Inhibitory Effect of Alopecurus aequalis Sobol Ethanol Extracts on LPS-induced Inflammatory Response in RAW 264.7 Cells (LPS로 유도한 RAW 264.7 세포의 염증반응에서 뚝새풀 에탄올 추출물의 억제 효과)

  • In the present study, we investigated the anti-inflammatory effects by Alopecurus aequalis Sobol on the lipopolysaccharide (LPS)-induced nitric oxide (NO) production by RAW 264.7 cell line. Consistent with these observations, DS reduced the LPS-induced expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) at the protein levels in a concentration-dependent manner. In addition, the release of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interleukin-6 (IL-6) were also reduced by DS. Moreover, LPS increased expression phosphorylation of $I{\kappa}B{\alpha}$, but DS showed inhibitory effect by reducing LPS-inducible p-$I{\kappa}B{\alpha}$ expression level. These results suggest that the down regulation of iNOS, COX-2, TNF-${\alpha}$, and IL-6 expression by DS are achieved by the downregulation of NF-${\kappa}B$ activity, a transcription factor necessary for pro-inflammatory mediators, and that is also responsible for its anti-inflammatory effects.

  • The anti-inflammatory effects of the ethanol extract of Undaria pinnatifida root (UPREE) were investigated using the lipopolysaccharide (LPS)-induced inflammatory response in RAW 264.7 cells by measuring the production of nitric oxide (NO), interleukin (IL)-6, tumor necrosis factor (TNF)-${\alpha}$, and IL-$1{\beta}$, and cell proliferation. We found that NO levels were reduced by 34% at $100{\mu}g/mL$. Moreover, the production of IL-6 and TNF-${\alpha}$ was suppressed by the UPREE treatment. In particular, the IL-6 production was inhibited by more than 30% at $100{\mu}g/mL$ UPREE. The proliferation of RAW 264.7 cells was measured by MTT assay, and we found no cytotoxicity in those cells treated with UPREE compared to the control. Our results suggest that UPREE shows promise as a therapeutic anti-inflammatory treatment.

Anti-fatigue effect of fermented porcine placenta through the regulation of fatigue-associated inflammatory cytokines

  • Fatigue is a common complaint and affects the quality of life in modern people. Physical stress may induce activation of certain immune cells. Fermented porcine placenta (FPP) has been used to alleviate fatigue. Inflammatory cytokines are produced by physical stress and results in symptoms of fatigue. However, the role of FPP on fatigue-associated inflammatory cytokine production has not been elucidated yet. Thus, we estimated the anti-fatigue effect of FPP and its active components, leucine (Leu) and lysine (Lys) in activated RAW264.7 macrophages and forced swimming test (FST) fatigue animal model. Pretreatment with FPP, Leu, or Lys significantly inhibited the lipopolysaccharide (LPS)-induced tumor necrosis factor-α, interleukin (IL)-1β, and IL-6 production without inducing cytotoxicity on LPS-stimulated RAW264.7 macrophages. FPP, Leu, or Lys inhibited the production of nitric oxide and downregulated the expression of inducible nitric oxide synthase on LPS-stimulated RAW264.7 macrophages. Furthermore, caspase-1 activities increased by LPS were significantly reduced by FPP, Leu, or Lys. In the FST, inflammatory cytokine levels of the mice administrated with FPP, Lys, and Leu were significantly reduced compared with the control group at 21 days. Collectively, these results show that anti-fatigue effect of FPP and its active components, Leu and Lys might be derived from the down-regulating of inflammatory mediators.

Screening of Herbal Medicines for Synergistic Effects of Metformin and Herbal Extracts Combination in RAW 264.7 Cells (RAW 264.7 세포에서 Metformin과 병행투여 시 상승효과를 나타내는 한약재의 선별 연구)

  • Objectives: This study was performed to evaluate the effects of Metformin and Lonicerae Flos, Agrobacterium Rhizogenes, Coptidis Rhizoma, Atractylodis Rhizoma Alba, Houttuyniae Herba extracs combinations on hypoglycemia in RAW 264.7 cells. Methods: Expressions of Sirt1, p-adenosine monophosphate-activated kinase (p-AMPK), AMPK-alpha, peroxisome proliferator activated receptor (PPAR)-alpha, PPAR-gamma, X-box binding protein 1 (XBP-1), tumor necrosis factor (TNF)-alpha and interleukin (IL)-6 were analyzed by real time polymerase chain reaction and Western blotting analysis. Results: The level of gene expression of Sirt1, p-AMPK, AMPK-alpha, PPAR-alpha and XBP-1 in relation to that of beta-actin were increased or decreased significantly with the Metformin and Lonicerae Flos, Agrobacterium Rhizogenes extracts combination groups. The level of gene expression of TNF-alpha and IL-6 were increased significantly with the Metformin and Houttuyniae Herba, Coptidis Rhizoma extracts combination groups. Conclusions: Metformin and Lonicerae Flos, Agrobacterium Rhizogenes extracts combination groups showed synergistic hypoglycemic effects by increasing AMPK and PPAR gene expression in RAW 264.7 cells.

