Oxidative stress impairs the meat quality of broiler by damaging mitochondrial function, affecting calcium metabolism and leading to ferroptosis

  • Objective: This work was conducted to investigate the effects of oxidative stress on meat quality, mitochondrial function, calcium metabolism and ferroptosis of broilers. Methods: In this study, a total of 144 one-day-old male Ross 308 chicks were divided into 3 groups (control group, saline group, and hydrogen peroxide [H2O2] group) with 6 replicates of 8 broilers each. The study lasted for 42 d. The broilers in the saline and H2O2 groups were intraperitoneally injected with 0.75% saline and 10.0% H2O2 on the 16th and 37th day of the experimental period respectively, the injection volumes were 1.0 mL/kg of broiler body weight. On the 42nd day of the experimental period, two chicks were randomly selected from each cage, a total of thirty-six chicks were stunned by electric shock and slaughtered to collect breast muscle samples. Results: The H2O2 exposure reduced pH value, increased drip loss and shear force of breast meat (p<0.05), impaired the ultrastructure and function of mitochondria. The H2O2 exposure damaged the antioxidant system in mitochondria, excessive reactive oxygen species carbonylation modified calcium channels on mitochondria, which impaired the activities of key enzymes on calcium channel, resulted in the increased calcium concentration in cytoplasm and mitochondria (p<0.05). In addition, the H2O2 exposure increased the iron content and lipid peroxidation (p<0.05), which induced ferroptosis. Conclusion: Oxidative stress could impair meat quality by causing mitochondrial dysfunction, resulting in calcium metabolism disorder and ferroptosis.

Effect of acid whey and freeze-dried cranberries on lipid oxidation and fatty acid composition of nitrite-/nitrate-free fermented sausage made from deer meat

  • Objective: This study evaluated the effect of acid whey and freeze-dried cranberries on the physicochemical characteristics, lipid oxidation and fatty acid composition of nitrite-free fermented sausage made from deer meat and pork fat. Antioxidant interactions between acid whey and cranberry compounds were also explored. Methods: Four formulations of fermented venison sausage were prepared: F1 (control), F2 (with 5% liquid acid whey), F3 (with 0.06% of freeze-dried cranberries), and F4 (with 5% liquid acid whey and 0.06% of freeze-dried cranberries). Each sample was analyzed for pH, water activity ($a_w$), heme iron content, 2-thiobarbituric acid reactive substances (TBARS) value and conjugated dienes at the end of the manufacturing process and at 30 and 90 days of refrigerated storage. Fatty acid composition was measured once at the end of the manufacturing process. Results: At the end of ripening, all samples presented statistically different values for a pH range of 4.47 to pH 4.59. The sum of the unsaturated fatty acids was higher, while the conjugated diene and the TBARS values were lower in sausages with freeze-dried cranberries as compared to the control sausage. The highest content of heme iron (21.52 mg/kg) at day 90 was found in the sausage formulation with the addition of freeze-dried cranberries, which suggests that the addition of cranberries stabilized the porphyrin ring of the heme molecule during storage and thereby reduced the release of iron. The use of liquid acid whey in combination with cranberries appears to not be justified in view of the oxidative stability of the obtained products.   • The effects of alloying elements on the structure and mechanical properties of compacted/vermicular graphite cast irons containing copper, tin and molubdenum for producing pearlite matrix, and also containing phosphorus and boron for increasing wear resistance, were investigated. The Brinell hardness and ultimate tensile strength of the specimens with the range of compositions, [1.5% Cu-0.05% Sn-(0.2-0.4)% Mo-(0.2-0.6)% P-(0.035-0.070)% B], was found to fall within in the following range, depending on their composition; Brinell hardness of BHN 250-315, ultimate tensile strength of $28.1-40.3kg/mm^2$. It was also found within this experiment that CV graphite cast irons possessing higher amount of phosphide eutectic exhibit better wear resistance, but the wear resistance became deteriorate when the area fraction of phosphide eutectics exceed more than 10%. From these experimental results, it could be concluded that the CV graphite cast iron containing 1.5% Cu, 0.05% Sn, 0.4% Mo, 0.2% P and 0.035% B showed good mechanical and wear resistance properties.

