Prionchulus oleksandri (Nematoda: Mononchida) from Korea

  • The genus Prionchulus Cobb, 1916 represents a group of predaceous nematodes belonging to the family Mononchidae Chitwood, 1937, and is found worldwide. However, only five species have been reported thus far from Korea. Prionchulus oleksandri Winiszewska and Susulovsky, 2003 is reported for the first time from Korea, from sediments collected from the Nakdong River. This species is distinguished from other Prionchulus species by its truncated lip region with small cephalic papillae and refringens vaginae. In this study, morphological characters(detailed morphometrics) of P. oleksandri are described and illustrated using optical microscopy. DNA barcode sequence information (the D2-D3 region of 28S rDNA, 18S rDNA, and internal transcribed spacer rDNA) is also provided for the molecular identification of the species.

Molecular and Morphological Confirmation of Three Undescribed Species of Mortierella from Korea

  • Three fungal isolates designated as CNUFC-YR329-1, CNUFC-PTS103-1, and CNUFC-PTS2-1 were discovered during a survey of fungal diversity of the order Mortierellales from freshwater and pine tree rhizosphere soil samples in Korea. The strains were analyzed morphologically and phylogenetically based on the internal transcribed spacer (ITS) and large subunit (LSU) of ribosomal DNA gene sequences. Based on their morphology and phylogeny, the three isolates were identified as Mortierella elongata, M. horticola, and M. humilis, respectively. To the best of our knowledge, M. elongata, M. horticola, and M. humilis, belonging to an undiscovered taxon, have not been previously described in Korea.

Pestalotiopsis kaki sp. nov., a Novel Species Isolated from Persimmon Tree (Diospyros kaki) Bark in Korea

  • During the screening of Korean microflora, a fungal strain (KNU-PT-1804) belonging to the genus Pestalotiopsis was isolated from persimmon tree (Diospyros kaki) bark collected from North Gyeongsang Province, Korea. The strain, KNU-PT-1804, produced smaller conidia compared with related species P. kenyana, P. neglecta, and P. telopeae. The novelty of the strain was confirmed based on phylogenetic analysis using molecular datasets of internal transcribed spacer (ITS) regions, β-tubulin (TUB2), and translation elongation factor 1-alpha (TEF1α) genes. Molecular phylogeny strongly supports that the strain is distinct from previously known Pestalotiopsis species, and we proposed the novel species, Pestalotiopsis kaki sp. nov., and provide a detailed description and illustration.

Report on a New Truffle Species, Tuber koreanum sp. nov., from Korea

  • The truffle and ectomycorrhizal roots formed by Tuber sp. were collected from the rhizosphere of Quercus aliena in Korea. The morphological characteristics of the ascoma, and molecular phylogenetic analysis using sequences from the internal transcribed spacer (ITS) and large subunit (LSU) of ribosomal DNA, translation elongation factor 1-alpha (TEF), and RNA polymerase second largest subunit (RPB2) regions confirmed the distinct morphology of the truffle. This truffle belongs to a monophyletic clade among the other Tuber species in the phylogeny. This study describes the truffle, Tuber koreanum, as a new species reported from Korea.

Puccinia klugkistiana, a Rust Fungus Occurring on Cleistogenes hackelii in Korea

  • Rust samples at both uredinial and telial stages were collected from Cleistogenes hackelii (Poaceae) in 2017 and 2019 in Korea. Based on the morphological examination and molecular phylogenetic analyses of the internal transcribed spacer (ITS) and large subunit (LSU) rDNA, it was identified as Puccinia klugkistiana. This rust at aecial stage has previously been recorded on Ligustrum japonicum (Oleaceae) in Korea. Puccinia diplachnicola has been previously recorded on C. hackelii in Korea; however, it is believed to be unrelated to the rust collected in the present study. Conclusively, this is the first study to report P. klugkistiana occurrence on C. hackelii in Korea.

Re-Examination of Several Elsinoë Species Reported from Japan

  • Elsinoë are plant pathogenic fungi that cause scabs, spotted anthracnose, and some morphological distortions on various plants, including woody plants, economically important crops, and ornamental plants. Taxonomical reexamination of Elsinoë species in Japan has not yet been conducted based on the modern species criteria. In this study, several Japanese isolates were reexamine based on the morphological and molecular-phylogenetic analysis of the internal transcribed spacer region (ITS), large subunit gene (LSU)m and protein-coding gene such as RNA polymerase II subunit (rpb2) and Translation elongation factor 1-alpha (tef). Japanese isolates were divided into four clades and three new species, Elsinoë hydrangeae, E. sumire, and E. tanashiensis were proposed. One species, Sphaceloma akebiae, was transferred to the genus Elsinoë.

