The effects of heavy metals on the growth rate and phosphate metabolism of Chlorella elliposidea cells were investigated. Chlorella cells were cultured in the media treated with Hg(0.3, 0.7, 0.9 ppm), Cd(1, 5, 15ppm), and Zn(1, 5, 50ppm) for 6days. Aliquots cells were taken out at the inoculation and at intervals during the culture, and measured packed cell vlolume and optical density. The inhibitions of heavy metals on the growth rate and chlorophyll contents were traced. Also after 6 days culture, the amounts of inorganic phosphate and organic compounds of various fractions in Chlorella cells were observed. The turbid effects of heavy metals on the growth rate and chlorphyll contents of Chlorella cells were in order of Hg>Cd>Zn. Because heavy metals depressed the biosynthesis of inorganic polyphosphates and nucleic acids and turn over of inorganic phosphates, the amounts of various phosphate compounds were decreased. The inhibitory effect of photosynthesis by heavy metals resulted in lower contents of carbohydrate. Due to the turbidity of biosynthesis of amino acids by heavy metals, contents of protein were reduced in comparison with those of control. It is suggested conciusively that the minimum concentrations affected by heavy metals on the growth rate and phosphate metabolism of Chlorella cells were 0.7 ppm Hg, 15ppm Cd, 50ppm Zn.
Arsenic (As) usually is bound to amorphous iron oxides in the soils, and it can be removed via dissolution of iron oxides. Inorganic acid and chelating agent are widely used to extract As in the soil washing. However, the overall performance is highly dependent on the state of As fractionation. In this study, oxalic acid and inorganic acids (HCl, $H_2SO_4$, and $H_3PO_4$) were applied to enhance the dissolution of iron oxides for remediation of As-contaminated soils. Oxalic acid was most effective to extract As from soils and removal of As was two times greater than other inorganic acids. Additionally, 75% of As bound to amorphous iron oxides was removed by 0.2 M oxalic acid. Arsenic removal by oxalic acid was directly proportional to the sum of labile fractions of As instead of the total concentration of As. Therefore, the oxalic acid could extract most As bound to amorphous iron oxides.
In current animal production in Japan, a large surplus of nitrogen and phosphorus is given to animals as their feed which are mostly imported from outside of our own country. Today, an excess of nitrogen and phosphorus from animal manure has been spread out of the area of animal production and the surroundings. These components have become the major reason for eutrophication of ground, surface and inland water. Nutritional studies for the reduction of nitrogen and phosphorus from animal waste has been done by many researchers. The reduction of excess protein in animal feed and the supplementation of deficient essential amino acids to feed have a possibility to increase the biological value of feed and to reduce nitrogen excretion, especially, via urine. The use of phytase activity to degrade phytate and to release utilizable inorganic phosphorus make it possible to cut an excess supply of feed additive inorganic phosphorus and to reduce phosphorus excretion from animal waste.
This study was carried out to investigate the compositions of fatty acids, inorganic components and volatile organic acids from Korean valerian roots, Valeriana fauriei var. dasycarpa Hara and Valeriana officinalis var. latifolia Miq. The contents of total lipids ranged from 3.7 % to 4.5% and the major fatty acids were linoleic, linolenic and palmitic acid. Ash contents ranged from 4.3% to 6.3%. and the contents of Na, Fe, Zn and Cu showed some quantitative differences depending upon grown region or species. Fourty components were identified from acidic fraction of dichloromethane extract of V. fauriei var. dasycarpa Hara, of which the major components were 3-methyl butanoic, dimethoxy-2-propenoic. 3, 4-dimethoxy benzoic and 3-methyl pentanoic acid.

