Metallo-collagenase production by Arthrobacter creatinolyticus KP015744

  • Amongst 27 isolates from deteriorated leather samples, Arthrobacter creatinolyticus KP015744 zzx28 was found to be an efficient collagenase producer. Collagenase production of 13.33 µmoles/min was shown at an optimum temperature at 37℃ after 72h and at pH 7.5 by using 2 ml/dL inoculum in 10 mg/ml collagen peptide type I as a substrate. In presence of Hg2+, EDTA and 𝛽-mercaptoethanol the collagenase production by the isolate was strongly inhibited however Fe2+, Ca2+and DMSO enhanced production of the enzyme. Specific activity was found to be 19.46×103 U/mg and molecular weight 66 kD by SDS PAGE. Isolate also has potential to hydrolyze keratin which is another important protein found in leather. Experimental results propose that collagenase can be effectively used as a tool for collagen and keratin rich solid waste treatment.

고추 탄저병균(Colletotrichum acutatum)의 분생포자 비산과 과실병반에 형성된 전염원 밀도 (Conidial Disperse of the Pepper Anthracnose Fungus Colletotrichum acutatum and Its Density on Infected Fruits)

  • 본 연구는 고추 탄저병균인 Collerorichum acutaum의 강우에 따른 비산량과 비산거리 및 포장에서 감염 후 병증상의 발현시기 그리고 병반에 형성된 전염원의 밀도 등을 알아보기 위해 수행하였다. 고추탄저병균의 분생포자는 비가 오는 날에 99.2%가 비산되었고 맑은 날에는 단지 0.8%의 포자만이 비산하였다. 포자의 비산량은 전염원과의 거리와 정의 상관이 있었다. 고추 탄저병균 포자의 93.3%는 점염원과 30 cm 수평거리에서 60 cm 높이 이하로 전파되었으며 120 cm 높이까지 비산하는 것으로 조사되었다. 노지에서 상대적으로 감수성이 높은 품종의 고추 과실과 낮은 품종의 과실은 각각 4일과 6일 후에 최초로 외부 병 증상을 나타내었다. 하지만 10일 후에 병 증상을 나타내기도 하여 기상환경에 따라 병원균의 잠복기간이 10일 이상일 가능성도 있을 것으로 판단되었다. 고추 병반에 형성된 전염원인 분생포자의 수는 병방의 크기가 클수록 많았는데 병반의 길이가 1.5 cm 이상일 경우에는 병반 당 천만 개 이상의 포자가 형성되었으며 여러 개의 병반 당 천만 개 이상의 포자가 형성되었으며 여러 개의 병반이 합쳐서 병반 길이가 4 cm가 넘을 경우 1억개가 넘는 포자가 형성되기도 하였다. 이상의 결과로 고추 탄저병균인 C. acutatum이 빗물에 튀어서 전파되고 비바람의 강도에 따라 상당히 먼 거리로 전파될 수 있으며 기상환경에 따라 감염 후 10일 이상의 잠복기를 가질 수 있으며 한 개의 큰 병반에는 수 천만개의 전염원인 형성되는 것을 확인되었다. 따라서 고추 탄저병의 가장 효과적인 방제수단은 빗물을 직접 맞지 않도록 하는 비가림 시설이며, 노지에서는 병든 과실을 조기에 제거하고 병 발생 초기에 적절한 약제를 살포하여 전염원의 밀도를 낮추는 것이 무엇보다 중요하다.