Effects of Caraganae Sinicae Radix on the osteoporosis in ovariectomized rats (금작근(金雀根)이 난소적출로 유발된 흰쥐의 골다공증에 미치는 영향)

  • Objective : This study was carried out to investigate the effects of the hormones and cytokines associated with bone metabolism in ovariectomized rats. Method : Twenty-four Female-Sprague-Dawley rats were divided into sham operated(normal) group, ovariectomized(control)group, ovariectomized and treated with extract Caraganae Sinicae Radix(treated) group. Each group was investigated the changes of body weight at 3,5,7weeks after treatment, and femur weight, femur/body weight, thickness of compact bone of body of femur, area of cancellous bone of distal epiphysis of femur, serum estrogen, serum calcitonin, serum parathormone, serum Tumor Necrosis $Factor-{\alpha}$, serum $lnterleukin-1{\beta}$ at 7weeks after treatment. The results were summerized as follows; 1. The treated group showed significant change in body weight compared with the control group at 5,7weeks after treatment. 2. The treated group revealed significant increases in femur/body weight compared with the control group. 3. The treated group showed a little thicker compact bone of body of femur than the control group. 4. The area of cancellous bone of distal epiphysis of femur in treated group was increased significantly compared with control group. 5. The level of serum estrogen showed no change compared with control group. 6. The level of serum calcitonin showed no change compared with control group. 7. The level of serum parathormone was decreased in treated group significantly compared with control group. 8. The level of serum $TNF-{\alpha}$ was decreased in treated group significantly compared with control group. 9. The level of serum $interleukin-1{\beta}$ showed no change compared with control group. The results indicate that Caraganae Sinicae Radix inhibits bones resorption in ovariectomized rats by it's lowering effects on serum parathormone and $TNF-{\alpha}$.

Immuno-Modulatory Activities of Polysaccharides Separated from Jubak in Macrophage Cells (주박(酒粕)에서 분리된 다당류의 대식세포 면역조절 활성)

  • Activating macrophage cells play an important role in the host immune defense system. In this paper, immuno-modulatory activities of polysaccharides separated from Jubak (JPS) in macrophage cells were investigated. Immuno-modulatory activities were estimated based on cell proliferation, nitric oxide (NO) and cytokine production, degree of mitogen-activated protein kinases (MAPKs), and nuclear factor (NF)-${\kappa}B$ phosphorylation in RAW264.7 macrophage cells. JPS (62.5 to $250{\mu}g/mL$) did not induce a cytotoxic event. Additionally, NO and proinflammatory cytokines (tumor necrosis factor-${\alpha}$ and interleukin-6) production significantly increased in a dose-dependent manner. Similarly, phosphorylation of MAPKs and NF-${\kappa}B$ increased upon JPS treatment. Therefore, our results suggest that polysaccharides separated from Jubak can induce macrophage activation through MAPK and NF-${\kappa}B$ signaling and induction of Th1 polarization.

  • SunBangHwalMyungEum (SBH) has the effects of subduing swelling, resolving masses and alleviating pain in traditional oriental medicine. Recent studies showed that SunBangHwalMyungEum produced anti-cancer, anti-metastasis and immuno-modulatory effects. However there is lack of studies regarding the effects of SBH on the immunological activities. The present study was conducted to evaluate the effect of SBH on the regulatory mechanism of cytokines and nitric oxide (NO) in Raw 264.7 cells. Methods : After the treatment of SBH, cell viability was measured by MTT assay, NO production was monitored by measuring the nitrite content in culture medium. Inducible nitric oxide synthase (iNOS) was determined by immunoblot analysis, and levels of cytokine were analyzed by sandwich immunoassays. Results : Results provided evidence that SBH inhibited the production of NO, iNOS, $interleukin-1{\beta}$ ($(IL-1{\beta})$), IL-6, and the activation of phospholylation of inhibitor ${\kappa}B{\alpha}$ in Raw 264.7 cells activated with lipopolysaccharide. Conclusions : These findings suggest that SBH can produce anti-inflammatory effect, which may play a role in adjunctive therapy in Gram-negative bacterial infections.

