금은화가 LPS로 유발된 급성 폐 손상에 미치는 영향 (Effects of Lonicerae Flos Extracts on LPS-induced Acute Lung Injury)

  • Objective : The object of this study was to observe the effects of Lonicerae Flos (LF) aqueous extracts on lipopolysaccharide (LPS)-induced rat acute lung injury. Method : Five different dosages of LF extracts were orally administered once a day for 28 days before LPS treatments, and then all rats were sacrificed after 5 hour-treatment of LPS. Eight groups of 16 rats each were used in the present study. The following parameters caused by LPS treatment were observed ; body weights, lung weights, pulmonary transcapillary albumin transit, arterial gas parameters (pH, $PaO_2$ and $PaCO_2$) bronchoalveolar lavage fluid (BALF) protein lactate dehydrogenase (LDH), and proinflammatory cytokines tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-$1{\beta}$ (IL-$1{\beta}$) contents, total cell numbers, neutrophil and alveolar macrophage ratios, lung malondialdehyde (MDA), myeloperoxidase (MPO), proinflammatory cytokines TNF-${\alpha}$ and IL-$1{\beta}$ contents. In addition, the histopathologic changes were observed in the lung in terms of luminal surface of alveolus, thickness of alveolar septum, number of polymorphonuclear neutrophils. Result : As results of LPS-injection, dramatical increases in lung weights, pulmonary transcapillary albumin transit increases, increases in $PaCO_2$, decreases in pH of arterial blood and $PaO_2$, increases of BALF protein, LDH, TNF-${\alpha}$ and IL-$1{\beta}$ contents, total cells, neutrophil and alveolar macrophage ratios, TNF-${\alpha}$ and IL-$1{\beta}$ contents increases were detected with decreases in LSA and increases of alveolar septum and PMNs numbers, respectively as compared with intact control. These are means that acute lung injuries (resembling acute respiratory distress syndrome) are induced by treatment of LPS mediated by inflammatory responses, oxidative stress and related lipid peroxidation in the present study. However, these LPS-induced acute lung injuries were inhibited by 28 days continuous pretreatment of 250 and 500mg/kg of LF extracts. Because of lower three dosages of LF treated groups, 31.25 and 62.5 and 125mg/kg did not showed any favorable effects as compared with LPS control, the effective dosages of LF in LPS-induced acute lung injuries in the present study, is considered as about 125mg/kg. The effects of 250mg/kg of LF extracts showed almost similar effects with ${\alpha}$-lipoic acid 60mg/kg in preventing LPS-induced acute lung injuries. Conclusion : It seems that LF play a role in protecting the acute respiratory distress syndrome caused by LPS.

미세분진이 흰쥐의 폐포대식세포에서 TNF-α와 IL-1β의 형성에 미치는 효과 (The Effects of Air-borne Particulate Matters on the Alveolar Macrophages for the TNF-α and IL-1β Secretion)

  • 연구 배경: 대도시의 대기오염은 점차 악화되어 시민의 건강을 위협하고, 심 폐 질환의 발병률과 이 로 인한 사망률을 증가시키고 있다. 서울시 도로가의 PM이 $TNF-{\alpha}$$IL-1{\beta}$의 생성에 직접적으로 어떠한 영향을 미치는지와 PM의 노출이 LPS의 $TNF-{\alpha}$$IL-1{\beta}$의 생성효과에는 어떠한 영향을 미치는지를 평가하고자 하였다. 방 법: 폐렴이 있는 흰쥐와 SPF 흰쥐의 폐포대식세포 각각에 PM을 농도별로 처리하여 분비되는 $TNF-{\alpha}$$IL-1{\beta}$의 농도를 측정하였다. 측정 방법으로는 western blot, ELISA 및 세포면역화학염색법을 이용하였다. 또한 동일 PM 농도에서 배양시간을 달리하여 위와 같이 측정하였다. 결 과: SPF인 흰쥐에서 분리된 페포대식세포에 PM을 단독으로 투여하였을 때 대조군에 비해서 $TNF-{\alpha}$$IL-1{\beta}$의 생성도가 모든 투여군에서 유의하게 증가하였으나, 투여용량의 증가에 따른 유의성은 없었다. 그러나 염증성인 쥐에서 분리된 폐포대식세포에서는 모든 투여군에서 대조군에 비하여 통계학적으로 유의하게 증가하였으며, PM 투여농도의 증가에 따른 생성량도 유의하게 증가하였다. 결 론: PM을 장기간 혹은 일정 농도 이상으로 흡입할 경우 폐포대식세포의 $TNF-{\alpha}$$IL-1{\beta}$의 분비에 영향을 미쳐 새로운 폐질환을 유발할 수 있다. 그러므로 기존에 염증성 폐질환이나 기관지천식이 있는 환자가 미세먼지를 흡입할 경우에는 $TNF-{\alpha}$$IL-1{\beta}$의 생성에 커다란 영향을 미쳐 호흡기 질환을 더욱 악화시킬 가능성이 있을 것으로 추정된다.

