Morphological evaluation of the small intestine mucosa and apoptosis activity (caspase-3) is necessary to assess the severity of damage to the small intestine. At the same time, proliferative index based on Ki-67 can be used to assess the regenerative potential of the small intestine. Fragments of small intestine of Wistar rats (n=60) of three groups: I) control (n=20); II) experimental group (n=20; local single electron irradiation at a dose of 2 Gy), III) experimental group (n=20; local single electron irradiation at a dose of 8 Gy) were studied by light microscopy using hematoxylin and eosin staining and immunohistochemical reactions with antibodies to Ki-67 and caspase-3. In all samples of the experimental groups, a decrease in all morphometric indices was observed on day 1 with a tendency to recover on day 3. Small intestinal electron irradiation led to disturbances in the histoarchitecture of varying severity, and an increase in cell apoptosis was observed (increased expression of caspase-3 and decrease in Ki-67). In addition, modulation of the PI3K/AKT and MAPK/ERK signaling pathways was detected. The most pronounced destructive changes were observed in the group of 8 Gy single electron irradiation. Local irradiation of the small intestine with electrons at a dose of 2 and 8 Gy results in a decrease in the number of enterocytes, mainly stem cells of the intestinal crypts.
The present study was designed to clarify the antidiabetic activity and mechanism of Dioscorea rhizome in diabetic db/db mice. Mice were administered Dioscorea rhizome and rosiglitazone orally for 7 weeks and the effects of these compounds on fasting blood glucose, glucose tolerance and intestinal disaccharidase activity in db/db mice were evaluated. The fasting serum glucose of the D. rhizome treated group was reduced when compared with that of the db/db control group. In addition, the disaccharidase activities in homogenates of the proximal, middle and distal segment of the small intestine were significantly decreased response to D. rhizome treatment, especially in the middle segment. These results suggest that D. rhizome decreases blood glucose via a decrease in the activity of disaccharidase in the mucosa of the middle region of the small intestine in db/db mice.
Five hundred and forty-eight samples of pig heart muscle were collected from the abattoirs of many regions in Korea to reveal the frequency of Sarcocystis infections and to identify the species from June 1988 to April 1989. Heart muscle of the pigs was inspected for sarcocysts by the direct detection technique and for bradyzoites by the trypsin digestion technique. For examination of development of the parasites in the final host, 5 cross bred mature dogs, 5 puppies and 5 kittens were fed 100g, 50g and 50g of the infected meat respectively, four times in 2 days. Of 402 fattened and 146 older culled breeding pigs, 3 fattened pigs and 39 culled pigs were positive for Sarcocystis. Sarcocystis cysts from heart muscle measured an average of $425{\times}169{\mu}m$ and bradyzoites an average of $15.6{\times}3.5{\mu}m$. Of 15 animals, only 2 puppies were infected with Sarcocystis. The prepatent period was 11 to 12 days and patent period was not examined since the puppies were infected with some another infections and one died on day 11 and another died on day 12 after ingestion of the meat. The sporulated oocysts were detected 11 days after ingestion of the meat and sporocysts 12 days from the puppy feces. The sporulated oocysts measured an average of $16.5{\times}11.5{\mu}m$ and sporocysts an average of $12.6{\times}7.9{\mu}m$. On scraping examination of the intestinal mucosa, fully sporulated oocysts were detected in the tip of the intestinal villi. Considering above all descriptions, Sarcocystis in pig heart muscle in Korea was identified with Sarcocystis suicanis.
Purpose: Alimentary tract duplication (ATD) is a rare congenital condition that may occur throughout the intestinal tract. Clinical symptoms are generally related to the involved site, size of duplication, or associated ectopic mucosa. This study aimed to identify clinical implications by anatomical locations and age group and then suggest a relevant management according to its distinct features. Methods: We retrospectively reviewed the clinical data of pediatric patients who received a surgical management due to ATD. Furthermore, data including patients' demographics, anatomical distribution of the duplication, clinical features according to anatomical variants, and outcomes were compared. Results: A total of 25 patients were included in this study. ATD developed most commonly in the midgut, especially at the ileocecal region. The most common clinical presentation was abdominal pain, a sign resulting from intestinal obstruction, gastrointestinal bleeding, and intussusception. The non-communicating cystic type was the most common pathological feature in all age groups. Clinically, prenatal detection was relatively low; however, it usually manifested before the infantile period. A laparoscopic procedure was performed in most cases (18/25, 72.0%), significantly in the midgut lesion (p=0.012). Conclusion: ATD occurs most commonly at the ileocecal region, and a symptomatic one may usually be detected before the early childhood period. Surgical management should be considered whether symptom or not regarding its symptomatic progression, and a minimal invasive procedure is the preferred method, especially for the midgut lesion.

