The Optimization of the Composition of Nitrogen Source in the Medium of Alcohol Fermentation of S. cerevisiae (S. cerevisiae에 의한 알콜배지에서 질소원 조성의 최적화 연구)

  • The effect of concentration of yeast extract and NH4Cl in the mediun of alcohol fermentation of S. cerevisiae ATCC 24858 on the fermentation characteristics, specific growth rate, sugar conversion, alcohol productivity was experimentally investigated. Regardless of initial sugar concentrations, the values of the above three characteristics increased with augument of concentration of yeast extract. However, the increasing tendency ceased above a certain concentration. The concentration of NH4Cl had little effect on the change of the three characteristics. The functional relationships between the concentration of yeast extract and the characteristics were different according to the initial sugar concentrations, but those between the ratio of yeast extract concentration to initial sugar concentration and the characteristics could be expressed as same forms respectively regardless of initial sugar concentrations. Also the values of the three characteristics approached to the maximum values around 0.085 of the ratio, but did not increase any more above 0.1 of the ratio. We have come to conclusion that the optimum ratio of the yeast extract concentration to the initial sugar concentration was about 0.085 and the ratio should not be decided as greater than 0.1 in the medium of alcohol fermentation of S. cerevisiae ATCC 24858.

Electron Donor Determination and Comparisons of Reaction Rates for Bioremediation of Nitrate Contaminated Groundwater (질산성 질소로 오염된 지하수의 생물복원을 위한 적정 전자공여체의 결정 및 반응속도 비교 연구)

  • Groundwater contamination by nitrate exceeding water quality criteria (10 mg $NO_3{^-}-N/L$) occurs frequently. Fumarate, acetate, formate, lactate, propionate, ethanol, methane and hydrogen gas were evaluated for their nitrate removal efficiencies and removal rates for in situ bioremediation of nitrate contaminated groundwater. Denitrification rate for each substrate was in the order of: fumarate > hydrogen > formate/lactate > ethanol > propionate > methanol > acetate. Microcosm studies were performed with fumarate and acetate. When fumarate was used as a substrate, nitrate was removed 100 percent with rate of 0.66 mmol/day while conversion rate from nitrate to nitrogen gas or another by-product was 87 percent. 42 mg of fumarate was needed to remove 30 mg $NO_3{^-}-N/L$. When using acetate as carbon source, 31 percent of nitrate was removed during initial adjustment period. Among removed fraction, however, 83 percent of nitrate removed by cell growth. Overall nitrate removal rate was 0.37 mmol/day. Acetate showed longer lag time in consumption compared to that of nitrate, which implying that acetate would be better carbon source compared to fumarate as more amount was utilized for nitrate removal than cell growth.

Effect of Degradative Products on Dibenzothiophene Desulfurization Activity of Gordona sp. MS6 (Gordona sp. MS6균주의 dibenzothiophene 탈황능에 미치는 분해산물의 영향)

  • Gordona sp. MS6, desulfurizing petroleum, can convert dibenzothiophene (DBT) to 2-hydroxybiphenyl (2-HBP) and sulfate. In this study, the effect of DBT degradation product on DBT desulfurization activity was investigated. When $Na_2$SO$_4$ and DBT were simultaneously added in the medium sulfur sources, specific growth rate of strain MS6 was doubled compared to not adding $Na_2$SO$_4$. But, sulfate inhibited DBT desulfurization rate, furthermore, when 0.05 g/L $Na_2$SO$_4$ was supplied DBT desulfurization rate decreased down to 40%. When 2-HBP and its derivative, 2, 2'-dihydroxybiphenyl (DHBP) were not added, DBT desulfurization rate was 5.0 $\mu$mol L$^{-1}$ㆍh$^{-1}$. With the increase of 2-HBP and DHBP concentration, DBT desulfurization rate was decreased. No DBT desulfurization activity of Gordona sp. MS6 was observed when initial concentration was over 0.15 mM 2-HBP and 0.8 mM DHBP [Gordona sp., Desulfurization, Dibenzothiophene, 2-Hydroxybiphenyl, Sulfate].

