세균액 및 세균단백질 추출물이 배양 세포에 미치는 영향 (EFFECTS OF HEAT-KILLED AND SONIC EXTRACTS OF MICROORGANISM ON CULTURED CELLS)

  • Dental pulp infection is most commonly caused by extensive dental caries, and some bacterial species invade root canals; bacterial components and products are thought to be associated with the pathogenesis of periapical periodontitis. A principle driving force behind pulpal disease response appears to lie in the host immune system's to bacteria and their products. We examined the production of interleukin $1{\beta}$ (IL-$1{\beta}$) and tumor necrosis factor ${\alpha}$(TNF-${\alpha}$) from human peripheral mononuclear cells, lymphocytes and monocytes stimulated by heat-killed Acitnobacillus actinomycetemcomitans (ATCC 29523), Porphyromonas gingivalis (ATCC 33277) and Prevotella intermedia (ATCC 25611), and also by their sonicated bacterial extracts (SBE), respectively. The effects of three strains of heat-killed bacteria and their SBEs on the morphology of cultured blood cell lines HL-60 (KCLB 10240) and J774A.1 (KCLB 40067) were observed under the inverted microscope. Ultrastructural changes of J774A.1 exposed to heat-killed P. intermedia and its SBE were investigated using transmission electron microscopy. Production of IL-$1{\beta}$ was reduced in human peripheral mononuclear cells after stimulation by sonic bacterial extracts of A. actinomycetemcomitans, P. gingivalis, and P. intermedia. Heat-killed and sonic extract of P. gingivalis inhibited the production of TNF-${\alpha}$ in peripheral mononuclear cells. Production of TNF-${\alpha}$ was inhibited in peripheral monocytes after stimulation by sonic extracts of A. actinomycetemcomitans, P. gingivalis, and P. intermedia. HL-60 and J 774A.1 cells showed granular degeneration after treatment with heat-killed and sonic extracts of A. actinomycetemcomitans, P. gingivalis, and P. intermedia Chromatin margination and shrinkage were observed in 774A.1 treated with heat-killed P. intermedia. Cell wall structure and organelles were destroyed and vacuoles were formed in cytoplasm in J774A.1 treated with P. intermedia sonic extract. These results suggest that A actinomycetemcomitans, P gingivalis and P intermedia may have an important role in the formation and progression of pulpal diseases via both modulation of production of IL-$1{\beta}$ and TNF-${\alpha}$ from blood mononuclear cells and cytopathic effects.

Effects of α-lipoic acid on LPS-induced neuroinflammation and NLRP3 inflammasome activation through the regulation of BV-2 microglial cells activation

  • Microglial cells are known as the main immune cells in the central nervous system, both regulating its immune response and maintaining its homeostasis. Furthermore, the antioxidant ${\alpha}-lipoic$ acid (LA) is a recognized therapeutic drug for diabetes because it can easily invade the blood-brain barrier. This study investigated the effect of ${\alpha}-LA$ on the inflammatory response in lipopolysaccharide (LPS)-treated BV-2 microglial cells. Our results revealed that ${\alpha}-LA$ significantly attenuated several inflammatory responses in BV-2 microglial cells, including pro-inflammatory cytokines, such as tumor necrosis $factor-{\alpha}$ and interleukin (IL)-6, and other cytotoxic molecules, such as nitric oxide and reactive oxygen species. In addition, ${\alpha}-LA$ inhibited the LPS-induced phosphorylation of ERK and p38 and its pharmacological properties were facilitated via the inhibition of the nuclear factor kappa B signaling pathway. Moreover, ${\alpha}-LA$ suppressed the activation of NOD-like receptor pyrin domain containing 3 (NLRP3) inflammasomes, multiprotein complexes consisting of NLRP3 and caspase-1, which are involved in the innate immune response. Finally, ${\alpha}-LA$ decreased the genes accountable for the M1 phenotype, $IL-1{\beta}$ and ICAM1, whereas it increased the genes responsible for the M2 phenotype, MRC1 and ARG1. These findings suggest that ${\alpha}-LA$ alleviates the neuroinflammatory response by regulating microglial polarization.

