Control Effects of Some Insecticides on Different Stages of the Stone Leek Leafminer, Liriomyza chinensis Kato (Diptera: Agromyzidae) (파굴파리의 충태별 약제방제 효과)

  • Control effects of some insecticides were evaluated against the stone leek leafminer, Liriomyra chinensis Kato (Diptera: Agromyzidae) with the some different treatment methods. Insecticidal activities effects were estimated on the different development stages of the insects on welsh onion. The insecticides that controlled L. chinensis eggs with over 83% efficacy were spinosad, dimethoate, emamectin, and cartap. The insecticides that showed over 87% of larvicidal activity were dimethoate and cartap. Dimethoate showed 93.3% insecticide residual activity for 3 days a(ter treatment as a foliar spray. For control of pupae, the insecticides that showed over 88% of contact insecticidal activity were terbufos GR and cartap GR. Both dimethoate and cartap had high adulticidal activity with over 95% control efficacy.

국내 식물시료에서 분리한 Bacillus thuringiensis 균주의 다양성

  • We collected 3,237 plant samples, mainly leaves of various trees, from many provinces in Korea and a total of 1,925 Bacillus thuringiensis isolates were obtained and characterized. The isolates were characterized in terms of crystal morphology, PAGE pattern of the toxin proteins, plasmids pattern, biochemical characteristics, and bioassay. The microscopic observation showed that 49.1% of the isolates have bipyramidal shape crystals, 7.1% of spherical shape crystals, 1.4% of rhomboidal shape crystals, and others have small or amorphous inclusions. The insecticidal activities of the spore-crystal mixtures of isolates were tested against Plutella xylostella, Bombyx mori, Culex pipiens, and Agelastica coerulea. Bioassay showed that 51.3% of the isolates were shown to be active; lepidopteran-specific (44.8%), dipteran-specific(4.9%) and coleopteran-specific (1.6%). The remainder(48.8%) did not show any activity against the insects we tested. Interestingly though, some of these non-active isolates were shown to have bipyramidal crystals. By serotyping 22 isolates of our collection, we found that there are various kind of subspecies such as aizawai, amagiens, canadensis, darmstadiensis, galleriae, finitimus, kurstaki, morrisoni and neoleonensis, and three isolates have been classified into a new serotype, H49, and one of them, the type strain, named subsp. muju. From this study it was found that phylloplane is a good source for the isolation of Bacillius thuringiensis, and Bacillus thuringiensis is distributed widely in Korea.

Physiological and Molecular Characterization of a Newly Identified Entomopathogenic Bacteria, Photorhabdus temperata M1021

  • The present study concerned the identification and characterization of a novel bacterial strain isolated from entomopathogenic nematodes collected from different regions in Korea. The bacterial isolate M1021 was Gramnegative, bioluminescent, and produced red colonies on MacConkey agar medium. A rod-shaped structure was confirmed by the electron micrograph. Fatty acid composition was analyzed by using the Sherlock MIDI system. The identification was further supported by 16S rDNA sequence analysis, which revealed 96-99% sequence homology with strains of Photorhabdus temperata. The location of the isolated strain of P. temperata in the phylogenetic tree was confirmed and it was named P. temperata M1021. P. temperata M1021 exhibited catalase, protease, and lipase activities when grown on appropriate media supplemented with respective substrates. The culture of P. temperata M1021 exhibited insecticidal activity against the larvae of Galleria mellonella and the activity was the highest after 3-4 days of cultivation with agitating at $28^{\circ}C$ under 220 rpm. Antibacterial activity was also observed against Salmonella Typhimurium KCTC 1926 and Micrococcus luteus KACC 10488.

Preliminary studies on Insecticidal activities of 5-substituted pyrazole oxime ether derivatives (새로운 pyrazole oxime ether 유도체의 살충활성 연구)

  • The pyrazole family was discovered as effective insecticides/acaricides, and fenpyroximate and tebufenpyrad were introduced in the market. In this study, new series of pyrazole oxime ethers were designed and synthesized. The pyrazolone 1 was prepared by the condensation of ethyl acetoacetate with methylhydrazine, and then subsequently subjected to the Vilsmeier-Haack chloroformylation yielding 4-formyl-5-chloro-pyrazole 2. The nucleophilic aromatic substitution of 2 generally allowed the introduction of a wide range of heterocycles into the pyrazole ring. The resulting pyrazole aldehydes 3a-i were readily converted to the corresponding pyrazole oxime derivatives 4a-i, and subsequently to 5-substituted pyrazole oxime ether derivatives 6a-i. The screening assay results clearly show that the activities of 6d were comparable to those of fenpyroximate (6a) against BPH, DBM, and TSSM. It indicates that 6d has a potential to be developed as an insecticidal agent.

  • This experiment was conducted to select prominent microorganisms with a good insecticidal activity among the ten species, which isolated from soil at the near of Chung-buk, Chung-nam, and Gang-won provinces and made protein crystal endotoxin. As a result, GB-413 strain was finally selected, which showed the high insecticidal activity against susceptible diamondback moth (Plutella xylostella), beet army worm (Spodoptera exigua) and tobacco cutworm (Spodoptera litura) as well as resistant diamondback moth strains. By modifying the cultivation process f.g. lowing the glucose concentration at early cultivation stage and adding the carbon after inducing the spores, the percentage of making spore as well as the number of active spore were increased and the time for cultivation and spore forming was reduced without a reduction of insecticidal activity. These results were not only applied successfully for the optimized cultivation process for a fermentation tank containing five tons capacity, but also improved the possibility of mass cultivation for commercial production.

