Antioxidative Effects of Korean Bamboo Trees, Wang-dae, Som-dae, Maengjong-juk, Jolit-dae and O-juk (한국산 왕대, 솜대, 맹종죽, 조릿대 및 오죽의 항산화 효과)

  • Bamboo trees have been used for a long time as folk remedies for treatment of hypertension and stroke symptoms in Oriental regions. These pharmaceutical effects of bamboos look like to be related to its antioxidant capacity and phytochemicals in bamboos. To evaluate the antioxidative effects of bamboo trees, five kinds of bamboo varieties dominant in Korean peninsular were chosen and determined its total antioxidaive activities, free radical scavenging activities and nitrite scavenging activities by TEAC (Trolox Equivalent Antioxidant Capacity) assay, DPPH and Griess reagent assay using in vitro system, respectively. To evaluate the correlation between antioxidative activities and Maillard reaction during hot water extraction, contents of reducing sugar and total nitrogen and brown color intensity at 420 nm were determined. When total antioxidative activities, free radical scavenging activities and nitrite scavenging activities of five kinds of bamboo trees were compared, wang-dae (Phyllostachys bambusoides S. et Z.) showed the strongest effect among samples, although all kinds of extracts showed relatively strong effects against oxidation. The bamboo culms extract showed stronger antioxidative effects than that of bamboo leaves. In each fraction obtained from 70% ethanol extract, antioxidative effect were increased in order of dichloromethane>ethyl acetate>butanol>water>hexane fraction. In reducing sugar analysis of extracts, reducing sugar contents of water extracts were higher than that of 70% ethanol extracts and wang-dae water extract showed the highest level which was 708.92 mg/g. Total nitrogen contents of the extracts were $1.785{\sim}2.605\;mg%$ and contents in water extracts were lower than that in 70% ethanol extracts. Brown color intensity at 420 nm showed similar tendency with results in reducing sugar contents.

Acute Cerebral Infarction in a Rabbit Model: Perfusion and Diffusion MR Imaging (가토의 급성 뇌경색에서 관류 및 확산강조 자기공명영상)

  • Purpose : The present study was undertaken to evaluate the usefulness of cerebral diffusion (DWI) and perfusion MR imaging (PWI) in rabbit models with hyperacute cerebral ischemic infarction. Materials and Methods : Experimental cerebral infarction were induced by direct injection of mixture of Histoacryl glue, lipiodol, and tungsten powder into the internal cerebral artery of 6 New-Zealand white rabbits, and they underwent conventional T1 and T2 weighted MR imaging, DWI, and PWI within 1 hour after the occlusion of internal cerebral artery. The PWI scan for each rabbit was obtained at the level of lateral ventricle and 1cm cranial to the basal ganglia. By postprocessing using special imaging software, perfusion images including cerebral blood volume (CBV), cerebral blood flow (CBF), and mean transit time (MTT) maps were obtained. The detection of infarcted lesion were evaluated on both perfusion maps and DWI. MTT difference time were measured in the perfusion defect lesion and symmetric contralateral normal cerebral hemisphere. Results : In all rabbits, there was no abnormal signal intensity on T2WI. But on DWI, abnormal high signal intensity, suggesting cerebral infarction, were detected in all rabbits. PWI (rCBV, CBF and MTT map) also showed perfusion defect in all rabbits. In four rabbits, the calculated square of perfusion defect in MTT map is larger than that of CBF map and in two rabbits, the calculated size of perfusion defect in MTT map and CBF map is same. Any rabbits do not show larger perfusion defect on CBF map than MTT map. In comparison between CBF map and DWI, 3 rabbits show larger square of lesion on CBF map than on DWI. The others shows same square of lesion on both technique. The size of lesion shown in 6 MTT map were larger than DWI. In three cases, the size of lesion shown in CBF map is equal to DWI. But these were smaller than MTT map. The calculated square of lesion in CBF map, equal to that of DWI and smaller than MTT map was three. And in one case, the calculated square of perfusion defect in MTT map was largest, and that of DWI was smallest. Conclusion : DWI and PWI may be useful in diagnosing hyperacute cerebral ischemic infarction and in e-valuating the cerebral hemodynamics in the rabbits.