The Effect of Dredged Soil Improvement on Soil Chemical Conditions and Plant Growth at the Slope of Saemangeum Sea Dike

  • This study was conducted to determine the changes in soil chemical properties and the growth of seedling according to the different dredged soil improving methods at slope of Saemangeum sea dike. Undersea dredged soil was improved by five different methods. Seedlings of Ulmus davidiana var. japonica, Chionanthus retusa, Celtis sinensis, and Pinus thunbergii were planted after 9 month of experience site installation, then soil pH, NaCl concentration in soil, soil organic matter (SOM), and survival rate and height of seedling was measured. Initial soil pH was highest in the control plot but it decreased to the similar level with other soil improving plots after 35 months. There were no differences in NaCl concentration between the control and soil improving plots, and it showed decreasing tendency during the study period. In the control plot, initial SOM was lowest among that of other plots during the study period. The survival rate of 36 months after planting of P. thunbergii was highest among the species. The gap of the tree growth of P. thunbergii between the control plot and the soil improving plots was small, however, other species showed relatively higher tree height in the soil improving plots than the control plots. Creation forest with P. thunbergii might be a cost effective afforestation in coastal reclaimed land since it rarely needs additional improvement of dredged soil.

Growth Characteristics of Rhoodotorula sp. Y-55 on Ethanol, Acetic acid, and Acetaldehyde Substrates (Ethanol, Acetic acid, Acetaldehyde 기질에서의 Rhoodotorula sp. Y-55의 증식 특징)

  • The growth characteristics of Rhodotorula sp. Y-55 were examined on minimal medium containing ethanol, acetic acid or acetadehyde as a sole carbon source by batch culture. The increased concentration of substrate reduced overall growth yield and prolonged lag time. The specific growth rate of the yeast was changed, depending upon the initial concentrations of ethanol and acetaldehyde during the exponential period, but was constant on acetic acid without regard to the initial substrate concentrations, giving a value of 0.l07h-1. The highest ${\mu}$ value was obtained on ethanol and acetadehyde substrates and the respective values were 0.270 at 20g/L and 0.041h-1 at 0.2g/L. The maximum overall growth yields were appeared to be 32.6% for ethanol of 10g/L, 25.6% for acetic acid of 20g/L, and 45% for acetaldehyde of 0.2g/L. The respective cellular contents of crude protein and nucleic acids were determined to be 41.5 and 4.9wt% on ethanol and 40.2 and 4.7wt% at the concentration revealing maximal growth yield.

Effects of Nutrient Solution on Growth and Amount of Ginsenoside of Two Year Old Ginseng Grown under Hydroponic Culture (수경재배 양액조건이 2년생 인삼의 생육 및 진세노사이드 함량에 미치는 영향)

  • Background: Electrical conductivity (EC) and pH are important features of nutrient solution, affecting both growth and quality of crops by altering nutrient uptake. Methods and Results: The pH values of nutrient solutions were controlled at 5.0, 5.5, 6.0, 6.5 and EC values were controlled at 0.68, 0.84, 1.23, 1.41 dS/m. Gingesng root weights were higher during the initial growth period when the plants were treated with low pH and low EC nutrient solutions. However, the higher pH and EC levels, the greater the increase in the rate of root weight between the initial and middle growth periods. The highest ginsenoside amount changed during growth period. The total ginsenoside amount was highest in the root, and the lowest in leaves at 45 and 90 days after treatment, respectively, with solution at a pH of 6.0. After 135 days of treatment, the highest total ginsenoside amount was detected in root treated with soluton with EC values of 1.23 dS/m. Conclusions: For the cultivation of ginseng using a nutriculture system, the pH and EC values of nutrient solutions should to be controlled based on the stage of growth and targeted plant organ (root or leaves).

Evaluation for Soil Moisture Stabilization and Plant Growth Response in Horizontal Biofiltration System Depending on Wind Speed and Initial Soil Moisture (풍속과 초기 토양수분에 따른 평면형 바이오필터 내 토양수분 안정화 및 식물 생육반응 평가)

  • The final aim of this study is to develop a biofiltration system integrated with plant vegetation for improving indoor air quality effectively depending on indoor space and characteristics. However, to approach this final goal, several requirements such as constant pressure drops (PDs) and soil moisture contents (SMCs), which influence the capacity design for a proper ventilation rate of biofiltration system, should be satisfied. Thus, this fundamental experiment was carried out to adjust a proper wind speed and to ensure a stabilization of initial SMCs within biofilter for uniform distribution of SMCs and PDs, and for normal plant growth, especially avoiding root stress by wind. Therefore, we designed horizontal biofliter models and manufactured them, and then calculated the ventilation rate, air residence time, and air-liquid ration based on the biofilter depending on three levels of wind speed (1, 2, and $3cm{\cdot}s^{-1}$). The relative humidity (RH) and PD of the humidified air coming out through the soil within the biofilter, and SMC of the soil and plant growth parameters of lettuce and duffy fern grown within biofilter were measured depending on the three levels of wind speed. As a result of wind speed test, $3{\cdot}sec^{-1}$ was suitable to keep up a proper RH, SMC, and plant growth. Thus, the next experiment was set up to be two levels of initial SMCs (low and high initial SMC, 18.5 and 28.7%) within each biofilter operated and a non-biofiltered control (initial SMC, 29.7%) on the same wind speed ($3cm{\cdot}sec^{-1}$), and measured on the RH and PD of the air coming out through the soil within the biofilter, and SMC of the soil and plant growth parameters of Humata tyermani grown within biofilter. This result was similar to the first results on RHs, SMCs, and PDs keeping up with constant levels, and three SMCs did not show any significant difference on plant growth parameters. However, two biofiltered SMCs enhanced dry weights of the plants slightly than non-biofiltered SMC. Thus, the stability of this biofiler system keeping up major physical factors (SMC and PD) deserved to be adopted for designing an advanced integrated biofilter model in the near future.

