Comparison of 12 Isoflavone Profiles of Soybean (Glycine max (L.) Merrill) Seed Sprouts from Three Different Countries

  • The levels of 12 isoflavones were measured in soybean (Glycine max (L.) Merrill) sprouts of 68 genetic varieties from three countries (China, Japan, and Korea). The isoflavone profile differences were analyzed using data mining methods. A principal component analysis (PCA) revealed that the CSRV021 variety was separated from the others by the first two principal components. This variety appears to be most suited for functional food production due to its high isoflavone levels. Partial least squares discriminant analysis (PLS-DA) and orthogonal projections to latent structures discriminant analysis (OPLS-DA) showed that there are meaningful isoflavone compositional differences in samples that have different countries of origin. Hierarchical clustering analysis (HCA) of these phytochemicals resulted in clusters derived from closely related biochemical pathways. These results indicate the usefulness of metabolite profiling combined with chemometrics as a tool for assessing the quality of foods and identifying metabolic links in biological systems.

Variations of Isoflavone Contents in Seeds and Sprouts of Sprout Soybean Cultivars (나물용 콩 품종의 종실 및 콩나물 함유 Isoflavone 변이)

  • Biofuctionality of soybean seeds and soy-bean products have been fortified by the uncovering of the multifuctional beneficial effects of isoflavones. As one way to fully utilize beneficial effects of isoflavones in soybean sprout is through the enhancement of isoflavone contents in soybean seeds, genetic selection for higher isoflavone and cultivational measures to increase isoflavone content in soybean seeds were attempted. Isoflavones (daidzein, gemstein) contents in soybean seeds and soybean sprouts were determined by high performance liquid chromatography. Total isoflavone contents in soybean seeds ranged from 756 to $1,682{\mu}g/g$ and Iksan #13 $(1,682{\mu}g/g)$ showed highest content among the 21 germplasms analyzed. Onetime treatment of soybean plants with Antipol or Piaster at the $V_4$ stage yielded seeds with higher isoflavones as $2,472{\mu}g/g\;or\;2,052{\mu}g/g$, respectively, which were higher by 37% and 14% than that of seeds in the control plants, respectively. In Eunhakong, Isoflavone contents of soybean sprout changed during sprouting. Daidzein content in hypocotyl increased to maximum on the third day of cultivation and decreased there-after, whereas the content changed little in cotyledon. In sprouts of Pungsannamulkong, daidzein content in hypocotyl showed a maximum level on the first day and decreased gradually thereafter but, the content changed little in cotyledon. Total isoflavone contents in lateral roots which developed on the 6th day after sprouting ranged from 4,416 to $5,232{\mu}g/g$ DW.

Effects of Soy Isoflavones Supplementation and Exercise on Urinary Calcium, Magnecium, Copper and Zinc Excretion in Postmenopausal Women (대두 이소플라본 보충과 운동이 폐경 후 여성의 소변 칼슘, 마그네슘, 구리, 아연 배설량에 미치는 영향)

  • This study assessed the effects of soy isoflavones supplementation with exercise on urinary mineral (calcium, magnesium, copper, zinc) excretion as an index of bone resorption rates in 67 postmenopausal women. A total subjects were assigned to Isoflavone (90 mg/day) or placebo groups. These groups were further divided into groups that undergone a regular exercise or a rather sedentary state performing daily activity only. We conducted study eight week period. Result showed urinary zinc excretion was more significantly decreased in the isoflavone-sedentary group ($-180.76\;{\pm}\;171.30\;ug/day$) than in the placebo-sedentary group ($-31.23\;{\pm}\;146.60\;ug/day$), placebo-exercise group ($40.93\;{\pm}\;193.44\;ug/day$) and isoflavione-exercise group ($-1.21\;{\pm}\;160.61\;ug/day$) (p < 0.05), but no significant changes in the differences between the values of the pre and post study values in urinary calcium, magnesium and copper excretion. These results suggest that Isoflavone supplementation decrease urinary zinc excretion rate in postmenopausal subjects.