Isolation and characterization of a novel gossypol-degrading bacteria Bacillus subtilis strain Rumen Bacillus Subtilis

  • Objective: The aim of the study was to isolate gossypol-degrading bacteria and to assess its potential for gossypol degradation. Methods: Rumen liquid was collected from fistulated cows grazing the experimental pasture. Approximately 1 mL of the rumen liquid was spread onto basal medium plates containing 2 g/L gossypol as the only source of carbon and was then cultured at $39^{\circ}C$ to isolate gossypol-degrading bacteria. The isolated colonies were cultured for 6 h and then their size and shape observed by microscope and scanning electron microscope. The 16S rRNA gene of isolated colonies was sequenced and aligned using National Center for Biotechnology Information-Basic Local Alignment Search Tool. The various fermentation conditions, initial pH, incubation temperature, inoculum level and fermentationperiod were analyzed in cottonseed meal (CSM). The crude protein (CP), total gossypol (TG), and free gossypol (FG) were determined in CSM after fermentation with isolated strain at $39^{\circ}C$ for 72 h. Results: Screening results showed that a single bacterial isolate, named Rumen Bacillus Subtilis (RBS), could use gossypol as a carbon source. The bacterium was identified by 16S rDNA sequencing as being 98% homologous to the sequence of Bacillus subtilis strain GH38. The optimum fermentation conditions were found to be 72 h, $39^{\circ}C$, pH 6.5, moisture 50%, inoculum level $10^7cell/g$. In the optimum fermentation conditions, the FG and TG content in fermented CSM decreased 78.86% and 49% relative to the control. The content of CP and the essential amino acids of the fermented CSM increased respectively, compared with the control. Conclusion: The isolation of a gossypol-degrading bacterium from the cow rumen is of great importance for gossypol biodegradation and may be a valuable potential source for gossypol-degradation of CSM.

  • 미생물제재(mycoherbicide)라는 용어는 1970년대부터 사용되기 시작하였다. 하지만 미생물 제재에 대한 관심은 화학제재의 비용증가로 인해 최근 주목을 받게 되었다. 고전적인 생물학적방제재(Biological Control Agent, BCA)는 생태적으로 안전하다는 이유로 생태학적인 관점에서 관심을 끌고 있다. 반면, 현대적인 의미의 생물학적방제재는 인공적으로 배양이 가능하고, 또한 그 잡초방제 효과도 화학제초제와 같은 수준이 요구된다. 현재 미생물제재는 7개국에서 곰팡이로부터 만들어지는 26종이있다. 이들은 재배작물에는 안전한 기주특이성을 갖는다. 그러나 대부분의 약제전달체계는 살아있는 미생물을 활력을 유지한 상태로 장기간 보존할 수 있는 능력을 가지고 있어야 함에도 불구하고 대부분이 조건을 충족시키질 못하였다. 또한, 실험실 수준에서는 방제효과가 잘 나타나지만, 실무적으로 사용할 경우 사용년도 및 포장에 따라 방제효과의 발현성과 지속성이 다르게 나타났다. 이 밖에 미생물제재 사용시 습도, 이슬, 온도 및 이들 요인이 잡초방제에 미치는 효과에 대한 연구결과도 미비한 상태이다. 따라서 잡초방제효과가 뛰어난 미 생물제재를 선별해야한다. 선행된 연구에서 제안된 효과적인 미생물제재의 조건은 첫째, 인공 배양법으로 내구성이 있는 접종원(inoculum)의 대량 생산이 가능하고, 둘째, 유전적으로 안정하면서 기주 특이성이 있고, 셋째, 다양한 범위의 잡초방제효과 있어야 한다. 미생물제재의 생산과 보급은 잡초의 생물학적인 방제에 지대한 역할을 할 수 있다. 액상보다는 입상 형태의 미생물제재가 가벼워서 사용이 간편하며, 젤(gel) 타입의 미생물제재도 사용되는 것으로 알려져 있다.

양돈분뇨의 열전처리에서 기질과 접종액의 비율이 메탄생산 퍼텐셜에 미치는 영향 (Effect of Substrate to Inoculum Ratio on Biochemical Methane Potential in the Thermal Pretreatment of Piggery Sludge)

