A Study on Homeostasis in Albino Rats by Feeding on Imbalanced Protein Diet (불균형식이(不均衡食餌)에 의(依)한 백서체내(白鼠體內) Homeostasis에 대(對)한 연구(硏究))

  • This Study was carried out to observe the effect of nutritional condition on the change of protein metabolism in the animal body by feeding on imbalanced protein diet. A total 242 growing male albino rats, weighing $115{\sim}120$ gm, were used for the experimental animals. The rats were fed on the standard diet(st), protein flee diet(pf) and imbalanced protein diet(ib) for twelve weeks respectively. Hemoglobin, packed cell volume in blood, and total nitrogen, amino acid nitrogen, urea-nitrogen, creatinine, transaminases(GPT, GOT) in liver and serum, and total nitrogen in small intestine, and total nitrogen, urea-nitrogen In small intestine, and total nitrogen, urea-nitrogen, creatinine, urea-nitrogen/creatinine ratio in urine were measured. The results obtained are as follows; 1. The gained body weight were lower in pf group and ib group than those of st group. The gained body weight fed for 12 weeks, were 80% lower in pf group than those of st group, and the body weight of pf group for $50{\sim}75$ days feeding were $40{\sim}60%$ decreased, compared with the stating weight, and then all of them died. 2. The change of the brain, liver, kidney, spleen and small intestine by feeding on imbalanced diet for 12 weeks were no remarkable difference with the starting weight, but those of protein free diet group were half or more decrease and those were significantly lower in spleen and small intestine especially than the other organ 3. The contents of hemoglobin in pf group for 8 weeks feeding, and the packed cell volume in pf group for 8 weeks feeding and in ib group for 12 weeks feeding were decreased. but those of the other feeding group were almost same value. 4. The total nitrogen in the liver, small intestine and serum of each diet group were no remarkable difference respectively. The contents of amino acid nitrogen in pf group for 2 and 6 weeks feeding were increased. 5. On transaminases: a) The cycle of increase and decrease of GPT activities were come periodically and the interval of cycle were fast in the early stage of feeding and slow there-after. b) The GPT activities were decreased gradually in pf group after feeding and those were increased in ib group for 6 weeks feeding but decreased there-after. The frequency of cycle were more GPT than GOT and specially those of GPT in early stage of feeding were two or three times while GOT was one. c) The interval of increase and decrease in GOT and amino acid nitrogen cycle were similar tendency. 6. The contents of total nitrogen, creatinine and urea-nitrogen of pf group in urine were decreased very sharply from sharting feeding to one week but increased dully from six weeks to eight weeks feeding. The contents of urea-nitrogen of ib group were increased dully by feeding on ten weeks but decreased by feeding on twelve weeks. From the above results, it is concluded that the trend of the metabolic change is maintained equally by homeostatic mechanism using the endogenous protein source during a certain period by imbalanced protein diet feeding. The homeostatic mechanism is come peridically, very fast in early stage of feeing and than slow there-after.

An Experimental Study on the Mucosal Epithelization of the Digestive Tract in the Dog (개의 소화관점막 재생에 관한 외과적 연구)

  • This study was performed for the purpose of investigating the possibility of epithelization on the defect of digestive tract after covering it with serosal wall of the cecum in the dog. The results obtained were as fallows: 1. Each defect of the digestive tract revealed almost complete formation of the mucous membrane and the muscle. Especially it was most fast in the jejunum. 2. Fistula formation was not found in all the experimental cases. The defects of small intestine and large intestine near the cecum were healed more fastly than the other portion. 3. The stomach and the duodenum appeared intestinal adhetion more frequently than the jejunum and the colon. Therefore, it is considered that covering with serosal wall of the cecum in the lower intestine is far better than in the upper intestine.

  • In order to investigate the efficacy of Siljangsan by using the experimental animals, the action of gastric juice secretion, antiulcer. transport rates in the intestine and anti-cathartic action were studied. The following results were obtained. 1. Preventive effects on ulcer induced by pylorus-ligated and Ethanol-HCI were recognized 2. Suppression effects on gastric juice secretinon, free of acidity and total acidity were recognized, but pepsin output was not recognized. 3. For the transport ability of intestine of mice, the ability of small intestine was activated and large intestine was suppressed in the group administrated 4. Anti-cathatic action of mice induced by castor oil was recognized. According to above results, effect based on oriental medical reference was consistent with the actual experimental effect.