Locally Advanced, Unresectable Pancreatic Cancer Treated by Stereotactic Radiation Therapy (국소적으로 진행된, 절제 불가능한 췌장암에서 정위 방사선 치료)

  • Puroose: In order to find out whether stereotactic radiation therapy (RT) using CyberKnife (CK) could improve survival rate and lower acute toxicity compared to conventional RT. Materials and Methods: From April 2003 through April 2004, 19 patients with Eastern Cooperative Oncology Group (ECOG) performance status ${\leq}3$ and locally advanced pancreas cancer without distant metastasis, evaluated by CT or PET/CT, were included. We administered stereotactic RT consisting of either 33 Gy, 36 Gy or 39 Gy in 3 fractions to 6, 4 and 9 patients, respectively, in an effort to increase the radiation dose step by step, and analyzed the survival rate and gastrointestinal toxicities by the acute radiation morbidity criteria of Radiation Therapeutic Oncology Group (RTOG). Prognostic factors of age, sex, ECOG performance score, chemotherapy, bypass surgery, radiation dose, CA 19-9, planning target volume (PTV), and adjacent organ and vessel invasion on CT scan were evaluated by Log Rank test. Results: The median survival time was 11 months with 1-year survival rate of 36.8%. During follow-up period (range $3{\sim}20$ months, median 10 months), no significant gastrointestinal acute toxicity (RTOG grade 3) was observed. In univariate analysis, age, sex, ECOG performance score, chemotherapy, bypass surgery, radiation dose, CA 19-9 level, and adjacent organ and vessel invasion did not show any significant changes of survival rate, however, patients with PTV (80 cc showed more favorable survival rate than those with PTV>80 cc (p-value<0.05). In multivariate analysis, age younger than 65 years and PTV>80 cc showed better survival rate. Conclusion: In terms of survival, the efficacy of stereotactic radiation therapy using CK was found to be superior or similar to other recent studies achieved with conventional RT with intensive chemotherapy, high dose conformal RT, intraoperative RT (IORT), or intensity modulated RT (IMRT). Furthermore, severe toxicity was not observed. Short treatment time in relation to the short life expectancy gave patients more convenience and, finally, quality of life would be increased. Consequently, this could be regarded as an effective novel treatment modality for locally advanced, unresectable pancreas cancer. PTV would be a helpful prognostic factor for CK.

Study on nutrition, dietary and health status of middle-aged Korean men according to sedentary hours: based on the 2019 Korea National Health and Nutrition Examination Survey (중장년 한국 남성의 좌식 시간에 따른 영양, 식이 및 건강행태 연구: 국민건강영양조사 제8기 1차년도(2019년) 자료를 이용하여)

  • Purpose: Adult Korean men belonging to the main economically active population are known to have long sedentary hours. This study was undertaken to determine the difference and relevance of sedentary hours on the nutrition, diet, and health status of adult men, and to suggest how to prevent health risk factors. Methods: Subjects (n = 1,068) were classified into 4 groups based on their sedentary hours, ranging from the first quartile (Q1) having the least hours spent sitting, to the fourth quartile (Q4) spending the longest hours. Results: Subjects belonging to Q4 had the lowest average age, the largest waist circumference, and the highest level of education. Among those engaged in economic activities, the ratio of white-collar workers was significantly higher in Q4. Accordingly, the rate of not doing high-intensity or moderate-intensity physical activity while working was also the highest in Q4. A significant difference was obtained in the drinking frequency between groups, but this was found to be associated with the average working hours rather than sedentary hours. The proportion of not doing aerobic exercise was higher with longer sitting hours. The highest diagnosis of diabetes (8.8%) was obtained in the Q4 group. Among the factors related to cardiovascular disease, only low density lipoprotein-cholesterol showed a significant difference, with Q4 being significantly higher than Q1. Considering energy and nutrient intake, vitamin B1 and calcium intake were the lowest in the group with the longest sitting hours, as well as the least consumption of vitamin C than the recommended estimated average requirement. Conclusion: The results of this study suggest that the health and nutritional status of Korean adult men are affected by sedentary hours. This should be recognized as a health risk factor and guidelines need to be developed for sedentary lifestyle management.

Effect of thinning ratio on the forest environment and fruiting of ectomycorrhizal mushrooms in a Pinus densiflora stand (소나무림에서 간벌률이 산림 내 환경과 외생균근성 버섯 발생에 미치는 영향)

