  • The study has been carried out for the development of an inoculation method in screening resistant varieties and/or lines to bacterial leaf blight of rice with special consideration on plant ages and inoculum concentrations. A higher incidence of the disease was found on younger plants than on the older ones when the plants were inoculated with the same concentration of inoculum by clipping method under the same circumstances. Applications of extremly high concentration of inoculum resulted the death of younger seedlings from all varieties within short period after inoculation, and the inoculum that was lower than optimum concentration failed in distinguishing varietal characteristics in regard of resistance. The proper inoculum size for the screening of resistance among the varieties and/or lines depended on the age of plants. The optimum concentration of inoculum for the plant age of 14, 37, 48 and 58days was $10^6,\;10^7,\;10^7-10^{-9}\;and\; 10^9 cells/ml.,$ respectively.

  • The human granulocyte-macrophage colony-stimulating factor (hGM-CSF) was produced from cell suspension culture of transgenic tobacco which was transformed by using Agrobacterium harboring the hGM-CSF gene. To improve the production of hGM-CSF in batch culture system, the effects of initial sucrose concentration and inoculum size were investigated. The results show that the hGM-CSF production was not affected by small inoculum size in medium containing either low or high concentration of sucrose. However, the production of hGM-CSF was increased under increasing of the inoculum sizes and sucrose concentration. Under the combination of inoculum and sucrose concentration, the maximum hGM-CSF production of 720 $\mu$g/L was obtained at 90 g/L of initial sucrose concentration and 110 g/L of inoculum size.

  • Effect of inoculum concentration inoculation method and plant age on development of clubroot disease of Chinese cabbage seedling were examined in growth chambers. Root galls were developed at the concentration of 105 resting spore or above per ml of incoulum and as the inoculum concentration became higher rate of development of root galls was faster. In the plants with root gall development fresh weight of above ground parts was reduced to 30-44% of that of healthy plants but root weight increased by 4-10 times. Growth of diseased plants was greatly reduced as compared to healthy plants. Planting in the diseased soil as a inoculation method was most effective for disease development showing uniform infections but time of initial root gall development was delayed by root soaking inoculation. Some plants inoculated by soil drenching method did not develop root galls. However root gall enlargement after its initial formation did not differ greatly among inoculation methods. Nine-day-old seedlings showed poor development of root gall but 16-days-old seedlings was found to be most adequate for inoculation for gall development.

  • Optimum inoculum concentration for the production of taxol was determined in Taxus brevifolia and Taxus cuspidata cell suspension cultures. By fresh weight, 2.5, 5, 7.5, 10 g/flask of cells were inoculated and cell growth as well as taxol production were examined. In both Taxus cell cultures, the higher the inoculum concentration, the shorter the length of the lag period. The optimum inoculum concentration for taxol production was found to be 5 g/flask. To produce taxol in large quantity, utilization of proper medium was thought to be important. In case of using a production medium with 6% sucrose, taxol production was noticed. Its level reached the maximum at the 9th day of culture and decreased afterwards. However, taxol was not detected from cell cultures in growth medium.

  • This study was performed to investigate the inhibiting activity of etridiazole and thiophanate-mthyl on mycelial growth of Rhizoctonia solani and Pythium ultimum and the effect of inoculum density of each pathogen on the control efficacy of the mixture of etridiazole and thiophanate-methyl in a seedling assay test. In mycelial growth inhibition test, $EC_{50}$ values of etridiazole and thiophanate-methyl against R. solani were 15.87 and 9.34 ${\mu}g\;mL^{-1}$, while those were 0.2 and more than $500\;{\mu}g\;mL^{-1}$ against P. ultimum, respectively. Controlling activity of the mixture of etridiazole and thiophanate-methyl against damping-off of pepper and cucumber, caused by R. solani and P. ultimum was tested in a greenhouse. With the inoculation of R. solani, disease incidences of seedling in pepper were 73% and 95% at 0.5% and 1.0% of inoculum concentration, while in cucumber those were 55% and 62% at 1.0% and 2.0%. When P. ultimum was inoculated into soil by 2.0% of inoculum concentration, those in pepper and cucumber were 66.7% and 96.8%, respectively. The efficacy of the mixture was somehow affected by the concentration of R. solani. While each control value of the mixture was 94.4% and 90.7% in pepper and cucumber at low inoculum concentration (0.05%), the efficacy of the fungicide decreased in pepper and cucumber by 70.7% and 72.9% at high concentration of R. solani (0.1% in pepper and 0.5% in cucumber). However, the control value of the mixture was 100% in pepper and cucumber, irrespective of the inoculum concentration of P. ultimum, however, the increase of inoculum concentration in soil did not result in the decrease of the fungicide efficacy.

