Effects of Dietary Glutamine and Glutamate Supplementation on Small Intestinal Structure, Active Absorption and DNA, RNA Concentrations in Skeletal Muscle Tissue of Weaned Piglets during d 28 to 42 of Age

  • Seventy-four piglets were used to investigate the effects of dietary glutamine (Gln) and glutamate (Glu) on the mucosal structure and active absorption of small intestinal, DNA and RNA concentrations of skeletal muscle tissue in piglets during d 28 to 42 of age. Postweaning piglets were fed for 14 d corn- and soybean meal-based diets supplemented with 0.0 or 1.0% L-Gln or L-Glu. On d 7 and 14 postweaning, pigs' small intestinal sections and longissimus dorsi were collected, at the same time, the D-xylose absorption test was conducted. The results suggested that in comparison to control piglets, jejunal atrophy during the first week postweaning was prevented by the glutamine and glutamate supplementation (1%) and the capability of small intestine to absorb Dxylose was improved. Furthermore the RNA concentration in skeletal muscle tissue was increased. These results provide an experimental basis for use of glutamine and glutamate on alleviating the weaning stresses and improving piglets' growth performance.

Sulfanilamide유도체의 동(II)착화합물에 대한 생물약제학적 연구 (Biopharmaceutical studies on copper(II) chelates of sulfanilamide derivatives)

  • Cu(II) chelates of several sulfanilamide derivatives (Sulfa-Cu) were prepared and their effects on solubility, absorptivity in intestinal lumen, biding tendency with serum protein and erythrocytes, concentration in rabbit blood, and acetylation rate were studied in comparison with their free ligand forms. For solubility concerned, the partition coefficients of Sulfa-Cu are decreased as following order: Sulfadimethoxine Copper chelate (SDM-Cu), Sulfamethoxypyridazine Copper chelate (SD-Cu), Sulfamerazine Copper chelate (SM-Cu), Sulfaisoxazole Copper chelate (SIX-Cu). The partition coefficients of SDM-Cu and ST-Cu were much greater than those of ligands. this phenomenone acounts for the rapid absorption of SDM-Cu and ST-Cu in the rat small intestine (in situ). The Sulfa-Cu were absorbed at the intestinal lumen of a rat in the rate of first order and there was no difference between long acting sulfa drugs and their Cu0chelates in biological half lives. In binding experiments, sulfa-Cu binded with serum protein in lower ratio than their ligands except SIX-Cu. On other hand, acetylation rates of sulfa-Cu were higher than those of free sulfa drugs and the acetylation rate were higher than those of free sulfa drugs and powder. In a experiment on Sulfa-Cu concentration in rabbit blood, the half lives of SD-Cu, SIX-cu, ST-Cu, and SM-Cu were longer than those of their ligands. Above all, the half life of SD-Cu appeared to be approximately 3.5 times logner than that of corresponding ligand, SD. When absorption of sulfa drugs or sulfa-Cu at the small intestinal lumen of a rat and the concentration in rabbit blood after absorption were compared, it was found that there was not always conrrelated.

베르베린 제제의 생체 이용율에 관한 연구 (Studies on the Bioavailability of Berberine Preparations)

  • In order to measure the bioavailability of Coptidis Rhizoma preparations, berberine was extracted by the mixture (1:1 v/v) of methanol and water from Coptidis Rhizoma and formulated into pills with some excipients. Disintegration time, dissolution rate, intestinal absorption rate, antibacterial effect, and serum concentration were measured. All pills were disintegrated about 60 minutes in gastric and intestinal juices and disintegration time did not change upon storage at $40^{\circ}C$ for 30 days. The pills manufactured using the extract which was extracted with the solvent containing dilute sulfuric acid showed faster dissolution and absorption from the small intestine than the other samples. It also showed not only the largest area under the serum concentration time curve and relative bioavailability, but also more potent antibacterial effect against gram-positive and gram-negative bacterias than the other samples. The correlation of the dissolution rate with absorption rate constant or $t_{max}$ showed a significant linear relationship.

Comparisons between White Ginseng Radix and Rootlet for Antidiabetic Activity and Mechanism in KKAy Mice

  • The mechanisms responsible for the antidiabetic activity of both the white ginseng radix (Ginseng Radix Alba, GRA) and the rootlet (Cinseng Radix Palva, GRP) were investigated. After a four week oral administration, the fasting blood glucose levels in the GRA- and GRP-treated groups were lower when compared to the control group. To elucidate the hypoglycemic mechanism(s) of the ginseng radices, glucose absorption from the small intestine, hepatic hexokinase and glucose-6-phosphatase activities, in addition to PPAR-${\gamma}$ expression in adipose tissue were examined. The results strongly suggest that GRA can improve hyperglycemia in KKAy mice, possibly by blocking intestinal glucose absorption and inhibiting hepatic glucose-6-phosphatase, and GRP through the upregulation of adipocytic PPAR-$\gamma$ protein expression as well as inhibiting intestinal glucose absorption.

