Intraspecific Variation in the Light Intensity Niche Component of the Diatom Skeletonema Costatum from Korean Coastal Waters (한국연안 산 규조 Skeletonema Costatum 의 조도에 대한 생태적지위 성분의 종내 변이)

  • Final biomass yields (peak optical density) and growth rates (divisions/day) of seven clones of Skeletonema costatum from Korean coastal waters were measured to understand their intraspecific variations in the light intensity niche component under 25$^{\circ}C$ condition. Daily growth rates of 6 out of 7 S. costatum clones were maximum at 6000 lux while that of YS4, a neritic clone, was maximum at 9000 lux. The final biomass yields of 4 out of the 7 s. costatum clones were maximum at the lowest light intensity of 2000 lux. Minimum final biomass yields were found at 9000 lux in all the S. costatum clones other than an estuarine clone, HDC9. The intraspecific variations of the mean growth rate and mean final biomass yield under each of the three different light intensities in terms of the coefficient of variation were not greater than 10% in any of the 7 S. costatum clones.

  • In Vietnam, Celastrus hindsii Benth, a medicinal plant rich in secondary metabolites, has been used to alleviate distress caused by ulcers, tumors, and inflammation for generations. The occurrence of two phenotypes, Broad Leaf (BL) and Narrow Leaf (NL), has raised questions about the selection of appropriate varieties for conservation and crop improvement to enhance medicinal properties. This study examined molecular differences in C. hindsii by comparing protein profiles between the NL and BL types using 2D-PAGE and MS. Peptide sequences and proteins were identified by matching MS data against the MSPnr100 databases and verified using the MultiIdent tool on ExPASy and the Blast2GO software. Our results revealed notable variations in protein abundance between the NL and BL proteomes. Selected proteins were confidently identified from 12 protein spots, thereby highlighting the molecular variation between NL and BL proteomes. Upregulated proteins in BL were found to be associated with flavonoid and amino acid biosynthesis as well as nuclease metabolism, which probably attributed to the intraspecific variations. Several bioactive proteins identified in this study can have applications in cancer therapeutics. Therefore, the BL phenotype characterized by healthier external morphological features has higher levels of bioactive compounds and could be better suited for medicinal use.

Growth Characteristics of Rhizophagus clarus Strains and Their Effects on the Growth of Host Plants

  • Arbuscular mycorrhizal fungi (AMF) are ubiquitous in the rhizosphere and form symbiotic relationships with most terrestrial plant roots. In this study, four strains of Rhizophagus clarus were cultured and variations in their growth characteristics owing to functional diversity and resultant effects on host plant were investigated. Growth characteristics of the studied R. clarus strains varied significantly, suggesting that AMF retain high genetic variability at the intraspecies level despite asexual lineage. Furthermore, host plant growth response to the R. clarus strains showed that genetic variability in AMF could cause significant differences in the growth of the host plant, which prefers particular genetic types of fungal strains. These results suggest that the intraspecific genetic diversity of AMF could be result of similar selective pressure and may be expressed at a functional level.

Identification of DNA polymorphisms in the field bean ( Glycine soza S. and Z. ) using RAPD markers (RAPD 표지인자를 이용한 돌콩 DNA 다형현상 분석)

  • Six field bean (GI-vcine soza S and Z ) plants were examined for their genetic polymorphisms and intraspecific variations using randomly amplified polymorphic DNA(RAPD) markers. In RAPD analysis of 5 random primers (Rp-1, Rp2, Rp-3, Rp-4, Rp-5), 30 of total 155 bands obtained kom 5 primers were polymorphic and sizes of polymirphic band ranged between 0.5 and 3.0 kb. Number of bands amplyfied per primer was varied from 2 to 11 and average number was 6.0. Genetic variation of intraspecies in the samples of six region was ranged behveen 11 to 25 percent, and genetic similarity among intraspecies was ranged from 0.69 to 0.78. In pairwise genetic similarity test of six field bean plants, Mun and Hoj showed highest coefficient of genetic similarity as 0.67, whereas Sin and Hoj was lowest as 0.45. According to the genetic similarity, the level of intraspecific variation is higher than that of regional distance in GI-vcine soza.

