Effect of pH on Growth and Cultural Characteristics of Bacillus sp. SH-8 and Bacillus sp. SH-8M (Bacillus sp. SH-8과 Bacillus sp. SH-8M의 생육 및 배양 특성에 미치는 pH의 영향)

  • The growth and cultural characteristics of Bacillus sp. SH-8 and SH-8M were investigated at various pH conditions. Bacillus sp. SH-8 showed normal growth pattern above pH 9.0. However, with the pH adjusted below 7.7, 0.$D_{550}$ decreased rapidly with concomitant reduction in viable cell numbers. In contrast, Bacillus sp. SH-8M demonstrated growth capability at pH 7.7, but with slightly reduced growth rate at pH 6.9. Similar results were obtained when those two strains were cultivated on the solid medium. Both of them showed short rod shapes at pH 10.2. However, at pH 7.7 only Bacillus sp. SH-8 was observed to have elongated rod shape. Extracellular pH of both the strains, when cultured at initial pH of 10.2, reached to 9.0 after the incubation of 28 hours. At the initial pH of 9.0 and 9.6, the extracellular pH was reduced at the beginning of cultivation, but elevated after 12 hours. When cultured at initial pH of 6.9 and 7.7, extracelluar pH of Bacillus sp. SH-8M increased to 8.0 and 8.7, respectively, while that of Bacillus sp. SH8 remained constant pH 7.0. The highest sporulation rate of Bacillus sp. SH-8 and SH-8M was obtained at the initial pH of 10.2 and after the incubation of 3 days with the sporulation rate of 95% and 85%, respectively.

The Production of Sodium Gluconate by Aspergillus niger (Aspergillus niger를 이용한 글루콘산 나트륨의 생산)

  • Sodium gluconate was produced by neutralization of gluconic acid formed during the submerged culture fermentation of glucose with Aspergillus niger ACM 7. The fermentation characteristics of Aspergillus niger ACM 7 were investigated quantitatively according to the change of the initial glucose concentrations and the initial pHs of fermentation broth. The maximum specific growth rate was estimated to be $0.20hr^{-1}$ at 95g/$\ell$ of initial glucose concentration. The maximum fermentability of sodium gluconate was 95% at the initial glucose concentration of 26g/$\ell$. However, the maximum sodium gluconate productivity was 1.18g/$\ell$/hr when the initial glucose concentration was 110g/$\ell$. The optimum pH was found to be 5.5 for both the cell growth and the sodium gluconate production. With optimized culture conditions, the productivity of sodium gluconate in a fed-batch culture(production fermentor, 16,000$\ell$) increased up to 7.1g/$\ell$/hr.

  • This study is to assess the effects of increasing wall thickness on the safety margin of pressure tube in operating and of lowering initial hydrogen concentration on the DHC growth in respect to the improvement of the reliability of pressure tube in CANDU reactors. The pressure tube with thicker wall of 5.2 mm shows much higher safety margin for flaw tolerance by 25% than the current 4.2mmm tube. The thicker pressure tubes have a great benefit in LBB assessment including the initial crack depth at which DHC occurs, the crack length at onset of leaking and the available time for action. The resistance for the pressure tube ballooning at LOCA accident is also increased with the thicker tube. The calculations for Heq concentration after 20 years of operation as a function of wall thickness and initial hydrogen concentration show that the 5.2 mm nil thickness tube with 5 ppm initial hydrogen concentration is the most resistant to DHC. with the lower initial hydrogen concentration, TSS temperature for the precipitation or hydride decreases and the crack growth during cooldown reduces.

  • Variation of sintered density of BaTiO3 powder calcined at 120$0^{\circ}C$ and 135$0^{\circ}C$ was investigated with respect to the grain growth behavior. It was found that BaTiO3 powder, which was calcined at 120$0^{\circ}C$, showed abnormal grain growth behavior during sintering process. At initial stage of sintering process, the densification rate of specimen was accelerated with rapid grain growth caused by the abnormal grain growth. But with the increase of sintering time, abnormally grown grain met each other and the density of specimen decreased drastically due to coalescence of pores located in triple junction. On the contrary, BaTiO3 powder calcined at 135$0^{\circ}C$ showed normal grain growth behavior and gradually densified with the increase of sintering time.

  • Previous studies have confirmed that fermented whey produced by Leuconostoc mesenteroides CJNU 0147 or Lactobacillus casei CJNU 0588 display bifidogenic growth stimulator (BGS) activity. The present study sought to determine if the strain itself can improve the growth of bifidobacteria in co-cultures. In reinforced clostridial medium (RCM), both strains stimulated the growth of a Bifidobacterium lactis strain during the exponential phase and also stimulated the growth during almost all growth phases in whey broth. Fermented whey containing viable Leu. mesenteroides CJNU 0147 and L. casei CJNU 0588 cells maintained viability of the B. lactis strain stored at $10^{\circ}C$ in MRS broth. Viable cell count of the B. lactis strain without the fermented whey was decreased to 5.6 log cfu/mL after 15 days, whereas that of the strain with the fermented whey was slightly increased to 7.1 log cfu/mL as compared with initial viable cell count of 6.9 log cfu/mL.

