Clinical and Ultrasonographic Changes of the Breast after Use of Soy Isoflavones

  • Background: Phytoestrogens may be an alternative therapy in control of menopausal symptoms but their definite effects on breast tissue must be determined. Our study aimed to define the clinical and ultrasonographic changes of the breast after use of soy isoflavones in menopausal women. Materials and Methods: Menopausal women with hot flashes were randomly grouped as cases and controls and cases received soy isoflavones for 12 weeks. Breast examination (BE) and ultrasonography (US) were done at 0, 6 and 12 weeks. Tenderness and nodularity on BE were graded 1-4 by breast surgeons. Results: There were 30 women in the case and 26 in the control group. The mean age was 51.3 years and the mean age of menopause was 49.2 years. There was no change in the BE and US at 6 weeks in controls. In the case group, 10% had grade 1 tenderness and 13.3% grade 2 tenderness and grade 1 nodularity in BE accompanied with diffuse small cysts in US. At 12 weeks, there was no change in BE and US in the 2 groups. Conclusions: There was no statistically significant difference in the BE of the 2 groups at 6 and 12 weeks (p value=0.36 and 0.41 for nodularity and tenderness respectively) and in the US results. Although the literature contains many facts concerning PEs and the breast, further prospective studies are needed to identify structural breast changes produced by PEs in order to identify the appropriate dosage and indications of use.

Stability of Soybean Isoflavone Isomers According to Extraction Conditions

  • Stability of soybean isoflavone isomers according to extraction conditions such as temperature, pH, and extracting solvents was investigated. Heating induced three chemical reactions to occur for malony1 derivatives of isoflavones, namely decarboxylation of malony1 groups into acety1 derivatives, deesterification of malony1 residues, and hydrolysis of $\beta$-glycosidic bonds. Among the twelve isoflavone isomers, change in concentrations of acety1glycosides were most pronounced: Acety1 derivatives were present only in trace amounts in unheated hypocotyls, but the content increased dramatically during heating. As for the glycosides, concentrations of daidzin and glycitin increased due to heat treatment, though that of genistin remained almost unchanged. Heat decomposition rates and the patterns differed among the three malony1 derivatives. After 120 min at $80^{circ}C$, the relative concentrations of daidzin, glycitin and genistin were increased from $9.2\%$, $12.4\%$ and $3.3\%$ to $19.3\%$, $21.9\%$ and $6.2\%$, respectively. When crude isoflavones were solubilized in glycine buffer (pH 10.0) and incubated at $80^{circ}C$, deesterification occurred faster than at pH 7.0. When the pH of isoflavone solution was increased, the malony1glycosides were hydrolyzed to their respective glycosides at increased rate. Both acetyl and aglycone forms were unchanged and only de-esterification reactions occurred. At the acidic pH, malonylglycosides were much stable both at 60 and $80^{circ}C$. However at pH 10, $80^{circ}C$ and 1 hr, $75-80\%$ of malonylglycosides were transformed to their deesterified glycosides. When isoflavones were extracted with $60\%$ aqueous ethanol at $60^{circ}C$, isoflavone isomers were stable and the deesterification reactions did not occur in these conditions. However, at $80^{circ}C$ deesterification of malonyiglycosides occurred significantly with $15-20\%$ of malonylglycosides being hydrolyzed into their respective glycosides. This experiment showed that malonylglycosides undergo decomposition when heated or exposed to alkaline conditions. Also, aqueous ethanol was preferred to aqueous methanol as solubilizing media for obtaining extract with minimum degradation of malonylglycosides.

