Study on coexistence through interference evaluation between 2.1GHz band and adjacent band for LTE-Advanced service (2.1GHz대역에서 LTE-Advanced 서비스를 위한 인접대역과의 간섭평가 및 양립방안 연구)

  • Currently, the three major Korean mobile operators hold a total of 390MHz of bandwidth, but at the current data traffic increase of almost 6 times per year, more frequency bandwidth should be secured in order to meet the exploding data traffic in the future. It is believed that 2.1GHz frequency band is suitable for mobile communication in the light of frequency characteristics and continuity of the band. In this paper, we perform compliance analysis with the international radio regulation for coexistence with the adjacent region in order to use 2.1GHz band for LTE-Advanced. In addition, we verify that 2.1GHz band can coexist with the adjacent band by conducting an interference evaluation.

Performance Comparison and Its Verification of Spectrum Sharing Technologies Using Interference Load Concept (간섭부하 개념을 이용한 주파수 공동사용 기술의 성능비교 및 검증)

  • In this paper, we proposed a concept of interference load to analyze the performance of spectrum sharing technologies in unlicensed frequency bands. The interference load can quantitatively compare the technical properties of various spectrum sharing technologies, such as frequency hopping(FH), duty cycle(DC), listen-before-talk(LBT). Therefore, it can help to evaluate whether a local regulation about spectrum sharing is reasonably established or not. In order to verify the suggested concept, we applied it to 2.4 GHz frequency bands. Also, we demonstrated a real-time test-bed. Two bit error rate(BER) curves for FH with 10 random channels and DC of 10 %, show such good agreement that our proposed concept is expected to be widely used to assess various spectrum sharing technologies.

Function of the pentose phosphate pathway and its key enzyme, transketolase, in the regulation of the meiotic cell cycle in oocytes

  • Objective: Previously, we identified that transketolase (Tkt), an important enzyme in the pentose phosphate pathway, is highly expressed at 2 hours of spontaneous maturation in oocytes. Therefore, this study was performed to determine the function of Tkt in meiotic cell cycle regulation, especially at the point of germinal vesicle breakdown (GVBD). Methods: We evaluated the loss-of-function of Tkt by microinjecting Tkt double-stranded RNAs (dsRNAs) into germinal vesicle-stage oocytes, and the oocytes were cultured in vitro to evaluate phenotypic changes during oocyte maturation. In addition to maturation rates, meiotic spindle and chromosome rearrangements, and changes in expression of other enzymes in the pentose phosphate pathway were determined after Tkt RNA interference (RNAi). Results: Despite the complete and specific knockdown of Tkt expression, GVBD occurred and meiosis was arrested at the metaphase I (MI) stage. The arrested oocytes exhibited spindle loss, chromosomal aggregation, and declined maturation promoting factor and mitogen-activated protein kinase activities. The modified expression of two enzymes in the pentose phosphate pathway, Prps1 and Rbks, after Tkt RNAi and decreased maturation rates were amended when ribose-5-phosphate was supplemented in the culture medium, suggesting that the Tkt and pentose phosphate pathway are important for the maturation process. Conclusion: We concluded that Tkt and its associated pentose phosphate pathway play an important role in the MI-MII transition of the oocytes' meiotic cell cycle, but not in the process of GVBD.

Knockdown of Radixin by RNA interference Suppresses the Growth of Human Pancreatic Cancer Cells in Vitro and in Vivo

  • Radixin, encoded by a gene on chromosome 11, plays important roles in cell motility, invasion and tumor progression. However, its function in pancreatic cancer remains elusive. In this study, radixin gene expression was suppressed with a lentivirus-mediated short-hairpin RNA (shRNA) method. We found that radixin shRNA caused down-regulation of radixin in PANC-1 cells, associated with inhibition of pancreatic cancer cell proliferation, survival, adhesion and invasive potential in vitro. When radixin-silenced cells were implanted in nude mice, tumor growth and microvessel density were significantly inhibited as compared to blank control cells or nonsense shRNA control cells. Thrombospondin-1 (TSP-1) and E-cadherin were up-regulated in radixin-silenced PANC-1 cells. Our results suggest that radixin might play a critical role in pancreatic cancer progression, possibly through invvolvement of down-regulation of TSP-1 and E-cadherin expression.