  • BMP 시험을 통한 최종메탄생산퍼텐셜 ($B_u$)의 측정은 바이오매스로부터 전환할 수 있는 바이오에너지 양을 추산하고 혐기소화조를 설계하는데 중요한 인자이다. 본 연구에서는 $B_u$의 측정에 있어 기질과 접종액의 비율 (S/I ratio)이 미치는 영향을 분석하기 위하여 기질의 유기물 특성이 다른 양돈 슬러리의 탈수여액 (LF), 양돈슬러지 탈수케이크의 열가수분해액 (TH), 탈수여액과 열가수분해액의 혼합액 (Mix)을 이용하여 BMP 시험을 실시하였으며, 각각의 시료의 이론적 메탄생산퍼텐셜 ($B_{th}$)과 혐기적 유기물 분해율을 구하여 비교하였다. TH 시료의 $B_u$는 S/I 비율 0.1, 0.3, 0.5에서 각각 0.27, 0.44, $0.46Nm^3\;Kg^{-1}-VS_{added}$로 나타났으며, 이론적메탄생산퍼텐셜 ($B_{th}$) 대비 최종메탄생산퍼텐셜 ($B_u$)의 비율 ($B_u/B_{th}$)로 나타낸 혐기적 유기물 분해율은 S/I 비율 0.1, 0.3, 0.5에서 각각 50.04, 82.46, 86.47%이었으며, LF 시료의 경우 S/I 비율 0.1, 0.3, 0.5에서 $B_u$은 각각 0.64, 0.53, $0.40Nm^3\;Kg^{-1}-VS_{added}$이었으며, 혐기적 유기물 분해율은 각각 152.07, 122.67, 95.71%로 나타났다. Mix 시료의 경우 최종 메탄생산 퍼텐셜과 혐기적 유기물 분해율에서 LF 시료와 유사한 경향을 보였다. 본 연구에서는 양돈슬러리의 BMP시험에서 S/I비율에 따라 상이한 최종 메탄생산 퍼텐셜이 나타나며, 낮은 S/I 비율에서 최종 메탄생산 퍼텐셜이 과대평가되었다.

Detection, isolation, and characterization of the cucumber mosaic virus in Pseudostellaria heterophylla from Korea

  • Weeds play an important role in the survival of viruses and are potential inoculum sources of viral diseases for crop plants. In this study, specimens of Pseudostellaria heterophylla exhibiting symptoms of the cucumber mosaic virus (CMV) were collected in Bonghwa, Korea. The characteristics of the disease were described and leaf RNA was extracted and sequenced to identify the virus. Three CMV contigs were obtained and PCR was performed using specific primer pairs. RNA from positive samples exhibiting CMV leaf symptoms was amplified to determine the coat protein. A sequence comparison of the coat protein gene from the CMV BH isolate shared the highest nucleotide identity (99.2%) with the CMV ZM isolate. Phylogenetic analysis showed that CMV-BH belonged to subgroup IA and that the most closely-related isolate was CMV-ZM. All test plants used for the biological assay were successfully infected with CMV and exhibited CMV disease symptoms such as blistering, mosaic, and vein yellowing. To our knowledge, this is the first report of CMV infection in P. heterophylla from Korea.

Relationship between the Population of Ralstonia solanacearum in Soil and the Incidence of Bacterial Wilt in the Naturally Infested Tobacco Fields

  • The population of Ralstonia solanacearum (Rs) in soil is very important as a primary inoculum source of bacterial wilt in tobacco fields. To investigate the population of Rs, physical properties and chemical components during the tobacco growing season, soil samples were taken from the fifteen fields which were located in the flue-cured tobacco growing area, Ansung, Kyunggi province and Wonju, Kangwon province. Two fields of the fifteen were bacterial wilt free. Six fields had less than 10% plants being diseased and seven over 10%. The Rs population level determined by using SMSA medium generally showed an up-and-down pattern being low in May, high in Jun and July and low in August. The soil population in May and June showed a positive correlation with the incidence of bacterial wilt (r=0.571$^*$, r=0.688$^{**}$), but P$_2O_5$, content of soil was negatively correlated with the disease incidence (r=-0.539$^*$). These results suggest that Rs population in soil examined in May or in June, and the P$_2O_5$ content in soil should be key factors to determine the bacterial wilt potential of tobacco fields.

