Endothelin-1 생성 저해와 멜라닌생성 저해에 의한 신선초 추출물의 미백 효과 (Whitening Effect of Angelica keiskei Koidzumi Extract by Inhibition of Endothelin-1 Production and Melanogenesis)

  • 신선초 추출물 및 그의 분획물의 미백 효과를 알아보기 위하여, B16 멜라노마와 정상 사람 표피 세포를 이용하여 멜라닌 생성에 관련된 다양한 실험을 실시하였다. 70% ethanol 용매로 추출한 신선초 추출물을 연속적으로 분획하여 hexane, ethyl acetate, n-butanol, 물 분획물을 얻었다. 버섯 티로시나제 활성 실험에서 신선초 추출물 및 그의 분획물은 저해 효과를 보이지 않았으나, B16 멜라노마를 이용한 멜라닌 생성 실험에서는 hexane 분획물과 ethyl acetate 분획물에서 억제 효과를 보였다. 세포내 멜라닌 생성의 초기 단계에 작용하는 티로시나제 단백질의 활성을 조사한 결과, hexane 분획($IC_{50}$ < $25{\mu}g/mL$)과 ethyl acetate분획($IC_{50}$ < $5{\mu}g/mL$)은 알부틴($IC_{50}{\leq}250{\mu}g/mL$)과 비교 시 더 낮은 농도에서 현저하게 티로시나제 활성을 저해시켰다. 따라서 신선초 추출물의 미백 작용 기전을 규명하기 위하여, 멜라노 사이트의 mitogen으로 작용하는 endothelin-1(ET-1)과 UVB에 의해 생성되어 염증을 매개하는 interleukin-$1{\alpha}$(IL-$1{\alpha})$의 생성량을 측정하였다. Hexane 및 ethyl acetate 분획물은 IL-$1{\alpha}$의 생성에 영향을 끼치지 못했으나, 표피세포에서 UVB에 의해 생성된 ET-1을 농도에 따라 감소시킴으로써 멜라노사이트의 melanogenesis를 저해하는 것으로 확인되었다. 따라서 신선초 추출물은 미백 효능을 갖는 화장품 소재로써 사용 가능하리라 판단된다.

The immune enhancing effects and characteristics of Bifidobacterium longum and Bifidobacterium breve for the probiotic use in humans and animals

  • The purpose of this study was to investigate probiotic characteristics and immune enhancing effects of Bifidobacterium (B.) longum KBB1-26 and BIF-4, B. breve KBB5-22 isolated from human intestine for probiotic use in humans and animals. We measured acid, bile and heat tolerance, antimicrobial activity against pathogenic bacteria, Escherichia (E.) coli, Salmonella (S.) Enteritidis, Staphylococcus (S.) aureus, and Listeria (L.) monocytogenes. Immune enhancing effects of B. longum and B. breve were investigated by measuring nitric oxide (NO), nuclear factor ($NF-{\kappa}b$), $interleukin-1{\beta}$ ($IL-1{\beta}$), interleukin-6 (IL-6), interleukin-12 (IL-12) and tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) in RAW 264.7 cells or RAW BLUE cells. B. longum KBB1-26 was survived at pH 2.0. B. longum KBB1-26 and BIF-4, B. breve KBB5-22 also showed tolerance to 0.3% of oxgall bile salt. B. longum KBB1-26 was able to survive at $70^{\circ}C$ and $80^{\circ}C$ for 20 min. KBB1-26 showed the antimicrobial inhibition zone to pathogenic bacteria such as E. coli (12 mm), S. Enteritidis (14 mm), S. aureus (14 mm) and L. monocytogenes (41 mm). The production of NO ($4.5{\pm}0.00{\mu}M/mL$) and $IL-1{\beta}$ ($39.7{\pm}0.55pg/mL$) of KBB1-26 significantly higher than BIF-4 and KBB5-22, respectively. In addition, KBB1-26 and KBB5-22 induce the production of high level of $TNF-{\alpha}$ and IL-6 in macrophages. Collectively, B. longum KBB1-26 have acid, bile, heat tolerance, antimicrobial activity and immune enhancing effects. These results suggest that KBB1-26 can be used as probiotics for humans and animals.

Anti-inflammatory Effect of Dactyloquinone B and Cyclospongiaquinone-1 Mixture in RAW264.7 Macrophage and ICR Mice

  • Sesquiterpene-quinone is a class of secondary metabolites frequently encountered from marine sponge. The present study was designed to examine the anti-inflammatory action of sponge-derived dactyloquinone B (DQB) and cyclospongiaquinone-1 (CSQ1) mixture using lipopolysaccharide (LPS)-induced inflammatory responses. We measured the production of nitric oxide (NO), tumor necrosis factor-alpha ($TNF-{\alpha}$), $interleukin-1{\beta}$ ($IL-1{\beta}$), and interleukin-6 (IL-6) and expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) protein. $TNF-{\alpha}$, $IL-1{\beta}$, and IL-6 production, which increased by treatment with LPS, were significantly inhibited by DQB and CSQ1 mixture. It also decreased the production of NO production, and iNOS and COX-2 expression. Furthermore, it reduced 12-O-tetradecanoylphorbol 13-acetate (TPA)-induced ear edema of ICR mice. These results demonstrate that sesquiterpene-quinone, DQB and CSQ1 mixture, might serve as a chemical pipeline for the development of anti-inflammatory agent.

