The Protective Role of Gleditsiae fructus against Streptococcus pneumoniae (폐렴 구균에 대한 조협의 보호 역할 연구)

  • Natural products have been used to mitigate the effects of cancer and infectious diseases, as they feature diverse bioactivities, such as antioxidant, antibacterial, anti-inflammatory, and immunomodulatory effects. Here, we chose 10 natural products that are well-known as pulmonary enhancers and investigated their bactericidal effects on Streptococcus pneumoniae. In the disk diffusion assay, the growth of S. pneumoniae was significantly regulated by G. fructus treatment regardless of extraction method used. We first adopted spraying as a novel delivery method for G. fructus. Interestingly, mice exposed to G. fructus three times a day for 2 weeks were resistant to S. pneumoniae intranasal infection (shown both through body weight loss and survival rates compared to the control group). Moreover, we confirmed that exposure to G. fructus regulated the colonization of the bacteria despite the sustained inflammation in the lung after exposure to S. pneumoniae, indicating that migrated inflammatory immune cells may involve a host defense mechanism against pulmonary infectious diseases. While a similar number of granulocytes (CD11b+Ly6C+Ly6G+), neutrophils (CD11b+Ly6CintLy6G+), and monocytes (CD11b+Ly6CintLy6G-) were found between groups, a significantly increased number of alveolar macrophages (CD11b+CD11chiF4/80+) was detected in BAL fluids of mice pre-exposed to G. fructus at 5 days after S. pneumonia infection. Taken together, our data suggest that this usage of G. fructus can induce protective immunity against bacterial infection, indicating that facial spray may be helpful in enhancing the defense mechanism against pulmonary inflammation and in evaluating the efficacy of natural products as immune enhancers against respiratory diseases.

  • This study is aimed at the development of a comprehensive web-based partial differential equation solver (WPDES) using multidimensional finite element method, which can be operated on the basis of world wide web. Overall issues of engineering and environmental information management and facility control could be implemented using this solver. This paper describes the development technique of the model, which is first part on development of partial differential equation solver. Conventional commercial general solver of computational fluid dynamics problems were investigated. All the relevant environmental models were analyzed to develop integrated environmental management system using WPDES. The governing equations and the parameters of investigated models were analyzed and integrated. Several numerical modules were invented for each partial differential term in partial differential equation of many related modeling problems. Each module was coded in the fashion of object oriented method, and was combined independently for the overall governing equation. WPDES has unique characteristic, which can analyze the problem through the suitable combination of modules without development of additional models for each environment problem with different governing equation, main variables, and parameters.

  • The rapid growth of algae occurs can induce the algae bloom when nutrients are supplied from anthropogenic sources such as fertilizer, animal waste or sewage in runoff the water currents or upwelling naturally. The algae blooms creates the human health problem in the environment as well as in the water resource managers including hypoxic dead zones and harmful toxins and pose challenges to water treatment systems. The algal blooms in the source water in water treatment systems affects the drinking water taste & odor while clogging or damaging filtration systems and putting a strain on the systems designed to remove algal toxins from the source water. This paper propose the emerging In-Situ self-diagnosable smart algae sensing device with wireless connectivity for smart remote monitoring and control. In this research, we developed the In-Site Algae diagnosable sensing device with wireless sensor network (WSN) connectivity with Optical Biological Sensor and environmental sensor to monitor the water treatment systems. The proposed system emulated in real-time on the water treatment plant and functional evaluation parameters are presented as part of the conceptual proof to the proposed research.

Compatibility of Entomopathogenic fungus Lecanicillium attenuatum and Pesticides to control Cotton Aphid, Aphis gossypii

  • Concerns of entomopathogenic fungi as alternative pest control agents are increasing even though chemical pesticides have been used as the main control agents for pests and diseases in crop production. This study was conducted to test the influence of fungicides and insecticides on an isolate of Lecanicillium attenuatum that was reported to have the pathogenicity against cotton aphid, because fungicides and/or insecticides can apply with mycopesticides simultaneous, before and/or after. Fungicides fenbuconazole+thiram and propineb inhibited the spore germination and mycelial growth of L. attenuatum CS625; dimethomorph and procymidone did not affect spore germination or mycelial growth. The insecticide abamectin, deltamethrin, imidachropride, and spinosad had no detrimental effects on spore germination or mycelial growth. Therefore, these results demonstrated that careful selection of pesticides and fungicides can be applied to the integrated pest and disease control with microbial pesticide.

Growth and Reproductive Performance of Small Ruminants under Integrated Livestock-Oil Palm Production System

  • The effects of supplementation with mixed fodder or concentrate were investigated on the performance of a) growth of male lambs and kids grazing extensively under oil palm trees and b) reproduction of ewes and goats stall-fed with native herbage. Supplemented kids and lambs had higher average daily weight gain than controls but the effect was only significant for those which received concentrate (p < 0.05). Final body weight was only significantly different from controls for lambs supplemented with concentrate (p < 0.05). The ages at puberty, first mating, first conception and first kidding of supplemented goats were about 110 days earlier than those for controls (p < 0.05 or better). Supplemented goats had first mating and conceived at lower body weights (p < 0.01) than those in control groups. Different feeding regimes had no effects (p > 0.05) on the reproductive performance of ewes apart from highest body weight of first lambing in animals supplemented with concentrate (p < 0.05). Native herbage available under oil palm trees of 5 years old was sufficient for growth and reproduction of sheep and goats. Concentrate was better than mixed fodder as supplement for enhancing the reproductive performance of goats but both appeared to have limited effects on the performance of reproduction in ewes and growth in male kids.

