  • This study aimed to compare a microparticle enzyme immunoassay (MEIA) with a liquid chromatography-tandem mass spectrometry (LC/MS/MS) technique for the measurement of tacrolimus concentrations in adult liver transplant recipients, to investigate how the assay choice influenced the population pharmacokinetics of tacrolimus and to identify patient characteristics that affected pharmacokinetic parameters in each assay. Tacrolimus concentrations from 29 liver (n=52 paired-samples) transplant recipients measured by both MEIA and LC/MS/MS were used to evaluate the performance of these methods in the clinical setting. Tacrolimus pharmacokinetics was studied independently using MEIA and LC/MS/MS data in 70 adult patients using a population approach performed with NONMEM. Patient characteristics which influenced pharmacokinetic parameters in each assay were compared. The relation between LC/MS/MS and MEIA measurements was best described by the regression equation MEIA=1.465*LC/MS/MS-1.336 (r=0.91). Multiple linear regression analysis showed significant inverse relationships between assay difference and hematocrit (Hct) (p<0.025) in liver graft recipients. In MEIA, the population estimate of tacrolimus CL/F and apparent volume of distribution (Vd/F) were found to be 10.1 L/h and 226 L, and in LC/MS/MS, 13 L/h and 305 L respectively. Neither patient's age, weight, gender, grafted hepatic weight, albumin concentration, nor markers of liver function influenced tacrolimus CL/F The final model of CL/F was found to be 10.1+(Hct/Hct mean)$^{12.0}$ in MEIA and 13+(1+Hct/578) in LC/MS/MS indicating that CL/F was influenced by hematocrit.

  • microRNA (miR)-612 shows anticancer activity in several types of cancers, yet its function in melanoma is still unclear. This study was undertaken to investigate the expression of miR-612 and its biological relevance in melanoma cell growth, invasion, and tumorigenesis. The expression and prognostic significance of miR-612 in melanoma were examined. The effects of miR-612 overexpression on cell proliferation, colony formation, tumorigenesis, and invasion were determined. Rescue experiments were conducted to identify the functional target gene(s) of miR-612. miR-612 was significantly downregulated in melanoma tissues compared to adjacent normal tissues. Low miR-612 expression was significantly associated with melanoma thickness, lymph node metastasis, and shorter overall, and disease-free survival of patients. Overexpression of miR-612 significantly decreased cell proliferation, colony formation, and invasion of SK-MEL-28 and A375 melanoma cells. In vivo tumorigenic studies confirmed that miR-612 overexpression retarded the growth of A375 xenograft tumors, which was coupled with a decline in the percentage of Ki-67-positive proliferating cells. Mechanistically, miR-612 targeted Espin in melanoma cells. Overexpression of Espin counteracted the suppressive effects of miR-612 on melanoma cell proliferation, invasion, and tumorigenesis. A significant inverse correlation (r = -0.376, P = 0.018) was observed between miR-612 and Espin protein expression in melanoma tissues. In addition, overexpression of miR-612 and knockdown of Espin significantly increased the sensitivity of melanoma cells to doxorubicin. Collectively, miR-612 suppresses the aggressive phenotype of melanoma cells through downregulation of Espin. Delivery of miR-612 may represent a novel therapeutic strategy against melanoma.

  • High temperature tensile test has been performed at $450^{\circ}C$ at different strain rate with various grain size due to different reduction rate of Al-4wt%Mg-0.4wt%Sc alloy which is known to be one of useful superplastic alloys. The grain size of Al-4wt%Mg-0.4wt%Sc alloy is $67~100\mu\textrm{m}$ which is courser than that of the alloy which is commonly used as the superplastic material. The total elongation of the Al-4wt%Mg-0.4wt%Sc alloy is strongly dependent on the average grain size, and is a linear function of the inverse average grain size for the present alloy.

  • In this paper, we propose an object tracking system which can be convinced of moving area shaped on objects through color sequential images, decided moving directions of foot messengers or vehicles of image sequences. In static camera, we suggests a new evaluating method extracting co-occurrence matrix with feature vectors of RGB after analyzing and blocking difference images, which is accessed to field of camera view for motion. They are energy, entropy, contrast, maximum probability, inverse difference moment, and correlation of RGB color vectors. we describe how to analyze and compute corresponding relations of objects between adjacent frames. In the clustering, we apply an algorithm of FCM(fuzzy c means) to analyze matching and clustering problems of adjacent frames of the featured vectors, energy and entropy, gotten from previous phase. In the matching phase, we also propose a method to know correspondence relation that can track motion each objects by clustering with similar area, compute object centers and cluster around them in case of same objects based on membership function of motion area of adjacent frames.