  • The chloroplast genome of Glycyrrhiza uralensis Fisch was sequenced to investigate intraspecific variations on the chloroplast genome. Its length is 127,689 bp long (34.3% GC ratio) with atypical structure of chloroplast genome, which is congruent to those of Glycyrrhiza genus. It includes 110 genes (76 protein-coding genes, four rRNAs, and 30 tRNAs). Intronic region of ndhA presented the highest nucleotide diversity based on the six G. uralenesis chloroplast genomes. A total of 150 single nucleotide polymorphisms and 10 insertion and deletion (INDEL) regions were identified from the six G. uralensis chloroplast genomes. Phylogenetic trees show that the six chloroplast genomes of G. uralensis formed the two clades, requiring additional studies to understand it.

  • Campsis grandiflora (Thunb.) K. Schum is an ornamental species with various useful biological effects. The chloroplast genome of C. grandiflora isolated in Korea is 154,293 bp long (GC ratio: 38.1%) and has four subregions: 84,121 bp of large single-copy (36.2%) and 18,521 bp of small single-copy (30.0%) regions are separated by 24,332 bp of inverted repeat (42.9%) regions including 132 genes (87 protein-coding genes, eight rRNAs, and 37 tRNAs). One single-nucleotide polymorphism and five insertion and deletion (INDEL) regions (40-bp in total) were identified, indicating a low level of intraspecific variation in the chloroplast genome. All five INDEL regions were linked to the repetitive sequences. Seventy-two normal simple sequence repeats (SSRs) and 47 extended SSRs were identified to develop molecular markers. The phylogenetic trees of 29 representative Bignoniaceae chloroplast genomes indicate that the tribe-level phylogenic relationship is congruent with the findings of previous studies.

  • 1990년 12월부터 1997년 7월까지 제주도의 8개 지점에서 4종(Phyllidia ocellata, Fryeria menindie, Phyllidiella pustulosa, Phyllidiella cooraburrama)의 혹갯민숭이과가 채집되어 이들의 색채변이 및 분포범위에 대하여 논하였다. 또한 원색 사진과 함께 재기재를 하였다. 혹갯민숭이류는 한국해역에서 새로이 기록되는 과이다.

  • Enzyme activities of Cenococcum geophilum isolates were examined on enzyme- specific solid media. Deoxyribonuclease, phosphatase, and urease were detected in all isolates, whereas cellulase was not detected in any of the isolates. Variations in enzyme activities of amylase, caseinolysis, gelatinase, lipase, and ribonuclease were observed among isolates.

Inter-and Interspecific Variation in Smooth(D. ischaemum) and Large Crabgrass (D. sanguinalis) (잔디밭 잡초 바랭이(Digitaria sp.)의 종내 및 종간 변이성)

  • A field trial was initiated to examine the range of inter- and intraspecific variations in morphological and phenological traits with five different accessions of smooth and large crabgrass. In addition, a controlled environment study was conducted to determine the phenotypic plasticity among the accessions of both species in response to 4 daily tempera-ture differentials. In the field experiment, significant inter- and intraspecific variations of smooth and large crabgrass were observed in morphological traits such as leaf length and width. However, most phenological traits were not substantially different between the species and among the accessions of each species. The first seedling emerged at the same time, requiring 9~ 10 days, regardless of the accessions and species. In a controlled environment study, all accessions of each species responded similarly to the 4 temperature differentials in seedling emergence, indicating seedling emergence was not a plastic trait. These results suggest that predicting crabgrass seedling emergence could be independent of geographical regions in the US.

A taxonomic study of Abeliophyllum Nakai (O1eaceae) based on RAPD analysis (RAPD분석에 의한 미선나무속의 분류학적 연구)

  • Abeliophyllum distichum Nakai (O1eaceae) is a monotype of Korea and is distinguished from related genus Forsythia Vahl by the morphological characters such as fruit shape, flower color and etc. Even though several intraspecific taxa were reported according to the color of flowers and shape of fruits, there have been many controversals on the taxonomic indentity and status of rank on those taxa. In the present study, we performed the RAPD analysis to delimit the infraspecific taxa of Abeliophyllum distichum and to investigate the genetic polymorphism and relationships among 12 populations. 212 scorable RAPD markers with 70 common markers were found from the PCR reactions with 16 random oligoprimers and were analyzed by Nei's genetic distance. From 0.108 to 0.321 of genetic variations were showed among the taxa. Some regional groups instead of same taxa were clustered from the phonogram of UPGMA analysis. Also, we could not find distinct lineage among intraspecific taxa. The result from RAPD analysis supported that the infraspecific taxa of Abeliophyllum distichum might be the individual variations and treated as the same taxa. RAPD analysis was very useful to confirm the high gene pool with diverse genetic polymorphism among Abeliophyllum distichum populations.