  • The relative frequency and distribution of bovine chromogranin(BCG)-, serotonin-, somatostatin(SOM)- and S-100 protein-immunoreactive cells in the intestinal tract of the duck (Anas platyrhynchos platyrhyncos, Linne) with ages were studied using immunohistochemical methods. BCG-immunoreactive cells were detected in duodenum on 23 days of incubation. Thereafter these cells were occurred throughout the intestine. SOM-immunoreactive cells were detected in the small intestine from hatching to 9 weeks. Thereafter they were also observed in colon. Serotonin-immunoreactive cells were detected throughout the intestinal tracts after hatching. These immunoreactive cells were increased with ages except that BCG-immunoreactive cells were decreased with ages from 6 weeks after hatching. BCG- and serotonin-immunoreactive cells were most frequently detected in colon regions and SOM-immunoreactive cells in duodenum regions, but no S-100 protein-immunoreactive cells were detected in this study.

  • The major objective of this study carried out was to compare the binding of Kintoki bean lectin (KBL) and Taro tuber lectin (TTL) to the mouse intestinal segments using $^3$H-labeled lectins and to assess the effect of such binding on the ability of the small intestine. Binding of $^3$H-KBL or $^3$H-TTL was studied under various conditions of time course, temperature, concentration, pH and additives of sugars, EDTA or unlabeled native lectin. The interaction of the lectins to intestinal tissue was stronger in KBL than in TTL, which was supposed to be the major reason for the stronger antinuritional enen of KBL. The optimal binding conditions were at 37$^{\circ}C$ for 60mins and at pH 7. The binding of both lectins were inhibited by fetuin and EDTA.

Characterization of the microbial communities along the gastrointestinal tract of sheep by 454 pyrosequencing analysis

  • Objective: The gastrointestinal tract of sheep contain complex microbial communities that influence numerous aspects of the sheep's health and development. The objective of this study was to analyze the composition and diversity of the microbiota in the gastrointestinal tract sections (rumen, reticulum, omasum, abomasum, duodenum, jejunum, ileum, cecum, colon, and rectum) of sheep. Methods: This analysis was performed by 454 pyrosequencing using the V3-V6 region of the 16S rRNA genes. Samples were collected from five healthy, small tailed Han sheep aged 10 months, obtained at market. The bacterial composition of sheep gastrointestinal microbiota was investigated at the phylum, class, order, family, genus, and species levels. Results: The dominant bacterial phyla in the entire gastrointestinal sections were Firmicutes, Bacteroidetes, and Proteobacteria. In the stomach, the three most dominant genera in the sheep were Prevotella, unclassified Lachnospiraceae, and Butyrivibrio. In the small intestine, the three most dominant genera in the sheep were Escherichia, unclassified Lachnospiraceae, and Ruminococcus. In the large intestine, the three most dominant genera in the sheep were Ruminococcus, unclassified Ruminococcaceae, and Prevotella. R. flavefaciens, B. fibrisolvens, and S. ruminantium were three most dominant species in the sheep gastrointestinal tract. Principal Coordinates Analysis showed that the microbial communities from each gastrointestinal section could be separated into three groups according to similarity of community composition: stomach (rumen, reticulum, omasum, and abomasum), small intestine (duodenum, jejunum, and ileum), and large intestine (cecum, colon, and rectum). Conclusion: This is the first study to characterize the entire gastrointestinal microbiota in sheep by use of 16S rRNA gene amplicon pyrosequencing, expanding our knowledge of the gastrointestinal bacterial community of sheep.

Clinical and Histopathogical Studies of Experimentally Induced Listeriosis in Korean Native Goats (한국재래산양의 Listeria병에 관한 임상 및 병리조직학적 연구)

  • These studies were taken experimentally to clarify the clinical and histopathological findings of listeriosis in Korean native goat(KNC). Four KNGs of 4 to 5 months of age were inoculated orally or in-travenously (IV) with Listeria monocytogenes isolated from a field case of KNG. 1. On the clinical findings, depression, anorexia and fever were observed in all inoculated goats, and nasal discharge, keratoconjunctivitis and diarrhea in 3 of 4 goats. Highest rectal temperature after in-oculation was 2.5$^{\circ}C$ higher in IV inoculated goats and 1.9$^{\circ}C$ higher in orally inoculated than normal rectal temperature observed before inoculation. Durations of clinical course after inoculation in IV and orally inoculated goats were 5 days and 8 days, respectively. 2. On the gross lesions, swelling of the lymph nodes, hemorrhage and .inflammation of the small intestine and rigor mortis were observed in 4 of 4 goats, and keratoconjunctivitis, hemorrhage and inflammation of the large intestine, swelling of the spleen, pneumonia and hydropericardium in 3 of 4 goats. Congestion of the visceral organs and ecchymosis of the sin in a fetus were observed. Keratoconjunctivitis, hemorrhage of the abomasum, swelling of the lymph node, hemorrhage and inflammation of the small intestine, swelling of the spleen, necrosis of the liver and pneumonia were observed as severe lesions. These lesions were more severe in IV inoculated goats than those in orally inoculated goats. 3. On histopathological findings, focal necrosis found throughout the livers occurred mainly on peripheral areas of hepatic lobules. These necrotic foci consisted of neutrophils, lymphocytes, macrophages, short rod bacteria and necretic hepatic cells. Suppurative pneumonia of the lung, hyperemia, congestion, epithelial necrosis and sloughing of the small and large intestine, swelling in periventricular regions of the central nervous system, swelling of the kidney, spleen and lymph node were observed as listerial lesions.

