Protection and Detoxification Effects of Oriental Herb Extract Mixture on TCDD-Induced Oxidative Stress (TCDD로 유발된 Oxidative Stress에 대한 생약재 추출물의 방어 및 해독효과)

  • This study was carried out to investigate the protective effects and detoxification of oriental herb extracts mixture (Saururus chinensis, Taraxacum platycarpum, Ulmus macrocarpa, Glycyrrhiza glabra, Rhynchosia nulubilis) on TCDD-induced oxidative stress. Thirty five male rats were divided into 5 groups: normal control group received saline; vehicle control group received DMSO and acetone; only TCDD-treated group received multiple intraperitoneal injection of TCDD ($100{\mu}g/kg$) and saline; post-treated group of OHEM (400 mg/kg/day) received oral administration for 5 weeks after TCDD treatment; and pre-treated group of OHEM (400 mg/kg/day) received oral administration for 6 weeks from 1 week before TCDD treatment. The elevated serum activities of alanine transaminase (AST), aspartate transaminase (ALT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH) and atherorganic index due to TCDD were significantly decreased by the treatment of OHEM (p<0.05); the pre-treatment of OHEM was especially effective. Hydropic lesions and cytoplasmic vacuolizations in the liver of TCDD-treated rats were inhibited by the treatment of OHEM. Also, OHEM treatment reduced edemas in small intestine villus of TCDD-treated rats. These results suggest that OHEM from various oriental herb extracts might be a useful protective and detoxification agent against TCDD.

  • The purpose of this article was to compare 'The Discourse on the Constitutional Symptoms and Disease' of ${\ulcorner}$Dongyi Soose Bowon${\lrcorner}$ written in 1894(Old Edition(舊本)) with that of ${\ulcorner}$Dongyi Soose Bowon${\lrcorner}$ published in 1901(In Edition(印本)), and to find the idea of pathologic mechanism and classification of 'the Exterior and Interior disease'. the conclusions were as follows. 1. The classification of constitutional symptoms and disease of Soeumin and Soyangin in 'Old Edition(舊本)' was almost equal to that in 'In Edition(印本)' 2. In pathological mechanism of constitutional symptoms and disease of Soeumin and Soyangin, 'The Exterior Disease' could be explained as the disease resulted from fight between 'Yang-chi(陽氣)(Hot-chi(熱氣))'of 'Thoracic vertebrae' and 'Yin-chi(陰氣)(Cold-chi(寒氣))' of 'Bladder' and 'The Interior Disease' between 'Hot-chi(熱氣)(Stomach-chi(胄氣))' of 'Stomach' and 'Cold-chi(寒氣)' of 'Large intestine'. 3. 'The Exterior Symptoms and Disease of the Exterior and the Interior Disease(表裏之表病)' could be explained as the disease occurring at the Branch portion(large portion)(標) by overcoming of Pathogenic factors but Vital energy still sufficient, and 'The Interior Symptoms and Disease of the Exterior and the Interior Disease(表裏之裏病)' occurring at Root portion(small portion)(裏) by invasion of Pathogenic factors and Vital energy almost exhausted. 4. In the classification of constitutional symptoms and disease of Taeumin, 'The Exterior Symptoms and Disease of the Exterior and the Interior Disease(表裏之表病)' in 'Old Edition(舊本)' were rearranged to 'The Exterior Disease' in 'In Edition(印本)', 'The Interior Symptoms and Disease of the Exterior and the Interior Disease(表裏之裏病)' to 'The Interior Disease'. 5. It was assumed that 'The Exterior and the Interior Disease' of Taeumin could be explained in relation between the exterior and e interior, based on the Healthy energy(保命之主) and e concept of the Branch and the Root portion

Alleviating Effects of Mulberry Fruit Extract on Postprandial Hyperglycemia in Streptozotocin-induced Diabetic Mice (STZ으로 유도된 당뇨 마우스에서 오디열매추출물의 식후 고혈당 완화 효과)