귤피 유래 면역활성 람노갈락투로난류의 신속 분리방법 (Rapid Isolation Method for Preparation of Immuno-Stimulating Rhamnogalacturonans in Citrus Peels)

  • 다당의 생물활성은 다당의 구조적인 특징과 분자량 분포에 의해 중요한 영향을 미치기 때문에, 특정 다당의 정제는 다당 연구를 위해 필수적이다. 따라서 본 연구에서는 서로 다른 특성을 소유한 다당의 분획을 위한 신속 분리 방법을 개발하고 대표 화합물로 한국산 귤피로부터 조제한 다당 혼합물을 이용, 본 분리법을 최적화 하였다. 귤피는 펙티나아제 처리 후 에탄올 침전법을 통해 조다당 획분인 CPE로 조제되었으며, CPE는 재차 농도별로 연속 희석된 에탄올 용액(EtOH:DIW=8:1, 4:1, 3:1, 2:1, 1.5:1, 1:1, 및 0.5:1)을 이용하여 7가지 획분(CPE8-CPE0.5)으로 분획되었다. CPE8-CPE1획분은 구성당 분석 결과 람노갈락투로난-I과 람노갈락투로난-II 다당의 지표 구성당인 총 11종의 서로 다른 당으로 구성되어 있었으며, arabino-${\beta}$-3,6-galactan 잔기를 함유하고 있는 것으로 확인되었다. 그러나 CPE0.5 획분에서는 람노갈락투로난-II 및 arabino-${\beta}$-3,6-galactan 잔기를 함유하고 있지 않았다. 한편, CPE8-CPE1 획분을 처리한 mouse 복강 대식세포에서는 농도의존적으로 IL-6의 생산 증가가 관찰된 반면, CPE1 및 CPE0.5 획분에서는 활성이 급격히 감소됨을 확인 할 수 있었다. 따라서 이상의 결과로부터 분리방법이 다양한 특성을 갖는 다당의 혼합물로부터 생물활성을 갖는 람노갈락투로난류를 신속히 분리하는데 매우 유용한 것으로 판단되었다.

Inhibitory effects of honokiol on LPS and PMA-induced cellular responses of macrophages and monocytes

  • The regulatory effects of honokiol on the cellular responses of macrophages and monocytes were evaluated. Specifically, we investigated the effects of honokiol with respect to lipopolysaccharide (LPS)-induced cytotoxicity, LPS- or phorbol-12-myristate-13-acetate (PMA)-mediated morphological changes, and relevant events (FITC-dextran-induced phagocytic uptake). Honokiol blocked the LPS-induced cytotoxicity of RAW264.7 cells in a dose-dependent manner. In addition, honokiol appeared to block the production of cytotoxic cytokines such as interleukin (IL)-$1{\beta}$ and tumor necrosis factor (TNF)-$\alpha$, nitric oxide (NO), and reactive oxygen species (ROS). Moreover, honokiol strongly prevented the morphological changes in RAW 264.7 and U937 cells that were induced by LPS and PMA. The surface levels of marker proteins, which are up-regulated under the morphological changes of RAW264.7 and U937 cells, were also diminished. The data presented here strongly suggest that the honokiol modulates various cellular responses managed by macrophages and monocytes.

  • We studied the effect of methanol extract of Carpopeltis affinis (CA) on atopic allergic reaction. CA dose-dependently inhibited interleukin (IL)-8 and tumor necrosis factor (TNF)-$\alpha$ secretion from the PMA- plus A23187- stimulated HMC-1. CA also dose-dependently inhibited the histamine and $\beta$-hexosaminidase release from mast cells. CA had no cytotoxic effect. (omitted)

Effects of Ethanol Extract of Ligularia fischeri Leaves on Freund's Complete Adjuvant-Induced Model of Chronic Arthritis in Mice

  • The aim of this study was to investigate the anti-inflammatory and anti-oxidant activity of Ligularia fischeri leaf extract on adjuvant induced arthritis in experimental mice. The oral administration of the L. fischeri leaf extract (LF), at doses of 100 and 200 mg/kg body weight once a day for 3 weeks, significantly reduced hindpaw swelling and the production of inflammatory cytokines (tumor necrosis factor(TNF)-${\alpha}$, interleukin(IL)-$1{\beta}$, and IL-6). Treatment with LF (100 mg/kg) also decreased the serum levels of triglyceride and low density lipoprotein(LDL)-cholesterol, and increased high density lipoprotein(HDL)-cholesterol contents compared with those of a control group. The induction of arthritis significantly increased oxidized proteins such as protein carbonyl, advanced oxidation protein products, and advanced glycation end-products in the lung, heart, and brain. Treatment with LF for 3 weeks reduced the levels of oxidized proteins. These results suggest that L. fischeri extract might be beneficial in the treatment of chronic inflammatory disorders.