  • To investigate the effect of thinning intensity on environmental factors and ectomycorrhizal mushroom fruiting in forest ecosystems, we studied canopy closure, throughfall, soil temperature, soil moisture, light response of understory vegetation, and ectomycorrhizal mushroom fruiting in a 10-year-old pine forest after 34%, 45%, and 60% thinning. Canopy closure was significantly higher in the 34% treatment and control plots, ranging from 80-85% in April. However, in November, all thinning treatment plots showed a decrease of approximately 5-10% compared with the control plot. The 60% treatment plot had over 200 mm of additional throughfall compared with the control plot, and monthly throughfall was significantly higher by more than 100 mm in October. The soil temperature in each treatment plot increased significantly by up to 1℃ or more compared with the control plot as the thinning rate increased. The soil moisture increased by more than 5% in the thinning treatment plots during rainfall, particularly in the 34% treatment plot, where the rate of moisture decrease was slower. The photosynthetic rate of major tree species (excluding Pinus densiflora)was highest in Quercus mongolica, with a rate of 7 µmolCO2·m-2·s-1. At a lightintensity of 800 μmol·m-2·s-1, Q. mongolica showed the highest photosynthetic level of 6 ± 0.3 μmolCO2·m-2·s-1 in the 45% treatment. The photosynthetic rate of Fraxinus sieboldiana and Styrax japonicus increased as the thinning intensity increased. The Shannon-Wiener index of mycorrhizal mushrooms did not significantly differ among treatments, but the fresh weight of mushrooms was approximately 360-840 g higher in the 34% and 45% treatments than in the control. Additionally, the fresh weight of fungi in the 60% treatment was 860 g less than that in the control. There were more individuals of Amanita citrina in the control than in the thinning treatment, while Suillus bovinus numbers increased by more than 10 times in the 34% thinning treatment compared with the control.

Modeling the Effects of Forest Management Scenarios on Aboveground Biomass and Wood Production: A Study in Mt. Gariwang, South Korea (산림경영활동에 따른 수종별 지상부생물량 및 목재생산량 변화 모델링: 가리왕산 모델숲을 대상으로)

  • The forest protection policies implemented in South Korea have resulted in the significant accumulation of forest. Moreover, the associated public interest has also been closely evaluated. As forests mature, there arises a need for forest management (FM) practices, such as thinning and harvesting. It is therefore essential to perform a scientific analysis of the long-term effects of FM. In this study, conducted in Mt. Gariwang, the effect of FM on forest succession and wood production (WP) were evaluated based on changes in aboveground biomass (AGB) using the LANDIS-II model. The FM consists of three scenarios (Selection, Shelterwood, and Two-stories), characterized based on the harvest intensity, frequency, and period. The model was applied to changes in the forest over 200 years. All scenarios show that the total AGB decreased immediately after thinning and harvesting. However, AGB recovery time differed among scenarios, with recovery to preharvest level occurring from 15 to 50 years after harvest; further, after 200 years, harvested forests had a greater total AGB than forests without FMs In particular, the changes in AGB of each species was different depending on its shade tolerance. The AGB of currently dominant shade-intolerant and mid-tolerant species decreased dramatically after harvesting. However, shade-tolerant species, dominant in the understory, continued to grow but were not harvested due to their small size. The cumulative WP for each scenario was estimated at 545.6, 141.6, and 299.9 tons/ha in Selection, Shelterwood, and Two-stories, respectively. The composition of WP differed according to harvest intensity and period. Most WP originated from shade-intolerant and mid-tolerant species in the early period. Later, most WP was from shade-tolerant species, which became dominant. The modeling approach used in this study is capable of analyzing the long-term effects of FM on changes in forests and WP. This study can contribute to decision making to guide FM methods for a variety of purposes, including WP and controlling forest composition and structure.

Emulsion Polymerization of Octamethylcyclotetrasiloxane under Ultrasonic Irradiation (고강도 초음파를 이용한 Octamethylcyclotetrasiloxane의 에멀전 중합)

  • Emulsion polymerization of octamethylcyclotetrasiloxane (OMCTS) was conducted under ultrasonic irradiation. Two sources of ultrasound with different intensities and frequencies of 20 KHz and 40 KHz were used for horn and bath type reactor, respectively. A combined process of horn and bath was also investigated. The effectiveness of the reaction systems was investigated by measuring conversion as well as intrinsic viscosity of the products. The influence of reaction temperature and sonication time on the progress of sonochemical polymerization was examined. It was found that conversion of greater than 80% and high viscosity were achieved within a few minutes of sonication in a horn type reactor, however, conversion and viscosity showed maximum values depending upon the sonication time. In a bath type reactor where a relatively weak intensity was maintained, longer duration time of more than one hour of sonication was required to reach a high level of conversion and viscosity. Compared with the horn type system, the conversion and viscosity in the bath type reactor were increased along with the sonication time. When the polymerization was carried out in a combined system of horn and bath, the evolution of conversion and molecular weight was quite different from the other cases. For the given geometry of reaction system, acoustic analysis using a commercial software was carried out and the results were correlated with experimental observation.

