녹반(綠礬)의 항빈혈작용(抗貧血作用)에 관한 연구(硏究) (Studies on the Anti-anemic Action of Melanterite)

  • Melanterite(Green vitriol) is a kind of mineral crude drug which has been used for a hemostatic and hematic, and it contained iron, zinc, magnesium, copper, calcium and manganese etc., and the contents of those matals were 14.34%, 1.21%, 0.91%, 0.41%, 0.37% and 0.15%, respectively. In the acute toxicity in mice its $LD_{50}$ was over 3,000 mg/kg and 2,000 mg/kg by the oral and subcutaneous administration, respectively, but the $LD_50$ by the intraperitoneal administration was 1,810 mg/kg. On the experimental anemia induced by the free bleeding in rabbits and by the phenylhydrazine in rats the reduced RBC, hemoglobin and hematocrit were rapidly recovered.

A Preliminary Study on Hypocholesterolemic and Hypoglycemic Activities of Some Medicinal Plants

  • Total cholesterol level in mice with hypercholesterolemia was determined after intraperitoneal administration of the methanolic extract of some medicinal plants. From the data obtained, it was suggested that the methanolic extract of Elaeagnus crispa, Ixeris dentata, Prunus davidiana, Eriobotrya japonice, Aralia elata and Phragmites communis produced a significant hypocholesterotemic effect. In the case of the extract of Saussurea diamantiaca, on the other band, the total cholesterol level was markedly increased. The methanolic extract of Ixeris dentata, Prunus davidiana and Phragmites communes also decreased the level of blood glucose in alloxan-diabetic male albino mice while that of Eriobotrya japonica, Allium tuberosum, Houttuynia cordata and Eucommia japonica did not produce this effect.

  • Spirodela polyrrhiza L. Schleid. (Lemnaceae), also known as 'duckweed', is a traditional medicine in Korea. The whole plant is used to treat many diseases, including the common cold, edema, acute nephritis, and urticaria. The present study investigated antinociceptive properties of the EtOAc soluble fraction of S. polyrrhiza (ESP). The antinociceptive activities of ESP were studied using experimental models of pain, including thermal nociception methods, such as the tail immersion test and the hotplate test. Moreover, we studied chemical nociception induced by intraperitoneal acetic acid and subplantar formalin in mice. ESP exhibited dose-dependent antinociceptive activity in both thermal and chemical pain models. In a drug combination test using the opioid receptor antagonist naloxone, diminished analgesic activities of ESP were observed, indicating that the antinociceptive activity of ESP is mediated by opioid receptors.

  • The clastogenic effects of the diesel emission particle extract (DEPE), mercuric chloride and lead acetate were examined by the mouse bone marrow micronucleus test. DEPE had a potent clastogenic effect by intraperitoneal injection with dose-response between 100 and 300mg/kg b.w.. Mercuric chloride and lead acetate also gave a clastogenic effects but mercuric chloride only had a dose-response between 1 and 3mg/kg b.w.. When DEPE was administrated with mercuric chloride or lead acetate, the frequency of micronucleated cells was slight but not significant increase in comparision to a single treatment with DEPE alone.

  • $\beta$-Immunan was proteoglycan obtained from mycelium of Ganoderma lucidum IY009. In this study, the protective effects of $\beta$-Immunan, against the CDDP induced in vitro cytotoxicity and in vivo renal toxicity, was measured. Concentration dependent cytotoxicities of CDDP in normal kidney cells (Vero, TCMK-l) were reduced by $\beta$-Immunan treatment. Increased renal toxicity factors, such as elevation of blood urea nitrogen (BUN) and serum creatinine, reduction of kidney weight and malonidialdehyde (MDA), by intraperitoneal administration of CDDP in rats was improved. These results indicated that $\beta$-Immunan have a protective effects against the CDDP induced renal toxicity, however, it needed to confirm the detailed mechanism for therapeutic effects.

