본 연구에서는 산양유 발효유(F-GM)의 급여가 면역 활성과 체력증진에 미치는 영향을 조사하였다. F-GM을 경구 투여한 마우스의 비장세포는 림프구 유사분열 촉진인자인 ConA와 LPS에 대한 증식반응의 상승을 나타내었다. ConA를 처리한 비장세포로부터 분비되는 cytokine을 ELISA법에 의해 정량한 결과 F-GM 투여에 의해 IL-2와 IFN-${\gamma}$의 생성은 유의하게 증가하였으나, IL-4 및 IL-10의 생성은 별다른 변화를 보이지 않았다. 또한 RT-PCR을 이용한 이들 cytokine mRNA의 정량실험에서도 F-GM 투여로 IL-2와 IFN-${\gamma}$의 mRNA가 증가하는 것을 확인하였다. 한편 KLH(20 mg/mouse)를 정상 대조군 마우스 혹은 F-GM을 급이한 마우스에 각각 면역하고 5주째에 KLH에 대한 항체가를 측정한 결과 F-GM을 급이한 마우스에서 항체가가 유의하게 상승하는 것으로 나타났다. KLH에 대한 항체의 isotype을 조사한 실험에서는 KLH에 대한 IgG1, IgG2a 그리고 IgM 특이항체의 상승이 관찰되었다. F-GM 경구 투여가 세포성 면역에 미치는 영향을 측정하기 위하여 면역개시 7주째의 마우스를 이용하여 KLH에 대한 지연형 과민반응(DTH)을 측정하였다. 그 결과 F-GM을 경구 투여한 마우스에서 DTH의 유의한 상승효과가 확인되었다. 한편 수영시험을 통해 F-GM의 경구 투여가 체력증강에 미치는 영향을 측정한 결과 F-GM을 투여한 마우스에서 매우 유의한 수영시간의 연장 효과가 관찰되었다. 이들 결과로부터 F-GM은 면역세포의 활성화를 통한 면역력 강화는 물론 체력을 증가시키는 생리활성을 갖는 것으로 확인되었다.
Genetic polymorphisms associated with aggressive periodontitis have previously been reported. Interleukin-10 is an immunoregulatory cytokine that plays a role in the pathogenesis of periodontitis. Individual capacity for IL-10 production appears to be under genetic influence, The aim of present investigation was to explore possible genetic association of IL-10 gene promoter polymorphisms with generalized aggressive periodontitis. The study population consisted of 37 generalized aggressive periodontitis patients from the Department of Periodontology, Chonnam National University Hospital and 27 control subjects, all the subjects were non-smokers, Genomic DNA was obtained from buccal swab. The IL-10promoter -597, -824, -1082 positions were genotyped by amplifying the polymorphic regions using polymerase chain reaction (PCR) , followed by restriction enzyme digestion and gel electrophoresis. IL-10-597 C (allele 1) to A (allele 2) and IL-10-824 C (allele 1) to T (allele 2) and IL-10-1082 G (allele 1) to A (allele 2) polymorphisms were examined. The results were as follows. 1. In patients, the distribution of genotypes C/C, C/A and NA at Il-10-597 was determined to be 13.5%, 37.8% and 48.7%, respectively and the distribution of genotypes at IL-10-824 was the same as that of IL-10-597. The distribution of genotypes G/G, G/A and NA at IL-10-1082 was found to be 2.7%, 16.2% and 81. 4%, respectively. No statistical difference in genotype distribution was found between the patient and control groups. 2. Allele 2 carriage rate at the three position of the IL-10 promoter region was higher in the control group than the patient group. 3. Allele 2 frequencies at IL-10-597 and -824 positions were higher in female group than male group and its difference was statistically significant(p<0.05). No significant difference in genotype distribution between the control and patient groups. Allele frequency between control and patient groups was not significantly different although allele 2 frequency at the three positions in the IL-10 promoter region appeared to be higher in control group. In conclusion, no clear association between IL-10 gene promoter polymorphisms and generalized aggressive periodontitis in Korean was observed.

