Effects of Swainsonine on the Cell-mediated Immune Responses of Lipopolysaccharide (리포포리사카라이드의 세포성 면역반응에 미치는 스와인소닌의 영향)

  • Effects of swainsonine (SW: 8${\alpha}$, ${\beta}$-indolizidine-1alpha, 2${\alpha}$, 8${\beta}$-triol from Locoweed) on the cellular and nonspecific immune responses of lipopolysaccharide (LPS) wer e studied in ICR mice. Mice were divided into 4 groups (10mice/group), and LPS was given to each mouse 1 hr after i.p. injection with 3.7mg/kg of SW by i.p. injection twice a week for 14 days at a dose of 2mg/kg. Immune responses of the delayed-type hypersensitivity response (DTH) to sheep red blood cells (s-RBC), phagocytic activity and natural killer (NK) cell activity were evaluated. LPS treatment didn`t affect NK cell activity, phagocytic activity, DTH to s-RBC compared with those in controls, and phagocytic activity of sareoma 180 tumor bearing mice. However, circulating leukocytes were significantly decreased. Combinaton of LPS and SW increased circulating leukocytes significantly compared vath that in LPS alone, and DTH to s-RBC, NK cell activity and phagocytic activities of normal and sarcoma tumor bearing mice were not affected. These findings indicate that SW didn`t affected the cellular immune responses suppressed by LPS but significantly increased circulating leukocytes.

Analysis of Medication Errors of Nurses by Patient Safety Accident Reports (환자안전사고 보고서를 통한 간호사 투약오류 분석)

  • Purpose: The purpose of this study was to identify and analyze the characteristics of nurses' medication errors during three years. Methods: Retrospective survey study design was used to analyze medication errors by nurses among patient safety accidents. Data were collected for three years from January, 2017 to December, 2019. Data were analyzed using frequency, percentage, 𝑥2-test, and logistic regression with SPSS 26.0 program. Results: Of a total 677 medication errors, 40.6% were caused by nurses. Among the medication errors, near miss (n=154, 56.0%), intravenous bolus injection (n=170, 61.8%), wrong dose (n=102, 37.1%) and carelessness for repetitive work (n=98, 35.6%) were the most common. Medication errors differed by department, and nurses' career, and patient safety accident type. The results of the logistic regression analysis showed that the risk factors of adverse events were medication of fluids (OR=3.93, 95% CI: 1.26~12.27), insulin subcutaneous injection (OR=39.06, 95% CI: 4.58~333.18), and occurrence of extravasation/infiltration (OR=7.26, 95% CI: 1.85~28.53). Conclusion: The simplest and most effective way to prevent medication errors is to keep 5 right, and a differentiated education program according to department and nurse career is needed rather than general education programs. Hospital-level integrated interventions such as a medication barcode system or a team nursing method are also necessary.

Effects of Ginseng Total Saponin on methamphetamine-induced Hyperactivity and Striatal Dopamine Increase in Mice

  • The present study was undertaken to investigate the behavioral and biochemical effects of ginseng total sponin (GTS) on methamphetamine-treated mice. GTS (50 or 100 mg/kg) was administered intraperitoneally two times with 2 hour interval. Two hours after the second injection of GTS, methamphetamine (2 mg/kg) was administered subcutaneously. The ambulatory activity of mice was measured by the ti1ting-type ambulometer every 10 min. for 1 hour. Methamphetamine-induced hyperactivity was reduced by GTS. in a dose-dependent manner. To study the neurochemical mechanism underlying the GTS effects, monoamine contents were measured from brain tissues. After 45 min. of methamphetamine injection. mice were sacrificed and monoamine contents were determined from the striatum. Biochemical analysis revealed that GTS reduced the methamphetamine- induced increase in striatal dopamine contents. These observations indicate that inhibition of methamphetamine-induced hyperactivity by GTS is mediated by the modulation of dopaminergic nervous system, and it could be helpful for the therapy of hyperactivity.

