Effects of Concentration of Glucose and By-product on the Growth of Bacillus amyloliquefaciens 23350 (B. amyloliquefaciens 세포 성장에 미치는 포도당 및 부산물 농도의 영향에 관한 연구)

  • In this study, an attempt was made to investigate optimum cell growth and products by Bacillus amyloliquefaciens 23350 in batch culture by varing carbon soures. Maximum dry cell density increased with the increase of initial glucose concentration. Maximum dry cell density was obtained with the highest value of 5.2g/l at 30g/l of initial glucose concentration. By adding acetic acid at 20g/l of initial glucose concentration, the cell growth rate decreased with the increase of initial acetic acid concentration. Among the various carbon sources, maximum $\alpha$-amylase production was obtained with 225unit/ml at 10g/1 of initial glucose concentration. Optimum production of $\alpha$-amylase was obtained with 376unit/ml at 2g/l of initial acetic acid concentration and 20g/l of initial glucose concentration. By 10g/1 of initial glucose concentration, both good maximum specific cell growth rate and maximum $\alpha$-amylase production rate were obtained. In view of the results studied optimum production and specific production rate of $\alpha$-amylase, acetic acid was initially added 2~4g/l with 20g/1 of initial glucose concentration in batch culture.

Settling Mode of the Dredged Soil (준설토의 침강형태에 관한 연구)

  • The settling of the dredged soil may vary with mineral composition, grain size distribution, initial water content and salt concentration of suspension of the site. A series of settling column test was performed to investigate the behaviour of solid suspension material from dredging and reclamation. Settling mode was divided into four types from the observation of interface and settling curves of clay minerals and marine clay samples, and the relationship charts of salt concentration and the initial water content were established to use in the dredging operation with any salt concentration. The critical initial water content which was defined as a threshold of zone settling and the consolidation settling was varied with salt concentration of water and was proportional to the plasticity of soil in sea water.

  • In this experiment, we studied about concentration characteristics of $NO_x$ and generation of ozone in the reactor of corona discharge type by using mixed gas of $NO/N_2$ and $N_2/O_2$. In the case of the initial NO concentration increased, decrease rate of NO concentration was weakened and discharge input power of minimum NO concentration became high. When NO concentration was high, NO decomposition limit was appeared. And NO reduction rate was decreased, when initial NO concentration and discharge input power increased. When discharge input power was 5W, we could know the most proper energy value for treatment of NO. When the concentration of initial NO increased, generation of ozone decreased and in the case of same concentration of NO, according to discharge input power increase, generation of ozone increased.

Studies on the Water Purification Using Water Parsley (미나리 (Oenanthe javanica(Blume) DC)를 이용한 수질정화에 관한 연구)

  • The results of the water purification studies using water parsley (Oenanthe javanica) were obtained as follows. The removal rate of nutrient salts increased with longer plant growth periods. The results of this study use the assumption, log(T+1) = $K(C_0-C)^A$, based on Prakish's Theory. The initial concentration was calculated from experimental data. A and K are closely related to the initial concentration. It is possible to model the concentration of residual salts, as time goes by, if concentration is constant. It was observed that water parsley neutralizes acid and alkali substances promptly. The maximum suitable neutralization period is 48 hours. But water parsley withered up in strong acid and alkali solutions within a week. The removal efficiency of Cd progresses in 2 steps, which are unrelated to the initial concentration of Cd. The first part of the curve shows the concentration rapid rate of Cd removal, followed by a levelling off. The removal rate of $NO_x-N$ in the sample water tank containing 0.5 ppm Cd was between 50~80% but the removal rate was less than 20% for the higher concentration. On the other hand, increased amounts of $PO_4-P$ in the sample water tank from the third day on suspected that $PO_4-P$ was desorbed from the water parsley. The accumulation efficiency of Cd in plant was increased in proportion to the initial concentration of Cd. The accumulation phenomenon was observed in the tanks more than 50~100 times.

  • In predicting oxidants concentration, the most important fact is to select a suitable photochemical reaction mechanism. Sensitivity analysis of $O_3$ and other important photochemical oxidants concentrations was conducted by using CBM-IV model. The predicted oxidants concentration was considerably related with the initial concentration of formaldehyde, $[NO_2]/[NO],\;NO_x$, RH and RCHO. As the initial concentration of formaldehyde increased, concentration of $NO_2$ increased. $O_3$ concentration was proportional to the $[NO_2]/[NO]$ ratio. When the initial concentrations of RH and RCHO were high, photochemical reaction was more reactive, including more rapid conversion of NO to $NO_2$ and increased oxidants. Also, the sensitivities of ozone formation to rate constants, $K_l,\;K_2\;and\;K_3$ in the $NO_2$ photolysis were studied.