Purpose: Beyond the limited scope of non-specific polyclonal regulatory T cell (Treg)-based immunotherapy, which depends largely on serendipity, the present study explored a target Treg subset appropriate for the delivery of a novel epitope spreader Pep19 antigen as part of a sophisticated form of immunotherapy with defined antigen specificity that induces immune tolerance. Methods: Human polyclonal $CD4^+CD25^+CD127^{lo-}$ Tregs (127-Tregs) and $na\ddot{i}ve$$CD4^+CD25^+CD45RA^+$ Tregs (45RA-Tregs) were isolated and were stimulated with target peptide 19 (Pep19)-pulsed dendritic cells in a tolerogenic milieu followed by ex vivo expansion. Low-dose interleukin-2 (IL-2) and rapamycin were added to selectively exclude the outgrowth of contaminating effector T cells (Teffs). The following parameters were investigated in the expanded antigen-specific Tregs: the distinct expression of the immunosuppressive Treg marker Foxp3, epigenetic stability (demethylation in the Treg-specific demethylated region), the suppression of Teffs, expression of the homing receptors CD62L/CCR7, and CD95L-mediated apoptosis. The expanded Tregs were adoptively transferred into an $NOD/scid/IL-2R{\gamma}^{-/-}$ mouse model of collagen-induced arthritis. Results: Epitope-spreader Pep19 targeting by 45RA-Tregs led to an outstanding in vitro suppressive T cell fate characterized by robust ex vivo expansion, the salient expression of Foxp3, high epigenetic stability, enhanced T cell suppression, modest expression of CD62L/CCR7, and higher resistance to CD95L-mediated apoptosis. After adoptive transfer, the distinct fate of these T cells demonstrated a potent in vivo immunotherapeutic capability, as indicated by the complete elimination of footpad swelling, prolonged survival, minimal histopathological changes, and preferential localization of $CD4^+CD25^+$ Tregs at the articular joints in a mechanistic and orchestrated way. Conclusions: We propose human $na\ddot{i}ve$$CD4^+CD25^+CD45RA^+$ Tregs and the epitope spreader Pep19 as cellular and molecular targets for a novel antigen-specific Treg-based vaccination against collagen-induced arthritis.
본 연구는 꿀벌 꽃가루 열수 추출물(BPW)의 면역 활성에 관하여 알아보기 위하여, 선천면역과 적응면역에서 중심 역할을 수행하는 큰포식세포에 BPW를 처리하여 세포 생존율, NO 분비능, 사이토카인($TNF-{\alpha}$, IL-6, $IL-1{\beta}$) 분비능과 메커니즘 분석을 통한 신호전달에 관하여 관찰하였다. BPW를 큰포식세포에 처리하여 면역활성에 관하여 알아봤을 때, 큰포식세포 면역활성의 대표적인 바이오마커인 NO, 사이토카인의 분비능과 iNOS의 세포내 발현이 BPW 처리구에서 유의적으로 증가되는 것으로 관찰되었다. 또한 활성화된 탐식 세포의 세포 표면에서 발현되는 CD80과 CD86의 발현과 탐식세포의 항원제시에 밀접한 관련이 있는 주조직적합성 복합체(MHC class I과 II)의 발현이 BPW 처리구에서 유의적으로 증가되는 것으로 관찰되었다. 기전분석 결과, BPW의 처리는 MAPKs의 인산화와 $NF-{\kappa}B$의 핵 내 이동성을 증가시켜 면역활성을 증가시키는 것으로 관찰되었다. 따라서 BPW의 처리는 큰포식세포의 활성을 유도시켜 면역활성을 조절하는 것으로 관찰되었다.
국내에서 발생하는 돼지호흡기복합증후군(Porcine respiratory disease complex, PRDC)의 발생 상황 및 원인체의 검출 방법의 비교와 PRDC에서 cytokine의 발현변화 여부를 확인하기 위해서 481건의 시료에 대해 PRDC 발생상황을 조사하였다. 총 481건 중 단독감염은 113건(23.5%), 2종 이상의 원인체에 의한 발병이 348(72.3%)건으로 나타났다. PRDC 발생상황을 주령별로 분석한 결과, 총 348건에서 3주령에서 10주령미만의 돼지에서 258건(74.1%)으로 가장 많이 나타났다. PRDC의 주 원인체로 알려진 PRRSV, PCV2, SIV에 대한 감별진단을 위해 면역조직화학염색법(IHC)과 PCR에 의한 검출을 원인체 검출을 비교한 결과, 결과 PCR 방법이 IHC보다 원인균인 PRRSV, PCV-2의 검출에 효과적인 것으로 나타났으며 이러한 결과를 근거로 PRDC를 유발하는 원인체에 대해서는 임상적으로는 PCV-2는 감염되더라도 병리소견을 발현하지 않는 예가 있어 임상증상을 나타내는 PRDC의 주요 원인체는 PRRSV로 확인되었다. PRDC로 진단된 시료 중 2종 및 3종의 혼합감염군을 대상으로 폐와 림프절에서 cytokine의 발현의 변화를 조사한 결과에서는, IL-6을 제외한 조사된 모든 cytokine들이 2종 및 3종 복합 감염군에서 대조군에 비해 감소되었다.
