Chemical Cleaning of Iron Stains on Ceramics (화학세척제를 이용한 도자기의 철산화물 제거 특성 연구)

  • To remove metal stains of the ceramics, chemical cleaning is essential case by case. This study investigated the removal characteristics of iron stains by oxalic acid and citric acid including their application methods of soaking and poultice. The soaking method in cleaning agents showed removal process by color difference and released iron contents from iron stains on ceramics. Iron stains were removed successfully from ceramics, which soaked in oxalic acid for 60 hours. However, it is recommendable to soak in 0.25M oxalic acid for one to three hours because most iron stains were disappeared in 3 hours soaking. Citric acid is less effective than oxalic acid in removing iron stains because of heavy molecular weight and low acidity. Poultices (bentonite, sepiolite, activated carbon fiber and celite) with oxalic acid were applied on contaminated ceramics. After ten hours, iron stains on ceramics were removed successfully by poultice. Among them, bentonite and sepiolite have better application. Therefore, sepiolite with 0.25M oxalic acid was applied on the iron stains of whiteware and celadon from Ma Island, and then stains were removed. However, it is judged that the application methods can be varied according to the form and depth of contaminant. In addition, the residues of poultice on the ceramics will be considered for preventing contamination.

Prevention of Oil Stain on Steel Induced by Fat and Oil Compounds (유지화합물에 의해 생성된 철판의 기름얼룩 방지)

  • An oil stain on the steel induced by metal working oil was studied on the six kinds of fat and oil compounds(rapeseed oil, oleic acid, methyl oleate, oleyl alcohol, lauryl amino, and lanolin) by MIL-C-22235A test. The samples used for the printing on the oil stain plates were prepared by the addition of fat and oil compounds(10wt% ) and antioxidants(B. H. T, B. T, T. C. P; 0.5wt%) to base oil(spindle oil; 1.5wt% of sulfur). An analysis based on the experimental data indicated that B.T was more effective than B. H. T and T. C. P as antioxidant for inhibiting oil stains. And acid values exerted a bad influence upon the preventation of oil stains. As expected by the chemical structure of fat and oil compound, oleic acid that could react with iron ions produced more stains on the steel pannel.

An investigation on the infection of Encephalitozoon cuniculi and pathological changes in laboratory rabbits (실험용 토끼에서 Encephalitozoon cunuculi의 감염과 병리학적 변화)

  • Eighteen NZW rabbits used for local skin irritation study were examined grossly and microscopically for natural infection with Encephalitozoon cuniculi. For microscopic tissue evaluation histochemical techniques such as PAS Gram iron hematoxylin and HE stain were used. Although rabbits in the study had no abnormal clinical signs 7/18(38.9%) animals were microscopically infected with E. cuniculi. The affected rabbits had gray-whitish and depressed approximately 0.1∼0.6 cm diameter lesions in the kidneys. All other organs examined were grossly unremarkable. Histopathologically however in addition to segmental interstitial nephritis focal lymphocytic myocarditis and granulomatous inflammatory reaction in portal areas of the liver multifocal granulomatous foci with vasculitis were present in the brain kidney and lungs. Aggregates of minute oval organisms were observed in brain and kidney sections frequently within the granulomatous foci and sometimes without any inflammatory reaction particularly in the renal tubules. in histochemical stains the organisms were gram positive stained with iron hematoxylin and had PAS positive granule at one pole. They measured approximately 1.5×2.5μm consistent with E. cuniculi. Histochemical characterization is important to differentiate E. cuniculi from other common protozoal infection such as Toxoplasma gondii. This study demonstrate the importance of subclinical. E. cunuculi infection and the associated histological alterations may interfere with tissue evaluation in toxicologic studies.

