Plant Inositol Signaling - Biochemical Study of Phospholipase C and D-myo-inositol -1,4,5-trisphosphate receptor

  • It is now generally accepted that a phosphoinositide cycle is involved in the transduction of a variety of signals in plant cells. In animal cells, the hydrolysis of phosphatidyl-4,5-bisphosphate catalysed by phosphatidylinositol - specific phospholipase C yields to D-myo-inositol - 1,4,5-trisphosphate and diacylglycerol, which are well known second messengers. The binding of InsP$_3$to a receptor located on the endoplasmic reticulum triggers a calcium release from the endoplasmic reticulum. We have detected and partially characterised key components of phosphoinositide signaling. First, tobacco microsomal fraction and plasma membrane PI-PLC. Consecutively, using a radioligand binding assay we have identified a $Ca^{2+}$ -dependent high affinity InsP$_3$binding site in microsomal membrane fraction vesicle preparation and then we have measured inositol-1,4,5-trisphosphate induced calcium release from tobacco microsomal fraction. These findings suggest that phosphoinositide signaling system is present and operates in the tobacco suspension culture.e.

  • Nea-inositol is triclinic P???, with a =4.799(1), b=6.520(1), c=6.505(1) Å, α=70.61(1), β=69.41(1), γ=73.66(1)°, Z=1, molecular symmetry ???. Scyllo-inositol, from A, is monoclinic, P2₂/c, with a=5.089(1), c=11.948(2)Å, β=116.98(2)°, Z=2, molecular symmetry ???. Form B is triclinic, P???, with a=6.725(1), b=6.797(1), c=8.635(2)° Å, α=95.45(2), β=99.49(2), γ=99.19(2)°, Z=2, molecular symmetry ???. This crystal structure is pseudo-monoclinic, having two centrosymmetrical molecules with the almost identical conformation and orientation in the crystal lattice. The molecules have the expected chair conformations with puckering parameters of Q=0.609(2)Å for n대, 0.581(2)Å for Scyllo-A, and 0.566(2) Å for Scyllo-B. The bond lengths and angles are normal, C-C, 1.505 to 1.531 8A, C-O, 1.415 to 1.440 Å, C-C-C, 108.2 to 112.9°. The molecules are linked by systems of finite and infinite chains of hydrogen bonds.

The Effects of Inositol Extracted from Rice on the Skin

  • Inositol, is a water-soluble crystalline compound. It helps with people’s metabolism and decreases cholesterol levels. It is also known to have anti-cancer results. In order to find out the affects of Inositol on the skin, Inositol skin lotion was produced with each amount of Inisitol: 0, 0.5, 1.0, 2.0, 3.0 wt% and tested on the faces and the arm areas of women in all ages for 7 weeks. The moisture, sebum, change in elasticity, and improvement of wrinkles were measured. Corneometer, Sebumeter, Cutometer, and an image analyzer were used as measuring equipments. There are subtle differences in the subjects when 1-2% of Inositol is used the moisture of the skin improved 19%, elasticity by 17%, and the amount of sebum for dry and oily skin types adjusted to the amount of sebum of the neutral skin types. This influenced the length, width, the number of peak, and the height of the wrinkles to improve 12.4%. It is thought that Inositol would be an effective new raw material in cosmetics.

흰다리새우(Litopenaeus vannamei) 사료 내 Myo-inositol 요구량 (Dietary Myo-inositol Requirements of Pacific White Shrimp Litopenaeus vannamei)

  • We aimed to determine the dietary myo-inositol (MI) requirements of Pacific white shrimp Litopenaeus vannamei. A basal diet was formulated without myo-inositol (M0) and a negative control diet (M0-) was prepared by adding tetracycline hydrochloride to the basal diet to prevent intestinal inositol synthesis. Five MI diets were prepared by adding MI at 300, 600, 900, 1,200 and 1,500 mg/kg to the basal diet (designated as, M300, M600, M900, M1200 and M1500, respectively). Triplicate groups of shrimp (initial body weight, 0.55±0.01 g) were fed one of the experimental diets for 42 days. The growth performance of shrimp in M0- group was significantly lower when compared to that of shrimp in M0, M1200 and M1500 groups. Feed efficiency was significantly improved in M1200 and M1500 groups when compared to the M0 and M0- groups. GPx activity was significantly higher in M1200 and M1500 groups compared to that in M0 and M0- groups. Therefore, a practical diet (over 240 mg/kg) meets the minimum MI requirements of Pacific white shrimp. However, the optimum dietary MI level would be potentially above 1,200 mg/kg for better feed utilization efficiency and antioxidant capacity of Pacific white shrimp.