Down-Regulation of Mcl-1 by Small Interference RNA Induces Apoptosis and Sensitizes HL-60 Leukemia Cells to Etoposide

  • Background: Acute myeloid leukemia (AML) is a fatal hematological malignancy which is resistant to a variety of chemotherapy drugs. Myeloid cell leukemia-1 (Mcl-1), a death-inhibiting protein that regulates apoptosis, has been shown to be overexpressed in numerous malignancies. In addition, it has been demonstrated that the expression level of the Mcl-1 gene increases at the time of leukemic relapse following chemotherapy. The aim of this study was to target Mcl-1 by small interference RNA (siRNA) and analyze its effects on survival and chemosensitivity of acute myeloid leukemia cell line HL-60. Materials and Methods: siRNA transfection was performed with a liposome approach. The expression levels of mRNA and protein were measured by real-time quantitative PCR and Western blot analysis, respectively. Trypan blue assays were performed to evaluate tumor cell growth after siRNA transfection. The cytotoxic effects of Mcl-1 siRNA (siMcl-1) and etoposide were determined using MTT assay on their own and in combination. Apoptosis was quantified using a DNA-histone ELISA assay. Results: Transfection with siMcl-1 significantly suppressed the expression of Mcl-1 mRNA and protein in a time-dependent manner, resulting in strong growth inhibition and spontaneous apoptosis. Surprisingly, pretreatment with siMcl-1 synergistically enhanced the cytotoxic effect of etoposide. Furthermore, Mcl-1 down-regulation significantly increased apoptosis sensitivity to etoposide. No significant biological effects were observed with negative control siRNA treatment. Conclusions: Our results suggest that specific suppression of Mcl-1 by siRNA can effectively induce apoptosis and overcome chemoresistance of leukemic cells. Therefore, siMcl-1 may be a potent adjuvant in leukemia chemotherapy.

Feasibility Study on the Introduction of No Net Loss of Green (녹지총량제의 국외사례 및 국내 적용가능성)

  • Due to the increasing needs of land supply, green space has been continuously encroached and resulted considerable naturality deterioration in Korea. In order to overcome the problem, it is necessary to introduce a integrated system for managing the quantitative and qualitative aspects of green space. With this regard, this study is to derive the feasibility for the adoption of the 'no net loss of green' policy to Korea from German and Japanese experiences. In Germany, natural resources are protected and inevitable damages were compensated through the adoption of 'interference regulation'. In Japanese case, future green space and artificial green foundation should be specified on the green basic plan which pursuit the improvement of naturality. In order to introduce the 'no net loss of green' policy to Korea, not only awareness raising on integrated green management system but also amendment of related regulations are needed. In detail, restoration and recovery on the damaged nature should be clearly elucidated and indicators to evaluate the current naturality and calculation methods for the restoration should be developed. Should this integrated green management system introduced, deterioration of naturality by development activities could be minimized thorough the adoption of land development permission criteria and green space management methodologies.