Activity and control effects of insecticides to American serpentine leafminer, Liriomyza trifolii(Diptera: Agromyzidae) (아메리카잎굴파리 (Liriomyza trifolii)에 대한 살충활성과 방제효과)

  • These studies were carried out to investigate the toxicities of 33 registered insecticide to the American serpentine leafminer, Liriomyza trifolii. Insecticidal activities were evaluated by testing systemic action and residual effect in the laboratory, and control efficacy and phytotoxicity in the greenhouse. All insecticides used in this study did not effect on the egg of L. trifolii, although spinosad showed 70% of egg-hatch suppression. For L. trifolii larvae ($2{\sim}3$ instar), the insecticides with over 95% of insecticidal activity were abamectin, cartap, cyromazine, emamectin benzoate, diflubenzuron + chlorpyrifos. The Insecticide what showed over 90% of insecticidal activity or neonate larvae were abamectin, cartap, emamectin benzoate, diflubenzuron + chlorpyrifos and milbemectin. Only cartap + buprofezin showed over 95% insecticidal activity against L. trifolii pupae. Almost insecticides used in this study little or not effected on the adult of L. trifolii. Emamectin benzoate and milbemectin showed moderate foliar systemic effects on eggs of L. trifolii (53.3, 47.9%, respectively). However, other insecticides showed little systemic effect. For larvae and adults, all insecticides showed low systemic effects. Insecticides with over 90% residual effect for 10 days were abamectin, emamectin benzoate and milbemectin (91.4, 90.4, 91.9%, respectively). In the control efficacy test on L. trifolii 90% of control values were obtained at 14th day after treatment of the insecticides including abamectin, cyromazine, emamectin benzoate and milbemectin. Cartap and cartap + buprofezin showed slight phytotoxicity on kidney bean leaf, however, other insecticides showed no phytotoxic effects. These results indicate that abamectin, emamectin benzoate and milbemectin can be used for tile control of L. trifolii in field.

Toxicological and Electrophysiological Activities of Pyrethroids between Larvae of Diamondback Moth, plutella xylostella and Beet Armyworm, Spodoptera exigua (배추좀나방과 파밤나방의 pyrethroids약제에 대한 감수성 및 전기적 신경 반응 비교)

  • Based on the insecticidal efficacy by insecticide treatment methods, neurophysiological responses were compared with the larvae of diamondback moth (DBM) and beet armyworm (BAW) using pyrethroids working on nervous system. By body spray method, all pyrethroids were showed lower insecticidal activity below 50% on the larvae of DBM and BAW. By leaf dipping method, DBM larvae were showed the insecticidal activity as 100% at 50 ppm deltamethrin, 80.0% at fenvalervate and 63.3% at permethrin. However, BAW larvae were showed lower insecticidal activity as similar as control in all concentration. In order to examine electrophysiological response on nervous system on DBM and BAW larvae when treated three insecticides, we investigated the voltage and reaction degree. The voltage of DBM were responded as high as $10^{-7}M$ in deltamethrin, $10^{-5}M$ in fenvalerate and $10^{-3}M$ in permethrin. However, those of BAW were showed dull responses with small variation of voltage to all three insecticides.

Effects of Flower Thinning Formulation on Activities of Digestive Enzymes and Acetylcholine Esterase in Honey bee Apis mellifera

  • The effects of a newly developed flower thinning formulation (FTF) on the vitality of the honey bee Apis mellifera were examined by measuring the activities of various digestive enzymes in adult worker bees. First, direct spraying of the FTF solution did not cause any behavioral changes or lethal effects for the honey bees based on 24 h observation. Second, oral ingestion of a sugar solution containing the FTF did not produce any significant change in the activities of amylase, proteinases, lipase, or acetylcholine esterase (AChE) in the worker bees 6 h or 24 h after treatment. Meanwhile, a commercial formulation containing sulfur compounds showed slightly reduced activities for several digestive enzymes and AChE, although no behavioral disturbance. Thus, the results of the present study suggest that the FTF is not toxic for honey bees, in terms of contact and ingestion. Therefore, this newly developed FTF can be used for flower thinning without any detrimental effects on pollinating insects.

The Influence of the Substituents for the Insecticidal Activity of N' -phenyl-N-methylformamidine Analogues against Two Spotted Spider Mite (Tetranychus urticae) (두 점박이 응애(Tetranychus urticae) 에 대한 N'-phenyl-N-methylformamidine 유도체의 살충활성에 미치는 치환기들의 영향)

  • To understand the influences of the substituents ($R_1{\sim}R_4$) on insecticidal activity of N'-phenyl-N-methylformamidine analogues (1~22) against two spotted spider mite (Tetranychus urticae), comparative molecular field analysis (CoMFA) model and comparative molecular similarity indices analysis (CoMSIA) model as three dimensional quantitative structure-activity relationships (3D-QSARs) model were derived and discussed quantitatively. From the results, the correlativity and predictability ($r^2{_{cv.}}=0.575$ and $r^2{_{ncv.}}=0.945$) of the CoMFA 1 model were higher than those of the rest models. The the CoMFA 1 and CoMSIA 1 model with the sensitivity of the perturbation and the prediction produced ($d_q{^{2'}}/dr^2{_{yy}}=1.071{\sim}1.146$ & $q^2=0.545{\sim}0.626$) by a progressive scrambling analysis were not dependent on chance correlation. The insecticidal activities from the optimized CoMFA 1 model were depend upon the steric field (62.5%), electrostatic field (28.9%), and hydrophobic field (8.6%) of N'-phenyl-N-methylformamidine analogues. Therefore, the inhibitory activities with optimized CoMFA 1 model were dependent upon steric factor. From the contour maps of the optimized models, it is predicted that the structural distinctions that contribute to the insecticidal activity will be able to applied new potent insecticides design.
