Suppression of Migration and Invasion by Alnus hirsuta in Human Hepatocellular Carcinoma Cells

  • Hepatocellular carcinoma (HCC) has a poor prognosis and high metastasis and recurrence rates. Although extracts of Alnus hirsuta (Turcz. ex Spach) Rupr. (AH) have been demonstrated to possess potential anti-inflammatory and anti-cancer activities, the underlying mechanism of AH in HCC treatment remains to be elucidated. We investigated the effects and potential mechanisms of AH on migration and invasion of Hep3B cells. Within the non-cytotoxic concentration range, AH significantly inhibited motility and invasiveness of Hep3B cells in a concentration-dependent manner. Inhibitory effects of AH on cell invasiveness are associated with tightening of tight junctions (TJs), as demonstrated by an increase in transepithelial electrical resistance. Immunoblotting indicated that AH decreased levels of claudins, which form major components of TJs and play key roles in the control and selectivity of paracellular transport. Furthermore, AH inhibited the expression and activity of matrix metalloproteinase (MMP)-2 and MMP-9 and simultaneously increased the levels of tissue inhibitor of metalloproteinase (TIMP)-1 and TIMP-2. These effects were related to inactivation of the phosphoinositide 3-kinase (PI3K)/AKT pathway in Hep3B cells. Therefore, AH inhibits migration and invasion of Hep3B cells by inhibiting the activity of MMPs and tightening TJs through suppression of claudin expression, possibly by suppressing the PI3K/AKT signaling pathway.

사람 U2OS 골육종 세포에서 Matrix Metalloproteinase의 발현에 Ibandronate가 미치는 영향 (Effects of Ibandronate on the Expression of Matrix Metalloproteinases in Human U2OS Osteosarcoma Cells)

  • 배경: 골육종은 소아 및 청소년기에 가장 흔하게 발생하는 악성 골종양 중 하나이다. 최근 수술적 치료와 항암 화학요법을 병행하여 생존률이 증가하였지만, 아직까지도 항암제는 항암제 내성 및 이차성 악성 종양의 발생 등 여러 문제점을 가지고 있다. 일부 종양은 matrix metalloproteinase(MMPs)의 발현이 증가되어 있고, MMP inhibitor에 대한 연구가 진행되고 있다. 반면 bisphosphonate(BPs) 제제는 골흡수를 억제하는 능력이 있으며, 파골세포와 관계된 골 병변에 광범위하게 사용되고 있다. 또 bisphosphonate 제제는 직접적인 항암효과를 가지고 있는 것으로 알려져 있다. 대상 및 방법: 골육종 세포주(U2OS)를 ibandronate(0, 0.1, 1, 10M)를 이용하여 48시간 동안 처치하였다. 세포의 생존능은 MTT assay를 이용하여, MMP-2와 MT1-MMP의 mRNA level은 reverse-transciption polymerase chain reaction을 이용하여 측정하였으며, MMP-2와 MT1-MMP 단백의 양은 Westernblot을, MMP-2의 활성은 Gelatin zymography를 이용하여 측정하였다. 또, ibandronate 처치 전후의 골육종 세포주의 침습성은 Matrigel invasion assay를 이용하여 측정하였다. 결과: 48시간 ibandronate 처치 후 U2OS 세포주의 침습력은 ibandronate에 대해 용량 의존적으로 감소하였다. 특히 10M 이내의 ibandronate는 세포독성이 나타나지 않았다. 젤라틴 융해능과 MMP-2 및 MT1-MMP의 단백 및 mRNA 정도역시 ibandronate 농도가 증가할수록 감소하였다. 결론: Ibandronate는 골육종 세포주의 MMP-2 및 MT1-MMP의 발현을 억제하였으며, 종양세포의 침습력을 감소시켰다. Bisphosphate의 종양세포 침습 억제 능력은 새로운 전이 억제제의 개발에 도움이 될 것으로 사료된다.