Alkaline phosphatase (APase) was found to be inducible in Anabaena sp. strain CA Growth was less than control in presence of most amino acids except glycine and serine, but most amino acids enhanced APase activity. Highest APase activity was recorded in tyrosine supplemented culture followed by hydroxyproline, cystein, valine and glutamic acid. Threonine supplemented material showed lowest APase level (1.8 nmol/mg protein/min). Lactose, glucose, sodium pyruvate and succinate stimulated growth but not APase activity. APase activity was high in the presence of sucrose, mellibiose, mannitol, arabinose, maltose and sorbose, even though the growth in these supplements was less than in control. Organic phosphate sources supported good growth of the organism. Best growth occurred in presence of inorganic phosphate, adenosine diphosphate, fructose 1,6-diphosphate or ribulose 1,5-diphosphate, followed by other phosphorus sources tested. APase activity in presence of any of the organic phosphate sources was 3 to 5 fold low as compared to phosphate limited culture. Also, there was no APase activity in cultures grown on inorganic phosphate. These data indicate that most amino acids and a few carbohydrates (sucrose, mellibiose, arabinose and sorbose) are suitable for APase production. Lactose, glucose, pyruvate or succinate may be used as a carbon source during photoheterotrophic growth of the cyanobacterium. Glycine and serine are preferred nitrogen sources for its growth. Phosphate repressible APase activity has been found in Anabaena sp. strain CA.
Calocybe indica is an edible mushroom commercially cultivated in India and other tropical countries. In this study, the culture characteristics and optimal conditions of milky mushroom strains were determined. The growth temperature and pH range of milky mushrooms was extensively investigated between 15-35 ℃ and pH 3-11. For efficient cultivation, 20 types of nutrient sources were selected that consisted of one of 21 types of carbon sources, 6 organic nitrogen sources, 6 inorganic nitrogen sources, 13 amino acids, 6 organic acids and 12 inorganic salts. The impact of each of the selected nutrition sources and their concentration on growth was investigated. The optimum pH and temperature were determined to be pH 6.0 and 15 ℃, respectively. The optimum concentration of medium elements for the mycelial growth of C. indica was determined to be as follows: carbon source, 2% maltose; organic nitrogen source, 1% yeast extract; inorganic nitrogen source, 0.1% NaNO3; amino acid, 0.7% asparagine; organic acid, 0.07% acetic acid; inorganic salt, 0.07 mM MnSO4.
Three humic acids, two from uranium bearing coal shale and one from the neighbouring limey shale region, are extracted from soils by dissolution in 0.1 M NaOH followed by acid precipitation. After purification cycles, they are characterized for their elemental composition, contents of inorganic impurities, molecular size distribution and proton exchange capacities. The results are compared with the data of reference and aquatic humic acids characterized under the project MIRAGE Ⅱ at TUM and also with other literature data. The proton exchange capacity determined by direct titration, is found to be 3.60 and 2.01 meq/g for coal shale and limey shale humic acids, respectively.
In order to study the influence of nitrate reduction to ionic balance in tissue of tobacco plant, differneces in amounts of those cations and anions were determined and these balances were compared with contents of organic acids and activities of nitrate reductase, while they were fertilized with different nitrogen sources ($NO_3-N$, $NH_4-N$, $NO_3+NH_4-N$) in water culture. The results of studies are summerized as follows; 1. Total uptake of inorganic cations was the highest in nitrate-fed plants, whereas that of inorganic anions showed the highest level in the plants grown with the mixture ($NO_3+NH_4$). The amounts of inorganic cations and anions were comparable in two treatments containing $NH_4-N$, but in plants treated with nitrate only had much higher level of inorganic cations than others. 2. Deficiency in the amount of inorganic anions in nitrate-fed plants was balanced with organic acids, dominantly with malic acid among them. But another two $NH_4-N$ fed plant sustained equilibrium between inorganic cations and anions. 3. Reduction of nitrate was raised in tissues of nitrate-fed plants. By the results of nitrate reduction, cations maintained equilibrium with nitrate ion were let loose. The replacement of inorganic anions with organic anions could be a compensation process for the loss of uptaken nitrate ions which must be reduced to be incorporated into organic N compounds.
The hydroxide precipitation method is appropriate to distinguish free metal ions with complexed metal ions with amino acids. Optimum pH conditions of hydroxide precipitation were investigated using mixed amino acids which have similar composition ratio with hydrolyzed amino acids. When applied to soil samples immobilities of Hg, Cr, and Cu ion with mixed and hydrolyzed amino acids were reasonable. But those of Cd and Zn were not sufficient.