The Effects of Negative Emotionality and Mother's Social Parenting during Infancy on Peer Interaction at Age 3 : A Longitudinal Study Using Latent Growth Modeling (영아기 부정적 정서성과 어머니의 사회적 양육행동이 3세 유아의 또래 상호작용에 미치는 영향 : 잠재성장모형을 이용한 종단 연구)

  • The purpose of the present study was to examine the trajectories of children's negative emotionality and mother's social parenting over a 3-year period (for children at ages 1-3) and to then analyze the bidirectional effects between the two variables. The longitudinal casual relationship among children's negative emotionality, mother's social parenting and peer interaction at age 3 was also examined. The data, taken from the Panel Study on Korean Children, were analyzed using latent growth modeling. The results were as follows. First, the level of negative emotionality was seen to have increased by 2 years of age, whereas the level of mother's social parenting decreased by this point. Second, higher initial negative emotionality predicted decreases in mother's social parenting, However, higher initial mother's social parenting predicted increases in children's negative emotionality. The initial level and slope of mothers' positive parenting predicted peer interaction at age 3, while only the slope of negative emotionality predicted peer interaction. These findings suggest that temperament and parenting predict changes in each other and peer interaction.

Effects of Initial Concentration and Nutrients in Treatment of petroleum Hydrocarbon Contaminated Soils using a Slurry-Phase Bioreactor (슬러리상 생물반응기를 이용한 석유계탄화수소 오염토양의 처리에 있어서 초기농도 및 영양소의 영향)

  • The purpose of this study was to evaluate effects of initial concentration and nutrients in treatment of petroleum hydrocarbon contaminated soils. The reactor used in this study was slurry-phase bioreactor of in-vessel type. Performance results on treatment of diesel fuel contaminated soils and micorbial growth were generated at the bench-scale level. The fate of TPH(Total Petroleum Hydrocarbon) and the microbial growth were evaluated in combination with biodegradation rate. Effect of initial loading levels of 50,000 and 100,000mg TPH/kg soil was studied. Performance results with two reactors were showed at the total TPH removal rate of 90.5% and 90.8%, respectively. However, the reactor with the initial concentration of 50,000mg TPH/kg soil showed higher biological TPH removal efficiency except for removal by volatilization than the other Although the different amount of nutrients was applied in two reactors, there was no remarkable difference in microbial growth rate. However, considerable factor in this results was that applied different initial concentration to two reactors. Although initial concentration was two times higher than it applied to the reactor without addition of nutrients, in total and biological TPH removal rate the reactor with addition of nutrients showed a higher than the other.

Longitudinal Root Anatomy, Cell Dynamics, and Physiological Cell Responses in Root Growth Zones of Two Tall Fescue Genotypes at Two Nitrogen Levels (톨페스큐 뿌리생장부위의 종적해부구조, 세포역학 및 생리적 반응에 대한 질소효과)

  • Anatomical studies of sink tissues are required for better understanding the biological plant growth system and energy metabolism. Kinematics of root growth zones of two genotypes of tall fescue (Festuca arundinacea Schreb.) receiving 50 or 200 ppm N were determined. Longitudinal anatomy and cell dynamics of root growth zones were studied and calculated. The root growth zone is organized similarly to the leaf growth zone which has cell division, elongation, and maturation zones, but the root growth zone is only about 3.0 mm long compared to 25 to 30 mm for the leaf growth zone. The root cap extends about 0.4 to 0.5 mm from the apical initial, while the cell elongation zone for both cortical and metaxylem cells extends about 3.3 mm from the apical initial for both genotypes and N levels. Root cap cells elongate from an initial length of about 5$\mu{m}$ long to a final length of about 40$\mu{m}$ before being sloughed. Initial lengths of cortical and metaxylem cells were about 8.5 $\mu{m}$ and 13.0 $\mu{m}$, respectively. Elongation of cortex and metaxylem cell showed sigmoidal curves with final lengths of about 120 $\mu{m}$ for cortex cells and 650 $\mu{m}$ for metaxylem cells. Initial size and final size for both types were not affected by N level, but cell fluxes and cell elongation rates of cortical and metaxylem cells were about double in low N. Cell production rates were about 5 to 6 times higher in cortical cells than in metaxylem cells. Differences in N caused a larger change in cell production rate, duration of cell elongation, and relative cell elongation rate than did the genotypes. These data indicate that N application affects root growth longitudinally by changing cell production rate and elongation rate.

  • A one site chemotherapy agent-diffusion model is proposed which accounts for diffusion of chemotherapy agent, normal and cancer cells. It is shown that, by controlling the initial conditions, consequently an initial dose of the chemotherapy agent, the system is guaranteed to evolute towards a target equilibrium state. Or, growth of the normal cells occurs against decay of the cancer cells. Effects of diffusion of chemotherapy-agent and cells are investigated through numerical computations of the concentrations in square and triangular cancer sites.

An effect of initial disturbance on the breakup mechanism of liquid jet (초기교란이 액주의 분열기구에 미치는 영향)

  • The present experimental study investigates the effect of an initial disturbance on the breakup mechanism of a liquid column. With varying the maginitude of the inital disturbance, we measure the surface wave of liquid column with adopting laser shadow method and analyze the growth rate of liquid column and breakup frequency. The experimental results show that thebreakup characteristics of liquid column is significantly influenced by the frequency of the initial disturbance. We concluded that the most uniform droplet occurs when the frequency of initial disturbance coincides with the natural frequency of the liquid column.
