Establishing porcine jejunum-derived intestinal organoids to study the function of intestinal epithelium as an alternative for animal testing

  • Background: The small intestine plays a crucial role in animals in maintaining homeostasis as well as a series of physiological events such as nutrient uptake and immune function to improve productivity. Research on intestinal organoids has recently garnered interest, aiming to study various functions of the intestinal epithelium as a potential alternative to an in vivo system. These technologies have created new possibilities and opportunities for substituting animals for testing with an in vitro model. Methods: Here, we report the establishment and characterisation of intestinal organoids derived from jejunum tissues of adult pigs. Intestinal crypts, including intestinal stem cells from the jejunum tissue of adult pigs (10 months old), were sequentially isolated and cultivated over several passages without losing their proliferation and differentiation using the scaffold-based and three-dimensional method, which indicated the recapitulating capacity. Results: Porcine jejunum-derived intestinal organoids showed the specific expression of several genes related to intestinal stem cells and the epithelium. Furthermore, they showed high permeability when exposed to FITC-dextran 4 kDa, representing a barrier function similar to that of in vivo tissues. Collectively, these results demonstrate the efficient cultivation and characteristics of porcine jejunum-derived intestinal organoids. Conclusions: In this study, using a 3D culture system, we successfully established porcine jejunum-derived intestinal organoids. They show potential for various applications, such as for nutrient absorption as an in vitro model of the intestinal epithelium fused with organ-on-a-chip technology to improve productivity in animal biotechnology in future studies.

Comparison of organic acids supplementation on the growth performance, intestinal characteristics and morphology, and cecal microflora in broilers fed corn-soybean meal diet

  • Objective: The aim of this study was to compare the effects of three kinds of organic acid (OA) products on the growth performance, intestinal characteristics and morphology, and cecal microflora in broilers fed a corn-soybean meal meal diet. Methods: A total of 420 one-day-old male Cobb 500 broilers with an average initial body weight of 49.11±1.02 g were used in this 42-day experiment. Birds were randomly allotted to one of five treatments (7 replicates with 12 birds per replicate). Treatments consisted of negative control (NC), basal diet; positive control (PC), basal diet+100 mg/kg of Aviramycin; OA1, basal diet+500 mg/kg of OA product 1; OA2, basal diet+1,000 mg/kg of OA product 2; and OA3, basal diet+1,200 mg/kg of OA product 3. Results: The results indicated that OA product addition had no effect on growth performance parameters, such as body weight gain, feed intake, and feed conversion ratio, from days 1 to 14, 15 to 28, and 0 to 42, or on the pH values of the intestine, intestinal weight, or intestinal weight to body weight ratio. The intestinal morphology in terms of villus height and crypt depth were affected by dietary supplementation of OA products, respectively. Furthermore, dietary addition of OAs had positive influences on the maintenance of the cecal microflora based on the results of 16S rRNA analysis. Conclusion: Dietary inclusion of three kinds of OA products all benefit broilers, but the mode of action may be different. This study provides a basis for the application of OA products used in the poultry industry.

Prevalence and Co-infection of Intestinal Parasites among Thai Rural Residents at High-risk of Developing Cholangiocarcinoma: A Cross-sectional Study in a Prospective Cohort Study

  • Intestinal parasitic infections (IPIs) are still important to the health of Thai rural residents. IPIs are the cause of many chronic diseases with, for example, opisthorchiasis resulting in progression to cholangiocarcinoma (CCA). This cross-sectional study in a prospective cohort study aimed to examine the prevalence and co-infection of intestinal parasites among Northeastern Thai rural residents, recruited into the Khon Kaen Cohort Study (KKCS), and who were residing in areas of high-risk for developing CCA. On recruitment, subjects had completed questionnaires and provided fecal samples for IPI testing using the formalin ethyl acetate concentration technique. Data on selected general characteristics and the results of the fecal tests were analysed. IPI test results were available for 18,900 of cohort subjects, and 38.50% were found to be positive for one or more types of intestinal parasite. The prevalence of Opisthorchis viverrini (O. viverrini) infection was the highest (45.7%), followed by intestinal flukes (31.9%), intestinal nematodes (17.7%), intestinal protozoa (3.02%), and intestinal cestodes (1.69%). The pattern of different infections was similar in all age groups. According to a mapping analysis, a higher CCA burden was correlated with a higher prevalence of O. viverrini and intestinal flukes and a greater intensity of O. viverrini. Both prevention and control programs against liver fluke and other intestinal parasites are needed and should be delivered simultaneously. We can anticipate that the design of future control and prevention programmes will accommodate a more community-orientated and participatory approach.