The structural relationships among adolescents'mobile phone dependency, trajectories of depression, and self-regulated learning abilities (청소년의 휴대전화의존도, 우울의 변화 궤적 및 자기조절학습 능력 간의 구조적 관계)

  • The purpose of this study was to examine the longitudinal relationships between Korean adolescents'mobile phone dependency, trajectories of depression, and self-regulated learning abilities. To achieve these goals, structural equation modeling analysis was conducted, using the 3rd, 5th and 7th wave of the data on 4th graders taken from the Korean Children and Youth Panel Survey. The results can be summarized as follows. First, growth-curve longitudinal analysis indicates that depression in 6th through 10th grade has increased. Second, mobile phone dependency among adolescents at 6th grade has a significant effect on both the initial value and the rate of change in depression. Also, the initial value and the rate of change in depression have significant relationships with mobile phone dependency at 10th grade. Moreover, both increased levels of mobile phone dependency and the rate of change in depression significantly influence adolescents'self-regulated learning abilities at 10th grade. Based on a longitudinal data set, these findings demonstrate the causal relationships between Korean adolescents'trajectories of depression and their mobile phone dependency. The findings also provide a comprehensive framework with implications for adolescents'development through an understanding of the relationships between adolescents'depression and mobile phone dependency, which impact their self-regulated learning abilities.

Characteristics of Microalgal Growth on Anaerobic Effluent of Animal Waste (축산분뇨 혐기성 처리수에서 미세조류의 성장특성)

  • Characteristics of microalgal growth was investigated using anaerobic effluent from two-phase animal waste digestor as substrate. Batch experiments were carried out to investigate the effect of the initial nitrogen and phosphorus concentrations on growth of Microcystis aeruginosa, Chlorella sp. and Euglena gracilis. In 400 times diluted anaerobic effluent (TN 3 mg/L), single cell growth of the Euglena gracilis population increased twice without delay, although Chlorella sp. and Microcystis aerugenos take over 144 hours. Similar appearance with single cell growth was observed in mixed cultures. However, microalgae population did not increase under condition of 10 times diluted influent (TP 3 mg/L) in both pure and mixed cultures, which was affected by high organic and nitrogen concentration. Logistic growth model successfully fitted to determine biokinetic parameters such as ${\lambda}$: lag time, ${\mu}m$: maximal specific growth rate, A: asymptote of growth.

Agglomeration (Dis-) Economies and Regional Economic Growth as a Spatial Economy (집적 (불)경제와 공간경제로서의 지역 경제 성장)

  • A regional economy is characterized as a spatial economy. However the literature shows that it has been treated as a point economy since space is little recognized in regional modeling due to mathematical complication. This leads to the fact that regional model does not sufficiently represent regional characteristic. This paper attempts to construct a regional growth model in a partial equilibrium framework specifically taking into consideration land as a primary factor. The model is formulated largely neoclassical. Labor is assumed to move in response to differences in the wage rate, while capital is perfectly mobile across regions. The paper shows that two growth equilibrium points exist, one stable equilibrium point and the other unstable equilibrium point. The unstable growth equilibrium indicates the existence of minimum threshold that a region must overcome the minimum threshold to grow constantly. Consequently, directions of regional growth are characterized by two growth paths depending on the initial condition of a region. That is to say, a region below the minimum threshold is converging toward the lower stable equilibrium point over time. When a regional economy initially lies above the minimum threshold, it will grow forever. A regional economy is not thus necessarily converging a stationary is not thus necessarily converging a stationary equilibrium point through factor movement. Finally, the impacts of the presence of agglomeration economies and diseconomies are analyzed through the phase diagram. The paper also shows that agglomeration economies result in lowering the minimum threshold and in escalating the level of stable equilibrium However, when agglomeration diseconomies prevail, the results are opposite, i.e., rising the minimum threshold of growth and lowering the growth level of stable equilibrium.