The morphological studies on the rectal development in the 60-, 90- and 120-day-old fetuses and neonates of Korean native goats were investigated by light microscopy and scanning electron microscopy. The rectal wall appeared to be differentiated into the epithelium, lamina propria, tunica muscularis and serosa in 60-day-old fetus. The rectal epithelia were stratified in some areas and simple columnar in others at 60- and 90-day-old fetuses but was only observed simple columnar epithelium at 120-day-old fetus. The goblet cells and the intestinal glands appeared in the rectal mucosa at 90-day-old fetus and gradually increase in number during gestation period. The rectal villi appeared at 60-day-old fetus and increased in length and width until 90-day-old fetus but the villi appeared to be shorter and degenerated after 120-day-old fetus. Only the rudiment trace of the villi remained at birth. The tunica muscularis of the rectum were continuously developed during gestation period. The inner circular and outer longitudinal muscle layers of rectum were observed in the 120-day-old fetuses. Under scanning electron microsoopy, the rectal villi appeared at 60-day-old fetus. The pentagonal/hexagonal microridges and numerous microvilli were observed on the luminal surface of rectal wall. Low and thumb-like rectal villi were developed at 90-day-old fetus. The rectal villi became degenerated after 120-day-old fetus. Only the rudiment of villi was remained at birth. Many circular folds were well developed. Opening portions of intestinal glands appeared like slot.

Background: This study aimed to evaluate upper gastrointestinal polyps detected during esophago-gastroduodenoscopy tests. Materials and Methods: We conducted a retrospective analysis on data regarding 55,987 upper gastrointestinal endoscopy tests performed at the endoscopy unit of Istanbul Education and Research Hospital between January 2006 and June 2012. Results: A total of 66 upper gastrointestinal polyps from 59 patients were analyzed. The most common clinical symptom was dyspepsia, observed in 41 cases (69.5%). The localizations of the polyps were as follows: 29 in the antrum (43.9%), 15 in the corpus (22.7%), 11 in the cardia (16.7%), 3 in the fundus (4.54%), 3 in the second portion of the duodenum (4.54%), 2 in the bulbus (3.03%) and 3 in the lower end of the esophagus (4.54%). Histopathological types of polyps included hyperplastic polyps (44) (66.7%), faveolar hyperplasia (8) (12.1%), fundic gland polyps (4) (6.06%), squamous cell polyps (4) (6.06%), hamartomatous polyps (3) (4.54%), and pyloric gland adenoma (3) (4.54%). Histopathological analysis of the gastric mucosa showed chronic atrophic gastritis in 30 cases (50.84%), HP infection in 33 cases (55.9%) and intestinal metaplasia in 19 cases (32.20%). In 3 cases with multiple polyps, adenocarcinoma was detected in hyperplastic polyps. Conclusions: Among polypoid lesions of the upper gastrointestinal tract, the most common histological type is hyperplastic polyps. Generally, HP infection is associated with chronic atrophic gastritis and intestinal metaplasia. The incidence of adenocarcinoma tends to be higher in patients with multiple hyperplastic polyps.

Heparin lyase I was purified to homogeneity from Bacteroides stercoris HJ-15 isolated from human intestine, by a combination of DEAE-Sepharose, gel-filtration, hydroxyapatite, and CM-Sephadex C-50 column chromatography. This enzyme preferred heparin to heparan sulfate, but was inactive at cleaving acharan sulfate. The apparent molecular mass of heparin lyase I was estimated as 48,000 daltons by SDS-PAGE and its isoelectric point was determined as 9.0 by IEF. The purified enzyme required 500 mM NaCl in the reaction mixture for maximal activity and the optimal activity was obtained at pH 7.0 and $50^{\circ}C$. It was rather stable within the range of 25 to $50^{\circ}C$ but lost activity rapidly above $50^{\circ}C$. The enzyme was activated by $Co^{2+}$ or EDTA and stabilized by dithiothreitol. The kinetic constants, $K_m$ and $V_{max}$ for heparin were $1.3{\times}10^{-5}\;M$ and $8.8\;{\mu}mol/min{\cdot}mg$. The purified heparin lyase I was an eliminase that acted best on porcine intestinal heparin, and to a lesser extent on porcine intestinal mucosa heparan sulfate. It was inactive in the cleavage of N-desulfated heparin and acharan sulfate. In conclusion, heparin lyase I from Bacteroides stercoris was specific to heparin rather than heparan sulfate and its biochemical properties showed a substrate specificity similar to that of Flavobacterial heparin lyase I.