Apoptotic effect of $IP_6$ was not enhanced by co-treatment with myo-inositol in prostate carcinoma PC3 cells

  • Inositol hexaphosphate ($IP_6$) is a major constituent of most cereals, legumes, nuts, oil seeds and soybean. Previous studies reported the anticancer effect of $IP_6$ and suggested that co-treatment of $IP_6$ with inositol may enhance anticancer effect of $IP_6$. Although the anticancer effect of $IP_6$ has been intensively studied, the combinational effect of $IP_6$ and inositol and involved mechanisms are not well understood so far. In the present study, we investigated the effect of $IP_6$ and myo-inositol (MI) on cell cycle regulation and apoptosis using PC3 prostate cancer cell lines. When cell, were co-treated with $IP_6$ and MI, the extent of cell growth inhibition was significantly increased than that by $IP_6$ alone. To identify the effect of $IP_6$ and MI on apoptosis, the activity of caspase-3 was measured. The caspase-3 activity was significantly increased when cells were treated with either $IP_6$ alone or both $IP_6$ and MI, with no significant enhancement by co-treatment. To investigate the effect of $IP_6$ and MI of cell cycle arrest, we measured p21 mRNA expression in PC3 cells and observed significant increase in p21 mRNA by $IP_6$. But synergistic regulation by co-treatment with $IP_6$ and MI was not observed. In addition, there was no significant effect by co-treatment compared to $IP_6$ treatment on the regulation of cell cycle progression although $IP_6$ significantly changed cell cycle distribution in the presence of MI or not. Therefore, these findings support that $IP_6$ has anticancer function by induction of apoptosis and regulation of cell cycle. However, synergistic effect by MI on cell cycle regulation and apoptosis was not observed in PC3 prostate cancer cells.

참깨 myo-inositol 1-phosphate synthase 유전자의 특성과 기능분석에 관한 연구 (Characterization and functional analysis of a myo-inositol 1-phosphate synthase cDNA in sesame (Sesamum indicum L.))

  • 1845 bp의 SeMIPS cDNA를 발육종자에서 분리하고 이 cDNA의 구조와 특성을 분석하였다. 이 cDNA는 엽록체로 향하는 신호펩타이드의 아미노산 서열이 존재하지 않아서 세포질형 MIPS로 예상되었다. 또한 이 cDNA의 아미노산 서열의 유사성은 다른 MIPS와 비교한 결과 60-94%의 높은 아미노산 서열 유사성을 보여주었으며, 특히 식물끼리의 유사성이 훨씬 높았다. Northern blot분석에서 볼 때 참깨의 조직별 SeMIPS mRNA는 완숙종자, 줄기, 뿌리에서는 약하게 발현되었고, 잎에서는 비교적 강하게 발현되는 현상을 보여주었다. Yeast 돌연변이체를 통한 활성 시험에서는 SeMIPS가 myo-inositol 1-phosphate synthase의 효소활성을 가지고 있다는 실험적 증거를 얻었으며, C-말단 아미노산 20개가 효소활성에 필수적이라는 사실이 본 실험에서 검증되었다.

흰쥐 소장 점막 phytase의 특성 및 활성에 미치는 금속 이온의 영향 (Characterization and Effect of Metal Ion on Activity of Phytase from Rat intestinal Mucosa)

