A cDNA microarray composed of 2,028 different ESTs from two shrimp species, Penaeus monodon and Masupenaeus japonicus, was employed to identify yellow head virus (YHV)-responsive genes in hemocytes of P. monodon. A total of 105 differentially expressed genes were identified and grouped into five different clusters according to their expression patterns. One of these clusters, which comprised five genes including cathepsin L-like cysteine peptidase, hypothetical proteins and unknown genes, was of particular interest because the transcripts increased rapidly ($\leq$ 0.25 hours) and reached high expression levels in response to YHV injection. Microarray data were validated by realtime RT-PCR analyses of selected differentially expressed transcripts. In addition, comparative analysis of the hemocyte transcription levels of three of these genes between surviving and non-surviving shrimp revealed significantly higher expression levels in surviving shrimp.
A target bacterial strain of interest for use in Red-based recombineering may already encode resistance to antibiotic markers used with current Red recombination tools, such that the resistance cannot be removed. Such cases include those where markers are needed to maintain an unstable genetic element co-resident in the strain or those where the genetic source of resistance is not known. We report the availability of PCR templates with FRT-flanked mutagenesis cassettes and plasmids encoding Red recombination functions that contain marker combinations not currently available on widely disseminated lambda Red molecular reagents. The functionality of these convenient alternative tools is demonstrated.

The understanding of real-world application of scientific knowledge such as the evolutionary theory has been emphasized in science curriculum. Despite this, many students and biology educators misunderstand the use and practical application of the evolution theory. The purpose of this study is to explore variables related to the understanding of practical application of evolution. Methodologically, we employed large-scale survey that examined three independent variables (major, academic year, and religion) and five dependent variables (understanding of practical application of evolution, knowledge of, interest in, and acceptance of evolution, and religiosity). Two hundred and thirty three biology pre-service teachers and 405 non-biology pre-service teachers participated in the survey. The results demonstrate that the biology teacher education program does not help in increasing the understanding of the practical application of evolution. Rather, the understanding of practical application of evolution was associated with their religion. In addition, the understanding of the practical application of evolution was associated with the acceptance of and interest in evolution. The results of this study inform the development of new biology teacher education programs to improve their understanding of practical application of the evolution theory. We also suggest further areas of research for other scientific concepts that could be used to gauge teachers' understanding of the practical application of scientific knowledge.
Lammas growth, a rare phenomenon for Korean red pine (Pinus densiflora), occurred in 2006. Lammas shoots showed higher frequency and longer length in Seoul's hotter urban center than in urban boundary or suburban forest sites. Frequency and length showed a close correlation with urbanization density and vegetation cover expressed in NDVI. Air temperature in the late summer of 2006 was more than $1^{\circ}C$ higher than an average year. Of the predominant environmental signals that modulate bud flush, only temperature changed significantly during the year. Differences in temperature between the urban centers, urban boundaries and suburban forests correlated with varying land-use density. The rise in temperature likely spurred lammas growth of the Korean red pine. Symptoms of climate change are being detected throughout the world, and its consequences will be clearer in the future. Considerate interest in the responses of ecological systems to the variable changes is required to prepare for unforeseeable crises. Monitoring of diverse ecological phenomena at Long Term Ecological Research sites could offer harbingers of change.

The value of silkworms as functional health food materials has increased, as has the interest in its disease control for stable production, and in the economic value of entomopathogenic microorganisms. In this study, we isolated and identified disease-causing fungi from white muscardine silkworms, and confirmed whether this strain could produce white muscardine silkworms. For the analysis of the cause of white muscardine disease in the infected silkworms, the fungi and prokaryotes causing the disease were identified, isolated, and identified using metagenome analysis. Metagenomic analysis detected a large amount of the fungus Metarhizium rileyi in silkworms, and a large amount of the bacterium Enterococcus mundtii, which was presumed to be the causative agent of the disease. For accurate identification of the fungi, these were purified by culture medium, and sequencing and phylogenetic tree analyses were performed using an internal transcribed spacer. As a result, M. rileyi, Cladosporium cladosporioides, and C. tenuissimum were identified. In general, M. rileyi is known to form green conidia, but in this study, white-yellow conidia were formed, indicating that the exact causative agent of the fungal disease cannot be estimated by diagnosing the symptoms. Thus, a diagnostic method is necessary for the continuously collection of required pathogens, and identifying their morphological and genetic characteristics.