  • In this study, a new algorithm for canceling MRI artifact due to translational motion in image plane is described. Unlike the conventional iterative phase retrieval algorithm, in which there is no guarantee for the convergence, a direct method for estimating the motion is presented. In previous approaches, the motions in the x(read out) direction and the y(phase encoding) direction are estimated simultaneously. However, the features of x and y directional motions are different from each other. By analyzing their features, each x and y directional motion is canceled by different algorithms in two steps. First, it is noticed that the x directional motion corresponds to a shift of the x directional spectrum of the MRI signal, and the non-zero area of the spectrum just corresponds to the projected area of the density function on the x-axis. So the motion is estimated by tracing the edges between non-zero area and zero area of the spectrum, and the x directional motion is canceled by shifting the spectrum in inverse direction. Next, the y directional motion is canceled by using a new constraint condition, with which the motion component and the true image component can be separated. This algorithm is shown to be effective by using a phantom image with simulated motion.

  • This paper describes a method for the detection of shorted-turn in the cylindrical synchronous generator rotor windings based on the discrete wavelet transform. Multi-resolution analysis(MRA) based on discrete wavelet transform provides a set of decomposed signals in independent frequency bands, which contain independent dynamic information due to the orthogonality of wavelet function. In the proposed method, shorted-turn detection in rotor windings is based on the decomposition of the rotor currents, where wavelet coefficients of these signals have been extracted. Comparing these extracted coefficients is used for diagnosing the healthy machine from faulty machine. Experimental results are presented for healthy, and machines with 25%, 42%, 67%, 83%, 99% inter-turn short circuits in a rotor slot. Deviation of wavelet coefficients in healthy mode from faulty modes depicts the inverse proportion of shorted-turns. Experimental results show the effectiveness of the proposed method for shorted-turn detection in the cylindrical synchronous generator rotor windings.

  • The flow stress of zircaloy-4 used in the spacer grid supporting a nuclear fuel rod was determined by the Johnson-Cook model, and model parameters were determined using reverse engineering. Parameters such as A, B, n and $\dot{\varepsilon}_0$ were determined by the tensile test result. To obtain the parameters C and m, a slot milling test and numerical simulation were performed. The objective functions were defined as the difference between the experimental and the simulation results, and then, the parameters were determined by minimizing the objective function. To verify the validity of the determined parameters, cross-verification for each case was conducted through a shearing test and simulation. The results tend to show agreement with the experimental results, such as the features of sheared edges and maximum punch force, with the correlation coefficients exceeding at least 0.97.

  • Retrieval effectiveness is the principal criterion for measuring the performance of an information retrieval system. The effectiveness of a retrieval system depends primarily on the extent to which it can retrieve wanted documents without retrieving unwanted ones. So, ultimately, effectiveness is a function of the relevant and nonrelevant documents retrieved. Consequently, 'relevance' of information to the user's request has become one of the most fundamental concept encountered in the theory of information retrieval. Although there is at present no consensus as to how this notion should be defined, relevance has been widely used as a meaningful quantity and an adequate criterion for measures of the evaluation of retrieval effectiveness. The recall and precision among various parameters based on the 'two-by-two' table (or, contingency table) were major considerations in this paper, because it is assumed that recall and precision are sufficient for the measurement of effectiveness. Accordingly, different concepts of 'relevance' and 'pertinence' of documents to user requests and their proper usages were investigated even though the two terms have unfortunately been used rather loosely in the literature. In addition, a number of variables affecting the recall and precision values were discussed. Some conclusions derived from this study are as follows: Any notion of retrieval effectiveness is based on 'relevance' which itself is extremely difficult to define. Recall and precision are valuable concepts in the study of any information retrieval system. They are, however, not the only criteria by which a system may be judged. The recall-precision curve represents the average performance of any given system, and this may vary quite considerably in particular situations. Therefore, it is possible to some extent to vary the indexing policy, the indexing policy, the indexing language, or the search methodology to improve the performance of the system in terms of recall and precision. The 'inverse relationship' between average recall and precision could be accepted as the 'fundamental law of retrieval', and it should certainly be used as an aid to evaluation. Finally, there is a limit to the performance(in terms of effectiveness) achievable by an information retrieval system. That is : "Perfect retrieval is impossible."

  • Semantic Web is getting popular to next web environment. Additionally, ontology language research is also activating to represent semantic relation of resource in semantic web. Specially, Ontology language as RDF and DAML+OIL appear on start point of research. But Ontology Language limited to describing characters of resource and to making a clear definition of relation of resource. So W3C suggest OWL at the next standard language for describing resource. OWL supply the lack of representation for RDF and RDF Schema. In this paper, we make Ontology to implement Online Retrieval System using OWL and propose the structure of storing Ontology document at the RDB. The structure support characters of OWL that are equivalent relationship, heterogeneous relationship, inverse relationship, union relationship and one of relationship between classes or properties. In this paper, we classify the extended elements for OWL from RDF Schema. And we propose the method of storing OWL using RDB for interoperability with many applications based on RDB. Finally, implement the storage and retrieval system based on OWL to provide advanced search function.