Erysiphe magnoliicola Comprises the Powdery Mildew Found on Magnolia kobus in Korea

  • The powdery mildew found on Magnolia kobus was recorded as Microsphaera alni for the first time in Korea in 1975. After splitting M. alni into several distinct species, this mildew was regarded as Microsphaera magnifica, now Erysiphe magnifica. Since E. magnifica is known to be a North American species, the powdery mildew found on M. kobus in Korea was studied to clarify its identity. Based on morphological characteristics and sequencing results of the internal transcribed spacer and large subunit rDNA gene, the powdery mildew found on M. kobus in Korea was identified as Erysiphe magnoliicola.

Apiospora pseudosinensis (Apiosporaceae): an Unrecorded Endophytic Fungus Isolated from Pseudosasa japonica

  • Bamboo is host to diverse range of fungi, however, research on bambusicolous fungi remains insignificant in Korea. The genus Apiospora is prevalent in various environments, particularly as endophytic fungi in Poaceae, to which bamboo belongs. In this study, endophytic fungi were isolated from stems of Pseudosasa japonica collected from Gongjusi, Korea. To identify these endophytic fungi, we observed morphological characteristics and conducted phylogenetic analysis using internal transcribed spacer (ITS), large subunit ribosomal DNA (LSU), and β-tubulin (TUB2) DNA, identifying and describing an unrecorded fungus, Apiospora pseudosinensis.

Acrophialophora ellipsoidea, an Undescribed Species Isolated from Soil in Korea

  • A designated fungal isolate, KNU-US-1802E was isolated from the soil in Uiseong, Korea. To identify characteristics of the isolate, it was cultured on PDA media for 6 days at $35^{\circ}C$. Colonies on PDA are flat, light gray, dense, with entire margins; reverse dark gray to black, with white margins. Aerial mycelia were smooth-walled, hyaline and 40~42 mm diameter after 6 days at $35^{\circ}C$. Conidia were hyaline, one-celled, ellipsoidal to fusiform, forming long chains with average length ${\times}$ width of $5.0{\pm}0.3{\times}2.9{\pm}0.2{\mu}m$. Molecular analysis indicates that the internal transcribed spacer (ITS) region and partial beta-tubulin (tub2) gene sequence showed 100% and 99% similarities, respectively with Acrophialophora ellipsoidea CGMCC 3.15255 collected from China. Phylogenetic analysis by the neighbor-joining (NJ) method shows that the KNU-US-1802E was clustered with A. ellipsoidea CGMCC 3.15255 in a phylogenetic tree constructed using the concatenated sequences of ITS region and tub2 gene sequences with a high bootstrap value. Based on these findings, the isolate KNU-US-1802E was identified as Acrophialophora ellipsoidea, and this is the first report of this isolate in Korea.

분자생물학적 자료에 의한 한국산 개발나물속의 분류학적 고찰 (Taxonomic Review of the Umbelliferous genus Sium L. in Korea: Inferences based on Molecular Data)

  • 핵 리보좀 DNA ITS 구간의 염기서열 정보를 이용하여 한국산 산형과 개발나물속의 분자 계통학적 연구를 수행하였다. ITS 계통수는 한반도 고유종인 대암개발나물(S. heterophyllum)이 한반도에 생육이 보고된바 없는 러시아 우수리지역의 고유종인 S. tenue와 동일한 종임을 밝혔다. ITS 1과 ITS 2 구간의 염기서열 비교시 S. tenue와 인천 문수산에 생육하고 있는 대암개발나물이 100% 일치함을 확인할 수 있었다. 또한 ITS 구간의 염기서열은 최근 신종으로 발표된 세잎개발나물(S. ternifolium)이 개발나물속의 다른 분류군과 뚜렷하게 구별됨을 지지하고 있었다. 세잎개발나물은 유연관계가 가장 가까운 일본 고유종 S. serra의 ITS 염기서열 비교시 1.4와 1.6%의 차이점을 가지고 있음이 확인되어 종으로서의 처리가 적합함을 확인할 수 있었다.