  • In this study, a linear accelerator (LINAC) through 3 Gy of radiation per body irradiated mice of the small intestine and the liver to produce in order to protect the cells after radiation exposure that caspase (caspase 3 &caspase 9) and NO (nitric oxide), and looked like to know cytokine of IL-6 and TNF-${\alpha}$, the result is as follows. First, caspase 3 & caspase 9 showed a noticeable increase in the radiation group than in the control group both small intestine and liver tissues (P <0.001). Second, NO are both intestine and liver tissue showed a marked increase in the radiation group than in the control group (P <0.001). Third, one of Cytokine IL-6 and TNF-${\alpha}$ showed a significant increase in the irradiated group than the control group both small intestine and liver tissues (P <0.001).

  • From the study of acupuncture and external diseases on mammary diseases, the following conclusions are obtained. 1. Point Shanzhong, Shaoze and Rugen are often used for galactostasis in acupuncture. 2. Often used vessels for galactostasis in acupuncture are Stomach Meridian, Conception Vessel, Small Intestine Meridian, Bladder Meridian, Liver Meridian and Large Intestine Meridian in order. 3. The direct stimulations are used in external treatment for galactostasis, often accompanied by internal medical treatment, to promote the circulation of Qi and Blood. 4. Point Zuimup, Guangming of Gall bladder Meridian and Zusanli of Stomach Meridian is used for lactation pain in acupuncture. 5. Point Jianjing, Zusanli and Shanzhong are most often used for acuremastitis and breast cancer in acupuncture. 6. Often used vessels for acuremastitis and breast cancer in acupuncture are Stomach Meridian, Gall bladder Meridian, Conception Vessel, Small Intestine Meridian, Bladder Meridian, Pericardium Meridian in order. 7. The external treatment used for acuremastitis and breast cancer can be classed into its pathological period. When lactation is interrupted, Kumhwang Powder, Okro Powder, Robongbang, Allii Radix, Pinelliae Rhizoma, garlic moxibustion and Arusaenatus Rgizoma powder are used. When pus is piled up, Chunghwa Extracts, Dioscoreae Rhizoma, Leonuri Herba, Typa Latifolia, Taraxacum platycarpum, Phaseolus angularis, Manchong, Crvi Cornus latex, and Aucklandiae Radix are used. When pus flows down, Salviae Miltiorrhizae Radix Extract, nine-one pellet, Yongboo extract, eight-dampness extract, Tissue-regeneration Powder, Toad-net treatment, Kitten-hair, Hongseung pellet are used. 8. Stomach Meridian, Conception Vessel, Small Intestine Meridian, Gall Bladder Meridian, Bladder Meridian are most often used vessels for mammary diseases. From the conclusions above, It appears that the medical effect could be maximized by further studying and developing of the acupuncture and external treatment for mammary diseases according to the its nature, while accompanying internal medicine appropriately at the same time.

Differential Cytokine and Immunoglobulin Expressions in the Small Intestine of Echinostoma hortense Infected BALB/c Mice

  • Infections involving Echinostoma hortense (E. hortense) are considered to more severe than infections caused by other heterophyids. Although parasite expulsion by host immune responses attenuates the symptoms of infection, the detailed mechanisms of the host immune response need to be determined, especially in local immune responses involving cytokine and immunoglobulin expressions. We infected BALB/c mice with E. hortense and examined recovery rates together with expressions of multiple cytokines and immunoglobulins in the villi and crypts of the small intestine using immunohistochemistry. We observed a close correlation between worm expulsion rates and cytokine/immunoglobulin expressions in E. hortense infected mice. This study contributes to an understanding of the relationship between the immune response and parasite expulsion in hosts.