어린 병아리에서 isoflavones 급여가 골격의 회분 함량 및 물리적 성질에 미치는 영향

  • Considerable beneficial effects on osteoporosis from soy intake in postmenopausal women are being reported (Murkies et al.1998, Arjmandi et al. 1996). Isoflavones(IF), one of phytoestrogenic substances in the soybean, have been suggested for the effect. Our preliminary study showed that even the young chicks fed IF-rich diet tended to have higher bone ash content than those fed IF-poor diet. IF have been reported to decrease bone ash content than those fed IF-poor diet, IF have been reported to decrease bone loss by reducing bone resorption. Soybean meal has been one of the most important protein sources in poultry diets. We assume that the IF intake through dietary soybean meal could give meaningful influences on the birds. This study was carried out to determine effects of dietary IF from soybean sources on bone development in young chicks. Soy protein concentrate (SPC, IF-poor)and an IF concentrate(Phyto-Nutramin) were formulated together with purified-type ingredients to provide three different levels(25, 240 and 480 mg/kg) of total IF. Control diet(240 mg/kg) was prepared with isolated soyprotein(ISP, IF-rich). The diets were fed to 96, 3-day old, layer-type, male chicks, divided into 4 treatments with 3 replications for 3 weeks. Chicks fed the ISP diet had better gain/feed ratios than fed the SPC diets. Birds fed the diets with higher levels of IF tended to show higher values in serum total and ionized Ca% and tibial bone density, length % ash, stiffness and strain. This trend however, appeared less significant at the end of third week. No noticeable differences in sizes of comb and testicle and serum alkaline phosphatase activities were observed among the dietary groups. These results suggest that dietary isoflavones from soybean sources could be associated with chemical composition and physical properties of bone in sizes of comb and testicle and serum alkaline phosphatase activities were observed among the dietary groups. These results suggest that dietary isoflavones from soybean sources could be associated with chemical composition and physical properties of bone in young chicks.

Phytochemical isoflavones against diabetic foot bacteria

  • Wound swabs and pus samples were collected from diabetic foot ulcers, and control pus samples from non-diabetic cases. In 144 diabetic cases screened, Pseudomonas aeruginosa was isolated from 78 cases, in which 10.59% of the isolates were multidrug resistant (MDR), whereas the 60 control cases were not MDR. The isolated bacteria were decreasingly resistant to 6 clinically administrated antimicrobics such as ceftazidime, gentamicin, ciprofloxacin, tobramycin, piperacillin and amikacin. Therefore, it is demanded that new and more effective antimicrobials of phytochemical origins are sought after. Among 11 isoflavones (YS11-YS21) isolated from Sophora and Euchresta (Leguminosae; pea plant family), 2 (YS19 and YS21) prominently exhibited the high antibacterial activity both in vitro and in vivo. By the preliminary results, the object of this paper is to evaluate the in vitro antibacterial effect of YS19 and YS21 on the clinically isolated bactera of Ps. Aeruginosa in hospitals. All the isolates were sensitive to YS19 and YS21 and for both, minimum inhibitory concentration (MIC) values ranged from $2\;to\;50\;{\mu}g/mL$. The $MIC_{90}$ values of YS19 and YS21 were $50\;{\mu}g/mL$. It is suggested that these isoflavones might consist a basis phytochemical prevention and therapy for diabetic foot infections caused by pseudomonads.

조리방법을 달리한 쥐눈이콩의 항산화력 및 이소플라본 배당체·비배당체 함량 비교 (Component Changes in Antioxidant Activity and Isoflavones (β-glucoside & aglycone) Contents of Small Black Bean According to Different Cooking Methods)

  • Purpose: In this study, small black beans (Rhynchosia nulubilis) that were produced in the Jungsun province of South Korea were selected for use in various cooking recipes because they are known to contain higher isoflavones and excellent antioxidant effects, as compared to any other domestic soybeans. Methods: Physicochemical and antioxidant characteristics of small black beans were analyzed with uncooked beans and four other cooking methods of pan broiling, boiling, steaming, and pressure cooking. Results: Contents of ${\beta}$-glucosides (daidzin, glycitin, and genistin) and aglycone (daidzein, glycitein, and genistein) in small black beans were significantly different depending on the cooking methods (p<0.001). The results of the experiment indicated that the amount of total polyphenol in pressure cooked beans was highest, showing a value of 7.16 mg/g (p<0.001). Most contents of isoflavones (${\beta}$-glucoside, aglycone) in uncooked beans appeared to increase after cooking. In particular, the amount of glycitein was highest in pan broiled beans ($9.63{\mu}g/g$). The total isoflavone content differed among the different cooking methods, ranging from highest to lowest in the following order : pan broiled beans ($759.49{\mu}g/g$), pressure cooked beans ($725.12{\mu}g/g$), boiled beans ($591.05{\mu}g/g$), steamed beans ($511.61{\mu}g/g$), and uncooked beans ($180.80{\mu}g/g$) (p<0.001). Conclusion: Especially, the amount of isoflavones increased significantly in pan broiled beans and pressure cooked beans, suggestive of optimized cooking methods for increasing nutritional and functional contents in cooked food.