Porcine Somatotropin Improves the Efficiency of Digestible Protein Use for Protein Deposition by Growing Pigs

  • A study was conducted to clarify the impact of recombinant porcine somatotropin (pST) on the efficiency of absorbed nitrogen use for protein deposition in growing pigs. Three levels of dietary crude protein (9.0, 11.5, 14.0% CP) were used. Each had either a sub-optimum or near optimum lysine: CP concentration (Low-lysine, 3.8 g/100 g CP and High-lysine, 5.5 g/100g CP) in order to achieve different metabolic efficiencies for nitrogen deposition (ca. 45 vs. 60%). Twelve crossbred female pigs $(59{\pm}4kg\;BW)$ were placed in metabolism cages and fitted with bladder catheters. Each pig received an excipient injection daily for the first 10-d, a pST (5 mg/d) injection for the second 10-d, and then excipient for the last 10-d. Pigs were randomly assigned to one of six dietary treatments (2 pigs/diet) and fed 4 times per d at $92g/kg\;BW^{0.75}$ $(3{\times}maintenance)$. Means for the excipient period were compared to means for the pST period. Urinary nitrogen (N) output declined in pST-treated pigs (p<0.01) irrespective of dietary protein content or lysine level. Nitrogen retention increased by an average of 11% (p<0.01) with pST treatment (726 vs. $803mg\;N/kg^{0.75}\;BW/d$). Forty-eight percent of the absorbed N was retained with Low-lysine diets, but this increased to 53% with pST injection (+11%, p<0.01). Pigs fed High-lysine diets retained 62% of absorbed N which increased to 69% with pST (+11% p<0.01). the addition of lysine improved N use by 27% (High vs. Low, p<0.01), but the effect of lysine and pST was additive (+40%). Therefore, pST improves N retention and the efficiency of apparently absorbed N use in growing pigs (>60kg). It does so with diets having the potential for either low or high efficiencies of N use (48% and 62%). More work is needed to determine if the partial efficiency of N use improves in direct proportion to pST dose since the improvement in protein deposition is a function of pST dose.

Effects of Several Herbicides on Control of Creeping Bentgrass in the Kentucky Bluegrass and Its Recovery (켄터키블루그래스 포장에 침입한 크리핑벤트그래스의 제초제를 이용한 방제와 회복)

  • This study was conducted to evaluate the selective control of creeping bentgrass invaded in Kentucky bluegrass by applying several herbicides and recovery of Kentucky bluegrass by sand injection seeding method. Selective herbicides such as mecoprop, triclopyr-TEA, imazaquin, bentazone and penosulam pyrazosulfuro-ethyl and non-selective herbicides such as glyphosate, paraquat dichloride and glyphsate ammonium oxyflorfen were used. Selective suppression of creeping bentgrass in Kentucky bluegrass was evaluated by turf color, chlorophyll and visual control indexes. Control of creeping bentgrass was most effective with the double dose application of triclopyr-TEA (2 Tri-T) in the selective herbicides and the 1 / 5 dose application of glyphosate ammonium oxyflorfen (1 / 5 GAO) in the non-selective herbicides. Visual control indexes by 2 Tri-T in selective herbicides and 1 / 5 GAOin non-selective herbicides were investigated 6.0 and 7.4, respectively. Treated sites were covered completely in 50 days after seeding Kentucky bluegrass by sand injection method.

