The bioavailabilities of baicalin in water, oil, w/o and o/w emulsion were evaluated in rats. The dissolution rate of baicalin in o/w emulsion was smaller than those of w/o form in dilute hydrochloric acid solution (pH 1.2) and in PBS (pH 6.8). The absorption rate of baicalin from w/o emulsion was smaller than that of o/w emulsion in the different parts of rat intestine of the rats. Following oral administration in rats, the $C_{max}$ of baicalin from water phase, oil phase, o/w wand w/o emulsion were 2.11, 0.61, 1.57, and $1.35\;{\mu}g/ml$, respectively. The relative bioavailability of w/o emusion was 129 % when it was compared with water phase. This result suggests that the improvement of bioavailability for baicalin in w/o emulsion might be practically available.
The absorption rate of lysino-methylene-ampicillin from the rat small intestine, compared with ampicillin, was determined in vitro and in situ to establish the biopharmaceutical properties of lysino-methylene-ampicillin which is one of the new penicillinase-resistance antibiotics. The half of administered dose was absorbed rapidly within thirty minutes. The water-oil distribution coefficient of lysino-methylene-ampicillin was 0.03 in chloroform versus buffer system with $Na_{2}HPO_{4}-citric$ acid of pH5.2 at $37^{\circ}C$, and its dissolution rate reached the plateau in an hour and then represented 0.6 percent of equilibrium solubility
The effects of cimetidine or ranitidine pretreatment on the intestinal absorption, plasma and urine level of caffeine, gastric acidity of mouse, and sleeping time by hexobarbital sodium were investigated pharmacokinetically. Cimetidine and ranitidine pretreatments were found to increase both the rate and extent of absorption of caffeine in rats. Cimetidine pretreatment increased blood level of caffeine and decreased urine level, while ranitidine pretreatment had no effect on urine level of caffeine. Ranitidine pretreatment inhibited gastric secretion due to caffeine more than cimetidine pretreatment. Cimetidine pretreatment increased the action of caffeine and showed shorter sleeping time by hexobarbital sodium, comparing with ranitidine pretreatment.
(-)-epigallocatechin-3-gallate (EGGG), a flavonoid found in green tea, is known to have low bioavailability. In this study, we determine whether piperine, a natural bioenhancer, can increase the absorption rate of EGCG. Using a Caco-2 cell monolayer, permeability experiments were performed in Hanks' balanced salt solution (HBSS) and EGCG stability was adjusted to pH 6.5 and pH 5.5 by ascorbic acid treatment. When HBSS was adjusted to pH 6.5, EGCG remained at 94.78% for up to 2 h and remained at 86.04% after 4 h and the net efflux decreased compared to the control. As a result, uptake was significantly increased in the piperine co-administered group compared to the EGCG-alone group, showing that piperine increased the permeability of EGCG in the Caco-2 cell monolayer. These results suggest that piperine inhibits EGCG glucuronidation and efflux, allowing for greater absorption of EGCG.
The effort of proteolytic enzyme on the absorption and excretion of sulfadimethoxine are investigated in this paper. The rat small intestinal absortion of sulfadimethoxine in the presence of proteolytic enzymes such as chymotrypsin and trypsin are increasingly absorbed more than 20% during 3 hours. Blood levels of sulfadimethoxine following oral administration are not effected by proteolytic enzyme, but both on the high concentration of chymotrypsin and the low concentration of trypsin at only 3 and 4 hours are increased(P<0.05) from that of the control . Blood levels of sulfadimethoxine after duodenum injection are remakably enhensed to correspand to 18.5-25.0% (P<0.01) by the proteolytic enzyme concentration respectively. Proteolytic enzymes did not give influence on excretions of sulfadimethoxine after duodenum and oral administration.
Rifampicin-arginine complex was prepared to increase the solubility and dissolution rate of rifampicin. Solvation method was applied to make the complex and its formation was identified by the solubility method, powder x-ray diffractometry, differential thermal analysis and spectroscopic determination. The complex was formed in the molar ratio of 1 : 1 which was proved by the slope ratio method and continuous variation method. The complex was a non-crystalline form determined by the x-ray powder diffractometric analysis. The solubility of complex in water was significantly higher than that of rifampicin itself. The stability constant and thermodynamical properties of the complex were determined to investigate the solubilization phenomena. The thermodynamic data showed that the complexation between rifampicin and arginine was an exothermic and spontaneous reaction. Simulated absorption studies carried out through the artificial lipid barrier in artificial gastric and intestinal juices. The results showed that the complex had an enhanced absorption rate of rifampicin nearly twice compared with that of rifampicin itself.