A Comparison of the Intestinal Absorption of Amino Acids in Piglets When Provided in Free Form or as a Dipeptide

  • Three 28 day-old $Duroc{\times}Large$ $White{\times}Landrace$ litter mate gilts weighing an average of 6.5 kg were used to study the intestinal absorption of amino acids when provided in dipeptide form or in the form of a free amino acid mixture. The pigs were given one of three treatments. The control involved a duodenal infusion containing no amino-acids (phosphate buffer plus 5% sorbitol) while the remaining two treatments involved either a duodenal infusion containing a glycine-lysine dipeptide (1 g) or a mixture of the free amino acids glycine and lysine at the same concentration as in the dipeptide. Blood was drawn from a cannula inserted in the portal vein, at 5 to 20 minute intervals, for two hours following infusion. The concentration of intact dipeptide as well as free glycine and lysine in the portal blood was determined by high performance liquid chromatography. The intact dipeptide was never detected in the portal blood at any time after infusion. Lysine appeared in the portal blood more rapidly after infusion of dipeptide than after infusion of free lysine and the concentration of lysine in portal blood was higher in the pig infused with the dipeptide than after infusion of free lysine at almost all time points measured. The cumulative absorption of lysine and glycine from the intestine during the two hour period after infusion was greater in the pig infused with dipeptide than in the pig infused with free amino acids. The results suggest that although intact dipeptide did not reach he portal circulation, a special transport mechanism for absorption of dipeptide by intestinal cells appears to be present in pigs similar to that observed in other species.

Host-Microbe Interactions Regulate Intestinal Stem Cells and Tissue Turnover in Drosophila

  • With the activity of intestinal stem cells and continuous turnover, the gut epithelium is one of the most dynamic tissues in animals. Due to its simple yet conserved tissue structure and enteric cell composition as well as advanced genetic and histologic techniques, Drosophila serves as a valuable model system for investigating the regulation of intestinal stem cells. The Drosophila gut epithelium is in constant contact with indigenous microbiota and encounters externally introduced "non-self" substances, including foodborne pathogens. Therefore, in addition to its role in digestion and nutrient absorption, another essential function of the gut epithelium is to control the expansion of microbes while maintaining its structural integrity, necessitating a tissue turnover process involving intestinal stem cell activity. As a result, the microbiome and pathogens serve as important factors in regulating intestinal tissue turnover. In this manuscript, I discuss crucial discoveries revealing the interaction between gut microbes and the host's innate immune system, closely associated with the regulation of intestinal stem cell proliferation and differentiation, ultimately contributing to epithelial homeostasis.

생약재에 의한 중금속의 체내흡수 억제 효과 (Suppressing Effect of Medicinal Plants on the Intestinal Absorption of Heavy Metals)

  • 음용수 중에 납과 카드뮴이 오염되었을 때 치커리, 당귀, 영지버섯의 중금속의 장내 흡수억제 효과가 있는지를 알아보기 위해 장내 흡수 조건을 고려한 투과막을 이용하여 in vitro실험을 실시하였다. 납과 카드뮴이 음용수 수질기준의 10배 또는 50배로 오염된 음용수에서 단독오염의 경우 납은 치커리, 당귀, 영지버섯에서 $40{\sim}60%$의 제거효과를 보였고 카드뮴의 경우는 치커리, 당귀, 영지버섯에서 $20{\sim}40%$의 제거효과를 나타내었다. 납과 카드뮴이 혼합오염된 경우에 납은 치커리, 당귀, 영지버섯의 세 시료에서 비슷하게 $30{\sim}50%$의 제거효과를 나타냈으며 카드뮴은 $10{\sim}30%$의 제거효과를 보여주었는데 치커리, 영지버섯에 비해 당귀가 높은 제거효과를 보여주었다. 추출조건별로 비교해보면 $70^{\circ}C$, 2분 추출조건의 경우가 $95^{\circ}C$, 10분의 경우보다 더 제거효과가 높았음을 관찰할 수 있었다. 한편 여과지 및 투과막에도 상당량의 중금속이 흡착되어 제거된다는 사실을 알 수 있었는데 오염된 중금속의 농도가 높을수록, 음료 추출온도가 높을수록 더 많은 중금속이 흡착되었음을 관찰할 수 있었다. 이러한 결과는 투과막이 중금속에 흡착되기 쉬운 섬유질로 이루어져 있기 때문인 것으로 추측되며 치커리, 당귀, 영지는 음용수중에 오염된 중금속의 장내흡수 억제 효과가 있음을 알 수 있었다.

