fertilization in the management of male factor infertility as well as for isolation of subpopulation of sperm for basic research.

  • The purpose of this study is to examine how the workplace bullying recognition level of an employee in Korean foodservice industry, which more strongly depends on human services than other industries, influences on job satisfaction and turnover intention. To this end, we collected 250 samples of employees working in the kitchen of family restaurants. Among the collected 250 sample, 241 samples were usable for the analysis excluding 9. The result is as follows. Firstly, there is a difference in average of satisfaction in work according to each group of causes to recognize workplace bullying. That is, the group which vaguely recognizes bullying as for threat to professional status and isolation, and psychological harassment shows higher satisfaction in work than the group which recognizes the bullying well, which is statistically significant. The latter demonstrates more turnover intention than the former. Secondly, the research examining the effects of the causes of workplace bullying over the employees' satisfaction in work and turnover intention shows the following result. The causes to recognize workplace bullying of the employees have a negative effect(-) on job satisfaction on the whole. The causes to recognize workplace bullying of the employees have a positive influence(+) on turnover intention. In conclusion, manager in foodservice industry will need to provide employees with the environment which can help staff improve cooperation, unity and solidarity within the organization.

Isolation and Characterization of Paramyosins of Marine Gastropods (해산복족류(海産腹足類)의 Paramyosin의 분리(分離) 및 그 특성(特性)에 관(關)한 연구(硏究))

  • The muscle of abalone, Notohaliotis discus (REEVE), and top-shell, Turbo cornutus Solander, were examined for protein composition. Then paramyosins which are known as one of the important structural protein of the muscle fibrils were isolated from the both muscle and their physico-chemical properties such as solubility, salting-out behaviour, intrinsic viscosity, ATPase activity, etc. involving amino acid composition and N-terminal amino acid residues were investigated to elucidate phylogenie characteristics more intensively from the viewpoint of comparative biochemistry. The analysis of protein composition resulted in the following estimations: abalone muscle; water-soluble protein of 22 %, salt-soluble protein, 34%, alkali-soluble protein, 20%, and stroma protein, 24%, and top-shell muscle; water-soluble protein of 16%, salt-soluble protein, 30%, alkali-soluble protein, 29%, and stroma protein, 25%, respectively. It is demonstrated in sedimentation analysis that paramyosin and myosin-actomyosin account for approximately 65% and 35% of the salt-soluble protein of abalone, and that the composition of both sediments in top-shell was approximately 70% and 30%, respectively. The ultracentrifugally homogenous paramyosins isolated essentially according to Bailey's ethanol-dried method from both of the muscle showed a $S^{\circ}_{20,w}$ of 3. 14s for abalone and a $S^{\circ}_{20,w}$ of 3.50s for top-shell. The both paramyosins were commonly rich in arginine, aspartic acid, and glutamic acid, while scarcely contained proline and tryptophan, in rough accord with the other paramyosins thus far reported. It is clear that these gastropod paramyosins showed of having the characteristic N-terminal amino acid residues such as N-aspartic acid, N-valine, N-serine, and N-threonine in common. The abalone paramyosin completely salted in with KCl beyond $0.35{\mu}$ and the top-shell paramyosin beyond $0.30{\mu}$. The abalone paramyosin was salted-out between 18% and 30% saturation of ammonium sulphate and the top-shell paramyosin between 22% and 29% saturation. The intrinsic viscosities at abalone and top-shell paramyosins at $25^{\circ}C$ were estimated respectively to be 3.1 dl/g and 2.6 dl/g showing somewhat higher than the values for some other paramyosins from lamellibranchs. In regard with the ATPase activity, the para myosin specimens did not exhibit any significant activity over through the pH conditions of 5 to 9.5. irrespective of the presence of $Ca^{++}$ or $Mg^{++}$. So was the case with the abalone paramyosin prepared by a slightly modified Bailey's wet-extraction method.
