홍도고들빼기의 분류학적 실체 (Taxonomic identity of Crepidiastrum ×nakaii recorded on Hongdo Island)

  • 고들빼기속에 속하는 홍도산 홍도고들빼기의 학명은 이고들빼기와 절영풀의 잡종인 Crepidiastrum ×nakaii로 기록되어 왔다. 홍도 식물상을 조사하는 과정에서 홍도고들빼기의 분류학적 실체를 재검토하였다. 이를 위해 홍도고들빼기의 형태적 특징과 지리적 분포 및 internal transcribed spacer (ITS) 염기서열을 고들빼기속 근연종들과 비교하였다. 형태적 특성 조사에서, 홍도고들빼기는 짧고 굵은 아관목성 줄기와 8개의 내총포편 및 10 내지 12개의 소화수를 갖는 점에서 갯고들빼기와 유사한 반면, 개화기에 시드는 뿌리잎과 막질로 예 거치인 줄기잎 및 부리가 있는 수과 형태 등에서는 이고들빼기와 유사하였다. 한편 이고들빼기와 갯고들빼기는 홍도와 인근 섬 지역에 자라지만, 절영풀은 분포하지 않는다. 이러한 형태 및 분포 조사결과, 홍도고들빼기의 학명은 이고들빼기와 절영풀의 잡종인 C. ×nakaii가 아니라, 이고들빼기와 갯고들빼기의 잡종인 C. ×muratagenii로 판단된다. 하지만, ITS 염기서열은 조사된 종들을 구별할 수 있는 해상력을 보여주지 못하였다.

국내 침입외래식물 사막갓(Brassica tournefortii; Brassicaceae)의 보고 및 잠재 분포 예측 (New record and prediction of the potential distribution of the invasive alien species Brassica tournefortii (Brassicaceae) in Korea)

  • 침입외래식물인 Brassica tournefortii Gouan이 전북 군산시, 고창군, 제주도 제주시에서 발견되었다. 본 종은 식물체 기부에 강모가 밀생하고, 줄기가 기부와 말단에서 분지하며, 열매는 성숙시 일부분만 열개되고, 종자는 수분이 있을 때 점성이 띠는 특징으로 배추속의 갓, 유채와 쉽게 구분된다. 일부 학자들은 B. tournefortii의 열매와 종자의 특징으로 Coincya Rouy 속에 속하는 분류군으로 취급하기도 하나, 열매의 과피편 표면에 한개의 맥이 있는 특징과, ITS 구간 염기서열의 maximum-likelihood 분석 결과에 따라 배추속 식물로 분류하였다. 본 연구에서 B. tournefortii의 분포지, 화상자료 및 기재문을 제시하였으며, 또한 본 종의 잠재분포변화가 기후 시나리오에 의해 예측되었다. 예측 결과, B. tournefortii의 국내 확산 가능성은 낮은 것으로 나타났지만, 정확한 판단을 하기 위해서는 지속적인 모니터링이 필요할 것으로 판단된다. 본 연구의 결과들은 B. tournefortii의 위험성 평가 및 효과적인 관리방안을 제시하기 위한 자료로 활용될 수 있을 것이다.

Identification of Genes Suitable for DNA Barcoding of Morphologically Indistinguishable Korean Halichondriidae Sponges

  • The development of suitable genetic markers would be useful for defining species and delineating the species boundaries of morphologically indistinguishable sponges. In this study, genetic variation in the sequences of nuclear rDNA and the mitochondrial cytochrome c oxidase subunit 1 and 3 (CO1 and CO3) regions were compared in morphologically indistinguishable Korean Halichondriidae sponges in order to determine the most suitable species-specific molecular marker region. The maximal congeneric nucleotide divergences of Halichondriidae sponges in CO1 and CO3 are similar to those found among anthozoan cnidarians, but they are 2- to 8-fold lower than those found among genera of other triploblastic metazoans. Ribosomal internal transcribed spacer regions (ITS: ITS1 + ITS2) showed higher congeneric variation (17.28% in ITS1 and 10.29% in ITS2) than those of CO1 and CO3. Use of the guidelines for species thresholds suggested in the recent literature indicates that the mtDNA regions are not appropriate for use as species-specific DNA markers for the Halichondriidae sponges, whereas the rDNA ITS regions are suitable because ITS exhibits a low level of intraspecific variation and a relatively high level of interspecific variation. In addition, to test the reliability of the ITS regions for identifying Halichondriidae sponges by PCR, a species-specific multiplex PCR primer set was developed.

울릉도의 자생란과 공생하는 난균근균의 분자생물학적 동정 (Molecular Identification of Orchid Mycorrhizal Fungi of Native Orchids in Ulleung Island)

  • 울릉도에서 채집된 6종의 지생란의 뿌리에서 난균근균을 확인하였다. 난균근균의 감염이 확인되었다. 각 종의 뿌리로 부터 분리된 난균근균이 분자적인 분석을 통하여 확인되였다. 뿌리로부터 분리된 균에서 DNA를 추출하여, 담자균류에 특이적인 프라이머인 ITS1-OF와 ITS4-OF를 사용하여 ITS 지역을 증폭한 후 염기서열을 분석하여 두 식물의 뿌리에 공생하는 난균근균을 동정하였다. 분자적인 분석을 통해 Tulasnellaceae 와 Ceratobasidaceae에 속하는 난균근균이 확인되었다.