우면산 등산로 주변 서양등골나물의 분포 경향 (Distribution Pattern of Ageratina altissima Along Trails at Mt. Umyeon in Seoul, Korea)

  • 서양등골나물(Ageratina altissima)은 우리나라의 육상생태계를 교란시키는 외래종으로 알려져 있다. 교란 지역으로부터 그늘이 지는 숲 속으로 침입하는 것으로 알려져있으나, 한국에서 나타나는 서양등골나물의 분포 특성에 대해서는 양지와 음지에 대한 다양한 보고가 있다. 본 연구는 서울 우면산을 대상으로 광도 및 토양 특성과 서양등골나물의 분포의 관계, 등산로 주변 서양등골나물 분포 양상을 살펴보았다. 서양등골나물의 분포는 상대광도가 증가함에 따라 증가하고, 낙엽층이 깊어지면 감소하는 것으로 나타났다(p < 0.001). 서양등골나물은 우면산에서 주로 등산로 주변에서 분포하고 있었으며, 특히 등산로로부터 숲으로 10m 이내에 대부분이 분포하고 있었는데(p < 0.01), 이 지역은 빛과 임상 상태가 서양등골나물에게 유리한 지역이었다. 서양등골나물은 계곡보다는 능선에서 더 많이 분포하였으며, 지형에 따른 차이는 등산로로부터의 거리 10m를 전후로 한 9m, 12m 위치에서 가장 컸다(p < 0.05). 토양의 수분함량, 석력함량, pH 등은 서양등골나물의 분포와 뚜렷한 상관관계를 보이지 않는 것으로 나타났으며, 이는 서양등골나물이 다양한 토양 환경에 대한 적응성을 가지고 있다는 것을 알려주었다. 본 연구는 등산로가 외래종의 침입에 취약한 지역임을 보여주었으며, 향후 등산로 관리에서 외래종의 침입에 대한 대책의 필요성을 알려주었다.

세가시육질꼬리옆새우(단각류)의 생물교란이 시화호 퇴적물 내 황화합물과 산소 농도의 수직적 분포에 미치는 영향 (Effects of Bioturbation of the Amphipod Monocorophium uenoi on the Vertical Distribution of Sulphides and Oxygen within Sulphides-Rich Sediments of Lake Sihwa)

  • 시화호의 우점종 중의 하나인 세가시육질꼬리옆새우의 염분구배 (1, 5, 10, 15, 20, 25, 30, 35, 40‰)에 따른 생존율 및 튜브형성 개수 실험을 하였다. 그리고 수층 내 산소농도가 충분할 경우 황화합물 농도가 높은 퇴적물 속에서 대상 종의 생존율과 이들의 생물교란 활동이 퇴적물 내 산소농도와 황화합물의 수직구배에 미치는 영향을 실험하였다. 대상 종은 20~30‰ 염분 조건에서 생존율이 높았으며 튜브형성 개수 또한 많았다. 수층 내 산소 농도가 충분할 경우, 대상 종은 황화합물 퇴적물에서 높은 생존율을 나타내었다. M. uenoi의 생물교란 활동은 퇴적물 내로 산소를 확산시킴으로써 황화합물을 제거하였다. 산소 확산과 황화합물이 제거되는 깊이는 대상 종의 서식밀도가 높을수록 증가하였다. 시화호 퇴적물에는 황화합물의 농도가 높게 나타나며, 특히 여름철에는 밀도성층의 강화로 저층수에 빈산소층이 형성되어 저서동물이 서식하지 못하는 무생물대가 형성된다. 가을 무렵에는 성층의 약화로 저층수에도 산소가 충분히 공급되며 M. uenoi가 높은 밀도로 출현한다. M. uenoi의 생물교란 활동은 저서동물 군집이 재형성될 수 있는 서식지를 조성하는데 중요한 역할을 하는 것으로 판단되었다.

Immunomodulatory Properties of Lactobacillus plantarum NC8 Expressing an Anti-CD11c Single-Chain Fv Fragment

  • The lactic acid bacteria species Lactobacillus plantarum (L. plantarum) has been used extensively for vaccine delivery. Considering to the critical role of dendritic cells in stimulating host immune response, in this study, we constructed a novel CD11c-targeting L. plantarum strain with surface-displayed variable fragments of anti-CD11c, single-chain antibody (scFv-CD11c). The newly designed L. plantarum strain, named 409-aCD11c, could adhere and invade more efficiently to bone marrow-derived DCs (BMDCs) in vitro due to the specific interaction between scFv-CD11c and CD11c located on the surface of BMDCs. After incubation with BMDCs, the 409-aCD11c strain harboring a eukaryotic vector pValac-GFP could lead to more efficient expression of GFP compared with wild-type strains shown by flow cytometry analysis, indicating the enhanced translocation of pValac-GFP from L. plantarum to BMDCs. Similar results were also observed in an in vivo study, which showed that oral administration resulted in efficient expression of GFP in both Peyer's patches (PP) and mesenteric lymph nodes (MLNs) within 7 days after the last administration. In addition, the CD11c-targeting strain significantly promoted the differentiation and maturation of DCs, the differentiation of $IL-4^+$ and $IL-17A^+$ T helper (Th) cells in MLNs, as well as production of $B220^+$ $IgA^+$ B cells in the PP. In conclusion, this study developed a novel DC-targeting L. plantarum strain which could increase the ability to deliver eukaryotic expression plasmid to host cells, indicating a promising approach for vaccine study.
