  • Separation of plutonium species was studied by ion chromatography installed in a glove box for the determination of plutonium element. The plutonium species, $PuO_2^{2+},\; PC^{4+}\; and\; Pu^{3+}$, were stably separated on dynamically equilibrated cation exchanger using 1-octanesulfonate and ${\alpha}$-HiBA eluant after controlling the plutonium oxidation states with KI, $NaNO_2\;or=;KBrO_3$ based on the oxidation-reduction potentials. For the separation of plutonium from other matrix, $PuO_2^{2+}\; and\; Pu^{4+}$ were reduced to $Pu^{3+}$ with KI and $NaNO_2$ followed by cation exchange chromatography.

  • An ion chromatographic method based on indirect photometric detection of UV transparent anions was developed. Separation of anion was accomplished on strong anion exchange column (Waters SAX) using UV detector at 254 nm. Among examined UV-active additives (Dns-H, Dns-glu, Dnsser, Dns-val), Dns-glu showed the highest sensitivity. Studies on the effects of the pH and ionic strength of eluent revealed that the increase of pH and ionic strength of the eluent decreased capacity factor. The best eluent for the separation of acetate, fluoride, chloride, nitrate and bromide was $1\;{\times}\;10^{-4}M$ Dns-glu in 5 mM phosphate buffer (pH 6.30). The detection limit of chloride ion was 2.1 ng in this condition.

  • A fluorinated-sulfonated, hydrophobic-hydrophilic copolymer was planed subsequently synthesized using typical nucleophilic substitution polycondensation reaction. A novel AB and ABA (or BAB) block copolymers were synthesized using sBCPSBP (sulfonated 4,4'-bis[4-chlorophenyl)sulfonyl]-1,1'-biphenyl), DHN (1,5-dihydroxynaphthalene), DFBP (decafluorobiphenyl) and HFIP (4,4'-hexafluoroisopropylidenediphenol). All block copolymers were easily cast and made into clear films. The structure and synthesized copolymers and corresponding membranes were analyzed using GPC (gel permeation chromatography), $^1H$-NMR ($^1H$ nuclear magnetic resonance) and FT-IR (Fourier transform infrared). TGA (Thermogravimetric analysis) and DSC (differential scanning calorimetry) analysis showed that the prepared membranes were thermally stable, so that elevated temperature fuel cell operation would be possible. Hydrophobic/hydrophilic phase separation and clear ionic aggregate block morpology was confirmed in both triblock and diblock copolymer in AFM (atomic force microscopy), which may be highly related to their proton transport ability. A sulfonated BAB triblock copolymer membrane with an ion-exchange capacity (IEC) of 0.6 meq/g has a maximum ion conductivity of 40.3 mS/cm at $90^{\circ}C$ and 100% relative humidity.

  • The liquid chromatographic retention behavior of structural isomers of phenols was investigated by a change of the mobile phase properties. The retention behavior of structural isomer of phenols in reversed phase liquid chromatography was affected by eluent electrolyte added. It can be seen that this behavior is illustrated by a mechanism of Langmuir isotherm and ion exchange between phenolate and the reversed phase coated with ions. The retention behavior was represented as two different areas according to the concentration of the electrolytes. These areas can be explained as counter ion and co-ion effect, respectively. The maximum retention values were dependent not upon the kinds of organic modifier but upon the kinds of electrolyte.

  • U(IV) ion, the valance state of uranium presumed at in a deep-depth disposal of a spent fuel, was prepared and separated from U(VI) ion. In order to prepare U(IV) ion, tests were performed by adding several reducing agents into a uranyl solution or by dissolution of uranium oxide in a mixed acid added with a reducing agent. The valance states of the uranium in the prepared solutions were identified by separating two ions with a Dowex AG 50W-X8 cation exchange resins and measuring the solutions using a laser-induced fluorescence spectroscopy. However, U(IV) and U(VI) were not separated by a Lichroprep Si60 exchange resin in the same separation condition of Pu(IV) and Pu(VI).

A Comparative Study of Sulfate and Chloride Intrusion in Mortar Sections: An Approach Using Laser Induced Breakdown Spectroscopy and Ion Exchange Membrane (LIBS와 이온교환막을 활용한 모르타르 단면 침투 황산염과 염화물 분석)

  • This research aimed to conduct an empirical assessment of the penetration of chloride and sulfate ions into mortar sections using an anion exchange membrane(AEM) and laser-induced breakdown spectroscopy(LIBS). The study involved a simultaneous ion chromatography(IC) analysis and LIBS analysis performed on mortars immersed in varying concentrations of chloride and sulfate. The findings revealed that at the wavelengths specific to Chloride(837.59nm) and Sulfur(921.30nm), the LIBS intensity achieved using AEM surpassed that obtained with a paper substrate at equivalent penetration concentrations. A robust correlation was confirmed between LIBS intensity and chloride ion concentration. Furthermore, when juxtaposed with IC analysis concentration outcomes at identical depths, the AEM displayed a higher intensity. The research noted an enhancement in LIBS intensity and a diminution in errors within the low-concentration section when deploying AEM. However, for the Sulfur wavelength of 921.3nm, there remains a need to augment the sensitivity of the LIBS signal within the low-concentration section in future studies. The findings underscore the potential of employing AEM and LIBS for precise analysis of chloride and sulfate ion penetration into mortar sections. This strategy can aid in bolstering assessment precision and mitigating errors, particularly in regions with low concentrations. It is recommended to further research and develop methods to amplify the sensitivity of the LIBS signal for sulfur detection in low-concentration sections. In sum, the study accentuates the significance of employing advanced techniques like AEM and LIBS for efficacious and precise analysis in the domain of mortar section assessment.