레트로바이러스를 이용한 Tissue Inhibitor of Metalloproteinase-2 유전자 발현이 대장암 세포의 전이 및 종양형성에 미치는 영향 (Anti-tumorigenic and Invasive Activity of Colon Cancer Cells Transfected with the Retroviral Vector Encoding Tissue Inhibitor of Metalloproteinase-2)

  • Matrix metalloproteinases (MMPs) and tissue inhibitors of MMPs (TIMPs) playa key role in tumor invasion and metastasis. As an inhibitor of MMP-2, TIMP-2 is known to block both the invasive and metastatic behavior of cancer cells, and decrease tumor growth activity. We performed this study to investigate the effects of TIMP-2 over-expression induced by retroviral mediated gene transfer in vitro and in vivo. The human colon cancer cell line SW480 was transfected with the retroviral vector encoding TIMP-2. The effects of TIMP-2 over-expression were analyzed by invasion assay and gelatinase activity test in colon cancer cells and tumorigencity in nude mice. In evaluation of the transfection efficiency of the retroviral vector encoding TIMP-2 in colon cancer cells, we confirmed up-regulation of TIMP-2 expression dependent on the time of cell culture. In addition, inhibition of MMP-2 expression in SW480/TIMP-2 was shown by gelatin zymography. In the in vitro invasion assay SW480/TIMP-2 inhibited the invasiveness on matrigel coated with collagen. To determine whether TIMP-2 can modulate in vivo tumorigenicity and metastasis, SW480/TIMP-2 cells were injected subcutaneously in nude mice. The tumor mass formation of SW480/TIMP-2 cells in nude mice was markedly decreased compared to nontransfected cancer cells. These results showed that colon cancer cells transfected with the retroviral vector encoding TIMP-2 inhibits the invasiveness in vitro and tumorigenicity in vivo.

Protein tyrosine phosphatase controls breast cancer invasion through the expression of matrix metalloproteinase-9

  • The expression of matrix metalloproteinases (MMPs) produced by cancer cells has been associated with the high potential of metastasis in several human carcinomas, including breast cancer. Several pieces of evidence demonstrate that protein tyrosine phosphatases (PTP) have functions that promote cell migration and metastasis in breast cancer. We analyzed whether PTP inhibitor might control breast cancer invasion through MMP expression. Herein, we investigate the effect of 4-hydroxy- 3,3-dimethyl-2H benzo[g]indole-2,5(3H)-dione (BVT948), a novel PTP inhibitor, on 12-O-tetradecanoyl phorbol-13-acetate (TPA)-induced MMP-9 expression and cell invasion in MCF-7 cells. The expression of MMP-9 and cell invasion increased after TPA treatment, whereas TPA-induced MMP-9 expression and cell invasion were decreased by BVT948 pretreatment. Also, BVT948 suppressed NF-${\kappa}B$ activation in TPA-treated MCF-7 cells. However, BVT948 didn't block TPA-induced AP-1 activation in MCF-7 cells. Our results suggest that the PTP inhibitor blocks breast cancer invasion via suppression of the expression of MMP-9.

Cobalt Chloride-Induced Downregulation of Puromycin-Sensitive Aminopeptidase Suppresses the Migration and Invasion of PC-3 Cells

  • Cobalt chloride ($CoCl_2$) treatment of cells in vitro has been shown to induce cellular changes that are similar to those seen following hypoxia. To identify genes that are differentially expressed in response to treatment with $CoCl_2$, we compared the mRNA expression profiles of PC-3 cells that were treated with $CoCl_2$ with those of untreated PC-3 cells, using specific arbitrary primers and two anchored oligo(dT) primers provided in the ACP-based GeneFishing kits. The results of this study demonstrated that the puromycin-sensitive aminopeptidase (PSA) gene was down regulated in PC-3 cells that were treated with $CoCl_2$. This downregulation of PSA expression, in turn, suppressed the proliferation, migration, and invasion of PC-3 cells, as well as the secretion and expression of matrix metalloproteinase-9 (MMP-9).