Objectives: Selaginella bryopteris L. (family: Selaginaceae), is often used in traditional Indian systems of medicine for the prevention and cure of several disorders and for the treatment of patient with spermatorrhoea, venereal disease, constipation, colitis, urinary tract infections, fever, epilepsy, leucorrhoea, beri-beri and cancer. It is also used as a strength tonic. This study aimed to evaluate the mechanisms underlying the anti-inflammatory effects of topically administered aqueous, polar and non-polar methanolic fractions ($10mg/20{\mu}L$) of Selaginella bryopteris. Methods: An acute oral toxicity study of Selaginella bryopteris at doses from 250 to 2,000 mg/kg body weight (bw) was performed. Aqueous, polar and non-polar methanolic extracts ($10mg/20{\mu}L$) applied topically for 5 days were evaluated for their anti-inflammatory effects against 12-tetra-O-decanoyl phorbol acetate (TPA)- induced inflammation by using the redness in the ear, the ear's weight (edema), oxidative stress parameters, such as lipid-peroxide (LPO) and nitric oxide (NO), and the pro-inflammatory cytokines involved in inflammation, such as tumour necrosis factor $(TNF)-{\alpha}$, interleukin $(IL)-1{\beta}$ and IL-6. Indomethacine ($0.5mg/20{\mu}L$) was used for the positive control. Results: Selaginella bryopteris produced no mortalities when administered orally at doses from 250 to 2,000 mg/kg bw. Topical treatment with the non-polar methanolic fraction ($10mg/20{\mu}L$) significantly suppressed redness ($2.4{\pm}0.5$) and edema ($30.4{\pm}1$) and effectively reduced the LPO level ($32.3{\pm}3.3$). The NO level was ($8.07{\pm}0.55$), and the $TNF-{\alpha}$, $IL-1{\beta}$, and IL-6 levels were decreased to $69.6{\pm}15.5$, $7.7{\pm}4.8$ and $82.6{\pm}5.9$, respectively. Conclusion: This study demonstrated for the first time the mechanisms underlying the anti-inflammatory effect of medicinal plants like Selaginella bryopteris and quantified the pharmacological interactions between them. The present study showed this herbal product to be a promising anti-inflammatory phytomedicine for the treatment of patients with inflammatory skin diseases.
Objective : Oriental medicines have been combined oriental medical theory which based on the seven modes of emotions. Notopterygium incisum (N. incisum) and Clematis manshurica (C. manshurica) have been used as an anti-rheumatic and analgesic medicine for the treatment of rheumatism, headache, cold, etc. In this study, we evaluate the synergistic anti-inflammatory effect of N. incisum and C. manshurica. Method : To evaluate the synergistic anti-inflammatory effect of a herbal mixture N. incisum and C. manshurica, we examined the changed ear thickness in 12-O-tetradecanoyl-phorbol-13-acetate (TPA)-induced mouse ear edema model after topical application of herbal mixture. In addition, the levels of markers for inflammation, such as tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-$1{\beta}$, prostaglandin $G_2$ ($PGE_2$), and nitric oxide (NO) were measured by ELISA assay and Griess reagent in lipopolysaccharide-stimulated Raw 264.7 cells. Results : Our results showed that aqueous extracts of N. incisum and C. manshurica combination significantly inhibited the mouse ear edema induced by TPA. Moreover, the aqueous extracts of N. incisum and C. manshurica combination exhibited synergistic effects in down-regulating TNF-${\alpha}$, IL-$1{\beta}$, $PGE_2$ level, but not NO. Conclusions : This study suggested that combined treatment of N. incisum and C. manshurica, based on seven methods in prescription compatibility, has a synergistic effect in down-regulating inflammatory response both in vitro and in vivo models.