  • Postprandial hyperglycemia is an early defect of type 2 diabetes and one of primary anti-diabetic targets. The alpha-glucosidase inhibitors regulate postprandial hyperglycemia by impeding the rate of carbohydrate (such as starch) digestion in the small intestine. This study was designed to investigate the inhibitory actions of mulberry fruit extract (MFE) on α-glucosidase and α-amylase activities, and its alleviating effect on postprandial hyperglycemia activities in vitro and in vivo. Male four-week old ICR mice and streptozotocin (STZ)-induced diabetic mice were treated with mulberry fruit extract. MFE showed strong inhibitory effects against α-glucosidase and α-amylase activities, with half-maximal inhibitory concentration (IC50) values of 0.16 and 0.14 mg/ml, respectively, and was more effective than acarbose, which was used as a positive control. The increase in postprandial blood glucose levels was more significantly attenuated in the MFE-administered group mice than in the control group mice of both STZ-induced diabetic and normal mice. Moreover, the area under the glucose response curve significantly decreased following MFE administration in diabetic mice. These results indicate that MFE may be a potent inhibitor of α-glucosidase and α-amylase, and helpful in suppressing postprandial hyperglycemia in diabetic mice. The mulberry fruit extracts may be considered as a potential candidate for the management of diabetes.

NQO1-Knockout Mice Are Highly Sensitive to Clostridium Difficile Toxin A-Induced Enteritis

  • Clostridium difficile toxin A causes acute gut inflammation in animals and humans. It is known to downregulate the tight junctions between colonic epithelial cells, allowing luminal contents to access body tissues and trigger acute immune responses. However, it is not yet known whether this loss of the barrier function is a critical factor in the progression of toxin A-induced pseudomembranous colitis. We previously showed that NADH:quinone oxidoreductase 1 (NQO1) KO (knockout) mice spontaneously display weak gut inflammation and a marked loss of colonic epithelial tight junctions. Moreover, NQO1 KO mice exhibited highly increased inflammatory responses compared with NQO1 WT (wild-type) control mice when subjected to DSS-induced experimental colitis. Here, we tested whether toxin A could also trigger more severe inflammatory responses in NQO1 KO mice compared with NQO1 WT mice. Indeed, our results show that C. difficile toxin A-mediated enteritis is significantly enhanced in NQO1 KO mice compared with NQO1 WT mice. The levels of fluid secretion, villus disruption, and epithelial cell apoptosis were also higher in toxin A-treated NQO1 KO mice compared with WT mice. The previous and present results collectively show that NQO1 is involved in the formation of tight junctions in the small intestine, and that defects in NQO1 enhance C. difficile toxin A-induced acute inflammatory responses, presumably via the loss of epithelial cell tight junctions.

Effects of dietary supplementation of a lipid-coated zinc oxide product on the fecal consistency, growth, and morphology of the intestinal mucosa of weanling pigs

  • Background: Dietary supplementation of zinc oxide (ZnO) to 2000 to 4000 mg/kg is known to be effective for the prevention and treatment of post-weaning diarrhea in the pig. Such a 'pharmacological' supplementation, however, can potentially result in environmental pollution of the heavy metal, because dietary ZnO is mostly excreted unabsorbed. Two experiments (Exp.) were performed in the present study to determine the effects of a lipid-coated ZnO supplement Shield Zn (SZ) compared with those of ZnO. Methods: In Exp. 1, a total of 240 21-day-old weanling pigs were fed a diet supplemented with 100 mg Zn/kg as ZnO (ZnO-100), ZnO-2500, SZ-100, or SZ-200 in 24 pens for 14 days on a farm with its post-weaning pigs exhibiting a low incidence of diarrhea. Exp. 2 was performed using 192 24-day-old piglets as in Exp. 1 on a different farm, which exhibited a high incidence of diarrhea. Results: In Exp. 1, fecal consistency (diarrhea) score (FCS) was less for the ZnO-2500 and SZ-200 groups than for the SZ-100 group (P < 0.05), with no difference between the SZ-100 and ZnO-100 groups. Both average daily gain (ADG) and gain:feed ratio were less for the SZ-200 group than for the ZnO-2500 group, with no difference between the ZnO-100 group and SZ-100 or SZ-200 group. The villus height (VH), crypt depth (CD), and VH:CD ratio of the intestinal mucosa were not influenced by the treatment. In Exp. 2, FCS was lowest for the ZnO-2500 group, with no difference among the other groups. However, neither the ADG nor gain:feed ratio was influenced by the treatment. Conclusion: Results suggest that physiological SZ supplementation has less beneficial effects than pharmacological ZnO for the alleviation of diarrhea irrespective of its severity and for promoting growth without influencing their integrity of the intestinal mucosal structures with little advantage over physiological ZnO in weanling pigs with a small pen size.