Cryphonectria parasitica, chestnut blight fungus, has a characteristic of decreasing pathogenicity when infected with Cryphonectria hypovirus 1. C. parasitica is known to be one of the most representative model systems used to observe the interaction between viruses, plants and fungi. The mitogen-activated protein kinase (MAPK) pathway, which is well conserved in various organisms ranging from yeast to humans, functions in relaying phosphorylation-dependent signals within MAPK cascades to diverse cellular functions involved in the regulation of pheromone, cell wall integrity, and osmotolerance in filamentous fungi. Several genes in the MAPK pathway were revealed to be regulated by hypovirus, or to be involved in pathogenicity in C. parasitica. Among these pathways, the CWI pathway has aroused interest because CpBck1, an ortholog of yeast Bck1 (a CWI MAPKKK), was previously reported to be involved in cell wall integrity and sectorization. Interestingly, sporadic sectorization was observed in the CpBck1 mutant and sectored phenotypes were stably inherited in the progeny that were successively transferred from sectored mycelia. In this study, we analyzed the biological function of CpSlt2, downstream gene of CpBck1, to confirm whether the sectorization phenomenon occurred in the specific single gene or cell wall integrity (CWI) pathway. As results, the CpSlt2-null mutant exhibited marked changes in colonial growth, near absence of conidiation and aerial hyphae, abnormal pigmentation, CWI-related phenotypic defects, and dramatically impaired virulence. As cultivation of the mutant strains progressed, the majority of the colonies showed sporadic sectorization and mycelia from the sectored area stably maintained the sectored phenotype. These results suggest that the unique sectorization is CWI pathway-specific, though the components in the same CWI pathway have common and specific functions.
The outcome of plant-pathogen interactions is influenced significantly by endogenous small molecules that coordinate plant defence responses. There is currently tremendous scientific and commercial interest in identifying chemicals whose exogenous application activates plant defences and affords protection from pathogen infection. In this review, we provide a survey of compounds known to induce disease resistance in plants, with particular emphasis on how each compound was originally identified, its putative or demonstrated mechanism of defence induction, and the known biological target(s) of each chemical. Larger polymeric structures and peptides/proteins are also discussed in this context. The quest for novel defence-inducing molecules would be aided by the capability for high-throughput analysis of candidate compounds, and we describe some issues associated with the development of these types of screens. Subsequent characterization of hits can be a formidable challenge, especially in terms of identifying chemical targets in plant cells. A variety of powerful molecular tools are available for this characterization, not only to provide insight into methods of plant defence activation, but also to probe fundamental biological processes. Furthermore, these investigations can reveal molecules with significant commercial potential as crop protectants, although a number of factors must be considered for this potential to be realized. By highlighting recent progress in the application of chemical biology techniques for the modulation of plant-pathogen interactions, we provide some perspective on the exciting opportunities for future progress in this field of research.
Recently, the interest in natural products for the treatment of cancer is increasing because they are the pre-screened candidates. In the present study, we demonstrate the therapeutic effect of celastrol, a triterpene extracted from the root bark of Chinese medicine on gastric cancer. The proliferation of AGS and YCC-2 cells were most sensitively decreased in six kinds of gastric cancer cell lines after the treatment with celastrol. Celastrol inhibited the cell migration and increased G1 arrest in cell-cycle populations in both cell lines. The treatment with celastrol significantly induced autophagy and apoptosis and increased the expression of autophagy and apoptosis-related proteins. We also found an increase in phosphorylated AMPK following a decrease in all phosphorylated forms of AKT, mTOR and S6K after the treatment with celastrol. Moreover, gastric tumor burdens were reduced in a dose-dependent manner by celastrol administration in a xenografted mice model. Taken together, celastrol distinctly inhibits the gastric cancer cell proliferation and induces autophagy and apoptosis.

To investigate the degree of individuals' concept and awareness of insects, a survey study was conducted with students and adults living in Korea. The misconception rate for insects was about 50% for both students and adults, but it was lower for students and people who had experienced insect-related events than for adults and those who had not. The highest misconception rate was obtained in answer to a question about the basic structure of an insect. Most people had a high awareness of insects. Significant differences and correlations for the awareness of insects were found between students and adults, men and women, people who had experienced insect-related events and those who had not. The experience of an insect-related event most influenced awareness of insects. These results suggest that increasing people's interest in insects and utilizing insects in treatment situations may be beneficial for the field of mental healthcare.
Lichen is one of the most widely distributed eucaryotic organisms in the world. Lichen is the result of a symbiotic association between two unrelated organisms - a fungus and an alga (or cyanobacterium). In USA, Japan and European countries, there has been a resurgence of interest in lichens as source of novel, pharmacologically active molecules during the last 20 years. However, lichen researches in Korean lichens were mainly focused on investigation of Korean lichen flora and most of them were primitive and short-term based projects until 1990s. In this communication, general introduction of lichens was attempted to draw the attention of Korean researchers to Korean lichen bioresource. Research activities on Korean lichens during the past were also summarized. Recent progress in Korean lichen research is briefly discussed with emphasis of KoLRI (Korean Lichen Research Institute) activities and roles in national researches projects on bioresource development in Korea.