α-Asarone Ameliorates Memory Deficit in Lipopolysaccharide-Treated Mice via Suppression of Pro-Inflammatory Cytokines and Microglial Activation

  • ${\alpha}$-Asarone exhibits a number of pharmacological actions including neuroprotective, anti-oxidative, anticonvulsive, and cognitive enhancing action. The present study investigated the effects of ${\alpha}$-asarone on pro-inflammatory cytokines mRNA, microglial activation, and neuronal damage in the hippocampus and on learning and memory deficits in systemic lipopolysaccharide (LPS)-treated C57BL/6 mice. Varying doses of ${\alpha}$-asarone was orally administered (7.5, 15, or 30 mg/kg) once a day for 3 days before the LPS (3 mg/kg) injection. ${\alpha}$-Asarone significantly reduced TNF-${\alpha}$ and IL-$1{\beta}$ mRNA at 4 and 24 hours after the LPS injection at dose of 30 mg/kg. At 24 hours after the LPS injection, the loss of CA1 neurons, the increase of TUNEL-labeled cells, and the up-regulation of BACE1 expression in the hippocampus were attenuated by 30 mg/kg of ${\alpha}$-asarone treatment. ${\alpha}$-Asarone significantly reduced Iba1 protein expression in the hippocampal tissue at a dose of 30 mg/kg. ${\alpha}$-Asarone did not reduce the number of Iba1-expressing microglia on immunohistochemistry but the average cell size and percentage areas of Iba1-expressing microglia in the hippocampus were significantly decreased by 30 mg/kg of ${\alpha}$-asarone treatment. In the Morris water maze test, ${\alpha}$-asarone significantly prolonged the swimming time spent in the target and peri-target zones. ${\alpha}$-Asarone also significantly increased the number of target heading and memory score in the Morris water maze. The results suggest that inhibition of pro-inflammatory cytokines and microglial activation in the hippocampus by ${\alpha}$-asarone may be one of the mechanisms for the ${\alpha}$-asarone-mediated ameliorating effect on memory deficits.

Botulinum Toxin A Treatment for Cervical Dystonia Resulting in Endoscopic Thyroidectomy: A Case Report (내시경 갑상샘 절제술 후 발생한 경부 근긴장이상증의 보툴리늄 A 독소를 이용한 치료: 증례보고)

  • Purpose: Surgery for thyroid gland requires skin incisions that can result in postsurgical neck scar. To overcome this, many surgeons performed a endoscopic thyroidectomy. But, this approach had a some problems. One of postoperative problems, iatrogenic cervical dystonia (CD) may occur. At common, CD is defined as a syndrome characterized by prolonged muscle contraction causing twisting, repetitive movements or abnormal posture. Botulinum toxin A (BTA, Botox$^{(R)}$, Allergan, Irvine, CA, USA) is well known treatment agent in the treatment of CD. So, the authors applied BTA injection in rare case with iatrogenic CD resulting in endoscopic thyroidectomy. Methods: A 43-year-old female had endoscopic subtotal thyroidectomy operation 3 years ago. She had symptoms such as progressive cervical pain, abnormal neck posture, depression, and sleep difficulty. About 1 year later, the patient who had previous myomectomy of the clavicular head of sternocleidomastoid muscle, however, symptoms were not improved. And then the patient received BTA therapy in our department. The 2 units per 0.1 mL solution was administered in a 1 mL tuberculin syringe. Results: The dose of BTA used in the patient was 36 units for vertical platysmal bands, superficially, and 10 units for ipsilateral sternocleidomastoid muscle, intramuscularly. After 2 weeks, additional the dose of BTA used in the patient was 5 points for remained scar bands, superficially. Complications related to injection such as significant swallowing difficulties, neck muscle weakness, or sensory change were not observed. In 9 months follow-up, the patient maintained a good result from the method of BTA injection alone. Conclusion: The basic concept is selective denervation for the hyperactive individual muscles and scar bands. We conclude that BTA is an effective and safe treatment for CD despite the iatrogenic and complex presentation of this complication.