  • We describe here a novel micro total analysis system for the purification and identification of the affinity-captured proteins. Also we demonstrated the mass analysis of the Carcinoembrionic antigen (CEA) and Alpha femtoprotein which were chosen as the target cancer marker. For MALDI-TOF analyses, the proteins should to be separated from a protein mixture and be concentrated when needed. This procedure usually takes a long time even before protease-digested samples are to be obtained from them. Here, we describe integrated and efficient micro chip for protein purification and digestion for MALDI-TOF analyses. At first, disease protein is purified by passing the micro chamber from a protein mixture or human whole serum and released from the micro affinity beads by thermal heating. Purified protein is then transfer to the hole for trypsin digestion. The final sample is analyzed by MALDI-TOF. All the processes could be finished successfully within one hour, which renders MALDI-TOF analyses of a target protein quite simple.

  • Liquid chromatography based mass spectrometry (LC-MS) is a key technology for analyzing highly complex and dynamic proteome samples. With highly accurate and sensitive LC-MS analysis of complex proteome samples, efficient data processing is another critical issue to obtain more information from LC-MS data. A typical proteomic data processing starts with protein database search engine which assigns peptide sequences to MS/MS spectra and finds proteins. Although several search engines, such as SEQUEST and MASCOT, have been widely used, there is no unique standard way to interpret MS/MS spectra of peptides. Each search engine has pros and cons depending on types of mass spectrometers and physicochemical properties of peptides. In this study, we describe a novel data process pipeline which identifies more peptides and proteins by correcting precursor ion mass numbers and unifying multi search engines results. The pipeline utilizes two open-source software, iPE-MMR for mass number correction, and iProphet to combine several search results. The integrated pipeline identified 25% more proteins in mouse epididymal adipose tissue compared with the conventional method. Also the pipeline was validated using control and colitis induced colon tissue. The results of the present study shows that the integrated pipeline can efficiently identify increased number of proteins compared to the conventional method which can be a breakthrough in identification of a potential biomarker candidate.

BIOLOGICAL CONTROL OF GINSENG ROOT ROTS WITH SOIL AMENDMENTS

  • The phenomenon of 'soil sickness' is one of the most important limiting factors for ginseng(Panax ginseng) production in Korea. The principal cause is known to be due to the root rots caused by Cylindrocarpon destructans and Fusarium solani. Attempts were made to control the root rots with non-polluting cultural methods or soil amendments. Among the nine soil amendments tested, crab shell, cow bone and pig feces were selected for further testing. Each of the three amendments increased the populations or various actinomycetes in the range of 10-25 times over that of non-amended soil, whereas the population of C. destructans was reduced to about $50-70\%$ as compared with the control. Five isolates of Streptomyces with clear zones on chitin-agar medium were selected and then tested for their antagonistic effects on C. destructans. When anyone of the five isolates of Streptomyces and C. destructans was grown together in a modified peptone broth, growth of the latter was highly inhibited. When three levels of crab shell, cow bone, or pig feces were used to amend potted soil infested with C. destruetans, the root rot ratings of ginseng seedlings were reduced to less than one half in all the treatments as compared to the control. In another similar experiment, crab shell and cow bone amendments resulted in almost complete control of the seedling root rots in soil infested with C. destructans or F. solani. In conclusion, biological control with soil amendments of ginseng root rots caused by C. destructans and F. solani was successful. Further basic studies should be pursued using soil amendments for better control. In addition, field experiments are needed to complement the soil amendment control measures in an integrated pest control program.

Web Based Environmental Management System using Predictive Spatial Information Models (예측적 공간정보 모형을 이용한 Web 기반의 환경관리시스템의 개발 및 적용)

  • This study is aimed at the development of comprehensive environmental management system, which can be operated on the basis of world wide web, as a topic of G7 project. Even though there should be lots of works remaining to achieve this goal, preliminary products can be summarized as follows : 1) integrated environmental information management system, 2) web based control engine, 3) surface water environment management system, 4) subsurface water environment management system, 5) sewer and waterworks management system. The core methodology of the engine is the generalized multidimensional finite element matrices to depict the terms in the analysis of various partial differential equations. Spatial information management system (ArcView) and Visual Basic were extensively employed to construct GUI oriented web based engine. The developed systems were composed of very intense computer codes due to the necessity of combinatory management of environmental problems. The web based engine could be served as a decision tool for the integrated management of environmental projects in Korea.

Past and Future Epidemiological Perspectives and Integrated Management of Rice Bakanae in Korea

  • In the past, rice bakanae was considered an endemic disease that did not cause significant losses in Korea; however, the disease has recently become a serious threat due to climate change, changes in farming practices, and the emergence of fungicide-resistant strains. Since the bakanae outbreak in 2006, its incidence has gradually decreased due to the application of effective control measures such as hot water immersion methods and seed disinfectants. However, in 2013, a marked increase in bakanae incidence was observed, causing problems for rice farmers. Therefore, in this review, we present the potential risks from climate change based on an epidemiological understanding of the pathogen, host plant, and environment, which are the key elements influencing the incidence of bakanae. In addition, disease management options to reduce the disease pressure of bakanae below the economic threshold level are investigated, with a specific focus on resistant varieties, as well as chemical, biological, cultural, and physical control methods. Lastly, as more effective countermeasures to bakanae, we propose an integrated disease management option that combines different control methods, including advanced imaging technologies such as remote sensing. In this review, we revisit and examine bakanae, a traditional seed-borne fungal disease that has not gained considerable attention in the agricultural history of Korea. Based on the understanding of the present significance and anticipated risks of the disease, the findings of this study are expected to provide useful information for the establishment of an effective response strategy to bakanae in the era of climate change.