Objectives This study aimed to observe the effect of Taeumjowi-tang on the cisplatin-induced gastrointestinal dysfunctions in rats. Methods Four groups, each of 8 rats per group, were used in this study. Saline and distilled water treated control rats were intact vehicle control group. Delayed gastrointestinal motility was induced by intraperitoneal treatment of cisplatin 2mg/kg, once a week for 5 weeks(Cisplatin control group). Taeumjowi-tang aqueous extracts(TJ) were orally administered in a volume of 5ml/kg, once a day for 14 days from 4th cisplatin treatment(TJ group). Ondansetron 1mg/kg was subcutaneously treated, in a volume of 1ml/kg, as same as TJ(ondansetron group). We measured the body weights, intestinal charcoal transit ratio, fecal parameters, fundus MDA(malondialdehyde), GSH(glutathione) contents and SOD(superoxide dismutase), CAT(catalase) activities, TPH(tryptophanhydroxylase) and MAO(monoamine oxidase) activities, pyloric gastrin and serotonin contents with their immunoreactive cells, colonic serotonin-immunoreactive cells, the histopathology of pylorus, fundus mucosa and colon. Results 1) The body weight gains, the small intestinal charcoal transfer rates, the fecal parameters(numbers, weights and water contents) were increased in TJ, ondansetron group. 2) The inhibit of fundus antioxidant defense systems by cisplatin were decreased in TJ, ondansetron group. 3) The pyloric TPH activities were increased and the pyloric MAO activities were decreased in TJ group. 4) The pyloric gastric contents and the gastrin-immunoreactive cells were increased and the pyloric serotonin contents and the pyloric and colonic serotonin-immunoreactive cells were decreased in TJ group. 5) The pylorus atrophic changes and the gastric surface erosive damage regions by cisplatin were favorably inhibited by treatment of TJ group. Conclusions The results obtained in this study suggest that TJ favorably retarded the cisplatin related GI(gastrointestinal) dysfunctions and constipation through modulations of GI enterochromaffin cells, serotonin and gastrin-producing cells and antioxidative systems.
Objective: Microencapsulation is a technique to improve stability, bioavailability, and controlled release of active ingredients at a target site. This experiment aimed to investigate the effects of microencapsulated basil oil (MBO) on growth performance, apparent ileal digestibility (AID), jejunal histomorphology, bacterial population as well as antioxidant capacity of broiler chickens in a tropical climate. Methods: A total of 288 one-day-old female broilers (Ross 308) were randomly allocated into 4 groups (6 replicates of 12 birds), based on a completely randomized design. Dietary treatments were as follows: i) basal diet (NC), ii) basal diet with avilamycin at 10 ppm (PC), iii) basal diet with free basil oil (FBO) at 500 ppm, and iv) basal diet with MBO at 500 ppm, respectively. Results: Dietary supplementation of MBO improved average daily gain, and feed conversion ratio of broilers throughout the 42-d trial period (p<0.05), whereas MBO did not affect average daily feed intake compared with NC group. The broilers fed MBO diet exhibited a greater AID of crude protein and gross energy compared with those in other groups (p<0.05). Lactobacillus spp. and Escherichia coli populations were not affected by feeding dietary treatments. Both FBO and MBO had positive effects on jejunal villus height (VH), villus height to crypt depth ratio (VH:CD) and villus surface area of broilers compared to NC and PC groups (p<0.05). Superoxide dismutase level in the duodenal mucosa of MBO group was significantly increased (p<0.01), whereas malondialdehyde level was significantly decreased (p<0.01). Conclusion: Microencapsulation could be considered as a promising driver of the basil oil efficiency, consequently MBO at 500 ppm could be potentially used as a feed additive for improvement of intestinal integrity and nutrient utilization, leading to better performance of broiler chickens.
Eosinophilic gastrointestinal disease (EGID), a chronic allergic condition characterized by dense infiltration of eosinophils in the digestive tract, is classified into two types, eosinophilic esophagitis (EoE), which features dense infiltration of eosinophils in the esophageal epithelial layer, and eosinophilic gastroenteritis (EGE), in which the entire digestive tract including the esophagus may be involved. Patients with EoE only have esophageal symptoms, since the other parts of the digestive tract are not involved. On the other hand, 80% of EGE patients have lesions in the small intestine. The esophageal epithelial layer in healthy individuals has no or negligible infiltration by eosinophils, while the small intestinal mucosal layer, especially the distal small intestinal mucosa, can show dense eosinophil infiltration even in the absence of disease. Therefore, histological changes observed in cases of EGE are not qualitative but rather quantitative, as compared to EoE, which has qualitative histopathological changes, indicating important pathogenetic differences between the types. Comparisons of clinical, laboratory, and morphological characteristics between EoE and EGE have revealed several interesting differences. Both EoE and EGE patients are frequently affected by atopic diseases, such as bronchial asthma and allergic rhinitis, and elevated plasma levels of Th2 type cytokines and chemokines are also similarly seen in both. On the other hand, age at diagnosis differs, as the former is generally found in individuals from 30 to 50 years old, while the latter appears in all age groups. Additionally, 80% of patients with EoE are male as compared to only 50% of those with EGE. Furthermore, approximately 60% of patients with EoE respond favorably to proton pump inhibitor (PPI) administration, whereas EGE patients rarely show a response to PPIs. Nevertheless, both diseases show a similarly favorable response to a six foods elimination diet and glucocorticoid administration. These similarities and differences of EoE and EGE provide important clues for understanding the pathogenesis of these EGID types.
