Intraspecific variations in macronutrient, amino acid, and fatty acid composition of mass-cultured Teleaulax amphioxeia (Cryptophyceae) strains

  • To compare the nutritional quality of TPG (Teleaulax / Plagioselmis / Geminigera) clade species of cryptomonads with that of RHO (Rhodomonas / Rhinomonas / Storeatula) clade species 6 Teleaulax amphioxeia (TA) and 1 Rhinomonas sp. strains were mass-cultured in newly designed 500-L photobioreactors to the end of exponential growth phase. Intraspecific variations (IVs) in terms of one standard deviation among the 6 TA strains in the compositions of the three macronutrients were 41.5 (protein), 89.8 (lipid), and 15.6% (carbohydrate) of the mean. When harvested from stationary growth phase mean compositions of essential amino acids (EAAs, 47.3%) and non-EAAs (52.7%) of the 2 TA strains, CR-MAL07 and CR-MAL08-2, were similar to those of a Chroomonas strain. The IVs between the 2 TA strains in the composition of EAAs (10.3 and 2.4) and non-EAAs (8.5 and 2.1% of the mean) were rather smaller than those of saturated fatty acids (30.3 and 26.1) and unsaturated fatty acids (UFAs, 12.0 and 12.5% of the mean) in f/2-Si and urea-based compound fertilizer (UCF) culture media, respectively. Mean compositions of eicosapentaenoic acid (EPA, 17.9%) and docosahexaenoic acid (DHA, 12.7%) of total fatty acids of the 2 TA strains were higher than those that of a Chroomonas strain. EPA and DHA compositions exhibited similar level of IVs between the 2 TA strains in f/2-Si (14.6 and 11.0) and UCF media (12.6 and 13.5% of the mean). Thus, the nutritional quality in terms of amino acids, UFAs, EPA, and DHA in a TPG clade species, T. amphioxeia was comparable to those of RHO clade species with notable IVs. Practically, biotechnological targets for TPG clade cryptomonad strains might be subspecies or clone level.

Egg Production of Clonorchis sinensis in Different Strains of Inbred Mice (근교계 마우스에서 간흡충 기생기간과 산란력의 변동)

  • In order to compare the intraspecific variation in host-parasite relationship of Clonorchis sinensis, six strains of inbred mice, ICR, DDY, GPC, BALB/c, nude and DS, were infected orally with 20 metacercariae of C. sinensis. The biologic incubation period of C. sinensis was the shortest in DDY mice, 21.2 days in average, followed by GPC 21.4, BALB/c and DS 23.2, ICR and nude 23.4 days, respectively. The fertile period of the cuke was also the longest in the DDY strain, 164 days on average, followed by GPC 132, BALB/c 97, nude 37, DS 32 and ICR 28 days. The egg-laying capacity of the cuke in DDY and GPC was relatively high and stable compared with the other four strains of mice. It was found that there are intraspecific variations in biologic incubation period, fertile period, and fecundity of C. sinensis. The DDY mouse is likely to be the most suitable experimental animal among the six strains of the mice tested. Key words: Mouse strain, Clonorchis sinensis, egg-laying capacity.

Platynosomum fastosum (Trematoda: Dicrocoeliidae) from Cats in Vietnam: Morphological Redescription and Molecular Phylogenetics

  • The present study was performed to reveal the morphological characteristics and molecular phylogenetic position of Platynosomum fastosum Kossack, 1910. A total 167 specimens of P. fastosum were collected in 8 (4.9%) out of 163 sets of gall-bladders and bile ducts of cats. The number of worms was 1-105 per infected cat. This species was characterized by having a long and slender body, slightly larger ventral sucker than the oral sucker, indistinct prepharynx, small pharynx, short esophagus, bifurcation midway between 2 suckers, and ceca extending to the posterior end of the body. The length of the partial sequences of ITS1 and 5.8S rDNA of P. fastosum were 990 bp, GC-rich. AT/GC ratio was 0.9, there were 9 polymorphic sites, and intraspecific variations ranged from 0.1% to 0.9%. Phylogenetic analyses by neighbor-joining phylogram inferred from ITS1 rDNA sequences revealed that the genetic distance between P. fastosum specimens ranged from 0.3 to 1.5% while the smallest interspecific distance among dicrocoeliid species was 20.9 %. The redescription and genetic characters of P. fastosum are taxonomically important to recognize future different species of the genus Platynosomum showing high intraspecific and morphological variability.

  • To evaluate the intraspecific variations among the Kasiogalpi(Eleutherococcus senticosus Max.) collections. randomly amplified DNA polymorphisms were examined. Twenty primers from 90 primers applied were selected. The range of polymorphism was $7.1{\sim}90.9%$ in 113 randomly and specifically amplified DNA fragments. Collections were divided into two major groups at the similarity coefficient value of 0.65. A considerable degree of genetic diversity was also detected among plants within the same collections. Deokyu (1, 2, 3, 4, 6), Bukhaedo (7, 8) and Odae (9, 10) col1ections showed higher degree of genetic similarity with a value of $0.65{\sim}0.86$, while Deokyu 5 showed much lower genetic similarity than other col1ections.