Conjugated Linoleic Acid (CLA)가 인체 대장암 세포주에서 Matrix Metalloproteinase (MMP) 활성과 세포이동성에 미치는 영향 (Effects of Conjugated Linoleic Acid (CLA) on Matrix Metalloproteinase (MMP) Activity and Cell Motility in Human Colon Cancer Cell Lines)

  • Conjugated linoleic acid (CLA) consists of several geometric isomers of linoleic acid. CLA is found in foods derived from ruminants and exhibits strong anticarcinogenic effects in a variety of animal models. Matrix metalloproteinases (MMPs) play a key role in cancer progression. Specifically, MMP-2 and -9, which hydrolyze the basal membrane type IV collagen, are involved in the initial breakdown of collagen and basement membrane components during tumor growth and invasion. However, the effects of CLA on cancer cell motility and MMP expression and activity are not currently well known. Therefore, the present study examined whether CLA reduces the activity of MMP and cell motility in SW480 and SW620 cells, the human colon cancer cell lines. Gelatin zymography and Western blot analysis revealed that phorbol 12-myristate 13-acetate (PMA) induced the activity and protein expression of Mr 92,000 MMP-9 in both cell lines. To examine whether CLA inhibits the MMP activity, cells were incubated with 100 ngfmL PMA in the presence of various concentrations of CLA. PMA-induced MMP-9 activity was decreased by 20 $\mu$ M CLA in SW480 cells, and by 10 $\mu$ M and 20 $\mu$ M CLA in SW620 cells. Results from the Hoyden chamber assay showed that cell motility was increased by PMA and that PMA-induced cell motility was significantly decreased by 20 $\mu$ M CLA in SW480 cells. These results indicate that CLA may reduce the motility and MMP activity in human colon cancer cells.

Inhibitory Effect of Ligularia Stenocephala on the Cancer Metastasis

  • Ligularia stenocephala has been used as a traditional medicine for the treatment of asthma, arthritis, jaundice, and hyperpiesia. In this study, we investigated the anti-metastatic and hypnotic effects of the methanolic extract of L. stenocephala (MLS). Gelatin zymographic analysis revealed that MLS suppresses matrix metalloproteinase-2 (MMP-2) and MMP-9 activities in B16F10 cells. The gene expressions of MMPs were also down-regulated by MLS treatment in a dose-dependent manner. In addition, cancer cell invasion and migration were attenuated by MLS via suppression of NF-${\kappa}B$ activation. The in vivo lung metastasis of B16F10 melanoma cells was also inhibited by the treatment of MLS. These findings show that MLS has anti-metastatic properties, and, therefore, it might be applicable as a valuable anti-metastatic agent.

HCT116 인체 대장암 세포주에서 상백피 추출물에 의한 전이 억제 효과 (Root Bark extract of Morus alba L. Suppressed the Migration and Invasion of HCT116 Human Colorectal Carcinoma Cells)

  • The root bark of Morus alba L. (MA) used in traditional oriental medicine for the treatment of pulmonary diseases exerts various pharmacological activities including anticancer effects. In the current study, we investigated the effects of MA on the migration and invasion of colorectal carcinoma cells. Results from a transwell assay showed that the methylene chloride extract of MA (MEMA) suppressed the migration and invasion of HCT116 human colorectal carcinoma cells in a concentration-dependent manner. MEMA reduced both mRNA and protein levels of matrix metalloproteinase (MMP)-9, but did not suppress the expression of MMP-2 in HCT116 cells. As a molecular mechanism, MEMA inhibited the phosphorylation of mitogen-activated protein kinases (MAPKs), including ERK, JNK and p38, in a dose-dependent manner. In addition, MEMA dephosphorylated both Src and signal transducer and activator of transcription 3 (STAT3) in HCT116 cells. Taken together, we demonstrate that MEMA suppressed the migration and invasion capacity of HCT116 human colorectal cancer cells by downregulation of MMP-9 and inactivation of both MAPKs and Src/STAT3 signaling pathway.