In situ Recovery of hGM-CSF in Transgenic Rice Cell Suspension Cultures (형질전환 벼 현탁세포 배양에서 hGM-CSF의 in situ Recovery 연구)

  • Production of foreign proteins by transgenic plant cell cultures has several advantages such as post-translational modification, low risk of product contamination and low-cost production and purification. However, target proteins are degraded by extracellular proteases existing in the media. A solution to this problem is the use of perfusion culture and ion exchange chromatography for the application of integrated bioprocess using in situ recovery. With this method, production of human granulocyte-macrophage colony-stimulating factor (hGM-CSF) was investigated in this study. First, optimization of cell concentration during the induction phase for the production of hGM-CSF was examined. As cell concentration increased, the level of hGM-CSF was decreased due to the presence of extracellular proteases. Induction using sugarfree media produced 33% more hGM-CSF. The effects of pH on the binding of hGM-CSF to cationic and anionic exchange resins were also investigated. In terms of stability, optimal pH was found to be 5~7. In the case of using buffer exchange when CM-Sepharose was used as a cationic exchange resin, optimal pH for binding was 4.8 and adsorption yield was 77%. When DEAE-Sepharose was used as an anionic exchange resin, it was 5.5 (74%). Without buffer exchange, optimal pH was 4.6 (84%). From these results, an integrated bioprocess using in situ recovery with simultaneous production and separation of foreign protein in transgenic plant cell suspension cultures was found to be feasible.

Characteristics of Parathion Hydrolase by Pseudomonas rhodesiae H5 (Pseudomonas rhodesiae H5가 생산하는 Parathion Hydrolase의 특성)

  • The parathion hydrolase (OPH) produced by Pseudomonas rhodesiae H5 was purified by ammonium sulfate precipitation, DEAE-Toyopearl 650M ion exchange chromatography and Sephadex gel filtration chromatography. Parathion hydrolase from crude extracts of P. rhodesiae H5 has two components designated as OPH $I_1$ and OPH $I_2$, Optimum pH and temperature of OPH $I_1$and OPH $I_2$ were pH 7.2 and $30^{\circ}C$, and pH 7.6 and $37^{\circ}C$, respectively. The activation energy of OPH $I_1$ for the hydrolysis of parathion was 3.01 ㎉/I, II, III in the temperature range of $4^{\circ}C$ to $30^{\circ}C$, and Michaelis constant ($K_m$) for parathion was 69.2 ${\mu}M$. The activation energy of OPH $I_2$ for the hydrolysis of parathion was 4.07㎉/㏖ in the temperature range of $4^{\circ}C$ to $37^{\circ}C$, and Michaelis constant for parathion was 150.9${\mu}M$. Furthermore OPH $I_1$ was completely inhibited by 1 mM $Ca^2+$, $Cu^2+$, $Mg^2+$, $Ni^2+$, but OPH $I_2$ was less inhibited than OPH $I_1$ by the metals used in this study.

Cloning and Expression of Mammaglobin Gene (Mammaglobin 유전자 재조합 및 발현에 관한 연구)

  • In this study, I attempted to develope the expression and purification system of human mammaglobin proteins in Escherichia coli and to produce anti-human mammaglobin rabbit antibody for the detection of human mammaglobin protein in the peripheral blood of breast cancer patients. Human mammaglobin gene was cloned and sequenced from m-RNAs purified from donated breast cancer tissues using RT-PCR. The cloned gene was inserted into pET30, pET22, and pET32 plasmid. The cloned gene in pET30 yields insoluble proteins which was difficult to purify from the cells extracts. The mammaglobin gene in pET32 was strongly expressed soluble proteins which were isolated using Ni-NTA affinity chromagraphy and DEAE-ion exchange chromatography, followed by enterokinase digestion of the purified proteins. The isolated proteins had enough purity to use as a antigen for the production of anti-mammaglobin antibody in rabbits. The polyclonal antibody produced against the isolated mammaglobin showed a specificity to mammaglobin after Westernblot immuno assay. In conclusion, the isolated mammaglobin protein and the anti-mammaglobin rabbit antibody may be used for diagnosis of breast cancer as well as development of anti-breast cancer drug.

Purification and Characterization of Protease Produced by Aspergillus wentti Isolated from Korean Traditional Meju (한국 전통 메주 유래의 Aspergillus wentti가 생성하는 Protease 의 정제 및 특성)

  • The protease produced by a newly isolated Aspergillus wentti from Korean traditional Meju was purified and characterized. The optimal medium composition and culture conditions for maximum protease production were ; bran :1% glucose solution =1 : 1, pH 9.0, $30^{\circ}C$, and 4 days of fermentation. Protease was purified by QAE-Sephadex, SP-Sephadex ion exchange chromatography and Sephadex G-100 chromatography. The specific activity and the purification fold of the purified enzyme were 213 unit/mg protein and 27.3, respectively. The molecular weight of purified protease was found to be 32 kDa by SDS-PAGE. Km and Vmax value's for hammastein milk casein were $3.049{\times}10^{-4}\;M\;and\;151.1\;{\mu}g/min$, respectively. Kinetic parameters showed that the enzyme has higher affinity to casein than isolated soybean protein, hemoglobin and bovine serum albumin. Optimal pH and temperature for reaction of the purified enzyme were 9.0 and $50^{\circ}C$, respectively. The enzyme was stable at pH 4.0-11.0, below $40^{\circ}C$, and the activity was not stimulated by metal ions. 1mM phenylmethylsulfonyl fluoride inhibited the enzyme activity by 98.5%. It means that the enzyme is one of serine protease.