  • To understand effect of inoculum size, cell density, sucrose concentration and concentrations of MS basal on suspension culture and protoplast isolation of wild viola(Viola patrinii DC.) callus from petiole segments this experiment was conducted. In the lot of 30 mesh inoculum size, two observations were; One was that a considerable increase in the fresh and dry weight of callus was determined. Another was that the callus mass was relatively compact compared with others. A recommendable cell density was 0.4g for 20ml culture medium and the higher sucrose concentration, the higher fresh and dry weight were obtained. The dilution of MS basal salt was differently affected on fresh and dry weight; the highest fresh weight was found in 1x MS salt, while the higest dry weight was in 1/3x dilution. The addition of casein hydrolysate(3g/L) was more effective to increase of both fresh and dry weight. THe contents of protein was great in the inoculum lots with larger inoculum sizeand higher concentration of MS basal salts contenting 3g/L of casein hydrolysate and higher sucrose compared with others. The greatest protoplasts were isolated from the lot of 10 mesh size treated with 1%pectinase SE-150 and 2% cellulase YC. In general, for optimal protoplast isolation the followingconditions were recommended; 1) Use of smaller cell size cultured for 2-5 weeks, and 2) more than 5 hours incubation using the combined mixture of the enzymes with proper concentrations.

  • The effects of initial concentration and pulsed pressurization on the inactivation of Clostridium sporogenes spores suspended in deionized water were determined during thermal processing $(TP;\;105^{\circ}C,\;0.1MPa)$ and pressure-assisted thermal processing $(PATP;\;105^{\circ}C\;and\;700MPa)$ treatments for 40 min and 5min holding times, respectively. Different inoculum levels $(10^4,\;10^6\;and\;10^8CFU/ml)$ of C. sporogenes spores suspended in deionized water were treated at $105^{\circ}C$ under 700MPa with single, double, and triple pulses. Thermally treated samples served as control. No statistical significances (p>0.05) were observed among all different inoculum levels during the thermal treatment, whereas the inactivation rates $(k_1\;and\;k_2)$ were decreased with increasing the initial concentrations of C. sporogenes spores during the PATP treatments. Double- and triple-pulsed pressurization reduced more effectively the number of C. sporogenes spores than single-pulse pressurization. The study shows that the spore clumps formed during the PATP may lead to an increase in pressure-thermal resistance, and multiple-pulsed pressurization can be more effective in inactivating bacterial spores. The results provide an interesting insight on the spore inactivation mechanisms with regard to inoculum level and pulsed pressurization.

  • Two antifungal bacteria were selected from forest soils during the screening of microorganisms antagonistic to Cylindrocarpon destructans, a cause of ginseng root rot. The antifungal bacteria were identified as Bacillus subtilis (I4) and B. amyloliquefaciens (yD16) based on physiological and cultural characteristics, the Biolog program, and 16S rRNA gene sequencing analyses. Antagonistic activity of both bacterial isolates to C. destructans increased with increasing temperature. More rapid starch hydrolytic activity of the bacteria was seen on starch agar at higher temperatures than at lower temperatures, and in the higher density inoculum treatment than in the lower density inoculum treatment. The bacterial isolates failed to colonize ginseng root the root tissues inoculated with the bacteria alone at an inoculum density of $1{\times}10^6$ cfu/ml, but succeeded in colonizing the root tissues co-inoculated with the bacteria and C. destructans. Scanning electron microscopy showed that the pathogen was damaged by the low-density inoculum treatment with the bacterial isolates as much as by the high-density inoculum treatment. Both bacterial isolates were more effective in reducing root rot when they were treated at a concentration of $1{\times}10^6$ cfu/ml than at $1{\times}10^8$ cfu/ml. Also, only the former treatment induced prominent wound periderm formation, related to structural defense against pathogen infection. The results suggest that the bacterial antagonists may have high potential as biocontrol agents against ginseng root rot at relatively low-inoculum concentrations.
