보리누른모자이크바이러스(BaYMV)의 분리 및 동정 (Isolation and Identification of Barley Yellow Mosaic Virus in Korea)

  • Barley yellow mosaic virus (BaYMV-HN) occurring Haenam area was isolated by mechanical inoculation onto barley cultivars, purification and production of antibody. BaYV-HN were purified from infected plants a filamentous virus with 13 nm in diameter and 250∼300 nm and 500∼650 nm in length. Specific antibody made by injecting the purified virus to the muscle of a rabbit. In gel-diffusion tests antibody to BaYMV-HN did not make spur with tow Japanese BaYMV isolates BaYMV-II-1 or BaYMV-III. BaYMV-HN showed the symptom of yellowing and necrosis in host plants. Mechanical inoculation tests with Japanese barley cultivars showed that BaYMV-HN infected New Golden, Akagi Nijo and Tosan Kawa 73, but did not infect Amagi Nijo, Haruna Nijo, Ishukushirazu (ym3), Misato Golden (Ym1), Kashimamugi, Joshushiro Hadaka and Mokusekko 3 (ym1). In Korean barley cultivars, some of the naked barleys which are Hinssalbori, Kinssalbori, Saessalbori and Saechalssalbori were not infected by BaYMV-HN. However, it infected all the covered barley cultivars and the beer barley cultivars. BaYMV-HN had two RNAs, RNA 1 (7.6 Kb) and RNA 2 (3.5 Kb), and one coat protein (33 KDa).

남부지방에 발생하는 보리마일드모자이크바이러스(BaMMV)의 분리 및 동정 (Isolation and Identification of Barley Mild Mosaic Virus Occurring in Southern Korea)

  • Barley mild mosaic virus (BaMMV-Kor) was isolated from the southern part of Korea, and by mechanical inoculation onto barley cultivars, purification and production of antibody. BaMMV-Kor purified form infected plants were filamentous particle, with 13 nm in diameter and 250∼300 nm and 500∼650 nm in length. Antibody of BaMMV-Kor was made by injecting the purified virus to the muscle of a rabbit. In gel-diffusion test, antibody to BaMMV-Kor created spur with BaMMV-Kal and BaMMV-M, but did not make spur with BaMMV-Kor infected New Golden, Ishukushirazu, Joshushiro Hadaka and Misato Golden, but did not infect Kashimamugi, Chikurin Ibaraki 1 and Mokusekko 3. In Korean barley cultivars, BaMMV-Kor infected most of the covered barley cultivars, but did not infect Saeolbori. It also infected naked barley cultivars except Chalbori and Hinssalbori. And all the beer barley cultivars were infected by BaMMV-Kor. BaMMV-Kor had two RNAs, RNA 1 (7.5 Kb) and RNA 2 (3.5 Kb), and coat protein (33 KDa).

Cacao Bean Husk 추출물로부터 Glucosyltransferase 저해 활성 물질의 분리 및 구조 동정 (Purification and Structure Determination of the GTase Inhibitor from Cacao Bean Husk Extract)

  • 충치 예방 물질의 탐색 연구 일환으로, Cacao bean husk(CBH) 추출물로부터 얻어진, gluco-syltransferase(Gtase) 저해 활성 분획을 분리하였 다. 이를 다시 MCI-gel CHP-20와 Sephadex LH 20의 column chromatography법으로 분리, 정제하였으며, 이로부터 얻어진 2개의 GTase 활성 저해 물질에 대한 구조 해석을 행하였다. 분리.정제된 2개의 GTase 저해 활성 물질은 anisaldehyde-$H_2SO_4$ 및 FeCb에서 각각 갈색 및 청색 반응을 하며, 또 TLC상에서 dimer의 flavan-3-ol $R_r$값을 나타내는 단일 물질이었다. NMR(PMR, C CMR) 빛 mass spectrum으로 두 정제 분획에 대 한 화학 구조를 분석한 결과, G Tase 저해능을 나타 내는 단물질은 (-)-epicatechin-($\beta\rightarrow8$)-catechin 으로 결합된 procyanidin B-1 ($C_{30}H_{26}O_{12}$) 및 cate chin의 2량체인 procyanidin B一3($C_{30}H_{26}O_{12}$)이었다.

F2 Gel Matrix - a Novel Delivery System for Immune and Gene Vaccinations

  • Exploiting the immune system to abolish cancer growth via vaccination is a promising strategy but that is limited by many clinical issues. For DNA vaccines, viral vectors as a delivery system mediate a strong immune response due to their protein structure, which could afflect the cellular uptake of the genetic vector or even induce cytotoxic immune responses against transfected cells. Recently, synthetic DNA delivery systems have been developed and recommended as much easier and simple approaches for DNA delivery compared with viral vectors. These are based on the attraction of the positively charged cationic transfection reagents to negatively charged DNA molecules, which augments the cellular DNA uptake. In fact, there are three major cellular barriers which hinder successful DNA delivery systems: low uptake across the plasma membrane; inadequate release of DNA molecules with limited stability; and lack of nuclear targeting. Recently, a polysaccharide polymer produced by microalgae has been synthesized in a form of polymeric fiber material poly-N-acetyl glucosamine (p-GlcNAc). Due its unique properties, the F2 gel matrix was suggested as an effective delivery system for immune and gene vaccinations.