Long-Term Stability for Co-Electrolysis of CO2/Steam Assisted by Catalyst-Infiltrated Solid Oxide Cells

  • This study investigated the long-term durability of catalyst(Pd or Fe)-infiltrated solid oxide cells for $CO_2$/steam co-electrolysis. Fuel-electrode supported solid oxide cells with dimensions of $5{\times}5cm^2$ were fabricated, and palladium or iron was subsequently introduced via wet infiltration (as a form of PdO or FeO solution). The metallic catalysts were employed in the fuel-electrode to promote $CO_2$ reduction via reverse water gas shift reactions. The metal-precursor particles were well-dispersed on the fuel-electrode substrate, which formed a bimetallic alloy with Ni embedded on the substrate during high-temperature reduction processes. These planar cells were tested using a mixture of $H_2O$ and $CO_2$ to measure the electrochemical and gas-production stabilities during 350 h of co-electrolysis operations. The results confirmed that compared to the Fe-infiltrated cell, the Pd-infiltrated cell had higher stabilities for both electrochemical reactions and gas-production given its resistance to carbon deposition.

Iron sulfate and molasses treated anthocyanin-rich black cane silage improves growth performance, rumen fermentation, antioxidant status, and meat tenderness in goats

  • Objective: This study investigated the effects of feeding anthocyanin-rich black cane treated with ferrous sulfate and molasses on animal performance, rumen fermentation, microbial composition, blood biochemical indices, and carcass characteristics in meat goats. Methods: Thirty-two Thai-native×Anglo-Nubian crossbred male goats (14.47±2.3 kg) were divided equally into two groups (n = 16) to investigate the effect of feeding diet containing 50% untreated anthocyanin-rich black cane silage (BS) vs diet containing anthocyaninrich black cane silage treated with 0.03% ferrous sulfate and 4% molasses (TBS) on average daily gain (ADG) and dry matter intake (DMI). At the end of 90 d feeding trial, the goats were slaughtered to determine blood biochemical indices, rumen fermentation, microbial composition, and carcass characteristics differences between the two dietary groups. Results: Goats fed the TBS diet had greater ADG and ADG to DMI ratio (p<0.05). TBS diet did not affect rumen fluid pH; however, goats in the TBS group had lower rumen ammonia N levels (p<0.05) and higher total volatile fatty acid concentrations (p<0.05). Goats in the TBS group had a higher (p<0.05) concentration of Ruminococcus albus but a lower (p<0.05) concentration of methanogenic bacteria. The TBS diet also resulted in lower (p<0.05) thiobarbituric acid-reactive substances concentration but higher (p<0.05) total antioxidant capacity, superoxide dismutase, catalase, glutathione peroxidase, and glutathione reductase concentrations in blood plasma, while having no effect on plasma protein, glucose, lipid, immunoglobin G, alanine transaminase, and aspartate aminotransferase. Meat from goats fed the TBS diet contained more intramuscular fat (p<0.05) and was more tender (p<0.05). Conclusion: In comparison to goats fed a diet containing 50% untreated anthocyanin-rich black cane silage, feeding a diet containing 50% anthocyanin-rich black cane silage treated with 0.03% ferrous sulfate and 4% molasses improved rumen fermentation and reduced oxidative stress, resulting in higher growth and more tender meat.

전해 드레싱을 이용한 고품의 내면 연삭 가공에 관한연구 (A Study on the Internal Grinding with High Quality Using Interval Type Electrolytic Dressing Method)

  • The establishment of a practical ultra-precision grinding technique using Diamond and CBN wheels is one of the major key technolo-gies to improve production techniques for machine-to-difficult materials without finishing process such as lapping and polishing. But the special efficient dressing technique for ultra-fine grit type grinding wheels to stabilize the grinding ability was not developed. Recently electrolytic in-process dressing technique is proposed to ultra-fine grit type metal bonded diamond wheels to protrude abra-sives continuously from the tool surface. This technology can be widely used to surface grinding and cylindrical grinding but cannot be used efficiently to internal grinding because of the electrode attachment trouble. This paper describes the effect of interval type electrolytic dressing as proposed newly to cast iron bonded diamond wheel for efficient internal grinding with mirror type high quality ground surface.