Growth Properties of the Iron-reducing Bacteria, Shewanella putrefaciens IR-1 and MR-1 Coupling to Reduction of Fe(III) to Fe(II)

  • Shewanela, putrefaciene IR-1 and MR-1 were cultivated by using various combinations electron donor-acceptor, lactate-Fe(III) lactate-nitrate, pyruvate-FE(III), pyruvate-nitrate H$_2$ acetate-Fe(III) and H$_2$-acetate-nitrate. Both strains grew fermentatively on pyruvate and lactate but not on without and electron acceptor. In culture with Fe(III), both astrains grew on pyruvate and lactate but on H$_2$-acetate- CO$_2$. In cultivation with nitrate, both stains grew on pyruvate lactage and on H$_2$-acetate-CO$_2$ The growth yields of IR-1 pyruvate, pyruvate-Fe(III) and lactate-Fe(III) were about 3.4, 3.5, and 3.6(g cell/M substrate), respectively. From the growth properties of both strains on media with Fe(III) as an electron acceptor, the bacterial growth was confirmed not to be increased by addition of Fee(III) as an electron acceptor to the growth medium, which indicates a possibility that the dissimilatory reduction of Fe(III) to Fe(III) may not be coupled to free energy production.

Isolation of antibiotic resistant Lactobacillus spp from fermented milk and starters, and detection of transmissible R plasmids (유산균 발효유 및 starter에서 항생제내성 유산균 분리 및 전달성 R plasmids 분포조사)

  • Plasmids are covalently closed circular molecules of DNA that are stably inherited and replicate somewhat independently of the bacterial chromosome. Genes carried on plasmids can mediate a wide variety of important functions, including antibiotics (R plasmids) and heavy metals resistance, toxins production, cell penetration, iron chelation, complement resistance, and metabolic characteristics such as sucrose and lactose fermentation. Fifty strains of lactobacilli were isolated from 26 staters and 29 fermented milk products. They were classified 27 strains as Lactobacillus paracasei subsp paracasei, 11 stains as Lactococcus lactis subsp cremoris, 6 strains as L delbrueckii subsp lactis, 4 strains as L acidophius, and 2 strains as L delbrueckii subsp bulgaricus. All of these strains were examined for drug resistance and transferability of R plasmids. All of the isolates were sensitive to Am, C, CF, E, NB, P, T, and Te. But resistant to SXT 94% (47 strains), K 66% (33 strains), S 56% (28 strains), ENR 50% (25 strains), NOR 38% (19 strains) CIP 38% (19 strains), GM 16% (8 strains), and N 14% (7 strains), in order. And 32 different resistant patterns were found. The most frequently encountered patterns were CIP-ENR-K-NOR-S-SXT (5 strains). In vitro R plasmids transfer experiment, 57 antibiotic resistant strains which were not transfer to the recipient 2 Escherichia coli strains by conjugation, These results indicate that Lactobacillus in internal trade market' stater recognize R factor but transmissible R plasmid is not existed.

Histochemical Studies of the Intestinal Mucosubstances in Agramus agramus, Inimicus japonicus, Epinephelus chlorostigma, and Helirolenus dactylopterus (노래미, 쑤기미, 구실우럭 및 홍감펭 장관 점액질에 대한 조직화학적 연구)

  • The conventional histochemical stains were used to study the properties of mucosubstances of the intestine in four teleostean species, i. e., Agramus agramus, Inimicus japonicus, Epinephelus chlorostigma, and Helicolenus dactylopterus, all belonging to the order Perciformes. The following methods were used; periodic acid Schiff (PAS), alcian blue (AB) pH at 2.5, AB pH at 1.0, AB pH at 2.5-PAS, and aldehyde fuchsin (AF) pH at 1.7-AB pH at 2.5 stain. The mucosal folds of intestines show differences in species and regions. Widely distributed in all portions studied, goblet cells situated between cylinderical epithelial cells are round or ovoid in shape. They were most densely distributed in distal intestine. In the middle and. distal intestine of Epinephelus chlorostigma and all regions of intestines of Agramus agramus, Inimicus japonicus, and Helicolenus dactylopterus, the presence of both acidic and neutral mucin was confirmed. The property of acidic mucosubstance was sialomucin. Neutral mucin was only encountered in the proximal intestine of Epinephelus chlorostigma. The amount and property of mucin showed differences in species and regions. In the distal intestine of Inimicus japonicus, the amount of acidic mucin is similar to that of neutral mucin. In all regions of intestine of Agramus agramus, proximal and middle intestine of lnimicus japonicus, middle and distal intestine of Epinephelus chlorostigma, and distal intestine of Helicolenus dactylopterus, acidic mucin occured more frequently than neutral mucin. The proximal and middle intestine of Helicolenus dactylopterus have more neutral mucin than acidic mucin.