Quality Characteristics of Jeju Traditional Doenjang (제주 전통된장의 품질 특성)

  • In order to establish the quality standard of Jeju traditional Doenjang(soybean paste), general components, free sugar, organic acid, fatty acids, amino acids, and isoflavones contents were analyzed. Moisture, crude ash, crude protein, crude lipid, acid value, pH, and salinity were 37.33~47.43%, 12.75~15.97%, 11.06~14.28%, 0.08~0.53%, 17.83~27.55 mg/g, 4.82~5.12, and 10.66~13.91% respectively. The content of amino type nitrogen was 301.06~573.89 mg%. The compositions of free sugar were glucose (0.81~10.22 mg/g), maltose(0.64~5.28 mg/g), sucrose(0.78~3.86 mg/g), and fructose(0.72~1.11 mg/g). The major organic acid was lactic acid (89.30~180.39 mg/g). Total free amino acids were from 13.89 to 15.51%, which makes no difference between the test samples. Among amino acids, glutamic acid was abundant (2.12~2.55%). The contents of isoflavones were 274.87~431.96 mg/kg, less than those of commercial soybean paste. In fatty acid composition of soybean paste, the most abundant fatty acid was found to be linoleic acid as 61.54~66.79% in Jeju traditional Doenjang. This results might be caused by the differences of brewed periods, microflora, processing methods and used soybean cultivars.

Effect of Soymilk and Exercise on Bone Mineral Density and Bone Metabolism Related Markers in Underweight College Women with Low Bone Density

  • The objective of this study was to investigate the effect of soymilk and exercise on bone mineral density (BMD) in underweight college women of 19-22 years of age, who had lower bone mass. The BMD of the lumbar spine and femoral neck was measured for 52 underweight college women. Among them, 33 subjects, whose t-score value was below -1, were selected. Questionnaire survey, anthropometrical measurements, dietary recall, analysis of BMD, fasting serum osteocalcin and urinary deoxypyridinoline (DPD) were conducted before and after the 10 week study. The 33 subjects were divided into 2 groups: soymilk group (n=19), and soymilk + exercise group (n=14). The soymilk group was given 400$m\ell$ soymilk containing 60mg of isoflavones on a daily basis and the soymilk + exercise group exercised three times a week with a daily intake of 400$m\ell$ soymilk for 10 weeks. The average ages of the soymilk group and the soymilk + exercise group were 21.1 years and 20.4 years, respectively and, there were no significant differences between the soymilk group and the soymilk + exercise group in the areas of height, weight or Body Mass Index (BMI). At the baseline, the mean daily energy intake of the soymilk group and the soymilk + exercise group was 1,597.9kcal (79.43% of RDA) and 1,704.2kcal (85.2% of RDA), respectively. The mean calcium intake of the soymilk group (408.3mg) was not significantly different from that of the soymilk + exercise group (389.4mg). Despite the 400$m\ell$ soymilk supplementation, there were no significant changes of nutrient intake in either group after treatment. However, there were significant increases in BMD's of lumbar spine and femoral neck in both groups. There were some increases in the serum osteocalcin level and decreases in the urinary deoxypyridinoline level as well. BMD change of the soymilk group was not significantly different from that of the soymilk + exercise group. In conclusion, supplementary intake of soymilk (containing 60mg of isoflavones) resulted in a significant increase in the BMD's of the lumbar spine and femoral neck in underweight college women with low bone mass. However, exercise did not result in any significant changes in the BMD's, implying the necessity for more intensive and specific long-term physical training for any substantial changes. Further investigation is necessary to determine the exercise that most strongly affects BMD.

Variation of Chemical Components and Their Interaction with Isoflavones in Maturing Soybean Seeds

