A New Record of Penicillium antarcticum from Marine Environments in Korea

  • During a survey of marine fungi from the waters surrounding Jeju Island, Korea, several Penicillium strains were isolated from seawater and marine sponges. Based on morphological characteristics and phylogenetic analyses of the internal transcribed spacer and RNA polymerase subunit II, four strains were identified as Penicillium antarcticum, a fungus that, to the best of our knowledge, had not been previously reported in Korea. Here, we provide detailed descriptions of the morphological characteristics and extracellular enzyme activities of the four strains.

Selection of Lecanicillium Strains with High Virulence against Developmental Stages of Bemisia tabaci

  • Selection of fungal strains with high virulence against the developmental stages of Bemisia tabaci was performed using internal transcribed spacer regions. The growth rate of hyphae was measured and bioassay of each developmental stage of B. tabaci was conducted for seven days. All of the fungal strains tested were identified as Lecanicillium spp., with strain 4078 showing the fastest mycelium growth rate (colony diameter, 16.3 $\pm$ 0.9 mm) among the strains. Compared to strain 4075, which showed the slowest growth rate, the growth rate of strain 4078 was increased almost 2-fold after seven days. Strains 4078 and Btab01 were most virulent against the egg and larva stages, respectively. The virulence of fungal strains against the adult stage was high, except for strains 41185 and 3387. Based on the growth rate of mycelium and level of virulence, strains 4078 and Btab01 were selected as the best fungal strains for application to B. tabaci, regardless of developmental stage.

Aphanoascus fulvescens: A New Record from Crop Field Soil in Korea

  • Aphanoascus fulvescens KNU16-343 was isolated from crop field soil and identified based on morphological and molecular characteristics. The shape and size of conidia and conidiophores, as well as the internal transcribed spacer region of rDNA, confirmed that the isolate was A. fulvescens. This is the first report of this fungal species in Korea.

Secondary Structure and Phylogenetic Implications of ITS2 in the Genus Tricholoma

  • The internal transcribed spacer (ITS) region in the genus Tricholoma was analyzed, including for its primary nucleotide sequence and secondary structural characterization. The secondary structures of the ITS2 region in the genus Tricholoma were identified for use in bioinformatic processes to study molecular evolution and compare secondary structures. Ten newly sequenced ITS regions were added to the analysis and submitted to the GenBank database. The resulting structure from a minimum energy algorithm indicated the four-domain model, as previously suggested by others. The conserved secondary structure of the ITS2 sequences of the genus Tricholoma exhibited certain unique features, including pyrimidine tracts in the loops of domain A and a complete structure containing four domains, with motifs identified in other ITS2 secondary structures. A phylogenetic tree was derived from sequence alignment based on the secondary structures. From the resulting maximum parsimonious tree, it was found that the species in the genus Tricholoma had evolved monophyletically and were composed of four groups, as supported by the bootstrapping values and pileus color.

Age-dependent Distribution of Fungal Endophytes in Panax ginseng Roots Cultivated in Korea

  • Fungal endophytes were isolated from 1-, 2-, 3-, and 4-year-old ginseng roots (Panax ginseng Meyer) cultivated in Korea. The isolated fungal endophytes were identified based on sequence analysis of the internal transcribed spacer and morphological characterization by microscopic observations. A total of 81 fungal endophytes were isolated from 24 ginseng roots. Fungal endophytes were classified into 9 different fungal species and 2 unknown species. Ginseng roots that were 1-, 2-, 3-, and 4-years old were colonized by 2, 6, 8, and 5 species of fungal endophytes, respectively. While Phoma radicina was the most frequent fungal endophyte in 2-, 3-, and 4-year-old ginseng roots, Fusarium solani was the dominant endophyte in 1-year-old ginseng roots. The colonization frequencies (CF) varied with the host age. The CF were 12%, 40%, 31%, and 40% for 1-, 2-, 3-, and 4-year-old ginseng roots, respectively. We found a variety of fungal endophytes that were distributed depending on the age of ginseng plants.

Phylogenetic and Morphological Identification of the Novel Pathogen of Rheum palmatum Leaf Spot in Gansu, China

  • A new leaf spot disease was observed on leaves of Rheum palmatum (Chinese rhubarb) in Northwest China (Gansu Province) starting in 2005. A Septoria-like fungus was isolated and completion of Koch's postulates confirmed that the fungus was the casual agent of the leaf spot disease. Morphology and molecular methods were combined to identify the pathogen. The fungus produced conidiomata pycnidia and the conidia were 2~5 septate, $61.2{\sim}134.1{\mu}m$ in length and $3.53{\sim}5.3{\mu}m$ in width, which is much larger than the known Spetoria species that infects Polygonaceae species. Phylogenic analysis of the internal transcribed spacer region confirmed that this Septoria-like fungus is within the Septoria genus but distinct from known Septoria species. Together, these morphological and phylogenetic data support that the R. palmatum infecting Septoria strain is a newly-described plant pathogenic species.

Blastobotrys illinoisensis, an Unrecorded Anamorphic Yeast Strain Isolated from the Gut of the Earthworm Eisenia fetida

  • Strain E4, an unrecorded species of dimorphic fungi, was isolated from the gut of earthworms collected in Gyeonggi Province, South Korea. Nucleotide sequence analysis of the D1/D2 region of the large subunit (LSU) rRNA gene and the internal transcribed spacer (ITS) region revealed that this species is a member of the genus Blastobotrys, Blastobotrys illinoisensis. Strain E4 differed from its closest known species, B. mokoenaii and B. malaysiensis, by harboring 3-5 and 12-14 nucleotide substitutions in the D1/D2 and ITS regions, respectively. Phylogenetic analysis based on concatenated sequences of the D1/D2 region of the LSU rRNA gene and the ITS region also indicated that strain E4 belongs to the Blastobotrys clade and is distinct from other related species in the clade. The previously unreported isolate could be distinguished from closely related species by its inability to ferment carbon sources. To our knowledge, this is the first report on the isolation of Blastobotrys species from the gut of earthworms in Korea. The strain used was E4 (=KCTC 27831=JCM 33428).