YH439, a Hepatoprotective Agent, Suppresses Cytokines and Nitric Oxide Production in LPS-primed Rats Administered with $CCL_4$ ($CCI_4$와 Lipopolysaccharide로 유도한 흰쥐 간 독성에 대한 YH439의 방어작용 : cytokines 및 nitric oxide 생성의 억제)

  • The aim of the present investigation was to examine whether YH439, a hepatoprotective agent, exerts protective effect against hepatotoxicity and reduces the production of cytokines and NO in lipopolysaccharide (LPS)-primed rats with carbon tetrachloride ($CCl_4$). Administration of LPS following a single dose of CCl4 injection resulted in remarkable elevations of the serum $TNF{\alpha},{\;}IL-l{\beta$ and IL-6 level. The serum NO level was moderately elevated and severe liver damage was evidenced by increases in serum alanine aminotransferase (ALT) and sorbitol dehydrogenase (SDH) activities. YH439 decreased the levels of TNF, $IL-l{\beta}$, IL-6, ALT, SDH as well as NO in the serum elevated by CCl4+LPS in a dose-dependent manner. Inducible nitric oxide synthase (iNOS) level was decreased in the liver of rats treated with YH439. The increased iNOS activity induced by LPS and $interferon-{\gamma}$ was significantly decreased in RAW 264.7 cells by YH439 treatment. YH439 increased the GSH level decreased by $CCl_4+LPS$ and suppressed the ratio of GSSG/GSH. The reduction of hepatotoxicity by YH439 may associated with the decrease in the production of cytokines as well as suppression of iNOS protein in conjunction with an increase in the GSH level.

  • Calvert et al. formulated the hypothesis that carbon tetrachloride ($CCl_4$) acted on the central nervous system to produce and intensify sympathetic discharge which resulted in anoxic necrosis of the liver. Recknagel suggested that the essential feature of $CCl_4$ hepatotoxicity depended on the cleavage of it to $CCl_3$(free radical) and the peroxidative decomposition of cytoplasmic membrane structural lipids. And there are many reports which show the increase of adrenergic activity in hyperthyroidism. In this paper, the influence of thyroxine on the hepatotexicity of carbon tetrachloride was investigated in mice. The results obtained were summarized as follows; 1) Hepatic total lipid and lipid peroxide contents were slightly decreased by L-sodium thyroxine injection(4mg/kg/day for 4days or 6days), but hepatic glycogen content was significantly decreased. 2) Hepatic total lipid and lipid peroxide contents and serum lactic dehydrogenase activity were significantly increased by $CCl_4$ (4 ml/kg single dose or triple dose: 4ml/kg/day for 3days), but hepatic glycogen content was significantly decreased. 3) The increase of hepatic total lipid and lipid peroxide contents and serum lactic dehydrogenase activity induced by $CCl_4$ were significantly inhitited by the pretreatment of thyroxine. 4) The decrease of hepatic glycogen induced by $CCl_4$ was not affected by the pretreatment of thyroxine.

Effects of Characterization of Polymeric Al(III) Coagulants on Coagulation of Surface Water (고분자성 Al(III) 응집제의 특성이 상수원수의 응집특성에 미치는 영향)

  • This research explored the feasibility of preparing and utilizing a preformed polymeric solution of Al(III) for coagulation in water treatment. Slow base(NaOH) injection into supersaturated aluminum chloride solutions did produce high yields of the type of Al polymers useful to water treatment applications. PACl's characteristic analysis showed that the quantity of polymeric Al produced at value of $r(OH_{added}/Al)=2.2$ was 83% of the total aluminum in solution, as showing maximum contents and precipitate was dramatically increased when r was increased above 2.35. And PACl was stable during sitoring period so aging effect was negligible. Results of the coagulation of Nakdong river waters with three PACls showed that the effectiveness of the three coagulants can be considered as r = 2.2 > r = 2.0 > r = 2.35 which are also the order of higher polymeric aluminum contents. Coagulation results for synthetic water exhibited optimum dose of 0.25mM Al, for three PACls, but above optimum dose, r = 2.0 and 2.2 PACl impaired the coagulation and sedimentation of turbidity and humic acid because of the restabilization of particulate. The effect of pH for on coagulation of Nak Dong River water showed that it had much effect turbidity and TOC removal, especially near pH 7. But pH effect was little for turbidity and TOC removal when r = 2.35 PACl was used for coagulation, that PACl had much more precipitates content.
