Cell cycle evaluation of granulosa cells in the $\gamma$-irradiated mouse ovarian follicles (감마선에 조사된 생쥐 난포 과립세포의 세포주기 분석)

  • This study was carried out to evaluate the biochemical and morphological effects of ionizing radiation on mouse ovarian follicles. Immature mice (ICR, 3 week-old) were irradiated with a dose of $LD_{80(30)}$ at KAERI. The ovaries were collected after 6 hours, 12 hours, 1 day, and 2 days post irradiation. With the morphological basis of the histological staining with hematoxylin-eosin, immunohistochemical preparation using in situ 3'-end labeling was evaluated. Flowcytometric evaluation of DNA extracted from the whole ovary was performed. The percentage of $A_0$ (subpopulation of cells with degraded DNA and with lower DNA fluorescence than $G_0/G_1$ cells), apoptotic, cells in the cell cycle was significantly higher in the irradiated group than in the control group. The number of in situ 3'-end labeled follicles increased at 6 hours post irradiation. All the analyses represented that the ionizing radiation-induced follicular atresia was taken place via an apoptotic degeneration. Such a degeneration underwent very fast and acutely. Therefore, it is concluded that the radiation-induced follicular degeneration is, like the spontaneous atresia, mediated by an acute apoptosis of follicular granulosa cells. Flowcytometric evaluation of cell cycles can make the role for quantifying the atretic follicles and understanding the mechanism of the radiation-induced cell death.

Detection of Irradiated Milk Formulas using Electron Spin Resonance (전자스핀공명법(ESR)을 이용한 방사선조사 조제유류의 판별)

  • Electron Spin Resonance (ESR) spectroscopy has been used to detect the presence of radiation-induced free radicals in biological samples since the mid 1950s and to irradiate foods containing cellulose, crystalline sugar, and bone. Therefore, we analyzed the ESR spectrum of irradiated infant formula and its ingredients in this study. Samples were irradiated with 2 different radiation sources of $^{60}Co$ gamma rays and electron beams (EBs), and the absorbed doses were 0, 1, 3, 5, and 7 kGy. ESR measurements were performed under normal atmospheric conditions using a JEOL JES-FA100 spectrometer equipped with an X-band bridge. Irradiated infant formula showed anunsymmetrical spectrum ($g_1$=2.0050, $g_2$=2.0006); in contrast, non-irradiated samples showed asymmetrical spectrum. The ingredients of irradiated samples showed a multi-component ESR signal in glucose and lactose and a singlet-type spectrum in milk powder (g=2.0050). $R^2$ of the dose-response curve showed a fine linearity of over 0.95 across the entire sample. We also compared the spectra of identical samples irradiated with $^{60}Co$ gamma rays and EBs, because EBs can be used for food irradiation in foreign countries, although this is not permitted in Korea. However, we could not find any significant differences according to the types of radiation source. Thus, ESR spectroscopy can be used to detect irradiated infant formula and several types of primary ingredients in this formula.

  • The organic material is one of the most popular material for the satellites and the spacecrafts in order to perform the thermal management, and to protect direct exposure from the space environment. The present paper observes material property changes of organic material under the space environment by using ground facilities. One of the representative organic thermal management material of satellites, 2 mil ITO(Indium Tin Oxide) coated aluminized KAPTON was selected for experiments. In order to investigate the single parametric effect of protons in space environment, MC-50 cyclotron system in KIRAMS(Korea Institute of Radiological and Medical Science) was utilized for the ion beam irradiation of protons and ion beam dose was set to the Very Large August 1972 EVENT model, the highest protons occurrence near the earth orbit in history. The energy of ion beam is fixed to 30MeV(mesa electron volt), observed average energy, and the equivalent irradiance time conditions were set to 1-year, 3-year, 5-year and 10-year exposure in space. The procedure of analyses includes the measurement of the ultimate tensile strength for the assessment of quantitative degradation in material properties, and the imaging analyses of crystalline transformation and damages on the exposed surface by FE-SEM(Field Emission Scanning Electron Spectroscopy) etc.

Relationship between Radiosensitivity and Repair Capacity in Human Epithelial Cancer Cell Lines (인체 상피암 세포주에서 방사선감수성과 손상회복의 상관관계에 관한 연구)