Obesity has been reported to be associated with low grade inflammatory status. In this study, we investigated the inflammatory response as well as associated signaling molecules in immune cells from diet-induced obese mice. Four-week-old C57BL mice were fed diets containing 5% fat (control) or 20% fat and 1% cholesterol (HFD) for 24 weeks. Splenocytes ($1{\times}10^7$ cells) were stimulated with $10\;{\mu}g/mL$ of lipopolysaccharide (LPS) for 6 or 24 hrs. Production of interleukin (IL)-$1{\beta}$, IL-6, and TNF-${\alpha}$ as well as protein expression levels of nucleotide-binding oligomerization domain (NOD)2, signal transducer and activator of transcription (STAT)3, and pSTAT3 were determined. Mice fed HFD gained significantly more body weight compared to mice fed control diet ($28.2{\pm}0.6$ g in HFD and $15.4{\pm}0.8$ g in control). After stimulation with LPS for 6 hrs, production of IL-$1{\beta}$ was significantly higher (P=0.001) and production of tumor necrosis factor (TNF)-${\alpha}$ tended to be higher (P < 0.064) in the HFD group. After 24 hrs of LPS stimulation, splenocytes from the HFD group produced significantly higher levels of IL-6 ($10.02{\pm}0.66$ ng/mL in HFD and $7.33{\pm}0.56$ ng/mL in control, P=0.005) and IL-$1{\beta}$ ($121.34{\pm}12.72$ pg/mL in HFD and $49.74{\pm}6.58$ pg/mL in control, P < 0.001). There were no significant differences in the expression levels of STAT3 and pSTAT3 between the HFD and the control groups. However, the expression level of NOD2 protein as determined by Western blot analysis was 60% higher in the HFD group compared with the control group. NOD2 contributes to the induction of inflammation by activation of nuclear factor ${\kappa}B$. These findings suggest that diet-induced obesity is associated with increased inflammatory response of immune cells, and higher expression of NOD2 may contribute to these changes.
본 연구에서는 미야베 에탄올 추출물의 항염증 활성을 확인하기 위해 LPS로 활성화된 RAW 264.7 세포와 croton-oil로 유도된 귀부종 동물 모델을 이용하였다. 그 결과, SMYEE 50 및 $100{\mu}g/ml$ 농도처리 시, LPS로 유도된 염증반응에서 $NF-{\kappa}B$ 활성 억제와 더불어 MAPKs의 인산화를 효과적으로 억제함을 보였다. LPS에 의해 증가된 NO와 전염증성 사이토카인의 분비량도 농도 의존적으로 감소하였다. 또한 SMYEE는 croton oil로 부종을 유발한 마우스모델에서 귀부종 억제효과를 나타내었고, 조직의 진피 두께 및 mast cell의 수가 현저히 감소하였음을 확인하였다. 이를 통해 SMYEE는 염증 반응의 전사인자인 $NF-{\kappa}B$ 및 MAPKs의 활성을 조절함으로써 iNOS와 COX-2의 발현 및 전염증성 매개인자인 NO, IL-6, $TNF-{\alpha}$ 및 $IL-1{\beta}$의 분비를 억제하여 항염증 활성을 가지는 것을 확인하였다. 현재까지 미야베 모자반내의 항염증 효능 물질에 관한 연구는 보고되지 않고 있으며 향후 실험을 통해 미야베 모자반 에탄올 추출물로부터 항염증 효과를 가지는 유효성분을 밝히기 위한 분리 연구를 진행할 예정이다.
초미세 분쇄법은 식품의 가공방법 중 하나로 특히 이를 이용하여 음용으로 활용범위를 넓힐 수 있다. 본 연구는 이에 착안하여 감을 이용한 초미세 분말을 이용하여 항산화 활성과 자가면역질환인 아토피에 효과가 있는지를 확인하여 보았다. 감 초미세 분말을 이용하여 DPPH와 FRAP 항산화 활성 실험을 수행 하였으며, 마우스 동물모델을 이용한 항알러지 피부염 동물모델을 이용하여 확인하였다. 항산화 활성 실험 결과 감 초미세 추출물은 농도에 따른 활성 등가를 나타내었다. DNFB (dinitrofluorobenzene)를 이용한 아토피 유도 후 이의 억제 활성은 아토피 유도군에서 염증의 유도와 함께 약 1.7배 정도 귀의 두께가 증가하는 것을 알 수 있었다. 또한, 감 초미세 추출물 FA 1, 2, 3 추출물을 처리한 결과, FA3이 다른 두 그룹 보다 귀에 비후정도가 감소하는 것으로 나타났다. 또한, 귀의 표피나 진피에서 발견되는 면역관련 세포는 대조군에 비해 아토피를 유도한 군에서 약 7배 정도의 세포수가 증가하는 것을 알 수 있었으며, 감 초미세 분말 추출물을 처리한 경우, 수적으로 감소하는 결과를 얻을 수 있었다.

제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