Regulation of the Edwardsiella tarda Hemolysin Gene and luxS by EthR

  • Edwardsiella tarda is a pathogen with a broad host range that includes human and animals. The E. tarda hemolysin (Eth) system, which comprises EthA and EthB, is a noted virulence element that is widely distributed in pathogenic isolates of E. tarda. Previous study has shown that the expression of ethB is regulated by iron, which suggests the possibility that the ferric uptake regulator (Fur) is involved in the regulation of ethB. The work presented in this report supports the previous findings and demonstrates that ethB expression was decreased under conditions when the E. tarda Fur ($Fur_{Et}$) was overproduced, and enhanced when $Fur_{Et}$ was inactivated. We also identified a second ethB regulator, EthR, which is a transcription regulator of the GntR family. EthR represses ethB expression by direct interaction with the ethB promoter region. In addition to ethB, EthR also modulates, but positively, luxS expression and AI-2 production by binding to the luxS promoter region. The expression of ethR itself is subject to negative autoregulation; interference with this regulation by overexpressing ethR during the process of infection caused (i) drastic changes in ethB and luxS expressions, (ii) vitiation in the tissue dissemination and survival ability of the bacterium, and (iii) significant attenuation of the overall bacterial virulence. These results not only provide new insights into the regulation mechanisms of the Eth hemolysin and LuxS/AI-2 quorum sensing systems but also highlight the importance of these systems in bacterial virulence.

The Role of Caveolin-1 in Senescence and Ototoxicity of Differentiated Cochlear Hair Cell Line (UB/OC-1)

  • Caveolin may be a molecular target for modulation of aging process in cochlear hair cells and have association with oxotoxicity. First we investigated the basal expression of caveolin-1, caveolin-2, caveolin-3, nitric oxide synthase, and superoxide dismutase in UB/OC-1 cochlear hair cell line. By using a RNA interference technique, we investigated whether down-regulation of caveolin influenced telomerase activity and reactive oxygen species (ROS) production in cochlear hair cells. In addition, cisplatin and gentamycin, known ototoxic drugs, were administered to the cochlear cells to determine their impact on caveolin expression. Further attempts at elucidating cellular aging mechanism with caveolin and ototoxic drugs were carried out. The main discoveries were the presence of caveolin-1 in UB/OC-1 cells and that down-regulation of caveolin-1 reduced protein kinase A activity. Telomerase was activated by caveolin down-regulation and caveolin down-regulation inhibited oxidative stress at the mitochondrial level. When cisplatin and gentamycin were administered to the cochlear hair cells during a caveolin expression state, a decrease in telomerase activity and increase ROS activity was observed. Caveolin-1 may modulate the senescent mechanisms in cochlear cells. An increase in caveolin-1 levels can lead to ROS production in the mitochondria which may cause ototoxicity.

Analysis of Reference Signal for Immunity Test of Sound System and Television Broadcast Receiver (음향 기기 및 TV 방송 수신기의 내성 시험용 기준 신호 분석)

  • Intermittent or periodic signals with undulating amplitudes of mobile devices cause interference noise at audible frequency to the sound system which is installed nearby. In this regard, it is recently issued that the intermittently transmissions of LTE mobile phones cause the interference noises with sound systems, similar to that of GSM mobile phones. For retaining the immunity to noises, a national regulation KN20 suggests a 1 kHz AM 80 % and a CDMA modulation signal as reference signals for immunity test of sound and television broadcast receiver. In this paper, the effects of interference noises of AM and CDMA are compared with those of GSM and LTE, and the improvement of reference signal for national immunity test of sound systems is suggested. It is recommended to delete the CDMA modulation signal from reference signals for immunity test because it hardly cause interference noise with sound systems. On the other hand, it is desirable to select the modulation signal of LTE instead of CDMA as a reference signal for immunity test of sound system and television broadcast receiver because many people use the LTE service and it is analysed that the LTE signal causes considerable interference noises.

Improved Design Method of a EMI(Electro Magnetic Interference Noise for Wireless Video System in Vehicle (차량용 무선 비디오 시스템 내 EMI 노이즈 개선 방안)

  • In this paper, we analyzed various noise in the video stream system that have largely influence on EMI noise. We presented the remedy for these various noises and then designed the wireless video streaming system for a vehicle. To minimize EMI noise, we derived the improvement of noise characteristic from impedance matching, new design of the inner layer of the PCB line design and new design of high-speed data Interfaces. As a result, the final system showed 40[dBuV/m] and 47[dBuV/m] dB in the each regulation band.