  • To investigate the relationship between radiosensitivity and postirradiation recovery in human cancer cells, a study was performed using human cancer cell lines-A549, CaSki, SNU-C5 and PCI-13. For the study of radiosensitivity, single doses of 2, 4, 6, 8, 10, 12, and 14 Gy were given and for postirradiation recovery, two fractions of 4 Gy were separated with a time interval of 0, 0.5, 1, 1.5, 2, 2.5, 3, 4, 5, or 6 hours. Surviving fraction was estimated using colony forming ability. Surviving fractions at 2 Gy (SF2) were 0.496 (0.570-0.412) for A549, 0.496 (0.660-0.332) for CaSki, 0.386 (0.576-0.216) for SNU-C5, and 0.185 (0.247-0.123) for PCI-13. By statistical analysis the SF2 of PCI-13 was lower significantly than those of others (p<0.05). This difference was also observed at 4, 6 and 8 Gy dose levels. At 6 and 8 Gy the surviving fractions of SNU-C5 were also lower significantly than A549 and CaSki (p<0.05). By the analysis with linear quadratic model, the values of ${\alpha}$ for A549, CaSki, SNU-C5 and PCI-13 were 0.3016, 0.3212, 0.4327 and 0.8423, respectively, and those of ${\beta}$ were 0.02429, 0.02009, 0.03349 and 0.00059, respectively. So, the value of ${\alpha}$ showed increasing tendency with decreasing SF2. By the multitarget single hit model the values of Do for A549, CaSki, SUN-C5 and PCI-13 were 1.97, 1.97, 1.46 and 0.81, respectively, and those of n were 1.53, 1.50, 1.56 and 2.28, respectively. So, the value of Do decreased with decreasing SF2. Post-irradiation recovery reached plateau at around 2 hours. Recovery ratio at plateau phase ranged from 1.2 to 4.2; the value were 1.2 for PCI-13, 3.2 for CaSki, 3.3 for SNU-C5, and 4.2 for A549. Recovery rate well correlated with SF2, and increased with increasing Do and decreasing ${\alpha}$. According to above results, the intrinsic radiosensitivity was quite different among the tested cell lines; PCI-13 was the most sensitive and A549 and CaSki was similar. This difference of radiosensitivity is thought to be partly due to the difference in amount of postirradiation recovery. By linear quadratic model the difference of ${\alpha}$ values was very high, and by multitarget single hit model the difference of Do value was significantly high among four cell lines.

Detection of Irradiated Chicken, Pork and Beef by ESR Spectroscopy (닭고기, 돼지고기 및 쇠고기의 방사선 조사 유무 판별을 위한 ESR Spectroscopy의 활용)

  • To identify irradiated foods, studies have been carried out with electron spin resonance (ESR) spectroscopy on bone containing foods, such as chicken, pork, and beef. Bones cleaned, pieced and dried were irradiated with doses of 0, 1, 3, 5 kGys using a $Co^{60}$ irradiator. The bones were placed in a resonant quartz tube with an internal diameter of about 4.0 mm within the Bruker Win-ESR spectrometer, and the intensity of the ESR signal could be quantified by double integration of the first derivative spectrum. The irradiated bone presented an asymmetric absorption in shape, different from that of an unirradiated one. It could be possible to detect at doses lower than 1 kGy below the dose employed commercially (3 kGy) in the case of irradiated chicken bone. The signal intensity was greatest in the beef bone, intermediate in the pork bone and lowest in the chicken bone; it was normally lower for smaller animals than for larger species, and small variations were observed between samples of the same species. The intensity of the signal induced in bones increased linearly with irradiation doses in the range of 1.0 kGy to 5.0 kGy, and it was possible to distinguish between samples given low and high doses of irradiation. The signal stability for 6 weeks made them ideal for the quick and easy identification of irradiated meats.

Hydroxyurea with Radiation Therapy of the Carcinoma of the Cervix IIA, IIB (병기 IIa, IIb 자궁경부암에서 방사선치료와 Hydroxyurea 병합치료 결과)

  • Purpose : To evaluate the efficacy of hydroxyurea with radiation in carcinoma of the cervix, huge exophytic or endophytic stage IIA and IIb. Materials and Methods : Sixty four patients with carcinoma of the cervix stage IIA (29 patients) with exophytic ($\geq$3cm in diameter) or huge endophytic mass and IIB (35 patients) treated with radiation and hydroxyurea at the Department of Radiation Oncology, Dongsan Hospital, Keimyung University. School of Medicine from Aug, 1989 to May, 1991. The maximum and mean follow up durations were 68 and 57 months respectively. The radiation therapy consisted of external irradiation to the whole pelvis(3600-5400cGy) and boost parametrial doses (for a total of 4500-6300cGy) with midline shield ($4{\times}10$ cm), and combined with intracavitary irradiation (3000-3500 cGy to point A). Hydroxyurea was to be taken in a single oral dose of 1.0gm/day during radiation therapy. Results : The control rate was 89.1%. The actuarial overall five year survival rate was 78.8% for stage IIA and 72.8% for stageIIB. The overall recurrence rate was 25% (16/64). Tewnty-three percent of the patients developed leukopenia ($\geq$grade 3) and four percent of the patients developed grade 3 or greater thrombocytopenia. Grade 3 or greater GI, GU complication and anemia were not noted. There was no treatment related death noted. Conclusion : We considered that hydroxyurea and radiation therapy may improve survival rate in huge exophytic and endophytic stage IIa cervical carcinoma with acceptible morbidity.