Gastric anisakiasis cases in Cheju-do, Korea (고래회충유충증 107례 보고 및 어류감염 실태 조사)

  • Human anisakiasis may occur after ingestion of raw marine flesh infected with nematode larvae of Anlsi,hidae. Anisakiasis caused by the migration of the larva into the wall of stomach, small intestine and other portion has been reported in Korea. This prospective study was made of all cases referred to parasitological laboratory in Cheju-do between. Tune 1989 and June 1992. Gastric anisakiasis was confirmed if larvae invading the gastric wall were observed by gastrofiberscopy One hundred and seven cases were diagnosed, most of which were in 30-49 years old. Most of the patients complained acute epigastric pain with history of eating raw marine fish. This symptom usually occurred about 12 hours to 1 day after ingestion of infected marine fish. Edema, erosion or ulcer of the mucosa and hemorrhage from the gastric wall were observed in the involved areas. Ninety larvae removed from the stomach were identified; the larva of Anisqkis siwlex was the most prevalent species, and the Iarva of Pseudntewanoua decipien was also detected. The important species of marine fish from which the patients became infected was demonstrated as yellow corvina, sea eel, ling, cuttle fish, yellowtail and others. Five species of marine fish as a possible source of infection were examined, and Anisakis simplex larvae and Pseunotewqnoua decipirens larvae were collected from the mackerel and rock cod. This study demonstrates that anisakiasis is recognized as a public health problem in Korea.

Emergency preoperative angioembolization without computed tomography to treat unstable pelvic fractures with bowel perforation (장 천공을 동반한 혈역학적으로 불안정한 골반골 골절에서 전산화 단층촬영 전 시행한 혈관색전술의 지혈효과: 증례보고)

  • Hemodynamically unstable pelvic fractures show a remarkably high mortality rate of 40% to 60%. However, their standard of care remains controversial. We report here a case of a 78-year-old woman who was admitted to the Emergency Department with pelvic pain following a fall. Based on pelvic radiography, she was diagnosed with an unstable pelvic fracture. Her blood pressure was 60/40 mmHg, and owing to her unstable vital signs, emergency angiography was performed without computed tomography (CT). Both internal iliac arteries were embolized without sub-branch selection for prompt control of pelvic bleeding. Following embolization, her vital signs were stabilized. Subsequent CT revealed free intra-abdominal air, suggesting bowel perforation had occurred and necessitating emergency laparotomy. An approximately 1 cm-sized free perforation of the small intestine was identified intraoperatively, and primary closure was performed. A retroperitoneal hematoma identified intraoperatively was not explored further because it was a non-expanding and non-pulsatile mass. The patient was admitted to the Intensive Care Unit and transferred to the general ward on postoperative day 3. In this case, the hemodynamically stable pelvic fracture with bowel perforation was successfully and safely treated by prompt angioembolization without conducting CT.

Evaluating the effects of finishing diet and feeding location on sheep performance, carcass characteristics, and internal parasites

  • A 3 yr experiment was conducted to evaluate the influence of diet and feeding location on animal performance, carcass characteristics, whole blood counts, and internal parasite burden of lambs assigned to 1 of 4 treatments: 1) confinement fed 71% alfalfa, 18% barley pellet, 5% molasses, 0.013% Bovatec, 6.1% vitamin/mineral package diet (CALF), 2) confinement fed 60% barley, 26% alfalfa pellet, 4% molasses, 2.5% soybean-hi pro, 0.016% Bovatec, 7.4% vitamin/mineral package diet (CBAR), 3) field fed 71% alfalfa, 18% barley pellet, 5% molasses, 0.013% Bovatec, 6.1% vitamin/mineral package diet (FALF), and 4) field fed 60% barley, 26% alfalfa pellet, 4% molasses, 2.5% soybean-hi pro, 0.016% Bovatec, 7.4% vitamin/mineral package diet (FBAR). A year × location interaction was detected for ending body weight (BW), average daily gain (ADG), and dry matter intake (DMI); therefore results are presented by year. In all years, cost of gain and DMI were greater for CALF and FALF than for CBAR and FBAR feed treatments (p ≤ 0.03). In yr 2 and 3 field treatments had greater ending BW and ADG than confinement treatments. For all years, diet did not affect ending BW or ADG. In yr 1 dressing percent and rib eye area were greater for field finished lambs than confinement finished (p ≤ 0.02) and Warner-Bratzler shear force was greater for CALF and FALF (p = 0.03). In yr 2 lambs in FALF and FBAR treatments had greater leg scores and conformation than CALF and CBAR (p = 0.09). In yr 1, FALF had a greater small intestine total worm count than all other treatments. In yr 1, ending Trichostrongyle type egg counts were greater for FALF (p = 0.05). In yr 2, ending Nematodirus spp. egg counts were greater for FALF and lowest for CBAR (p < 0.01). Abomasum Teladorsagia circumcinta worm burden was greater in CALF than all other treatments (p = 0.07) in yr 2. While field finishing lambs with a grain- or forage-based diet we conclude that it is possible to produce a quality lamb product without adverse effects to animal performance, carcass quality or increasing parasite burdens.