  • This study was focuses on the variation of isoflavone contents during seed development and their interaction with major chemical components such as protein, amino acids, saccaharides, lipid and fatty acids. During maturing, lipid, protein, and amino acid contents in soybean seeds showed the highest values at R7 stages, but isoflavone contents were increased until R8 stage. It was noted that malonyl glucosides $(64.2\%)$ are predominant forms among conjugated isoflavones followed by glucosides $(30.7\%)$, acetyl glucosides $(4.1\%)$ and aglycones $(0.9\%)$. Sucrose and stachyose were presented as a major saccharide in soybean seeds. As maturing days progressed, they were constantly increased and the highest contents were observed at R8 stage. While small quantities of raffinose, fructose, glucose, maltose, DP3 (DP: degree of polymerization), DP6, and DP7 were detected. These results showed that saccharide composition at the beginning of seed development is primarily monosaccharides with little sucrose and oligosaccharides, but as maturing days proceeds, sucrose and starch increase with concomitant decrease in monosaccharides. Sucrose and stachyose were positively correlated with isoflavone (r=0.780, 0.764 at p<0.01, respectively), while fructose, glucose, maltose, and DP7 were negatively correlated (r=-0.651, -0.653, -0.602, and -0.586 at p<0.05, respectively). Soybeans at R8 stage were high in protein and amino acid, but low in free amino acid contents. Protein and amino acid contents showed positively significant correlations with isoflavone (r=0.571 and 0.599 at p<0.05, respectively), but free amino acid content were negatively correlation with isoflavone (r=-0.673, p<0.01). The lipid content reaches its final content relatively early stage of seed development and remains constant as compared with other chemical components. Among the fatty acids, although varietal difference was presented, stearic acid and linolenic acid were gradually decreased, while oleic and linoleic acid were increased as seed maturing progressed. Lipid was significantly correlated (r=0.754, p<0.01) with isoflavones. However, neither saturated fatty acid nor unsaturated fatty acids significantly affected the isoflavone contents of maturing soybean seeds.

Effect of Soyabean Isoflavones Exposure on Onset of Puberty, Serum Hormone Concentration and Gene Expression in Hypothalamus, Pituitary Gland and Ovary of Female Bama Miniature Pigs

  • This study was to investigate the effect of soyabean isoflavones (SIF) on onset of puberty, serum hormone concentration, and gene expression in hypothalamus, pituitary and ovary of female Bama miniature pigs. Fifty five, 35-days old pigs were randomly assigned into 5 treatment groups consisting of 11 pigs per treatment. Results showed that dietary supplementation of varying dosage (0, 250, 500, and 1,250 mg/kg) of SIF induced puberty delay of the pigs with the age of puberty of pigs fed basal diet supplemented with 1,250 mg/kg SIF was significantly higher (p<0.05) compared to control. Supplementation of SIF or estradiol valerate (EV) reduced (p<0.05) serum gonadotrophin releasing hormone and luteinizing hormone concentration, but increased follicle-stimulating hormone concentration in pigs at 4 months of age. The expression of KiSS-1 metastasis-suppressor (KISS1), steroidogenic acute regulatory protein (StAR) and 3-beta-hydroxysteroid dehydrogenase/delta-5-delta-4 isomerase ($3{\beta}-HSD$) was reduced (p<0.01) in SIF-supplemented groups. Expression of gonadotropin-releasing hormone receptor in the pituitary of miniature pigs was reduced (p<0.05) compared to the control when exposed to 250, 1,250 mg/kg SIF and EV. Pigs on 250 mg/kg SIF and EV also showed reduced (p<0.05) expression of cytochrome P450 19A1 compared to the control. Our results indicated that dietary supplementation of SIF induced puberty delay, which may be due to down-regulation of key genes that play vital roles in the synthesis of steroid hormones.

Isoflavones and biotransformed dihydrodaidzein production with in vitro cultured callus of Korean wild arrowroot Pueraria lobata (한국산 야생칡 캘러스에서의 이소플라본 및 생물전환에 의한 디하이드로다이드제인 분석)

  • Pueraria lobata, a medicinally important leguminous plant produces various isoflavones including puerarin, daidzin and daidzein which are metabolized to equol via dihydrodaidzein and tetrahydrodaidzein by the bacterial fermentation of natural isoflavone sources in human intestines. In this study, we described callus proliferation and isoflavone production in callus of Korean wild arrowroot and dihydrodaidzein biosynthesis in callus extract fermented with Pediococcus pentosaceus. Proliferation was the best at callus cultured in the medium containing 1.0 mg/L TDZ and 1.0 mg/L NAA at light condition for 12 days. Puerarin was significantly more produced at callus cultured in the medium containing 2.0 mg/L kinetin and 1.0 mg/L NAA at dark condition for 16 days, but daidzin and daidzein were not significant. Callus extract was successfully fermented with P. pentosaceus and dihydrodaidzein, which is one of equol precursors formed by biotransformation, was confirmed to be produced. These results will facilitate mass production of callus and isoflavones as equol precursors from Korean wild arrowroot and can be applied for the production of equol by biotransformation in vitro.