Potentiation of Innate Immunity by β-Glucans

  • $\beta$-Glucans have been known to exhibit antitumor activities by potentiating host immunity by an unknown mechanism. The C-type lectin dectin-1, a $\beta$-glucan receptor, is found on the macrophage and can recognize various $\beta$-glucans. Previously, we demonstrated the presence of $\beta$-glucan receptor, dectin-1, on the Raw 264.7 cells as well as on murine mucosal organs, such as the thymus, the lung, and the spleen. In order to investigate immunopotentiation of innate immunity by $\beta$-glucan, we stimulated a murine macrophage Raw 264.7 cell line with $\beta$-glucans from Pleurotus ostreatus, Saccharomyces cerevisiae, and Laminaria digitata. Then, we analyzed cytokines such as tumor necrosis factor (TNF)-$\alpha$ and interleukin (IL)-6 by reverse transcription-polymerase chain reaction (RT-PCR). In addition we analyzed gene expression patterns in $\beta$-glucan-treated Raw 264.7 cells by applying total mRNA to cDNA microarray to investigate the expression of 7,000 known genes. When stimulated with $\beta$-glucans, the macrophage cells increased TNF-$\alpha$ expression. When co-stimulation of the cells with $\beta$-glucan and lipopolysaccharide (LPS), a synergy effect was observed by increased TNF-$\alpha$ expression. In IL-6 expression, any of the $\beta$-glucans tested could not induce IL-6 expression by itself. However, when co-stimulation occurred with $\beta$-glucan and LPS, the cells showed strong synergistic effects by increased IL-6 expression. Chip analysis showed that $\beta$-glucan of P. ostreatus increased gene expressions of immunomodulating gene families such as kinases, lectin associated genes and TNF-related genes in the macrophage cell line. Induction of TNF receptor expression by FACS analysis was synergized only when co-stimulated with $\beta$-glucan and LPS, not with $\beta$-glucan alone. From these data, $\beta$-glucan increased expressions of immunomodulating genes and showed synergistic effect with LPS.

Anti-Inflammatory Activity of Antimicrobial Peptide Periplanetasin-5 Derived from the Cockroach Periplaneta americana

  • Previously, we performed an in silico analysis of the Periplaneta americana transcriptome. Antimicrobial peptide candidates were selected using an in silico antimicrobial peptide prediction method. It was found that periplanetasin-5 had antimicrobial activity against yeast and gram-positive and gram-negative bacteria. In the present study, we demonstrated the anti-inflammatory activities of periplanetasin-5 in mouse macrophage Raw264.7 cells. No cytotoxicity was observed at 60 ㎍/ml periplanetasin-5, and treatment decreased nitric oxide production in Raw264.7 cells exposed to lipopolysaccharide (LPS). In addition, quantitative RT-PCR and enzyme-linked immunosorbent assay revealed that periplanetasin-5 reduced cytokine (tumor necrosis factor-α, interleukin-6) expression levels in the Raw264.7 cells. Periplanetasin-5 controlled inflammation by inhibiting phosphorylation of MAPKs, an inflammatory signaling element, and reducing the degradation of IκB. Through LAL assay, LPS toxicity was found to decrease in a periplanetasin-5 dose-dependent manner. Collectively, these data showed that periplanetasin-5 had anti-inflammatory activities, exemplified in LPS-exposed Raw264.7 cells. Thus, we have provided a potentially useful antibacterial peptide candidate with anti-inflammatory activities.

The anti-inflammatory effects of Cicadidae Periostracum (선태(蟬蛻)의 항염효과(抗炎效果)에 대한 실험적(實驗的) 연구(硏究))

  • Objectives: The purpose of this study was to investigate the anti-inflammatory effects of aqueous extract from Cicadidae Periostracum(CP) on the RAW 264.7 cells. Method: We examined the cytokine productions including nitric oxide(NO), interleukin(IL)-1b, IL-6 and tumor necrosis factor-a(TNF-a) in lipopolysaccharide (LPS)-induced RAW 264.7 cells and also inhibitory mechanisms such as mitogen -activated protein kinases(MAPKs) and nuclear factor kappa BNF-kB) using Western blot. Results: CP inhibited LPS-induced production of NO, IL-1b and TNF-a but not of IL-6 in RAW 264.7 cells. CP respectively inhibited the activation of MAPKs such as extracelluar signal-regulated kinase(ERK 1/2), c-Jun $NH_2$-terminal kinase(JNK), p38 and NF-kB in the LPS-stimulated RAW 264.7 cells. Also oral administration of CP inhibited CLP - induced endotoxin shock. Conclusion: our results showed that CP down-regulated LPS-induced NO, IL-1b and TNF-a productions mainly through ERK, JNK, p38 MAPK and NF-kB pathway, which suggest the anti-inflammatory effects of CP.