  • Phytase(myo-inositol hexakisphosphate phosphohydro-lase;EC 3.1.3.8)는 흰주 소장 점막으로 부터 분리${\cdot}$정제 하였다. 이 정제된 효소를 Sephacryl S-200 gel filtration방법으로 측정한 분자량으니 160kDa이고, 순도 및 이 효소의 서브유니트를 SDS-polycrylamide gel전기영동법(SDS-PAGE)으로 조사한 결과 서브유니트 구조는 분자량이 10kDa와 90kDa으로 구성된 hetrodimer(이종이량체)임을 알 수 있었다. 그리고 $ MgCl_{2}$ 존재 하에서는 효소 활성이 증가하나 $ZnCl_{2}$, $MnCl_{2}$, 및 EDTA존재 하에서는 효소 활성이 저해되었다. 기질 트이성과 pH범위에서 기질인 phytic acid(inositol-hexakispho-sphate)에 대해 높은 친화력을 보였다. Phytic acid에 대한 Km값은 pH 7.4에서 0.31mM이다. 따라서 흰쥐의 소장 점막 phytase는 주로 inositol의 대사계에서 중요한 역할을 하는 것으로 생각된다.

Enterobacter sp. YB-46의 myo-Inositol dehydrogenase 유전자 클로닝과 특성분석 (Molecular Cloning and Characterization of myo-Inositol Dehydrogenase from Enterobacter sp. YB-46)

  • myo-Inositol (MI)을 대사하여 다른 물질로 전환하는 미생물을 과수원 토양으로부터 분리하였다. 분리균 YB-46은 유일한 탄소원으로 MI이 첨가된 배지에서 성장하였고 16S rDNA 염기서열에 따라 Enterobacter 속의 균주로 추정되었다. Fosmid pCC1FOS 벡터를 사용하여 제조된 거대 유전체 은행으로부터 MI을 미지의 대사 물질로 전환하는 Escherichia coli 형질전환주를 선발하였다. 이로부터 플라스미드를 분리하고 삽입된 유전자의 일부 염기서열을 결정한 결과 336 아미노 잔기로 구성된 myo-inositol dehytrogenase (IolG)를 암호화하는 iolG 유전자가 발견되었다. 분리균 YB-46의 IolG는 E. aerogenes와 Bacillus subtilis의 IolG와 약 50% 수준의 상동성을 보였다. 카르복실 말단에 hexahistidine이 연결되도록 제조한 His-tagged IoG (HtIolG)의 유전자를 재조합 대장균에서 발현하여 균체 파쇄액으로부터 HtIolG를 정제하였다. 정제된 HtIolG는 $45^{\circ}C$와 pH 10.5에서 최대 활성을 보였고 MI과 D-glucose에 대한 활성이 가장 높았으며 D-chiro-inositol, D-mannitol 및 D-xylose에도 90% 이상의 활성을 보였다. 최적 반응조건에서 MI을 기질로 하여 반응 동력학적 계수를 측정한 결과 $K_m$$V_{max}$가 1.83 mM과 $0.724{\mu}mol/min/mg$로 확인되었다. HtIolG의 활성은 $Zn^{2+}$에 의해 1.7배 증가하였으며, $Co^{2+}$와 SDS에 의해서는 크게 감소하였다.

Sodium molybdate가 납중독 랫드의 말초신경내 myo-inositol 인지질 대사 관련 효소에 미치는 영향 (Effects of sodium molybdate on myo-inositol phospholipid metabolism-related enzymes in peripheral nerves of lead-intoxicated rats.)

  • We have previously demonstrated that sodium molybdate(Mo) improved lead-intoxicated status by enhancing the metabolism of mao-inositol-related phospholipids in sciatic nerves isolated from rats. In this study, in order to address the reduction mechanism of Mo for lead toxicity, effects of Mo on cystidine-diglyceride transferase, phosphatidylinositol kinase, and phosphatidyl inositol-4-phosphate kinase, involved in mao-inositol metabolism of nerve, were investigated in vivo and in vitro. Mo significantly increased the activities of cystidine- diglyceride transferase and phosphatidylinositol kinase in lead-intoxicated rat, and the pattern of increase was dose-dependent manner. However, Mo did not affect the activity of phosp- hatidylinositiol-4-phosphate kinase in normal and lead-intoxicated rats. We also found that Mo affected the activities of phopholipid metabolism-related enzymes not by the indirect manner such as activation of another metabolic pathway but by the direct manner. These results suggest that the improvement mechanism of Mo for lead-intoxicated status might be a normalization of the activities of phospholipid metabolism-related enzymes in sciatic nerve.
