Immunomodulatory Function of Murine NK Cell Activity by Alginate

  • The in vivo immunomodulatory function of the activity of murine natural killer (NK) cells induced by high mannuronic acid-containing alginate (HMA) was examined. HMA was injected i.p at doses of 25 and 100 mg/kg. The NK activity was 3 times higher with 100 mg/kg HMA than the baseline. In addition, in vitro studies of splenocytes cultured with HMA for 20 h showed a significant increase in NK activity at E:T ratio of 100: 1; a 160$\%$ and 210$\%$ increase at 10 and 100 $\mu$g/mL, respectively. There was a six fold increase in interferon-$\gamma$ production in a postculture of splenocytes with 100 $\mu$g/mL HMA. HMA had no suppressive effects on the lymphocyte function in the presence or absence of mitogens. This suggests that HMA is useful in cancer immunotherapy.

  • In bioassay-guided search for inducible nitric oxide synthase (iNOS) inhibitory compounds from higher plants of South Korea, two $\beta$-carboline (2) have been isolated form the cortex of Melia azedarach var. japonica. The structures of these compounds were elucidated on the basis of spectroscopic data. Compounds 1 to 2 showed marked inhibitory activity of iNOS on LPS-and interferon-${\gamma}$-stimulated RAW 264.7 cells.

영지버섯 다당체의 Nitric Oxide 생성능 및 생성기전 연구 (Nitric Oxide Production Ability and its Formation Mechanisms in Macrophage TIB 71 Cell Line by Polysaccharide Extracted from Ganoderma lucidum)

  • This study was carried out to get infomation on the nitric oxide production ability and its formation mechanisms of polysaccharides extracted from Ganoderma lucidum(PSG) by using murine macrophage cell line. The cultured mycelial cells of Ganoderma lucidum were extracted by alkali, and than neutralized by acid. The extract were passed through the column of DEAE cellulose for more purification. The neutral fraction was concentrated and precipitated with 95% ethanol. The precipitate was lyophilized and PSG was obtained. The immunomodulating effects of PSG on macrophage were performed by using murine macrophage cell line ATCC TIB 71 cells with PSG 0.5mg. PSG alone could not induce the production of nitrite, but it had a significant potential effect on nitrite secretion when the cells were primed and triggered with BCG and Interferon(IFN)-${\gamma}$. Also it was prominent by using calcium channel blocker(verapamil) and adenylate cyclase activator(forskolin).

Diagnosis and Treatment of Extrapulmonary Tuberculosis

  • Extrapulmonary tuberculosis (EPTB) constitutes about 20% of all cases of tuberculosis (TB) in Korea. Diagnosing EPTB remains challenging because clinical samples obtained from relatively inaccessible sites may be paucibacillary, thus decreasing the sensitivity of diagnostic tests. Whenever practical, every effort should be made to obtain appropriate specimens for both mycobacteriologic and histopathologic examinations. The measurement of biochemical markers in TB-affected serosal fluids (adenosine deaminase or gamma interferon) and molecular biology techniques such as polymerase chain reaction may be useful adjuncts in the diagnosis of EPTB. Although the disease usually responds to standard anti-TB drug therapy, the ideal regimen and duration of treatment have not yet been established. A paradoxical response frequently occurs during anti-TB therapy. It should be distinguished from other causes of clinical deterioration. Surgery is required mainly to obtain valid diagnostic specimens and to manage complications. Because smear microscopy or culture is not available to monitor patients with EPTB, clinical monitoring is the usual way to assess the response to treatment.

Ceramide analogs inhibit inducible nitric oxide synthase expression and nitric oxide production in interferon-gamma and lipopolysaccharide-stimulated RAW 264.7 macrophages.

  • Nitric oxide (NO) production through the inducible nitric-oxide synthase (iNOS) pathway has been implicated in inflammatory diseases and cellular injury. Inhibition of various genes related to inflammation, including iNOS is one of the major roles of well-known anti-inflammatory drugs. In the present study, the effects of ceramide analogs on iNOS expression and NO production were evaluated to investigate how ceramide and its structurally related analogs modulate NO-mecliated cellular signals and inflammation.   • Objective : This study was performed to evaluate the effect of Forsythiae Fructus (FF) ethanol extract on inflammatory cytokine production and its underlying mechanisms in mouse macrophages. Methods : Peritoneal macrophages from thioglycollate medium-injected mice were cultured and stimulated with lipopolysaccharide(LPS) or LPS/interferon(IFN)-${\gamma}$ for cytokine measurement and cellular signaling molecule analysis. Results : FF ethanol extract decreased the levels of secreted tumor necrosis factor(TNF)-${\alpha}$ and interleukin(IL)-6 in IFN-${\gamma}$/LPS-stimulated cells in a concentration-dependent manner. FF extract reduced IFN-${\gamma}$/LPS-induced STAT1 phosphorylation and LPS-induced p38 and JNK activation, but not ERK1/2 activity. The extract also inhibited LPS-induced $I{\kappa}B{\alpha}$ degradation through suppression of $I{\kappa}B{\alpha}$ kinase. Conclusions : These results suggest that FF ethanol extract affects the production of TNF-${\alpha}$ and IL-6 through inhibition of activation of STAT-1, $I{\kappa}B{\alpha}$, p38, and JNK.