Caffeic acid phenethyl ester protects against photothrombotic cortical ischemic injury in mice

  • In this study, we aimed to investigate the neuroprotective effects of caffeic acid phenethyl ester (CAPE), an active component of propolis purified from honeybee hives, on photothrombotic cortical ischemic injury in mice. Permanent focal ischemia was achieved in the medial frontal and somatosensory cortices of anesthetized male C57BL/6 mice by irradiation of the skull with cold light laser in combination with systemic administration of rose bengal. The animals were treated with CAPE (0.5-5 mg/kg, i.p.) twice 1 and 6 h after ischemic insult. CAPE significantly reduced the infarct size as well as the expression of tumor necrosis $factor-{\alpha}$, hypoxiainducible $factor-1{\alpha}$ monocyte chemoattractant protein-1, $interleukin-1{\alpha}$, and indoleamine 2,3-dioxygenase in the cerebral cortex ipsilateral to the photothrombosis. Moreover, it induced an increase in heme oxygenase-1 immunoreactivity and interleukin-10 expression. These results suggest that CAPE exerts a remarkable neuroprotective effect on ischemic brain injury via its anti-inflammatory properties, thereby providing a benefit to the therapy of cerebral infarction.

  • In an ongoing investigation into anti-inflammatory compounds from natural products, the $CH_2Cl_2$ soluble fraction of Patrinia scabiosaefolia F. (Valerianaceae) was found to inhibit IL-6 production in TNF-$\alpha$ stimulated MG-63 cell line. By means of a bioassay-directed chromatographic separation technique, lappaol E (1), and nortrachelogenin (2) were isolated. These compounds have been isolated from this plant for the first time. Compounds $1{\sim}2$ showed potent antioxidative activity using NBT superoxide scavenging assay. Moreover, compound 1 decreased IL-6 production in TNF-$\alpha$ stimulated MG-63 cell line.

Differential Modulatory Effects of Cholera Toxin and Pertussis Toxin on Pain Behavior Induced by TNF-${\alpha}$, Interleukin-1${\beta}$ and Interferon-${\gamma}$ Injected Intrathecally

  • The present study was designed to characterize the possible roles of spinally located cholera toxin (CTX)- and pertussis toxin (PTX)-sensitive G-proteins in pro- inflammatory cy tokine induced pain behaviors. Intrathecal injection of tumor necrosis factor-a (TNF-${\alpha}$; 100 pg), interleukin-1${\beta}$ (IL-1${\beta}$ 100 pg) and interferon-${\gamma}$ (INF-${\gamma}$; 100 pg) showed pain behavior. Intrathecal pretreatment with CTX (0.05, 0.1 and 0.5 mg) attenuated pain behavior induced by TNF-${\alpha}$ and INF-${\gamma}$ administered intrathecally. But intrathecal pretreatment with CTX (0.05, 0.1 and 0.5${\mu}g$) did not attenuate pain behavior induced by IL-1${\beta}$. On the other hand, intrathecal pretreatment with PTX further increased the pain behavior induced by TNF-${\alpha}$ and IL-1${\beta}$ administered intrathecally, especially at the dose of 0.5 ${\mu}g$. But intrathecal pretreatment with PTX did not affect pain behavior induced by INF-${\gamma}$. Our results suggest that, at the spinal cord level, CTX- and PTX-sensitive G-proteins appear to play important roles in modulating pain behavior induced by pro-inflammatory cytokines administered spinally. Furthermore, TNF-${\alpha}$, IL-1${\beta}$ arid INF-${\gamma}$ administered spinally appear to produce pain behavior by different mechanisms.

중추신경계(中樞神經系)에서 숙지황(熟地黃)의 항염증작용(抗炎症作用)에 관(關)한 연구(硏究) (Studies on the anti-inflammatory action of steamed Rehmannia glutinosa in central nervous system)

  • We investigated whether an aqueous extract of Rehmannia glutinosa steamed root (RGAE) inhibits secretion of inflammatory cytolanes from primary cultures of mouse astrocytes. RGAE dose-dependently inhibited the $TNF-{\alpha}$ secretion by astrocytes stimulated with substance P (SP) and lipopolysaccharide (LPS). Interleukin-1 (IL-1) has been shown to elevate $TNF-{\alpha}$ secretion from LPS-stimulated astrocytes while having no effect on astrocytes in the absence of LPS. We therefore also investigated whether IL-1 mediated inhibition of $TNF-{\alpha}$ secretion from primary astrocytes by RGAE. Treatment of RGAE to astrocytes stimulated with both LPS and SP decreased IL-1 secretion to the level observed with LPS alone. Moreover, incubation of astrocytes with IL-1 antibody abolished the synergistic cooperative effect of LPS and SP. These results suggest that RGAE has an antiinflammatory activity on the central nervous system curing some pathological disease states.