Forsythiae Fructus and Its Active Component, Arctigenin, Provide Neuroprotection by Inhibiting Neuroinflammation

  • In this study, we found that Forsythiae fructus (FF) and one of its main compounds, arctigenin, significantly inhibited nitric oxide production in lipopolysaccharide (LPS)-stimulated BV-2 microglial cells. Arctigenin also suppressed the expression of inducible nitric oxide synthase and cyclooxygenase-2, and inhibited the activation of extracellular signal-regulated kinase, c-Jun N-terminal kinase and p38. Moreover, it also reduced levels of proinflammatory cytokines, interleukin $1{\beta}$, tumor necrosis factor ${\alpha}$ and prostaglandin E2, and inhibited neuronal death in LPS-treated organotypic hippocampal cultures. Therefore, we suggest that arctigenin may confer a neuroprotective effect via the inhibition of neuroinflammation.

Colchicine for steroid-resistant recurrent pericarditis in a child

  • Recurrent pericarditis is rare in children and is considered idiopathic in most cases. Its course is chronic, and preventing recurrences is important for the patient's quality of life. Although a treatment strategy in pediatric recurrent pericarditis has not yet been established, non-steroidal anti-inflammatory drugs (NSAIDs) are the most common treatment for management of this condition, followed by corticosteroids, colchicine, immunosuppressive agents, immunoglobulins, and interleukin-$1{\beta}$ receptor antagonists (e.g. anakinra). Herein, we report a case of recurrent pericarditis with pericardial effusion in a 5-year-old child who presented with fever and epigastric pain. He responded poorly to NSAIDs and corticosteroid therapy, but was successfully treated with colchicine.

  • Inflammatory mediators has been recognized as an important role in the pathogenesis of rheumatoid arthritis (RA). IL-17 is increasingly recognized as an important regulator of immune and inflammatory responses, including induction of proinflammatory cytokines and osteoclastic bone resorption. Evidence of the expression and proinflammatory activity of IL-17 has been demonstrated in RA synovium and in animal models of RA. However, the signaling pathways that regulate IL-17 production remain unknown. In the present study, we investigated the role of the phosphatidylinositol 3 kinase (PI3K)-Akt pathway in the regulation of IL-17 production in RA. PBMC were separated from RA (n=24) patients, and stimulated with various agents (anti CD3, anti CD28, PHA, ConA, IL-15). IL-17 levels were determined by sandwich ELISA and RT-PCR. The production of IL-17 was significantly increased in cells treated with anti-CD3 antibody, PHA, IL-15 or MCP-1 (P<0.05). ConA also strongly induced IL-17 production (P<0.001), whereas TNF-alpha, IL-1beta, IL-18 or TGF-beta did not. IL-17 was detected in the PBMC of patients with osteoarthritis (OA) but their expression levels were much lower than those of RA PBMC. Anti-CD3 antibody activated the PI3K-Akt pathway and activation of the PI3K-Akt pathway resulted in a pronounced augmentation of nuclear factor kappaB ($NF-{\kappa}B$). IL-17 production by activated PBMC in RA is completely or partially blocked in the presence of $NF-{\kappa}B$ inhibitor PDTC and PI3K-Akt inhibitor, wortmannin and LY294002, respectively. Whereas the inhibition of AP-1 and extracellular signal-regulated kinase (ERK)1/2 did not affect IL-17 production. These results provide new insight into that PI3K/Akt and $NF-{\kappa}B$ dependent signal transduction pathway could be involved in the overproduction of key inflammatory cytokine, IL-17 in rheumatoid arthritis.

국산 생약 추출물의 항염증 활성 스크리닝 (Screening for Anti-inflammatory Activities in Extracts from Korean Herb Medicines)

  • 화장품은 일반인이 장기간 연용하는 제품으로 인체 피부에 대한 안전성이 매우 중요하다. 화장품에 의한 접촉피부염은 자극성 물질이 피부에 접촉 후 침투하여 유발하는 염증반응으로 활성화된 면역세포에서 염증을 매개하는 다양한 인자를 분비함으로써 시작된다. 본 연구에서는 국산 생약 추출물을 이용하여 RAW264.7 대식 세포에서 염증 관련 인자에 대한 영향을 통해 항염증 활성을 스크리닝 하였다. 51종의 국산 생약 추출물 중 측백, 측백엽(초), 향부자, 형개, 동과자, 산약, 산약(초), 상지, 송절, 택사의 에탄올 추출물 10종이 lipopolysaccharide(LPS)에 의해 유도된 세포독성을 감소시킴과 동시에 염증 관련 인자인 Nitric oxide (NO), interleukin (IL)-1${\beta}$, IL-6, tumor necrosis factor (TNF)-${\alpha}$의 생성을 억제하는 것을 확인하였다. 또한, 이들 생약 추출물은 인체피부에서도 자극을 유발하지 않음을 인체 첩포시험을 통해 검증하였다. 따라서, 항염증 활성이 확인된 10종 국산 생약 추출물은 피부 자극을 예방할 수 있는 화장품 소재로서의 활용이 가능할 것으로 사료된다.