Dietary phosphorus deficiency impaired growth, intestinal digestion and absorption function of meat ducks

  • Objective: An experiment was conducted to investigate the effects of dietary non-phytate phosphorus (nPP) deficiency on intestinal pH value, digestive enzyme activity, morphology, nutrient utilization, and gene expression of NaPi-IIb in meat ducks from 1 to 21 d of age. Methods: A total of 525 one-d-old Cherry Valley ducklings were fed diets (with 7 pens of 15 ducklings, or 105 total ducklings, on each diet) with five levels of nPP (0.22%, 0.34%, 0.40%, 0.46%, or 0.58%) for 21 d in a completely randomized design. Five experimental diets contained a constant calcium (Ca) content of approximately 0.9%. Body weight (BW), body weight gain (BWG), feed intake (FI), and feed to gain ratio (F:G) were measured at 14 and 21 d of age. Ducks were sampled for duodenum and jejunum digestion and absorption function on 14 and 21 d. Nutrient utilization was assessed using 25- to 27-d-old ducks. Results: The results showed ducks fed 0.22% nPP had lower (p<0.05) growth performance and nutrient utilization and higher (p<0.05) serum Ca content and alkaline phosphatase (ALP) activity. When dietary nPP levels were increased, BW (d 14 and 21), BWG and FI (all intervals), and the serum phosphorus (P) content linearly and quadratically increased (p<0.05); and the jejunal pH value (d 14), duodenal muscle layer thickness (d 14), excreta dry matter, crude protein, energy, Ca and total P utilization linearly increased (p<0.05); however, the serum ALP activity, jejunal $Na^+-K^+$-ATPase activity, and duodenal NaPi-IIb mRNA level (d 21) linearly decreased (p<0.05). Conclusion: The results indicated that ducks aged from 1 to 21 d fed diets with 0.22% nPP had poor growth performance related to poor intestinal digestion and absorption ability; but when fed diets with 0.40%, 0.46%, and 0.58% nPP, ducks presented a better growth performance, intestinal digestion and absorption function.

INTESTINAL WALL PERMEABILITY STUDY OF RANITIDINE IN DOGS

  • Recently a novel in vivo approach in dogs, using a regional segmental intestinal perfusion technique, has been developed. The perfusion tube consists of a highly sophisticated multichannel tube with two inflatable occluding balloons, which are placed in 10cm apart. The tube was introduced orally from the stomach through the upper jejunum under the guidance of solid-state pH meter. In the present study, four healthy dogs were infused in the proximal jejunum on two periods. The two perfusion experiments used the same flow rate, 2 $m\ell$/min, and the same perfusion solution to determine the intrasubject variability. The mean (${\pm}$ S. E.) fractions of ranitidine absorbed calculated from the perfusion data were 21.32${\pm}$2.01% (n=3) (1st period), 27.88 ${\pm}$ 17.54% (n=4) (2nd period), respectively. The effective permeabilities (Peffs${\times}$10$\^$4/) of ranitidine were 1.51${\pm}$0.47cm/sec (n=3) (1st period), 1.50 ${\pm}$ 0.31 cm/sec (n=4) (2nd period), respectively. The pH and osmolarity of perfusion solution were 7.50 ${\pm}$ 0.03 and 300 ${\pm}$ 0.06 mOsm/L, There was no significant intrasubject variation. Mixing equilibrium (steady-state) was reached at about 50 min. l-Phenylalanine was absorbed almost completely. Intrinsic intestinal wall permeability of ranitidine showed low permeable characteristics, suggesting permeability-limited absorption. The absorption of 1-phenylalanine, an actively transported nutrient, was not inhibited by ranitidine. The low intestinal membrane permeability is one of the important factors responsible for the variable oral absorption of ranitidine. Supported by FDA Grant FD01462-04 and KOSEF Grant.

Effect of High-Molecular-Weight Poly-$\gamma$-Glutamic Acid from Bacillus subtilis (chungkookjang) on Ca Solubility and Intestinal Absorption

  • The bioavailability of Ca is currently one of the most important topics in nutrition research and is correlated with gastrointestinal solubility. Thus, to increase the solubility of calcium, this study was undertaken to examine the effect of $\gamma$-PGA on intestinal Ca solubility. The calcium solubility increased when the amount of $\gamma$-PGA was increased, due to the inhibition of the formation of an insoluble Ca complex with phosphate. Therefore, when $\gamma$-PGA-500 (avg. MW 5,000 kDa) was added at 0.5 mg/ml, $75\%$ of the total Ca was soluble. The amount of soluble Ca uptake in the small intestine was investigated using Balb/c mice as an animal model system. The soluble Ca uptake in the mice orally administered with $\gamma$-PGA-500 (avg. MW 5,000 kDa) was significantly higher than that in the $\gamma$-PGA-l00 (avg. MW 1,000 kDa)-administered mice (P<0.05). Accordingly, these results strongly support the notion that the molecular size of $\gamma$-PGA is correlated with Ca solubility. The effects of other factors, such as casein phosphopeptide and vitamin D, on intestinal Ca absorption have also previously been investigated. Therefore, it is hoped that the present observations will help clarify the role of $\gamma$-PGA in Ca solubility and its industrial application as an additive.
