Characteristics of Thermoluminescence and Electron Spin Resonance and Organoleptic Quality of Irradiated Raisin and Dried Banana During Storage (건포도와 건바나나의 감마선 조사와 저장기간에 따른 열발광 및 전자스핀공명 특성과 관능적 품질)

  • The characteristics of thermoluminescence (TL) and electron spin resonance (ESR) and organoleptic qualities of gamma-irradiated raisin and dried banana were investigated during storage at 4$^{\circ}C$ for 6 months. The minerals separated from non-irradiated raisins showed TL glow curve (TL$_1$) with very low intensity around 200~30$0^{\circ}C$, while the irradiated samples at 1 kGy or more showed glow curves with higher intensity around 18$0^{\circ}C$, with linear increase by irradiation dose ($R^2$=0.9684), which made it possible to identify irradiated samples during 6 months. Moreover, TL ratios (TL$_1$/TL$_2$) through the reirradiation step at 1 kGy enhanced confidence in the identification of irradiated raisins. The ESR signals of multicomponent lines resulted from crystalline sugar radicals were shown in irradiated banana, identifying irradiated samples. The ESR signal intensity was dependent on irradiation doses ($R^2$=0.8977) and the signals were stable enough to be detected by 6th month after storage. Considering tile marketability of irradiated dried fruits during 6 months at low temperature TL and ESR analyses were shown suitable for the identification of irradiated raisins and dried banana, respectively.

  • Male rats of Albino strain were divided into four groups, control group, X-irradiated group, WR-2721 treatment group and X-irradiated group treated with WR-2721. The radioprotective effect of treatment with S-2 (3-aminopropylamino)ethylphosphorothioic acid (WR-2721) in the dose of 200mg/kg by intraperitonial injection on rats 20min prior to wholebody X-irradiation (8Gy) was studied. Each group determined serum alkaline phosphatase (ALP), aspartate aminotransferase (ASI), alanine aminotransferase (ALT), lactate dehydrogenase (LDH) activities and contents of serum glucose after 1, 3, 7 and 10 days. The ALP and AST activities of X-irradiated group were significantly decreased (p<0.05) compared with that of control group, but X-irradiated group treated with WR-2721 less decreased those enzyme activities compared with the X-irradiated group. X-irradiated group was significantly increased (p<0.05) ALT and LDH activities compared with that of control group, but X-irradiated group treated with WR-2721 less increased those enzyme activities compared with the X-irradiated group. The concentration of serum glucose of X-irradiated group was significantly increased (p<0.05) compared with that of control group, but X-irradiated group treated with WR-2721 less increased compared with that of X-irradiated group. It may be considered that WR-2721 provided radioprotective effect of organs of body from X-irradiation.

Enact of Glutathione(GSH/GSSG) Contents of Fermented Ginseng on the ${\gamma}$-irradiated Liver of Mice (감마선을 조사한 생쥐 간에서 발효인삼이 Glutathione 함량에 미치는 영향)

  • Purpose: The radioprotective effects of white and fermented ginseng on liver damage induced by $^{60}Co\;{\gamma}$-ray were investigated. Materials and Methods: To one group of ICR male mice were given white(150 mg/kg/day for 7 days, orally) and fermented ginseng(150 mg/kg/day for 7 days, orally) before $^{60}Co\;{\gamma}$-ray irradiation. To another group were irradiated by 5 Gy(1.01 Gy/min) dose of $^{60}Co\;{\gamma}$-ray. Contrast group were given with saline(0.1 mL). The levels of reduced(GSH) and oxidized(GSSG) glutathione in liver tissue were measured. Results: In the fermented(150 mg/kg) and white ginseng(150 mg/kg) groups than irradiation group, the GSH levels were significantly increased, but the GSSG levels were significantly decreased. The ratio of GSSG/total GSH was significantly decreased in the fermented(150 mg/kg) and white ginseng(150 mg/kg) groups than irradiation group. Conclusion: In the fermented(150 mg/kg) groups than white ginseng(150 mg/kg) groups the GSH levels were significantly increased. The radioprotective effects of fermented(150 mg/kg) groups than white ginseng(150 mg/kg) groups were increased.