Carcinoma of the Uterine Cervix Treated with External Beam Irradiation Alone (자궁경부암의 외부방사선 치료 성적)

  • A retrospective analysis was peformed on 42 patients with carcinoma of the uterine cervix who were treated with external beam (EB) radiation therapy alone at the Department of Therapeutic Radiology, Seoul National University Hospital from March 1979 to December 1988. After whole pelvic field irradiation of 50Gy, all the patients received additional booster dose of 12-22Gy to the primary tumor Thirty one received EB radiotherapy alone because of poor geometry for intracavitary application,5 because of medical problems and 6 because of other reasons. Five year locoregional control rate and five year survival rate were $34.5\%\;and\;35.4\%,$ respectively. Five year survivals were $66.7\%,\;36.4%,\;32.8\%\;and\;25.0\%$ for stage IIA, IIB, IIIB and IVA, respectively. The response one month after treatment well correlated with prognosis. The incidence of grade 2 and 3 complication was $12\%\;and\;10\%,$ respectively. There was tendency of increased complication with advanced stage.

Effect of Photoperiod on Radiation-Induced Pink Mutations in Tradescantia Stamen Hairs (자주달개비 수술털에서 방사선에 의해 유발되는 분홍돌연변이에 대한 광주기의 영향)

  • The present study was carried out to investigate the combined effect of radiation and photoperiod (PP) regimes on Tradescantia 4430 somatic cell mutations. Potted plants were irradiated with 0.3, 0.5 and 1.0 Gy of gamma radiation from 60Co source. The plants irradiated only with gamma radiation were used as control group (CT). The somatic cell mutation rate in 0.5 Gy irradiated CT and PP20 group started to increase on the 6th day and reached a maximum value on the l0th day and 9th day after irradiation while the rate in the experimental group under 4 hours of photoperiod a day (PP4) started to increase on the l0th day and reached a maximal value on the 16th day post-irradiation. The slope of dose-response curve in CT was 5.99 ($r^2$=0.99), while it was 6.93 ($r^2$=0.98) in PP20 and 11.74 ($r^2$=0.99) in PP4, respectively. The biological efficacy of radiation in the induction of pink mutation increased by 15.7% in PP20 and 95.9 % in PP4, respectively. It is suggested that photoperiod regimes unfavorable to the plant have an additive effect on radiation-induced mutations and a delaying or inhibiting effect on cell damage repair, as well.

The Effects of Hydroxyl Radical Generation by Means of the Addition of $H_2O_2$ and $Fe^{3+}-EDTA$ in the Electron-beam Process (전자빔 공정에서 $H_2O_2$$Fe^{3+}-EDTA$의 첨가가 수산화라디칼 생성에 미치는 영향)

  • This study focuses both on the quantitative measurement of hydroxyl radicals formed by an electron beam (E-beam) process and on the decomposition of pentachlorophenol(PCP) in the presence of $H_2O_2$ and $Fe^{3+}-EDTA$ as additives. To attain this objective, the quantitative measurement of hydroxyl radical was performed with the hydroylation of benzoic acid (BA), producing hydroxybenzoic acid (OHBA). As a result, the concentrations of hydroxyl radical measured were lower than those of hydroxyl radical predicted. Probably, it indicates that the reactive species generated during E-beam irradiation are able to scavenge the hydroxyl radicals. In particular, the degradation of PCP was promoted by the addition of $H_2O_2$ (< 1mM). On the other hand, its degradation as well as the generation of chloride ions as a by-product was inhibited by the addition of $H_2O_2$ (> 1mM), and thus carbon yield(%) of oxalic acid as a by-product was increased. During E-beam irradiation the addition of $Fe^{3+}-EDTA$ effectively decomposed the PCP, thus increasing the G-values. Considering the formation of OHBA and the decomposition of PCP, these results suggest that the addition of $Fe^{3+}-EDTA$ in the E-beam process can produce the further hydroxyl radicals and enhance the efficiency of PCP decomposition at low dose.

Pretreatment of Kenaf Core by Combined Electron Beam Irradiation and Water Steam for Enhanced Hydrolysis (향상된 가수분해율을 얻기 위한 전자선 조사와 물찜의 복합 전처리공정을 이용한 케냐프 코어 전처리)

  • We have investigated the combined pretreatment of electron beam irradiation (EBI) and water steam as a kenaf core pretreatment process. After each sample was exposed to electron beam dose ranging from 50 to 1,000 kGy, the irradiated sample was treated by water steam using an autoclave for 5-h at $120^{\circ}C$. The pretreated samples were characterized using FTIR-ATR and XRD. FTIR spectra and XRD analysis of nonpretreated and pretreated samples confirm that crystallinity changes were observed before and after the pretreatment. The crystallinity index (CrI) was increased from 50.6% for nonpretreated sample 55.0% for 500 kGy exposed sample. And then, we analyzed sugar yield that is the amount of produced mono-saccharides in pretreated sample by enzymatic hydrolysis; an enzyme activity rate was 70 FPU/mL and 40 CBU/mL, and the loading time was 24, 48 and 72-h. The highest sugar yield was 83.9% at 500 kGy after 72-h for enzymatic hydrolysis. The sugar yield of enzymatic hydrolysis for pretreatment samples was increased as doses are subsequently changed to 100, 200 and 300 kGy, allowing to give 50.8%, 58.6% and 67.9%, respectively.