  • To analyse the genetic relationship and intraspecific variations among the Eleutherococcus senticosus population, the polymerase chain reaction(PCR) was performed total genomic DNAs of 10 E. senticosus collections by random 10 primers. The genetic diversity and genetic distance among 10 collections of Eleutherococcus spp. were used to describe the dendrogram showing phylogenic relationship. Ten collections were classfied into two group(group I, II) at the similarity coefficient value of 0.50. Group I included E. senticosus of Bukhado(Japanese), youngwal(Korea), E. seoulense, and E. chiisanesis while group II included several internal and Russia collection. The range of polymorphism was from 66.7 to 90.9% in 87 amplified DNA fragments. The similarity value of all collections ranged from 0.41 to 0.92. The average of genetic distance was 0.61.

  • This study deals with the characterization of three populations (two hatchery and one natural) of Indian major carps Catla catla and Labeo rohita from different locations in India. The genetics of Indian major carps has been completely obscure and this is the first report on comparative allozyme variations in natural and hatchery population. The total 10 biochemical genetic markers used to measure interspecific and intraspecific level of diversity. The allele frequency data indicate different level of genetic variability in three populations. The hatchery population exhibited least polymorphism, low level of heterozygosity and genetic diversity.

Structure and Reproduction of Grateloupia filicina (Halymeniaceae, Rhodophyta) from Indian Coast

  • The vegetative and reproductive features of Grateloupia filicina (Lamouroux) C. Agardh (Cryptonemiales, Halymeniaceae) from different parts of the Indian Coast were studied. The plants grow in wide range of habitats and showed a lot of morphological variations. The development of the thallus is multiaxial type and the medullary region is composed of irregular, branched and stellate shaped cells. The gametophytic plants are dioecious and the male plants are smaller compared to female plants. The carpogonial branch is two-celled and formed on an a accessory branch system known as ampulla. Cystocarps are spherical to sybspherical with distint ostioles and scattered on the thallus surface. Tetrasporangia are common and tetraspores are either cruciate or decussate. Bisporangia are occasionally encountered. Our study suggests occurrence of two intraspecific taxa of G. filicina i.e.: var. luxurians and var. filinina from India coast.

Ecotypic Variation in Salinity Responses of Ulva pertusa(Chlorophyta) from the Korean Coast

  • Salinity ecotypes in Ulva pertusa Kjellman were examined for the growth responses of the three isolates taken from different salinity regimes. All isolates showed a broad salinity tolerance, but growth patterns were correlated with the salinity regime of their original habitat. The germlings from Anin exhibited optimum growth at the native salinity of 32%. The germlings from Yongyon which had hypersaline habitats were tolerable to high salinity, i.e. growth rates peaked at 40%, whereas those from Samgando which had low salinities achieved maximum growth rate at 24\%. The germlings of inter-isolate cross demonstrated intermediate growth response between that of their respective parents. Our data also clearly indicated intraspecific differences among the three isolates, which was interpreted as development of different physiological ecotypes. We conclude that U. pertusa may consist of several ecotypes, each of which has some capacity for physiological adaptation to salinity variations.

Studies on the Intraspecific Variations on Geological Distributions of Acusta despecta sieboldiana in Korea (한국산 달팽이 ( Acusta despecta sieboldiana) 의 지리적 분포에 따른 종내 변이에 관한 연구)

  • 국내 11개 지역(경기도 옹진군 덕적도, 경기도 의정부시 소요산, 강원도 춘천시, 충청남도 태안군 안홍, 경상북도 울릉군 울릉도, 전라북도 부안군 변산, 전라남도 신안군 홍도, 흑산도 비금도, 진도, 제주도)에서 채집된 달팽이 (A. despecta sieboleiana)를 대상으로 외부형태 분석과 동위효소를 검출하여 각 지역 집단간의 형태적, 유전적 유연관계와 변이 정도를 연구한 결과는 다음과 같다. 외부형태에 의한 집단간 유연관계는 안홍과 덕적도집단(average taxonomic distance, D=0.358)이 형태적으로 가장 유사한 집단으로 나타났다. Polyacrylamide gel을 사용하여 11가지의 동위효소를 검출한 결과 AKP, ACP, AO, EST, GPD, HBDH, LDH, SDH, XDH의 동위효소에서 유전적 다형현상이 나타났다. 전 집단의 평균 다형형의 빈도는 59.19%이며 이형접합자 빈도는 0.263이고 각 집단의 유전적 차이치는 춘천과 울릉도 집단이 0.066(genetic distance)으로 가장 높은 유사성을 보였다. 지리적 분포에 따른 외부형태 변이와 유전적 변이와의 관계는 유의성이 없었다.

DNA Barcoding of Six Diogenid Species (Crustacea: Decapoda: Paguroidea) in Korea

  • In this study, cytochrome c oxidase subunit I(COI) sequences of 17 individuals from six Korean diogenid species(i.e., 2 Areopaguristes japonicus, 4 A. nigroapiculus, 3 Paguristes digitalis, 4 P. ortmanni, 3 Diogenes edwardsii, and 1 Ciliopagurus kempfi) were determined and analyzed. The DNA barcoding results of this study were consistent with the morphological identification of these six species. Interspecific variations of COI sequences within six Korean diogenid species exceeded the minimum interspecific variation of diogenid hermit crabs in previous studies. Little intraspecific variation exists except for P. digitalis. This study should facilitate further molecular taxonomy of East Asian diogenids.