  • Myasthenia gravis (MG) is relatively rare ouuring as one of important autoimmune disease to affect neuromuscular junction. This study was clinically to evaluate total body irradiation (TBI) against two patients including 33-year and 39-year females for chronic MG with thymoma who hospitalized in the St. Mary's Hospital, Catholic University since 1994 as well as who showed no response by thymectomy, immunotherapy and hormonal therapy. TBI designed by the dose of 150$\~$180 cGy consisting of 10 cGy per fraction, three times a week, for 5~e weeks using linear accelerator of 6 MV. During the treatment of TBI, they did complain acute side effect such as vomiting and also appear improved physical condition from 4$\~$6 weeks after TBI. Through the follow-up period of 18 or 42 months after TBI, they did not have any symptomatic recurrence. Consequently, the results suggest that TBI can be used as an alternative tool for the patients concurrently for MG with thymoma who had been refractory to various conventional therapies like thymectomy, immunotherapy and hormonal therapy.

Detection of Irradiated Agricultural Products by Thermoluminescence(TL) (Thermoluminescence(TL)를 이용한 농산물의 방사선 조사유무 확인)

  • A study was carried out to establish the detection method of irradiated agricultural products cultivated in Korea by Thermoluminscece(TL). Samples were irradiated using Co-60 gamma rays at various doses(0.05, 0.1, 0.2, 0.3 and 0.5 kGy). After irradiation, separated minerals of the samples measured by TL. TL intensity increased with increasing doses and the irradiated samples were higher than the non-irradiated samples. TL first and second glow curves showed maximum TL temperature point at $176.16{\sim}190.08^{\circ}C$ and $143.84{\sim}146.56^{\circ}C$, respectively. All the irradiated samples can be classified correctly by the shape of the glow curve and the glow curve ratio. Correlation coefficients of ginger, carrot, potato and sweet potato were 0.9968, 0.8522, 0.9612 and 0.9071, respectively, that showed very high correlation between irradiation dose and TL intensity. Therefore, these results suggest that TL measurement is an useful detection method for irradiated agricultural products.

  • In order to develop the commercial storage method of chestnut by irradiation combined with natural low temperature, a chestnut variety Ok-gwang was stored in a natural low temperature storage room ($450{\times}650{\times}250$ cmH; year-round temperature change, $2-17^{\circ}C$; R.H., 80-85%) on batch scale followed by irradiation with optimum dose level. Sprouting rate of chestnut was 100% after seven month storage in control whereas that of 20-25 Krad irradiated group was only 5-15%. In comparison of rotting rate, weight loss and texture, 25 Krad irradiated group was better than that of control. Moisture and reducing sugar were increased in contrast with the decrease of total sugar during nine month storage and these changes were more remarkable in control. Ascorbic acid content was slightly decreased both in control and irradiated group; more decrement was noticed in control.

  • Purpose: Irradiation in the oral cancer patients causes early and late complications such as intraoral mucositis and fibrosis, with a various expression of GM-CSF and TGF-${\beta}1$. The purpose of this study was to investigate the production of GM-CSF and TGF-${\beta}1$ by the irradiated human gingival fibroblasts cultivated with lipopolysaccharide. Materials and Methods: Irradiated (total dose, 60 Gy) human gingival fibroblasts were incubated with LPS. Culture supernatants that were collected at 24, 48, and 72 hours were assessed for GM-CSF and TGF-${\beta}1$ by enzyme-linked immunosorbent assay. Results: 1. GM-CSF production in nomal gingival fibroblasts was increased with incubation time, but decreased with incubation time in irradiated gingival fibroblasts. GM-CSF production in both normal and irradiated gingival fibroblasts induced with LPS was higher than the control. 2. TGF-${\beta}1$ production in normal gingival fibroblasts was decreased after 24 hours, but, it was increased until 48 hours in irradiated gingival fibroblasts. TGF-${\beta}1$ production in normal gingival fibroblasts exposed with LPS was higher than the control. Conversely, It was lower than the control in irradiated gingival fibroblasts exposed with LPS. Conclusion: This indicates that irradiation in gingival fibroblasts may play an important role in radiation-induced intraoral mucositis and fibrosis. However, LPS decreases the production of TGF-${\beta}1$ in the irradiated gingival fibroblasts.