Mariannaea samuelsii Isolated from a Bark Beetle-Infested Elm Tree in Korea

  • During an investigation of fungi from an elm tree infested with bark beetles in Korea, one isolate, DUCC401, was isolated from elm wood. Based on morphological characteristics and phylogenetic analysis of the internal transcribed spacer and 28S rDNA (large subunit) sequences, the isolate, DUCC401, was identified as Mariannaea samuelsii. Mycelia of the fungus grew faster on malt extract agar than on potato dextrose agar and oatmeal agar media. Temperature and pH for optimal growth of fungal mycelia were 25oC and pH 7.0, respectively. The fungus demonstrated the capacity to degrade cellobiose, starch, and xylan. This is the first report on isolation of Mariannaea samuelsii in Korea.

Characterization of Myrothecium roridum Isolated from Imported Anthurium Plant Culture Medium

  • During an investigation of microorganisms and pests in plant culture media from imported anthurium pots, a fungal isolate (DUCC4002) was detected. Based on its morphological characters including colony shape on potato dextrose agar, the microstructures of spores observed by light and scanning electron microscopy and the results of phylogenetic analysis using an internal transcribed spacer rDNA sequence, the fungal isolate was identified as Myrothecium roridum. Pathogenicity testing on anthurium leaves revealed that the fungus could colonize and produce sporodochia on the inoculated leaves. This is the first report of M. roridum detected in imported plant culture medium in Korea.

Spot Anthracnose Disease Caused by $Colletotrichum$ $gloeosporioides$ on Tulip Tree in Korea

  • The tulip tree ($Liriodendron$ $chinense$) has been widely cultivated in Korea as a street or garden tree for its large flowers, which have a superficial resemblance to tulips. Occurrence of anthracnose disease on the leaves of tulip trees growing on the campus of Gyeongsang National University, Jinju, Korea, has been observed. Based on mycological characteristics, pathogenicity, and internal transcribed spacer sequence, the causal fungus was identified as $Colletotrichum$ $gloeosporioides$. This is the first report on anthracnose disease caused by $C.$ $gloeosporioides$ on tulip trees in Korea.

Monostroma alittorale, a marine green algal species newly recorded in Korea

  • A marine green algal species (Chlorophyta) was collected from the eastern coast of Korea. It is morphologically characterized by monostromatic thallus, usually undulate and entire margins, cap-like chloroplast and several pyrenoids per cell. In a phylogenetic tree based on molecular data, the Korean alga nests in the same clade as Monostroma alittorale originally described from Japan, as a sister clade of M. grevillei from France. The genetic distance for ITS(Internal Transcribed Spacer) sequences among Monostroma species ranges from 2.3% to 38.2%. The value between the Korean entity and M. alittorale was calculated as 0.01%, considered to be intraspecific divergence. This Korean entity is identified as Monostroma alittorale based on morphological and molecular analyses. This is the first record of M. alittorale in Korea.

New record of Ulva sublittoralis (Ulvales, Chlorophyta) in Korea

  • A marine ulvalean species (Chlorophyta) was collected from the eastern coast of Korea. This species is morphologically characterized by a distromatic, dark to medium green and mostly irregularly orbicular or irregularly expanded thallus with entire or undulate margin without serrations. Vegetative cells are irregularly polygonal with distinctly rounded corners in shape, and have chloroplast completely covering the outer cell wall and one to two pyrenoids per cell. In a phylogenetic tree based on ITS (Internal Transcribed Spacer) sequences, this Korean alga nests in the same clade with Ulva sublittoralis, as a sister clade of U. californica, U. flexuosa and U. tanneri, which share the irregularly orbicular or expanded thallus normally without teeth cells. The genetic divergence between them is intraspecific within Ulva. Accordingly, it is identified as U. sublittoralis based on the morphological and molecular data. This is the first record of Ulva sublittoralis in the Korean marine algal flora.

A Novel Production Method for High-Fructose Glucose Syrup from Sucrose-Containing Biomass by a Newly Isolated Strain of Osmotolerant Meyerozyma guilliermondii

  • One osmotolerant strain from among 44 yeast isolates was selected based on its growth abilities in media containing high concentrations of sucrose. This selected strain, named SK-ENNY, was identified as Meyerozyma guilliermondii by sequencing the internal transcribed spacer regions and partial D1/D2 large-subunit domains of the 26S ribosomal RNA. SK-ENNY was utilized to produce high-fructose glucose syrup (HFGS) from sucrose-containing biomass. Conversion rates to HFGS from 310-610 g/l of pure sucrose and from 75-310 g/l of sugar beet molasses were 73.5-94.1% and 76.2-91.1%, respectively. In the syrups produced, fructose yields were 89.4-100% and 96.5-100% and glucose yields were 57.6-82.5% and 55.3-79.5% of the theoretical values for pure sucrose and molasses sugars, respectively. This is the first report of employing M. guilliermondii for production of HFGS from sucrose-containing biomass.