The aim of the present study was to evaluate the effect of commercial monostrain and multistrain probiotics in diets on growth performance, intestinal morphology and mucin gene (MUC2) expression in broiler chicks. Three hundred seventy-eight 1-d-old male Arian broiler chicks were allocated in 3 experimental groups for 6 wk. The birds were fed on a corn-soybean based diet and depending on the addition were labeled as follows: control-unsupplemented (C), birds supplemented with Bacillus subtilis (BS) and lactic acid bacteria (LAB) based probiotics. Each treatment had 6 replicates of 21 broilers each. Treatment effects on body weight, feed intake, feed conversion ratio and biomarkers such as intestinal goblet cell density, villus length, villus width, and mucin gene expression were determined. Total feed intake did not differ significantly between control birds and those fed a diet with probiotics (p>0.05). However, significant differences in growth performance were found. Final body weight at 42 d of age was higher in birds fed a diet with probiotics compared to those fed a diet without probiotic (p<0.05). Inclusion of Bacillus subtilis based probiotic in the diets also significantly affected feed conversion rate (FCR) compared with control birds (p<0.05). No differences in growth performance were observed in birds fed different types of probiotic supplemented diets. Inclusion of lactic acid bacteria based probiotic in the diets significantly increased goblet cell number and villus length (p<0.05). Furthermore, diets with Bacillus subtilis based probiotics significantly increased gene expression (p<0.05), with higher intestinal MUC2 mRNA in birds fed diet with probiotics compared to those fed the control diet. In BS and LAB probiotic fed chicks, higher growth performance may be related to higher expression of the MUC2 gene in goblet cells and/or morphological change of small intestinal tract. The higher synthesis of the mucin gene after probiotic administration may positively affect bacterial interactions in the intestinal digestive tract, intestinal mucosal cell proliferation and consequently efficient nutrient absorption.
This study was carried out to investigate the efficacy of sn-2 palmitic acid-fortified vegetable oil (Sn2PA) on calcium absorption and to confirm the synergistic effects of fructooligosaccharide on calcium absorption. Male SD rats were fed 6 kinds of casein based diets containing vegetable oil (control), sn-2 palmitic acid-fortified vegetable oil (Sn2PA) and Sn2PA with fructooligosaccharide(Sn2PAFO) in two levels of calcium (normal 0.5% and high 1.0%) for 3 weeks. Total lipids, cholesterol, triglyceride and calcium in blood were measured. Feces were collected using cages for 4 days. Serum concentrations of total lipids and calcium were not significantly different among groups. However, serum triglyceride was significantly decreased by fructooligosaccharide supplementation regardless of dietary calcium level. The lipid absorption was not significantly different among experimental groups. Calcium absorption was significantly higher in Sn2PAFO group than other groups. Calcium solubility of intestine was increased by sn-2 palmitic acid supplementation. These results suggest that sn-2 palmitic acid and fructooligosaccharide supplementation could be beneficial for baby foods including infant formula, with regard to increasing absorption of calcium by more soluble calcium in the small intestinal content.
The present study was designed to assess the influence of caeca on the availability of calcium, phosphorus, magnesium, manganese and copper from soybean, sunflower, rapeseed, sesame, fish and meat cum bone meal in adult roosters. The excretion of endogenous origin minerals viz., copper, magnesium, manganese and calcium was significantly (p<0.001) higher in caecectomized than in normal roosters. The difference in the endogenous excretion was 50; 60.45; 40.35 and 29.63 per cent for copper, magnesium, manganese and calcium, respectively, in caecectomized roosters. The caeca played a pivotal role in the reabsorption of endogenous origin calcium, magnesium, manganese and copper. The mechanism of phosphorus absorption by the caecal epithelium was negligible. The caecectomized roosters underestimated the bioavailability of copper in sunflower meal and manganese in almost all the test feedstuffs. The present investigation revealed that the caeca played a critical role in the absorption of minerals from vegetable protein feedstuffs which escape digestion and absorption in the small and large intestinal segments.
Microcapsules(Mc) of cephalexin (CEPH), using Eudragit RS, RL, L, S and polyethylene glycol 1540, were evaluated biopharmaceutically. The area under the curve of CEPH-Eudragit RS/RL Mc administered orally once was larger than that of cephalexin powder twice every 6 hrs. Controlledrelease effectiveness and the absorption rate effectiveness, two important parameters of Vallner's method, of CEPH-Eudragit RS/RL Mc indicate that these Mc can be good sustained-release preparations. And a simple pharmacokinetic model is introduced which allows the gastric emptying and intestinal-transit rates of a drug itself and a solid-state drug contained in Mc. Decreasing $K_r$, without change in $K_a$, showed that the rate-limiting step of absorption moved from absorption step to releasestep.