Generation of Highly Expandable Intestinal Spheroids Composed of Stem Cells

  • Many of early findings regarding intestinal stem cells (ISCs) and their niche in the human intestine have relied on colorectal cancer cell lines and labor-intensive and time-consuming mouse models. However, these models cannot accurately recapitulate the physiologically relevant aspects of human ISCs. In this study, we demonstrate a reliable and robust culture method for 3D expanding intestinal spheroids (InSexp) mainly comprising ISCs and progenitors, which can be derived from 3D human intestinal organoids (HIOs). We did functional chararcterization of InSexp derived from 3D HIOs, differentiated from human pluripotent stem cells, and optimization culture methods. Our results indicate that InSexp can be rapidly expanded and easily passaged, and show enhanced growth rates via WNT pathway activation. InSexp are capable of exponential cell expansion and cryopreservation. Furthermore, in vitro-matured HIO-derived InSexp proliferate faster than immature HIO-derived InSexp with preservation of the parental HIO characteristics. These findings may facilitate the development of scalable culture systems for the long-term maintenance of human ISCs and provide an alternative platform for studying ISC biology.

Preparation and Mucoadhesive Test of CSA-loaded Liposomes with Different Characteristics for the Intestinal Lymphatic Delivery

  • Drug delivery to the lymphatic system may be important in terms of the treatment with lymphatic involvement, such as tumor metastases and immunization. Especially, drug transport via the intestinal lymphatics after oral administration has been attracted lots of interests. The purpose of this study was to prepare cyclosporin A (CSA)-loaded liposomes, with different characteristics, and evaluate their mucoadhesivity. Three liposome preparations were formulated: cationic stearylamine liposomes (SA-Lip), anionic phosphatidylserine liposomes (PS-Lip), Polymer (chitosan)-coated liposomes (CS-Lip), and characterized. The liposome preparations were found to be spherical in shape, with PS-Lip being the smallest. The liposome preparations exhibited entrapment efficiencies in the order: PS-Lip $(52.5{\pm}2.9%)$ > SA-Lip $(48.8{\pm}3.3%)$ > CS-Lip $(41.7{\pm}4.2%)$. Finally, mucoadhesive tests were carried out using rat intestine, with SA-Lip (67%) showing the best adhesive rate of the three preparations (PS-Lip: 56%, CS-Lip: 61%). These results suggest that a positive charge on the surface of drug carriers may be an important factor for the intestinal drug delivery.

Effect of Laparoscopic Nerve-sparing Radical Hysterectomy on Bladder Function, Intestinal Function Recovery and Quality of Sexual Life in Patients with Cervical Carcinoma

  • Objective: To investigate bladder and intestinal function recovery and quality of sexual life after laparoscopic nerve-sparing radical hysterectomy (LNRH) for treatment of early invasive cervical carcinoma. Methods: Subjects included patients who underwent radical hysterectomy by laparotomy who were randomly assigned to 2 groups: 30 patients who underwent LNRH and 35 classical laparoscopic radical hysterectomy (LRH). We assessed the patients general clinical information, surgical characteristics, pathological findings, and adjuvant therapies. A urodynamic study was used to assess bladder function. Intestinal function recovery and quality of sexual life were evaluated by questionnaire. Results: No significant differences were found in age, surgery characteristics, pathological findings, adjuvant therapies, and main adverse effects between the 2 groups. The mean duration of the postoperative catheterization (DPC) in group LNRH was shorter than that in group LRH (P < 0.001). The maximum flow rate, maximum cystometric capacity, maximum detrusor pressure and urinary complications in group LNRH were better than those in group LRH. The quality of sexual life evaluated according to the female sexual function index (FSFI) was better in group LNRH than in those who underwent LRH. The intestinal function of patients in group LNRH also recovered better compared with patients in group LRH.