Effect of Dietary Supplementation of Enzyme and Microorganism on Growth Performance, Carcass Quality, Intestinal Microflora and Feces Odor in Broiler Chickens (효소제와 미생물제제의 첨가 급여가 육계의 생산성, 도체성적, 장내 미생물 및 계분 악취에 미치는 영향)

  • This experiment was conducted to investigate the effect of the addition of enzymes and microorganisms to broiler feed on productivity, carcass characteristics, intestinal microflora, and feces odor. A total of one-hundred eighty 180 chicks (Ross 308) were randomly assigned to 5 treatments with 3 replications each having 12 birds per pen. The experimental group was divided into 0.1% EZ group (0.1% metallo-protease added to the feed), 0.2% EZ group (0.2% metallo-protease added to the feed), M group (2.0% Bacillus veleznesis CE 100 added to the feed), and MW group (2.0% Bacillus veleznesis CE 100 added to the feed and drinking water). In the results, final body weight, body weight gain, the feed conversion ratio, protein efficiency, and energy efficiency were not significantly different among all treatments in across all periods. Carcass weight, proventriculus, gizzard, heart, small intestine, cecum, and rectum weight were not significantly different among all of the treatments. However the liver weight was significantly higher in the 0.1% EZ group than in the control, M and MW groups (P< 0.05). E. coli was significantly lower in MW than in the control and M (P<0.05), and it was significantly higher in the M than 0.2% EZ and MW (P<0.05). H2S emissions in feces was not significantly different among all treatments, but NH3 emissions was were significantly higher in 0.1% EZ than in MW (P<0.05). In conclusion, the addition of 0.1% of metallo-protease was effective in the development of the liver of broilers.

Genetic sequence and phylogenetic analysis of spike genes of porcine epidemic diarrhea virus (PEDV) in Jeonbuk province (전북지역 돼지유행성설사 바이러스 Spike 유전자 염기서열 및 계통분석)

  • Although many swine farms continuously vaccinated to sow to prevent Porcine epidemic diarrhea(PED), PED has occurred annually in swine herds in Jeonbuk province, Korea. In the present study, the small intestine and feces samples from 17 farms where severe watery diarrhea and death of newborn piglets occurred in 2019 were collected, amplified by RT-PCR and determined the complete nucleotide sequences of the spike (S) glycoprotein genes of nine Jeonbuk PEDV isolates. The spike (S) glycoprotein is an important determinant for molecular characterization and genetic relationship of PEDV. These nine complete S gene isolates were compared with other PEDV reference strains to identify the molecular diversity, phylogenetic relationships and antigenicity analysis. 9 field strains share 98.5~100% homologies with each other at the nucleotide sequence level and 97.3~100% homologies with each other at the amino acid level. The nine Jeonbuk PEDV isolates were classified into G2b group including a genetic specific signal, S-indels (insertion and deletion of S gene). In addition, comparisons the neutralizing epitopes of S gene between 9 field strains and domestic vaccine strains of Korea mutated 12-15 amino acids with SM-98-1 (G1a group) and mutated 0-3 amino acids with QIAP1401 (G2b group). Therefore, the development of G2b-based live vaccines will have to be expedited to ensure effective prevention of endemic PED in Korea. In addition, we will need to be prepared with periodic updates of preventive vaccines based on the PEDV variants for the re-emergence of a virulent strain.