대두로부터 Isoflavone추출 및 $\beta$-glucosidase를 이용한 Aglycone 형태로의 전환 최적 조건 (Optimum Conversion to the Aglycone Form Using $\beta$-glucosidase and Isoflavone Extraction from Soybean)

  • 본 연구는 항산화작용, 항암작용, estrogen 유사작용, 항골다공증작용 등 다양한 생리적 기능을 가진 isoflavones의 추출조전을 추출용매획 농도, 추출온도, pH 및 추출시간으로 나누어 단계적으로 isoflavones을 추출함에 있어 최척의 추출 조건올 찾고자 하였으며, 보다 기능이 잘 알려진 4종 isoflavones 추출에 대한 결과를 분석하여 최적추출 초건을 선정하였다. 그 결과 75% ethanol, $80^{circ}C$, pH4, 3시간의 추출 조건에서 $4,024\mu\textrm{g}/mL$의 가장 높온 총 isofla vones 추출량을 나타내었고, 대두추출물 중의 단백질 제거를 위해 이용된 염화칼융의 농도가 증가할수록 보다 높은 함량의 isotla yones윤 얻을 수 있었다. 위의 결과로 얻은 최적추출조건에서의 대두추춤물에는 거의 대부분이 체내에 홉수되지 않는 daidzin과 genistin 같은 배당체의 형태로 존재해 있음을 확인하였다. 따라서 $beta$-glucosidase라는 효소를 이용하여 이러한 배당채 들흘 daidzein이나 genistein과 같이 체내여] 홉수 가능한 aglycones 형태로 전환시키는데 있어 효소의 농도, 반응온도 및 pH, 반웅시간의 조건에서 aglycon$\xi$s 형혜익 최적 전환수 율 조건을 각각 선정하였다. 그 결과 효소농도 8.7 units, 온도 $40^{circ}C$, pHS의 초건에서 4 40 분 풍안 반응시켰을 때 90%이상 전환된 aglycones 형태를 확인할 수 있었다.

대두 가공 식품 중의 이소플라본 함량 (Isoflavone Content in Korean Fermented and Unfermented Soybean Foods)

  • 대두에 함유되어 있는 이소플라본은 항암, 골다공증 예방과 같은 기능을 수행하는 유용 생리활성물질이다. 국내의 주요 대두 가공 식품은 두부, 콩나물, 두유와 같은 비발효 식품과 간장, 된장, 청국장, 춘장과 같은 발효식품이 있다. 비발효 식품에서는 대두와 마찬가지로 대부분 배당체 형태로 존재하였다. 두유에는 평균 676 mg/kg의 이소플라본이 존재하였고, 두부는 종류에 따라 차이가 있었으며, 평균 1,151 mg/kg이 존재하였다. 콩나물에는 약 424 mg/kg이 존재하였다. 발효 식품 중에는 청국장(920 mg/kg)과 된장(627 mg/kg)에 많은 양의 이소플라본이 함유되어 있으며, 춘장에도 상당량이 존재하지만 간장에는 매우 적은 양이 함유되어 있었다. 또한 많은 양의 이소플라본이 aglycone의 형태로 존재하였다. 따라서 국내 전통 발효 식품인 청국장과 된장은 생체 이용성이 우수한 형태인 이소플라본 aglycone의 좋은 공급원이라고 할 수 있다.

Isoflavones Extracted from Sophorae fructus Upregulate IGF-1 and TGF-$\beta$ and Inhibit Osteoclastogenesis in Rat Born Marrow Cells

  • Isoflavones have been a central subject in research on the natural phytoestrogens found in Leguminosae. Their effects on bone formation and remodeling are important in that they can act like estrogen by binding on estrogen receptors on the target cell surface. We, therefore, believed that isoflavones may help in the treatment of patients with estrogen deficiency disease such as estrogen replacement therapy (ERT) for osteoporosis. As commonly known, osteoporosis is one of the hormonal deficiency diseases, especially in menopausal women. When estrogen is no longer produced in the body a remarkable bone remodeling process occurs, and the associated events are regulated by growth factors in the osteoblast lineage. In the present study, we investigated whether isoflavones (Isocal) extracted from Sophorae fructus affect the growth factors IGF-I and TGF-$\beta$ that have been known to be related with bone formation. In the study, we found that the active control (PIII) effectively enhanced the level of nitric oxide (NO) and growth factors, and thereby inhibited osteoclastogenesis. The most efficient concentration was $10^{-8}$% within five days, whereas the comparative control (soybean isoflavone) was not as effective even at a lower concentration. In conclusion, the products which contain enriched glucosidic isoflavone and nutrient supplements such as shark cartilage and calcium can be used for osteoporosis therapy by enhancing the production of IGF-I and TGF-$\beta$. Furthermore, the NO produced through endothelial constitutive NO synthase (ecNOS) may playa role in inhibiting bone reabsorption.