TIMP-2 유전자 재조합 아데노바이러스의 폐암세포 침윤 억제 효과 (TIMP-2 Gene Transfer Via Adenovirus Inhibits the Invasion of Lung Cancer Cell)

  • 연구배경 : 폐암은 진단 당시 이미 국소 침윤이나 원격 전이가 된 경우가 많고 이에 대한 적절한 치료법이 없기 때문에 예후가 불량하다. TIMP(tissue inhibitor of metalloproteinase)는 암세포의 침윤 및 전이에 중요한 역할을 하는 metalloproteinase를 억제하는 물질로서 생체 내에 존재하는 전이 억제 물질이다. 본 연구는 아데노바이러스를 이용한 TIMP 유전자 치료법을 개발하여 폐암의 치료에 응용하고자 하였다. 방법 : 폐암세포는 침윤 및 전이 능력이 큰 Calu-6를 사용하였다. TIMP-2 유전자를 pACCMVpLpA에 subcloning 한 후 pJM17과 함께 293 cell에 cotransfection 한 후 homologous recombination을 이용하여 Ad-TIMP-2를 제작하였다. Ad-TIMP-2를 Calu-6 cell에 이입하여 TIMP-2 protein 이 생산되는지를 TIMP-2 ELISA를 이용하여 확인하였고 TIMP-2의 생물학적 활성은 zymography로 확인하였다. Soft agar clonogenic assay로 종양형성능을 평가하였다. Ad-TIMP-2로 처리한 calu-6를 6주간 soft agar에서 키운 후 육안으로 보이는 colony 수를 측정하였다. Matrigel을 이용하여 invasion assay를 시행하여 calu-6의 침윤 능력의 변화를 평가하였다. 결과 : TIMP-2 ELISA 결과, 모세포 calu-6와 Ad-$\beta$-gal 이입 calu-6 그리고 Ad-TIMP-2 이입 calu-6는 각각 0.44, 0.43, 20.7 ${\mu}g/10^6$ cells/72hrs의 TIMP-2를 생산하였다. Zymography 결과 Ad-TIMP-2에 의해 생산된 TIMP-2는 matrix metalloproteinase-2의 gelatin 분해 효과를 억제하여 생물학적 활성을 확인할 수 있었다. Soft agar clonogenic assay 결과, 모세포인 calu-6는 453$\pm$53개, Ad-$\beta$gal과 Ad-TIMP-2 이입된 calu-6는 각각 332$\pm$35, 280$\pm$45개의 colony가 형성되어 유의한 감소를 보이지 못했다. Invasion assay 로 모세포 calu-6 에 대한 침윤율을 평가한 결과, Ad-$\beta$gal과 Ad-TIMP-2가 이입된 calu-6(10moi)는 각각 71$\pm$8.9%, 12$\pm$8.4%의 침윤율을 보였으며 $\beta$-gal 군에 비해 TIMP-2군이 유의하게 침윤율이 낮았다. 결론 : Ad-TIMP-2는 폐암 세포의 종양형성능을 억제하지 못하였으나 침윤은 억제하여 TIMP-2가 폐암 유전자 요법에 이용될 가능성을 제시해 주었다.

Inhibition of Proinflammatory Cytokine-induced Invasiveness of HT-29 Cells by Chitosan Oligosaccharide

  • The effect of chitosan oligosaccharide (COS, 1 kDa${\gamma}$, 10 ng/ml IL-$1{\alpha}$, and 25 ng/ml TNF-${\alpha}$) in HT-29 cells. Inducible nitric oxide synthase (iNOS) expression induced by these cytokines was inhibited by COS. COS pretreatment inhibited the invasiveness of cytokines-treated HT-29 cells through Matrigel-coated membrane in a dose-dependent manner. COS also inhibited cytokines-induced matrix metalloproteinase (MMP)-2 activity. This study shows that proinflammatory cytokines induce NO production, iNOS expression, and invasiveness of human colorectal adenocarcinoma HT-29 cells. COS pretreatment inhibited cytokines-mediated NO production, iNOS expression, and invasiveness of HT-29 cells. These results provide sufficient information for the further development of COS as an antitumor metastatic agent for the treatment of colon cancer.