아무르 불가사리(Asterias amurensis)의 관족으로부터 신경성 펩타이드의 분리 및 정제 (Isolation and Purification of Neuropeptides from the Tube Feet of the Starfish Asterias amurensis)

  • Two neuropeptides were purified from the acidified tube feet extract of the starfish Asterias amurensis by C18 reversed phase and size-exclusion high-performance liquid chromatography (HPLC). The tube feet extract and the purified peptides (AST-I and AST-II) showed potent contractile activity on dorsal retractor muscle (DRM) of the starfish Asterina pectinifera and intestine (smooth muscle) of the panther puffer Takifugu pardalis. Treatment of the purified peptides with dithiothreitol (DTT) for 60 min at $37^{\circ}C$ significantly altered their retention times, suggesting that these compounds contained disulfide bonds. The molecular weights of AST-I and AST-II were determined to be 2064.2 Da and 6137.2 Da, respectively, by MALDI-TOF mass spectrometry.

한국산 다시마 유래 Fucoidan의 정제 및 분해균의 분리 (Purification of Fucoidan from Korean Sea Tangle (Laminaria religosa) and Isolation of Fucoidan-Degrading Microorganisms)

  • The fucoidan from Laminaria relicollected at Wando in Korea was purified with the yield of 2.3% in mass. The monosaccharide composiof the purified fucoidan was nearly identical to that of the commercial standard: fucose 63.71 %, xylose 22.98%, galactose 6.62%, mannose 0.24%, and uronic acid 3.26%. Microorganisms capable of degrading the purified fucoidan were isolated from the colonies on the minimal medium containing 0.2% of purified fucoidan as a sole carbon source. Of these isolates, a strain showing a relatively higher capability to degrade fucoidan, up to 63%, was partially characterized as a Gram positive, aerobic, moderately halophilic marine bacterium.

  • An experiment was conducted to establish a large scale production method of anti-serum against chicken IgA and to profile the developmental changes of serum IgA levels during the feeding period(from hatching to 7 weeks of age) in broiler chicks. Blood samples were taken from Hubbard chicken at the age of hatching, 3 days of age, and weekly thereafter till to 7 weeks of age. The pure IgA was isolated from ammonium sulfate treated chicken bile juice by gel filtration chromatography ( Sepharose GL-6B) - The quantitative assay of serum IgA were carried by RID method. Developmental changes of serum IgA concentrations were 0.42 mg /mL at hatching, thereafter dicreased gradually, lowest at 1 week of age(0.17 mg /mL), and gradually increased to 7 weeks of age(2.73 mg /mL). There was no sexual difference in serum IgA level, but female chicks showed higher IgA levels than male chicks during the experimental period.

  • Penicillium oxalicum(HCLF-34)의 세포외 분비효소로부터 ultrafilration, gel filtration chromatograph와 anion exchange chromatography법을 이용하여 남조세균(Anabaena cylindrica) 분해효소를 분리하였다. 이 효소의 분자량은 약 22 kDa이며, renaturation SDS-PAGE에서 monomer로서 남조세균 분해 활성을 갖는다. 아미노산 서열은 N-말단부터$NH_(2)$-Glu-Ser-Tyr-Ser-Ser-Asn-Ala-Ala-Gly-Ala-Val-Leu-Ile---, 13개의 아미노산을 분석하였으며, 분석된 아미노산의 homology를 조사한 결과 aspergillopepsin II precursor(acid protease A)와 13개의 아미노산 중 11개(84%)의 유사도를 나타내었고, acid proteinase EapC precursor과 13개 중 10개(81%)의 유사도를 나타내었다.

  • 대롱새우껍질로부터 Neutrase처리로 단백질을 제거하고 0.1 M EDTA 2Na로 무기질을 제거하여 키틴을 분리 정제하였다. 키틴의 정제시 Neutrase 3 mg/ml을 첨가하여 pH 6.0, $50^{\circ}C$에서 처리하였을 때 단백질 제거 효과가 제일 좋았다. Neutrase처리에 의한 단백질 제거, 탈 아세틸화도 그리고 칼슘, 인함량은 시판 키틴과 거의 비슷하였다. 분자량은 $1.2{\times}10^{8}$ dalton이었고 키틴의 수율은 28.5%이었다.

In Vitro Inhibitory Effect of Triterpenoidal Saponins from Platycodi Radix on Pancreatic Lipase

  • In the process of investigating anti-obesity effect of Platycodi Radix, we found that aqueous extract of Platycodi Radix might inhibit intestinal absorption of dietary fat by inhibiting pancreatic lipase (PL) activity. In order to clarify the anti-obesity mechanism of Platycodi Radix, activity-guided isolation was performed to find active components. The total saponin fraction of Platycodi Radix appeared to have a potent inhibitory activity against the hydrolysis of triolein emulsified with phosphatidycholine by pancreatic lipase in vitro. Based on these results, further purification of active components yielded 10 known triterpenoidal saponins, among these compounds, platycodin A, C, D, and deapioplatycodin D exhibited significant inhibitory effects on PL at the concentration of $500\;{\mu}g/mL$ with 3.3, 5.2, 34.8, and $11.67\%$ pancreatic lipase activity vs control, respectively. Platycodin D was found to inhibit the PL activity in a dose-dependent manner. Therefore, the anti-obesity effect of Platycodi Radix might be due to the inhibition of pancreatic lipase by its saponins.