Fusarium oxysporum Protects Douglas-fir (Pseudotsuga menziesii) Seedlings from Root Disease Caused by Fusarium commune

  • Fusarium root disease can be a serious problem in forest and conservation nurseries in the western United States. Fusarium inoculum is commonly found in most container and bareroot nurseries on healthy and diseased seedlings, in nursery soils, and on conifer seeds. Fusarium spp. within the F. oxysporum species complex have been recognized as pathogens for more than a century, but attempts to distinguish virulence by correlating morphological characteristics with results of pathogenicity tests were unsuccessful. Recent molecular characterization and pathogenicity tests, however, revealed that selected isolates of F. oxysporum are benign on Douglas-fir (Pseudotsuga menziesii) seedlings. Other morphologically indistinguishable isolates, which can be virulent, were identified as F. commune, a recently described species. In a replicated greenhouse study, inoculating Douglas-fir seedlings with one isolate of F. oxysporum prevented expression of disease caused by a virulent isolate of F. commune. Moreover, seedling survival and growth was unaffected by the presence of the F. oxysporum isolate, and this isolate yielded better biological control than a commercial formulation of Bacillus subtilis. These results demonstrate that an isolate of nonpathogenic F. oxysporum can effectively reduce Fusarium root disease of Douglas-fir caused by F. commune under nursery settings, and this biological control approach has potential for further development.

Production of Cellulase and Xylanase by Aspergillus niger KKS

  • A fungal strain capable of producing extracellular cellulase was isolated from farmland. It was identified as Aspergillus niger, and named Aspergillus niger KKS. Production of cellulase and xylanase by the A. niger KKS was studied through a shake-flask culture. The effects of culture conditions such as inoculum size, temperature, pH, and medium composition on the cellulase and xylanase production were examined. The optimum temperature and pH for the enzyme production were $30^{\circ}C$ and pH 7.0, respectively. The optimized medium was composed of 2.0% (w/v) rice straw, 0.5% (w/v) proteose peptone, 0.5% (w/v) $KH_2 PO_4$, 0.05% (w/v) yeast extract, 0.01% (w/v) $CoSO_4 \cdot 7H_2O$, and 0.05% (w/v) $CuSO$_4$\cdot 5H_2O$. When the strain was incubated with the optimized medium, it gave the activities of endoglucanase, $\beta$-glucosidase, $\beta$-xylosidase, xylanase were 3.80, 4.20, 4.00, 80.0 (IU/mL), respectively. Filter paper and cotton activities were 0.68 and 0.045 (IU/mL), respectively. The results of this study show that A. niger KKS is a potential organism with a wide spectrum of enzyme activities, such as those of $\beta$-glucosidase, $\beta$-xylosidase, and xylanase.

Estimation of In vitro Digestibility of Barley Straw by Using a Homogenized Rumen Fluid and Artificial Saliva Mixed with Nitrogen and Energy Sources

  • A $2{\times}2{\times}4$ factorial study was conducted to examine the possibility of improving estimates of in vitro digestibility, using untreated (UBS) and ammonia-treated (ABS) barley straw, through homogenization of rumen fluid (RF) and by additions of urea (U) and casein (C) as N sources and Xylose + Glucose (XG) as energy sources into artificial saliva. Digestibiltiy of ABS was significantly greater than that of UBS (p < 0.001). There was a significant decrease in digestibility when additions (U, UC, UCXG) were compared with the control (p < 0.001). A 2-way interaction between RF and straw type was significant (p < 0.05) for dry matter digestibility (DMD). Homogenization of RF increased DMD of ABS (p < 0.05) whereas it decreased DMD of UBS (p > 0.05). The study showed that addition of N alone or in combination with energy sources was not better than control, rather the reverse, digestion was inhibited by a combination of U and casein (UC). It was concluded that sufficient N and branched chain fatty acids were supplied in the inoculum from sheep fed high protein grass cubes to support the growth of cellulolytic microbes during in vitro incubation. Further studies are, however, required to explore the potential of homogenization in improving the in vitro method to estimate digestibility of cereal straws.