Comparative Proteomic Analysis for a Putative Pyridoxal Phosphate-Dependent Aminotransferase Required for Virulence in Acidovorax citrulli

  • Acidovorax citrulli (Ac) is the causative agent of bacterial fruit blotch disease in watermelon. Since resistant cultivars have not yet been developed, the virulence factors/mechanisms of Ac need to be characterized. This study reports the functions of a putative pyridoxal phosphate-dependent aminotransferase (PpdaAc) that transfers amino groups to its substrates and uses pyridoxal phosphate as a coenzyme. It was observed that a ppdaAc knockout mutant had a significantly reduced virulence in watermelon when introduced via germinated-seed inoculation as well as leaf infiltration. Comparative proteomic analysis predicted the cellular mechanisms related to PpdaAc. Apart from causing virulence, the PpdaAc may have significant roles in energy production, cell membrane, motility, chemotaxis, post-translational modifications, and iron-related mechanisms. Therefore, it is postulated that PpdaAc may possess pleiotropic effects. These results provide new insights into the functions of a previously unidentified PpdaAc in Ac.

Photocatalytic hydrogen production by water splitting using novel catalysts under UV-vis light irradiation

  • Photocatalytic hydrogen generation by water splitting ($H_2O_{(1)}{\rightarrow}H_2_{(g)}+1/2O_2_{(g)}$) has been studied on photocatalysts based on Zn, Cd, Fe and Cu, synthesized by coprecipitation. Iron and copper nanoparticles were incorporated as cocatalysts to enhance the photocatalytic activity of the ZnCd solid solution. The effect of the different synthesis parameters (temperature, elemental atomic ratios, amount of Cu and Fe incorporated in the catalyst and calcination temperature) on the photocatalytic production of hydrogen has been studied in order to determine the best experimental synthesis conditions. The catalysts have been characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), and BET. The experiments of photocatalytic water splitting were performed in aqueous solution of the photocatalysts previously dispersed in a soft ultrasound bath. The photocatalysts were irradiated under different lights ranging from 220 to 700 nm. The photocatalytic activity was found to be clearly dependent on the specific area of the photocatalyst.

Investigation of Sweet and Sour Corrosion of Mild Steel in Oilfield Environment by Polarization, Impedance, XRD and SEM Studies

  • Metallic structures in the oil and gas production undergo severe degradation due to sweet and sour corrosion caused by the presence of $CO_2$ and $H_2S$ in the fluid environment. The corrosion behavior of 304 austenitic stainless was investigated in the presence of varying concentrations of $CO_2$ or $H_2S$ and $CO_2+H_2S$ to understand the effect of the parameters either individually or in combination. Potentiodynamic polarization study revealed that a small amount of $CO_2$ aided in the formation of calcareous deposit of protective layer on passive film of 304 steel, while increase in $CO_2$ concentration ruptured the layer resulting in sweet corrosion. The presence of $S^{2-}$ damaged the passive and protective layer of the steel and higher levels increased the degradation rate. Electrochemical impedance studies revealed lower polarization resistance and impedance at higher concentration of $CO_2$ or $H_2S$, supporting the outcomes of polarization study. XRD analysis revealed different types of iron carbides and iron sulphides corresponding to sweet and sour corrosion as the corrosion products, respectively. SEM analysis revealed the presence of uniform, localized and sulphide cracking in sour corrosion and general corrosion with protective carbide layer amid for sweet corrosion.

Application of the Preliminary Displacement Principle to the Temper Rolling Model

  • A mathematical model for the analysis of roll gap phenomena in the strip temper rolling process is described. A new approach to solve the roll indentation and diverging problem in modeling of severe temper rolling cases is obtained by adopting the preliminary displacement principle of two contacted rough bodies to describe the friction behavior in the roll gap. The mechanical peculiarities of the temper rolling process, such as a high friction value with high roughness rolls and a non-circular contact arc, low reduction and non-negligible entry and exit elastic zones as well as central preliminary displacement zone etc., are all taken into account. The deformation of work rolls is calculated with the influence function method and an arbitrary contact are shape is permitted. The strip deformation is modeled by the slab method and the entry and exit elastic deformation zones are included. The preliminary displacement principle is used to determine the boundaries and to calculate the friction of the central preliminary displacement zone. The model is calibrated against the production mill data and installed in the setup computer of a temper rolling mill in POSCO. The validity and precision of the model have been proven through a comparison of the measured roll forces and the predicted ones.