Nitrite scavenging activity and protective effect of the Puerariae Radix and green tea extract on lead acetate and cadmium-induced liver damage in mice (갈근 및 녹차추출물의 아질산염 소거작용 및 간독성 보호효과)

  • This study was designed to evaluated the nitrite scavenging activity and protective effect of the Puerariae Radix and green tea extract on lead acetate and cadmium-induced liver damage in mice. The quantitative analytical method for major antioxidants, isoflavones, puerarin, catechine and caffeine in galgun (Puerariae Radix) and green tera extract were established by HPLC. Contents of isoflavones, such as daidzin, genistin, daidzein and genistein were 4.23g/100g, 0.13g/100g, 0.07g/100g, and 0.03g/100g, and puerarin contents was 8.99g/100g, respectively. The total catechins and caffeine contents of green tea extract were 49.24g/100g and 6.53g/100g. The nitrite scavenging ability of galgun extract (pH 1.2, 100mg/ml) was 98.07% and it was higher than those of other extracts. It was the highest at the pH 1.2 and more than 64% in 25~100mg green tea extract, and was dependents on pH and concentration of the samples. The hepatoprotective effects of an aqueous extract from the root of gal gun and standard puerarin were evaluated against lead acetate and cadmium-induced liver damage in mice. Galgun extract and standard at a dose of 100mg/kg and 10mg/kg, 50mg/kg were administered orally once daily for successive 5 days and then a lead acetate and cadmium were orally at 3 hrs after the every day administration of galgun. The substantially elevated serum enzymatic activities of alanine and aspartate aminotransferase were due to lead acetate and cadmium treatment was dose dependently restored to the near normal level. In addition, galgun extract also significantly prevented the elevation of hepatic malon-dialdehyde formation in the liver of lead acetate and cadmium intoxicated mice in a dose-dependent manner. The results of this study clearly indicated that green tea and galgun extracts had nitrite scavenging ability and galgun extract had potent hepato-protective effects against lead acetate and cadmium-induced hepatic damage in mice and standard puerarin was also showed similar to the results of the galgun extracts.

High Performance Liquid Chromatographic Analysis of Isoflavones in Soybean Foods (콩 종류와 대두 가공식품에 함유된 isoflavones의 정량)

  • Phytoestrogen has been used as a suppliment of estrogen in order to treat osteoporosis. The representative phytoestrogens, isoflavones, are daidzein, genistein and formononetin which were present highly in our traditional soybean foods. The quantitative analysis of the isoflavone was done with a high performance liquid chromatography(HPLC) using a UV/VIS detector for the contents of the isoflavones in Astragali Radix, soybean sprouts, bean-curd(Tofu), soybean, soybean oil, pea, kidney pea, black bean(Yak-kong), soybean sauce(Ganjang). soybean paste(Doenjang), and fermented soybean(Maejoo). The content of free daidzein in soybean sprouts was $43.49{\pm}3.41\;mg/kg$$ which was much higher than that in soybean, $14.52{\pm}0.58\;mg/kg$, although total daidzein of fermented soybean was lower than that in soybean (Table 2. P<0.01). The amounts of free genistein in soybean sprouts, fermented soybean. and soybean paste were $27.63{\pm}1.66\;mg/kg,\;291.52{\pm}6.81\;mg/kg,\;and\;18.75{\pm}1.33\;mg/kg$, respectively. The level of free formononetin in soybean paste was the highest among the soybean products (P<0.01). The content of formononetin in Aatragali Radix, $9629.73{\pm}0.57\;{\mu}g/kg$, was about 160 times higher than that in soybean. Thus Korean traditional soybean products, black bean(Yak-Kong) and Astragali Radix can be a good choice of phytoestrogens.