대두 이소플라본 섭취수준이 난소절제한 흰쥐에서 Daidzein과 Genistein의 뇨와 변 중 배설에 미치는 영향 (Effects of Soy Isoflavone Intake on Urinary and Fecal Excretion of Daidzein and Genistein in Ovariectomized Rats)

  • 본 연구에서는 16주령 암컷 흰쥐를 대상으로 이소플라본의 함량이 다른 세 종류의 식이를 6주간 공급한 후 뇨와 변에서 daidzein과 genistein의 배설량을 측정하여 생체 이용성을 살펴보고 난소절제 여부에 따른 차이가 있는지도 알아보고자 하였다. 식이 내 이소플라본의 첨가량이 많을수록 변 중 daidzein 배설량은 유의적으로 증가하여 섭취량에 따른 반응의존성을 나타내었다. 그러나 난소절제여부에 따른 차이는 없었다. 변 중 genistein의 배설량도 이 소플라본의 첨가량이 많을수록 유의적으로 증가하였으나, HI군에서 비교해보면 난소절제시 sham수술보다 변 중 배설량이 유의적으로 낮았다. 변 중 회수율의 경우에는 배설량과 같은 섭취량에 비례적인 관계는 나타나지 않았으며 난소절제 여부에 따른 차이도 뚜렷하지 않았다. 뇨 중 daidzein의 배설량은 변 중 배설량과 마찬가지로 sham과 난소절제 모두에서 이소플라본의 첨가량이 많을수록 유의적으로 증가되었다. 난소절제 여부에 따른 뇨 중 이소플라본의 배설량을 보면 LI, MI, HI군에서 daidEein의 배설량은 sham에 비해 난소절제에서 유의적으로 낮았으며 genistein의 배설량도 LI군에서는 유의적으로 낮았다. 또한 뇨 중 회수율을 보면 sham에서 LI군에 비해 Ml와 Hl군에서 회수율이 유의적으로 낮았는데, 난소절제의 경우에는 이소플라본의 수준에 따른 회수율의 차이가 없었다. 그리고 회수율은 난소절제에서 sham에 비해 LI와 MI군에서 유의적으로 낮았다. 그러므로 이소플라본의 섭취수준이 증가하면 뇨와 변을 통한 이소플라본의 배설은 한국인의 일상적 섭취수준의 10배 범위에서는 섭취량에 의존적임을 알 수 있으며, 난소절제를 하였을 경우 뇨와 변을 통한 배설량을 감소시켜서 더 많이 체내에서 이용함을 알 수 있었다. 섭취량이 증가하면 sham수술의 경우에는 뇨 중 회수율이 감소함에 비해 난소절제에서는 회수율이 동일하고 LI와 MI군에서는 유의적으로 sham에 비해 회수율이 낮으므로 난소절제모델에서는 이소플라본의 생체이용성이 매우 높음을 확인할 수 있었다. 이러한 점으로 미루어볼 때 난소호르몬의 분비가 없는 폐경 후 여성에게는 이소플라본의 섭취가 호르몬으로서의 효과가 뚜렷할 것으로 짐작되며 이소플라본의 보충섭취를 권장한다.

청국장에 존재하는 Isoflavone의 Mass분석 (Mass Analysis of Isoflavones in Chungkookjang)

  • 청국장은 미생물, 효소, 다양한 생리활성물질을 포함하는 대두발효식품으로 정장, 핼액순환개선 효과 등이 알려져 있다. 청국장에 존재하는 isoflavone이 유방암, 전립선암의 억제와 관련이 있을 수 있다. 본 연구에서는 HPLC와 Mass 분석을 통해 분말청국장 100% ethanol추출물에 aglycone 형태의genistein, daidzein, glyciten이 존재함을 확인했다. 당이 붙어 있는 genistin, daidzin, OH기가 추가적으로 붙어 있는 8-OH-genistein, 8-OH-daidzein은 존재하지 않았다. Estrogen 수용체에는 $ER{\alpha},ER{\beta}$가 있다. $ER{\alpha}$는 유방암, 전립선암의 촉진에, $ER{\beta}$는 이들 암의 억제와 관련이 있을 것으로 추정한다. 인간의 estrogen 수용체가 cloning되어 있는 효모의 전사촉진 assay를 이용하여, 청국장에 있는 isoflavone이 $ER{\beta}_{1}$를 선택적으로 자극할 수 있음을 확인했다.