Microbiological and Mutagenical Safety Evaluation of Gamma Irradiated Ready-to-Eat Foods of Animal Origin (즉석식품 제조를 위한 육가공제품의 감마선 조사에 따른 미생물 및 유전독성학적 안전성 평가)

  • The radio-sensitivity of pathogens and the effect of irradiation on microbiologican safety and mutagenicity of meat products such as seasoned and cooked beef and ham were investigated. Samples were radiation-sterilized and inoculated at 10/sup 7/ cfu/g with each of the four pathogens including Salmonella typhimurium, Escherichia coli, Staphylcoccus aureus, and Listeria ivanovii. No viable cells of pathogens were observed in the sample irradiated with 3 kGy. The D/sub 10/ value of inoculated pathogens in seasoned and cooked beef and ham were 0.24∼0.48 and 0.39∼0.45, respectively. Results of Ames test performed with non-irradiated and irradiated seasoned and cooked beef and ham were both negative at the level of 625, 1,250, 2,500, 50,000, and! 10,000 ㎍ sample/plate, respectively. Results indicate that low dose (2∼3 kGy) irradiation is effective to ensure safety for seasoned and cooked beef and ham with toxicological wholesomeness.

Detection Characteristics of TL, ESR and DNA Comet for Irradiated Peanuts by Origins (TL, ESR및 DNA Comet분석에 의한 원산지별 땅콩의 방사선 조사 검지 특성)

  • Gamma-irradiated peanuts, Korean and Chinese origins, were investigated on detection properties by thermoluminescence (TL), electron spin resonance (ESR), and DNA comet assay (single cell gel electrophoresis). TL measurement showed that the non-irradiated sample revealed a glow curve with low intensity at about 25$0^{\circ}C$, while the irradiated samples showed higher intensity around at 18$0^{\circ}C$. TL ratio (TL$_1$/TL$_2$) of area for TL$_1$ glow curve to TL$_2$ was below 0.05 for the non-irradiated sample and 0.2 or more for the irradiated ones, thus identifying each other. ESR spectroscopy for the irradiated peanuts using outer skin showed negligible signals induced by irradiation, indicating ESR is little applicable to the detection of irradiated peanuts. In DNA comet assay, the non-sample had no or very short tails, whereas the irradiated samples revealed the cells with long tails. Significance in the increase of their lengths depending on irradiation dose (r=0.761/Korean, r=0.768/Chinese) was also found. There was no remarkable difference in detection properties by origins of samples in all determinations, It is concluded that TL analysis or DNA comet assay is suitable for detection of irradiated peanuts and a combined method is recommendable for enhancing the reliability of detection results.

Effect of Prophylactic Cranial Irradiation in Acute Lymphoblastic Leukemia in Children (소아 급성 임파구성 백혈병에서 예방적 전뇌 방사선조사의 효과)

  • CNS prophylaxis with 18 or 24 Gy cranial irradiation plus intrathecal methotrexate was given to 134 childhood acute lymphoblastic leukemia patients who had got bone marrow remission (M1) after remission induction chemotherapy from August 1979 to December 1986. The rate of initial total CNS relapse was 14.2% (19/134), the rate of isolated CNS relapse was 5.2% (7/134), and the rate of CNS relapse concomittantly combined with bone marrow relapse or testicular relapse was 9% (12/134). Male sex or older age was accociated with higher CNS relapes and the initial peripheral leukocyte count over 50,000/ul had higher relapse rate. Relapse with radiation dose of 18Gy was somewhat lower than that with 24Gy. Within 4 years after CNS prophylaxis occurred 89% of the total CNS relapses, 100% of the isolated CNS relapses, and 83% of the combined CNS relapses. Adjusted to exposed cases to risk of CNS relapse, the total CNS relapse rate was 11.9% during maintenance chemotherapy and 4.9% after maintenance chemotherapy.