Neuroprotective Effects of Pinelliae Rhizoma Water-Extract by Suppression of Reactive Oxygen Species and Mitochondrial Membrane Potential Loss in a Hypoxic Model of Cultured Rat Cortical Cells. (배양대뇌신경세포 저산소증모델에서 유해산소생성억제 및 사립체막전위 소실방지에 의한 반하(半夏)의 신경세포사 억제 효능)

  • Oxidative stress by free radicals is a major cause of neuronal cell death. Excitotoxicity in hypoxia/ischemia causes an increase in reactive oxygen species (ROS) and a loss of mitochondrial membrane potential (MMP), resulting in dysfunction of the mitochondria and cell death. Pinelliae Rhizoma (PR) is a traditional medicine for incipient stroke. We investigated the effects of PR Water-Extract on the modulation of ROS and MMP in a hypoxic model using cultured rat cortical cells. PR Water-Extract was added to the culture medium at various concentrations (0.25${\sim}$5, 5.0 ${\mu}g/ml$) on day in vitro 12(DIV12), given a hypoxic shock (2% $O_2$/5% $CO_2$, $37^{\circ}C$, 3 hr), and cell viability was assessed on DIV15 by Lactate Dehydrogenase Assay (LDH assays). PR Water-Extract showed a statistically significant effect on neuroprotection (10${\sim}$15% increase in viability; p<0.01) at 1.0 and 2.5 ${\mu}g/ml$ in normoxia and hypoxia. Measurement of ROS production by $H_2DCF-DA$ stainings showed that PR Water-Extract efficiently reduced the number and intensity of ROS-producing neurons, especially at 1 hr post shock and DIV15. When MMP was measured by JC-1 stainings, PR Water-Extract efficiently maintained high-energy charged mitochondria. These results indicate that PR Water-Extract protects neurons in hypoxia by preventing ROS production and preserving the cellular energy level.

Effect of Several Herbal Medicines on Atopic Dermatitis Patients' SCORAD and Cytokine Levels (수종 한약 처방이 아토피 피부염 환자의 SCORAD 및 Cytokine 변화에 미치는 영향)

  • The purpose of this study is to determine whether the clinical activity of patients with atopic dermatitis(AD) correlate with their serum cytokine levels. 86 patients with AD were treated with KHS, YMH and GHT. On each visit to the clinic, their skin status was evaluated using the SCORAD index and serum IL-4, $IFN-{\gamma}$, IL-12, IL-13, IL-2 and IL-10 were measured by ELISA. After oral administration of herbal medicine, patients' skin status and subjective parameters such as pruritus and sleeplessness were significantly improved according to regression analysis. In particular, patients aged 0-9 showed stronger responses to treatment, which implies that the younger patients aged, the easier the treatment by herbal formula became. In analysis of the type of chosen herbal medicines, YMH treated group showed a stronger response in reduction of affected lesion, intensity of a lesion and subjective symptoms, while KHS treated group showed the least reduction. The levels of serum IL-4, $IFN-{\gamma}$ and IL-2 correlated with disease activity. Moreover, in patients under 0-9, the IL-4 level had a corrleation with disease activity; patients under 10-19 showed a correlation between the levels of IL-4 and IL-2 and disease activity; patients under 20-29 showed a correlation between the levels of IL-2, IL-12 and $IFN-{\gamma}$ and disease activity. In comparison with chosen herbal medicines, the groups treated with YMH or GHT treated group showed a correlation between the levels of IL-4, IL-12 and IL-2 and disease activity while KHS treated group did not. Our data suggest that serum levels of IL-4 and IL-13 are good indicators of clinical improvement in treating AD.

  • Today, companies exist amid unlimited competition and uncertain adminstration environment, so hands-on workers are being exposed to various job requirements and stress due to this. Especially, employees in the design firm are physically difficult because of heavy working intensity unlike other industrial field, and because the process is interrelated unlike working of a five-day workday of the other industry, it is difficult to guarantee periodic holidays and vacation. In addition, recently, while BIM is introduced in the construction industry, various changes in the individual role as well as organizational dimension such as business environment and scope & role, etc. are required, so it is the actual situation that the category and factors of job stress due to this are being also changed. Accordingly, this research has grasped important and weak sectors according to characteristics such as career, position, gender, education and existence/nonexistence of BIM template, etc. by confirming a job stress level and inducing factors of hands-on workers at the design firm. This research has a meaning in constructing a business environment of a design firm that is a main participant of BIM-based projects that are expected to have high utilization in the future.