  • This study was conducted to investigate the biochemical characteristics, the population and antibiotic susceptibility test of Clostridium perfringens isolated from intestinal contents of slaughter cattle in Kyung-ju and Po-hang. 1. In slaughter cattle Cl perfringens were isolated from intestinal contents of 51 of 101 cases(50.4%) and the population were $\leq$$10^5$cfu/ml of 44 cases(86.3%). 2. In antibiotic susceptibility test, ampicillin, bacitracin, cephalothin, penicillin polymyxin B were highly susceptible, ohloramphenicol, erythromycin, tetracycline were lowly susceptible, gentamicin, kanamycin, amikacin, streptomycin, sulfamethoxine, sulfamethazine were resistant. 3. In leaving test intestinal contents leaved for 0, 4, 8, 16, 32 hours in room temperature and population of Cl perfringens were gradually increased.

Effect of the supplementation of pig skin collagen on growth performance, organ weight, blood characteristics and intestinal microbiota in broilers

  • This experiment was conducted to investigate the effects of pig skin collagen supplementation on growth performance, organ weight, blood characteristics, and intestinal microbiota in broilers. A total of 50 Ross 308 broilers were used for 2 weeks. The five dietary treatments were as follows: NC) basal diet, PC) NC + fish collagen powder 0.1%, T1) NC + pig skin collagen 0.1%, T2) NC + pig skin collagen 0.5%, and T3) NC + pig skin collagen 1.0%. The body weight gain (BWG), feed intake (FI), and feed conversion ratio (FCR) were not affected (p > 0.05) by the dietary treatments in this experiment. Additionally, there were no significant differences (p > 0.05) in the organ weights among the treatments. Broilers fed T1, T2 and T3 diets had higher (p < 0.05) white blood cell (WBC) counts than the broilers fed the NC and PC diets. The Lactobacillus counts in the excreta were improved (p < 0.05) in the broilers fed the T1 and T2 diets. Moreover, the Salmonella counts in the excreta were decreased (p < 0.05) in the broilers fed the PC and T1 diets. In conclusion, supplementation of pig skin collagen in diets improved the white blood cells (WBCs) in the blood and Lactobacillus counts in the excreta, and reduced the Salmonella counts in the excreta. However, when pig skin collagen was increased in the diets, there were no significant differences (p > 0.05). Therefore, the addition of 0.1% pig skin collagen in the feed provided beneficial effects on the blood characteristics and the intestinal microbiota environment.

Estimation of Ruminal Degradation and Intestinal Availability of Crude Protein in the Animal-Origin Feedstuffs Using Mobile Nylon Bag Technique

  • Ruminal degradation characteristics and intestinal availability of crude protein (CP) in four animal-origin feeds (fish meal, meat meal, viscera meal, feather meal) were estimated by mobile nylon bag technique. Three ruminally and duodenally cannulated Holstein dairy cows (average body wt. 550kg) fed a diet containing 40% concentrate and 60% orchard grass hay on a dry matter (DM) basis. Assuming that the outflow rate of diet in rumen is 5% per hour (k =0.05), contents of quickly degradable CP (QDP), slowly degradable CP (SDP), and undegradable CP (UDP) in the rumen were 27.6%, 9.4%, 63.0% for fish meal, 34.3% 28.1%, 37,6% for meat meal, 43.9%, 12.5%, 43.6% for viscera meal, and 14.4%, 15.8%, 69.8% for feather meal, respectively. Intestinal CP degradability was 51.0% for fish meal, 27.2% for meat meal, 37.9% for viscera meal and 56.2% for feather meal. Available UDP in the intestinal tract was contained 288 g, 217 g, 246 g and 423 g per kilogram DM of diet in fish meal, meat meal, viscera meal and feather meal, respectively.

  • Lactic acid bacteria (LAB) Play an important role in the human diet and are used in the production of edible fermented products such as kimchi and yoghurt. LAB are regarded as safe food additives used to enhance the nutritive value of foods. Plant-origin lactic acid bacteria (PLAB) cultured in vegetal media are now widely used in food industries. PLAB have been found to activate intestinal immunity, modulate the balance of the intestinal bacterial from, and enhance intestinal function. They are known for their strong resistance to acid; this enables them to persist for a longer duration in the human intestine. PLAB can also survive in the intestinal environment under conditions of poor nutrition. They have stronger vitality as compared to LAB of animal origin. Due to the unique characteristics of PLAB, they are being widely used in Japan for processing foods such as yoghurt and beverages. Recently, PLAB has also been used as the culture for processing yoghurt in Korea. We expect further research on the functional effects of PLAB.