소양인(小陽人) 인동등지골피탕(忍冬藤地骨皮湯)이 중소합병증(中消合倂症)에 미치는 영향(影響) (Effecs of Soyangin Indongdeungjikolpitang on the Complication of Diabetes)

  • 소양인(少陽人) 인동등지골피탕(忍冬藤地骨皮湯)이 중소(中消) 합병증(合倂症)에 미치는 영향(影響)을 살펴보고자 하였고 결과(結果)는 다음과 같다. 1. 장관운동(腸管運動)을 억제(抑制)하였다. 2. Acetic acid에 의한 writhing syndrome을 억제(抑制)하였다. 3. Evans blue에 의한 모세혈관투과성(毛細血管透過性)을 억제(抑制)하였다. 4. Histamine에 의한 급성족부종(急性足浮腫)을 억제(抑制)하였다. 5. Cotton pellet에 의한 육아종(肉芽腫) 형성(形成)을 억제(抑制)하였다. 6. 흉선세포(胸線細胞) 및 비장세포(脾臟細胞)의 세포생존율(細胞生存率)을 증가(增加)시켰다. 7. 흉선세포(胸線細胞)로부터 ${\gamma}-IFN$ 및 IL-2의 분비(分泌)를 억제(抑制)하였다. 8. 비장세포(脾臟細胞)로부터 IL-4의 분비(分泌)는 증가(增加)시켰으나, $TNF-{\alpha}$$IL-1{\beta}$의 분비(分泌)는 억제(抑制)하였다. 9. 복강(腹腔) 대식세포(大食細胞)로부터 $TNF-{\alpha}$, $IL-1{\beta}$ 및 nitric oxide의 분비(分泌)를 억제(抑制)하였다. 10. 복강(腹腔) 대식세포(大食細胞)의 lucigenin chemiluminescence 및 FITC-conjugated E. cole engulfment를 촉진(促進)하였다. 이상의 실험(實驗) 결과(結果) IJTE는 장관운동(腸管運動)을 억제(抑制)하였으며, 진통(鎭痛) 및 항염작용(抗炎作用)을 나타냈고, 이들 작용은 면역세포(免疫細胞)로부터 $TNF-{\alpha}$, $IL-1{\beta}$ 및 nitric oxide의 생성(生成)을 억제(抑制)하기 때문인 것으로 추정된다. 따라서 이러한 기전(機轉)에 의하여 인동등지골피탕(忍冬藤地骨皮湯)은 중소(中消) 합병증(合倂症)의 하나인 염증(炎症)을 억제(抑制)하는데 유효(有效)한 것으로 사료(思料)된다.

Effect of Egg White Combined with Chalcanthite on Lipopolysaccharide induced Inflammatory Cytokine Expression in RAW 264.7 cells

  • Aim: Historically, mineral compound herbal medicines have long been used in treatments of immune-related diseases in Korea, China and other Asian countries. In this study, we investigated the anti-inflammatory effect of egg white combined with chalcanthite (IS4) on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Methods: RAW 264.7 cells cultured with LPS and various concentrations of IS4 were analyzed to determine the production of pro-inflammatory cytokines and mediators by using enzyme-linked immune sorbent assays (ELISAs). Results: IS4 concentration inhibited the production of interleukin-1beta (IL-$1{\beta}$), interleukin-6 (IL-6), and granulocyte-macrophage colony-stimulating factor (GM-CSF) induced by LPS. IS4 at high concentrations (25 and 50 ${\mu}g/ml$) inhibited, in concentration-dependent manner, the expression of tumor necrosis factor-alpha (TNF-${\alpha}$) stimulated by LPS. Conclusion: IS4 has shown an anti-inflammatory effect in RAW 264.7 cells.

  • We investigated whether an aqueous extract of Polygala tenuifolia root (PTAE) inhibits secretion of inflammatory cytokines from primary cultures of mouse astrocytes. PTAE dose-dependently inhibited the Tumor necrosis $factor-{\alpha}$ $(TNF-{\alpha})$ secretion by astrocytes stimulated with substance P (SP) and lipopolysaccharide (LPS). Interleukin-1 (IL-1) has been shown to elevate $TNF-{\alpha}$ secretion from LPS-stimulated astrocytes while having no effect on astrocytes in the absence of LPS. Reverse transcriptase-polymerase chain reaction analysis demonstrated the significantly reduced level of the $TNF-{\alpha}$ mRNA was expressed in astrocytes treated with PTAE.
