납(Lead)이 취외분비 기능에 미치는 영향 (Effect of Lead Acetate on Pancreatico-biliary Secretion)

  • No evidence has accumulated that lead compound is an essential component for biological function in animals. Lead is absorbed primarily through the epithelial mucosal cells in duodenum and the absorption can be enhanced by the substances which bind lead and increase its solubility. Iron, zinc and calcium ions, however, decrease the absorption of lead without affecting its solubility, probably by competing for shared absorptive receptors in the intestinal mucosa. Therefore, the absorption of lead is increased in iron deficient animals. Lead shows a strong affinity for ligands such as phosphate, cysteinyl and histidyl side chains of proteins, pterins and porphyrins. Hence lead can act on various active sites of enzymes, inhibiting the enzymes which has functional sulfhydryl groups. lead inhibits the activity of ${\delta}$-aminolevulinic acid dehydratase for the biosynthesis of hemoproteins and cytochrome, which catalyzed the synthesis of monopyrrole prophobilinogen from ${\delta}$-aminolevulinic acid. Accordingly lead decrease hepatic cytochrome p-450 content, resulting an inhibition of the activity of demethylase and hydroxylase in liver. Little informations are available on the effect of lead on digestive system although the catastrophic effects of lead intoxication are well documented. The present study was, therefore, attempted to investigate the effect of lead on pancreaticobiliary secretion in rats. Albino rats of both sexes weighing $170{\sim}230g$ were used for this study. The animals were divided into one control and three treated groups, i.e., control (physiologic saline 1.5ml/kg i.p.), lead acetate $(l0{\mu}mole/kg/day\;i.p.)$, $Pb(Ac)_2$ and EDTA$(each\;10{\mu}mole/kg/day\;i.p.)$, $Pb(Ac)_2$ and $FeSO_4(each\;l0{\mu}mole/kg/day\;hp)$. The pancreatico-biliary juice was collected under urethane anesthesia, and activities of amylase and lipase were determined by employing Sumner's and Cherry and Crandall's methods. The summarized results are follows. 1) In the experiment for acute toxicity of lead acetate, 20% of mortality was observed in rat treated with lead acetate as well as inhibition of the activity of amylase in the juice at the 3 rd day of the treatment. 2) No increases in body weight were observed in rats treated with lead acetate, while in control group the significant increases were observed. However, the body weights of animals were increased in the group lead acetate plus EDTA or $FeSO_4$. 3) Lead acetate decreased significantly the volume of pancreatico-biliary juice whereas additional treatment of EDTA and $FeSO_4$ prevented it. 4) Total activity of amylase was markedly reduced due to lead acetate treatment, but no change was showed following additional treatment with EDTA and $FeSO_4$. 5) No changes in the cholate and lipase output were observed in rats treated with lead acetate as compared with that of control rats. 6) Increase in bilirubin output in rats treated with lead acetate was shown on the 2nd and 3rd weeks treatment. 7) In the case of in vitro experiment, lead acetate also markedly inhibited release of amylase from pancreatic fragment. 8) Histologic finding indicated that acini vacuolation was induced in the pancreatic tissue of rat treated with lead acete. From the above results, it might be concluded that lead acetate decreases the volume of pancreatico-biliary secretion and inhibits the amylase activity, by acting directly on pancreatic cells.

  • 이 실험은 추백리의 병원성을 연구하고자 2006년 2월부터 12월까지 Salmonella enterica serovar. enterica subspecies (S.) Pullorum 야외주를 분리한 후, 이를 건강한 닭에 실험적으로 감염시킨 다음 임상증상, 여러 기관의 육안 및 조직병리학적 검색과 아울러 공격주의 재분리와 동정을 시도하였다. S. Pullorum에 혈청학적으로 음성인 12주령의 100수의 암탉을 $A{\sim}E$까지 20수씩 5그룹으로 구분하였다. $A{\sim}DS$. Pullorum을 $10^6\;CFU$, $10^7\;CFU$, $2{\times}10^7\;CFU$, $10^8\;CFU$로 각각 경구 감염시켰고, E는 비감염 대조군으로 삼았다. 실험 방법으로는 부검, 조직병리학적 검사, Salmonella에 대한 세균 배양, 염색, 생화학적 특성을 조사하고 그 결과를 기술하였다.