Effect of Polylysine on Scrapie Prion Protein Propagation in Spleen during Asymptomatic Stage of Experimental Prion Disease in Mice

  • Prion diseases are incurable neurodegenerative disorders. Our previous study demonstrated that polylysine was effective in prolonging the incubation period in a rodent model and in alleviating the scrapie prion protein (PrPSc) burden in the brain at the terminal stage of the disease. Here, we report that intraperitoneal administration of polylysine suppresses the accumulation of prions in the spleen during the early stages of the disease. This study supports the congruence of PrPSc inhibition by polylysine in both the spleen and brain.

  • Effects of water extracts of antler on blood sugar levels were studied in normal rats and KK-mice. a spontaneously diabetic strain of murine. Blood sugar levels were determined at 30, 60, and 120 minutes after intraperitoneal administration of 1 ml/kg or 2ml/kg AE into normal rats. A moderate but significant decrease in blood sugar level was noticed only in 30 minutes following 2 ml/kg AE administration. On the other hand no significant changes in blood sugar level was observed in KK-mice to which 1ml/kg/day AE had been given for 10 weeks. The results of present study strongly indicate that antler extract exerts no significant effect on blood sugar levels in experimental animals.

  • The enzyme-linked immunosorbent assay using the so-called "double-sandwich"technique was applied to determine Clostridium botulinum type F toxin. Polystyrene tubes were coated with horse anti-type F toxin serum and then toxin sample was added. The tubes were subsequently treated with rabbit anti-type F toxin IgG and sheep anti-rabbit serum IgG-horseradish peroxidase conjugate. By this technique, about 10 mouse intraperitoneal 50% lethal doses (ip LD/50/) of type F toxin could be detected. Low back-ground reading was achieved with the use of phosphate-buffered saline containing 0.05% Tween 20 and 1% bovine serum albumin as diluents of rabbit IgG and conjugate. Addition of EDTA in the diluents of toxin increased ELISA extinction value significantly. No cross-reaction was observed with botulinum type A and B toxin, but type E toxin gave sleight cross-reaction.

알코올 투여후 마우스 간 크산틴 산화효소 활성에 미치는 인삼의 영향 (Effect of Ginseng Saponins on Xanthine Oxidase Activity after Ethanol Treatment in Mouse Liver)

  • A dose, 1g/kg of ethanol produced experimental hyperuricemia in mouse. Ginseng saponins were tested for their ability to alter the hepatic xanthine oxidase activity and the blood level of uric acid in the ethanol-treated mouse. Intraperitoneal injection of ginseng saponin 4mg/kg markedly decreased the xanthine oxidase activity in the ethanol-treated mouse liver. It was also observed that ginseng saponin reduced the blood concentration of uric acid in experimentally induced hyperuricemia by alcohol treatment. In vitro, it was found that a low concentration of ginseng saponin in the reaction mixture incresed the hepatic xanthine oxidase activity, while a high concentration inhibited both enzyme preparations of normal and ethanol treated mice. In contrast with the xanthine oxidase, uricase activity was not influenced by ginseng saponin as well as in vivo. These results suggest there is a possibility that ginseng saponin may have some therapeutic effect on gout and other hyperuricemia syndrome.

백굴채가 마우스의 세포성 및 체액성 면역반응에 미치는 영향 (The Effects of Chelidonii Herba Extract on the Cell Mediate and Humoral Immune Responses in Mice)

  • The effects of Chelidonii Herba on the cell mediate and humoral immune responses were investigated in ICR mice. ICR mice received intraperitoneal injection of methanol extracts (ether fraction, buthanol fraction, water fraction) for 5 days before sensitization. The change of body weight (%), the organ weight ratio (%), the numbers of RBC and WBC, the ear swelling by dinitrofluorobenzene, the titers of hemagglutination (HA) and hemolysin (HY) to SRBC were determined. The results were as follows; 1. The change of body weight (%) showed a tendency of increasing generally. 2. The weight of spleen (%) revealed decreasing but that of thymus revealed increasing. 3. Ear swelling was maxium at 48 hours after challenge and was significantly decreased in the groups treated with Ether, Buthanol and Water. 4. The numbers of RBC and WBC were decreased generally. 5. HA and HY titers were decreased significantly.