알러지성 비염 병태 모델에 대한 방풍통성산(防風通聖散)의 항알러지 효과 (The Effect of Bangpungtongsung-San on Model of Allergic Rhinitis)

  • Background and Objective : Allergic rhinitis is a IgE-mediated hypersensitive reaction at nasal mucosa. In oriental medicine, Bangpungtongsung-San is clinically widely used for the treatment of the allergic rhinitis. The objective of this study is to investigate the effects of Bangpungtongsung-San on allergic rhinitis experimentally. Material and Methods : BALB/c mouse were divided into there groups: intact, control, experimental groups. Control and experimental group were induced allergic rhinitis by Ovalbumin as· the method of Levin and Vaz. Experimental group was orally administered the Bangpungtongsung-San for 28days. Total IgE, Interleukin-4, Interleukin-5 and $Interferon-{\gamma}$ were measured at three groups. The statistical significance was examined by Independent Samples T test. Results : 1. The experimental group shows increase of $IFN-{\gamma}$ by 17% compared with control group but there was no statistical significance. 2. The experimental group shows increase of IL-4 and IL-5 compared with control group but there was no statistical significance. 3. The experimental group shows increase of Total IgE and diminution of OVA-specific IgE by 32% compared with control group but there was no statistical significance. Conclusion : The effect of Bangpungtongsung-San on allergic rhinitis is not immunological but seems to be anti-imflammatory and anti-stress effect.

Immune-Enhancing Effect of Nanometric Lactobacillus plantarum nF1 (nLp-nF1) in a Mouse Model of Cyclophosphamide-Induced Immunosuppression

  • Nanometric Lactobacillus plantarum nF1 (nLp-nF1) is a biogenics consisting of dead L. plantarum cells pretreated with heat and a nanodispersion process. In this study, we investigated the immune-enhancing effects of nLp-nF1 in vivo and in vitro. To evaluate the immunostimulatory effects of nLp-nF1, mice immunosuppressed by cyclophosphamide (CPP) treatment were administered with nLp-nF1. As expected, CPP restricted the immune response of mice, whereas oral administration of nLp-nF1 significantly increased the total IgG in the serum, and cytokine production (interleukin-12 (IL-12) and tumor necrosis factor alpha (TNF-${\alpha}$)) in bone marrow cells. Furthermore, nLp-nF1 enhanced the production of splenic cytokines such as IL-12, TNF-${\alpha}$, and interferon gamma (IFN-${\gamma}$). In vitro, nLp-nF1 stimulated the immune response by enhancing the production of cytokines such as IL-12, TNF-${\alpha}$, and IFN-${\gamma}$. Moreover, nLp-nF1 given a food additive enhanced the immune responses when combined with various food materials in vitro. These results suggest that nLp-nF1 could be used to strengthen the immune system and recover normal immunity in people with a weak immune system, such as children, the elderly, and patients.

Inhibition of ERK1/2 by silymarin in mouse mesangial cells

  • The present study aimed to show that pro-inflammatory cytokines [tumor necrosis factor (TNF)-${\alpha}$, interferon (IFN)-${\gamma}$, and interleukin (IL)-$1{\beta}$] synergistically induce the production of nitric oxide (NO) production in mouse mesangial cells, which play an important role in inflammatory glomerular injury. We also found that co-treatment with cytokines at low doses (TNF-${\alpha}$; 5 ng/ml, IFN-${\gamma}$; 5 ng/ml, and IL-$1{\beta}$; 1.25 U/ml) synergistically induced NO production, whereas treatment with each cytokine alone did not increase NO production at doses up to 100 ng/ml or 50 U/ml. Silymarin, a polyphenolic flavonoid isolated from milk thistle (Silybum marianum), attenuates cytokine mixture (TNF-${\alpha}$, IFN-${\gamma}$, and IL-$1{\beta}$)-induced NO production. Western blot and RT-PCR analyses showed that silymarin inhibits inducible nitric oxide synthase (iNOS) expression in a dose-dependent manner. Silymarin also inhibited extracellular signal-regulated protein kinase-1 and -2 (ERK1/2) phosphorylation. Collectively, we have demonstrated that silymarin inhibits NO production in mouse mesangial cells, and may act as a useful anti-inflammatory agent.