위 편평선종 및 조기위암에서 내시경적 점막절제술 후 절제부위에서 발생한 융기형 병변에 관한 임상적 고찰 (A Clinical Study of Protruding Lesions That Arise at the Scar of an Endoscopic Mucosal Resection for an Early Gastric Carcinoma and a Gastric Flat Adenoma)

  • Purpose: Several studies of an endoscopic mucosal resection(EMR) have been reported, but reports about benign protruding lesions that arise at the scar of EMR for early gastric cancer (EGC) or a gastric adenoma are rare. The purpose of this study was to elucidate endoscopic and histological characteristics of benign protruding lesions which arise at the scar of an EMR for EGC and a gastric flat adenoma. Materials and Methods: In 101 lesions (73 gastric flat adenomas and 28 EGCs) from 96 patients, 16 lesions developed new protruding lesions that arose at the scar of the EMR. We retrospectively analyzed the endoscopic findings of initial and protruding lesions, and several other clinical factors (H. pylori infection, eradication therapy, and proton pump inhibitor (PPI) or H2-blocker use). Results: 1. The mean duration until detection of the protruding lesion was 8.9 months ($1.5\∼27$). Protruding lesions arose at the scar of the EMR in 1 of 28 EGCs ($3.6\%$) and from 15 of 73 gastric flat adenomas ($20.5\%$). All of the patients were men. 2. With respect to the endoscopic findings, the shapes of the protruding lesions were as follows: 10 Yamada (Y) I, 4 Y-II, 1 Y-III, and 1 flat lesion. Histological examination of the protruding lesions revealed regenerating hyperplasia in 5 lesions, intestinal metaplasia in 5, and both in 6. 3. The incidence of these lesions was higher in cases of tubular adenomas with focal high-grade dysplasia than in cases of tubular adenomas without dysplasia (p<0.05). 4. The incidence of H. pylori infection was higher in patients ($81.7\%$) who developed a protruding lesion than in those ($51.8\%$) who did not develop (p=0.029); also, the incidence of use of PPI was higher in those patients (p=0.045). However, eradication therapy for H. pylori and duration of use of PPI or H2-blocker showed no difference between groups. Conclusions: It may be possible that the potential hyperplasia that may reside in normal mucosa surrounding EGC or a gastric adenoma might awaken during the healing process of the EMR ulcer and develop to benign protruding lesions. And, H. pylori and PPI might also be related to the development of the protruding lesions.

아연 보충급여에 따른 한국재래계의 성장, 사료이용성, 장기무게, 혈액생화학적 성상, 장기무게 및 소화효소 활성도에 미치는 영향 (Effects of Dietary Zinc Supplements on Growth, Feed Efficiency, Organ Weight, Blood Biochemical Profiles, and Activity of Digestive Enzymes in Growing Korean Native Chicks)

  • 본 연구는 Zn 보충급여가 한국재래계의 생산성 및 소화기능에 미치는 영향을 조사하기 위하여 기초사료(100 ppm)에 ZnO 또는 Zn-methionine 형태로 각각 50 ppm을 첨가하여 체중, 사료이용성, 장기무게, 혈액생화학적 성상 및 소화효소 활성도를 조사하였다. 사양성적을 보면 대조군(CON)에 비해 산화아연(ZNO) 및 유기태(ZMT)군에서 체중은 각각 1.2% 및 2.4% 증가되는 경향을 보였지만 유의적 차이는 없었다. 사료섭취량과 사료요구율 역시 모든 처리군에서 차이가 없는 것으로 나타났다. 간, 비장 및 소장점막 무게는 아연의 급여 및 화학적 형태에 따른 차이가 없었으나, 췌장무게는 ZNO군에서 대조군과 ZMT군에 비해 유의하게 감소되었다(P<0.05). 혈액 생화학 성분인 glucose, total protein, triglyceride, total cholesterol, aspartate aminotransferase(AST)와 alanine aminotransferase(ALT) 등은 ZnO 또는 Zn-methionine 급여에 따른 차이가 없었다. 췌장 효소활성도를 조사한 결과 ${\alpha}$-amylase와 carboxypeptidase A 활성도는 아연의 급여에 따른 차이가 없었으나, trypsin은 ZnO와 Zn-methionine 급여에 따라 활성도가 현저히 증가되었다(P<0.05). 소장의 흡수상피세포에 존재하는 이당류분해효소(maltase 및 sucrase)는 Zn 급여원에 따라 차이를 보이지 않았다. 펩타이드 분해효소인 leucine aminopeptidase의 활성도는 ZnO 급여 또는 Zn-methionine 급여에 따라 증가하는 경향을 보였다(P=0.08). 이상의 결과로 보아 기초사료에 Zn의 보충급여(50 ppm)는 닭에서 단백질 소화작용에 관여하는 효소의 활성도 증진효과를 보이며, 단백질의 생체 이용성 향상에 긍정적인 영향을 미칠 수 있는 것으로 생각된다.