  • This study was investigated experimental condition which is able to evaluate photocatalytic activity of various commercial $TiO_2$. The experiments were performed for three representative substances (ethanol, phenol and methylene blue) and four kinds of commercial $TiO_2$, under the experimental conditions such as pH, reactant concentration, amount of $TiO_2$, reaction time and UV intensity. The optimum experimental conditions to evaluate photocatalytic activity were as follows : for ethanol, the initial concentration 1000 ppm, initial pH 8, $TiO_2$ loadings 0.1 wt%, and reaction time 90 minutes: for phenol, the initial concentration 200 ppm, initial pH 8, $TiO_2$ loadings 1 wt%, and reaction time 60 minutes: for methylene blue, the initial concentration 200 ppm, initial pH 4, $TiO_2$ loadings 0.5 wt%, and reaction time 30 minutes.

  • The purpose of study is to examine the sensitivity of the rate of degradation to initial TCE concentration and iron concentration in the solution. The batch tests were executed to assess the degradation rate at varying initial conditions. First order rate constants($k_a$) were more rapid with the lower initial TCE concentration, Howere the correleation was not always linear between $k_a$ and initial TCE concentration. $k_a$ was proportionally increased as the increasing surface area. It implied that the effective reactive surface area acted as the limiting factor on the reductive dechlorination of TCE by iron.

  • Characteristics of ethanol fermentation were investigated during the stationary culture of a killer yeast, Saccharomyces cerevisiae B15-1. Specific ethanol production rate reached the maximum level, 1.203 g-EtOH/g-cell-hr, at 150 g/l of the initial glucose concentration. No big differences were obtained in ethanol fermentability based on the initial sugar concentration below 150 g/l. When 200 g/l of sugar was used, fermentability dropped significantly. Although the final cell mass and the amount of ethanol produced were increased, their increase rates were declined according to the increase of initial sugar concentration. It was found that most of the sugar used below 150 g/l of concentration could be changed to ethanol. However, when 200 g/l of sugar was used, some of them remained in the media even after increase of cell mass and fermentation stopped. The ethanol yield was decreased when initial sugar concentration was high, and were increased when the amount of ethanol produced was increased and finally reached the plateau over 60 g/l of ethanol concentration.

  • Emulsion liquid membrane (ELM) process suffers from emulsion instability problem. So far, emulsion produced by mechanical methods such as stirrer and homogenizer has big size and high emulsion breakage. This paper discussed the application of emulsion produced by sonicator to extract cadmium in a batch ELM system. The emulsions consist of N,N-Dioctyl-1-octanamine (trioctylamine/TOA), nitrogen trihydride (ammonia/NH4OH), sorbitan monooleate (Span 80), and kerosene as carrier, stripping solution, emulsifying agent, and organic diluent, respectively. Effects of comprehensive parameters on extraction efficiency of Cd(II) such as emulsification time, extraction time, stirring speed, surfactant concentration, initial feed phase concentration, carrier concentration, volume ratio of the emulsion to feed phase, and pH of initial feed phase were evaluated. The results showed that extraction efficiencies of Cd(II) greater than 98% could be obtained under the following conditions: 15 minutes of emulsification time, 4 wt.% of Span 80 concentration, 4 wt.% of TOA concentration, 15 minutes of extraction time, 250 rpm of stirring speed, 100 ppm of initial feed concentration, volume ratio of emulsion to feed phase of 1:5, and initial feed pH of 1.53.

A Study on the Phosphorus Removal from Wastewater by Eggshell (난각을 이용한 폐수중의 인 제거에 관한 연구)

  • This study is a fundamental research to test the applicability of abandoned eggshell as seed material for crystallization reaction. Eggshell was calcinated at $850^{\circ}C$ and ground to lesser than 0.42mm. The calcination characteristics of eggshell were examined by X-ray diffraction (XRD). The effect of initial calcium concentration, alkalinity, reaction temperature condition, seed dosage were studied by batch test. For the low concentration sample(P concentration is under 50mg/L), more than 90% of P can be removed. The effect of initial calcium concentration(0~120mg/L) was performed. At the result of the test, more than 50mg/L calcium concentration has high removal efficiency. Alkalinity effect was studied for synthetic solution(100mg/L initial P, 50mg/L calcium, 0.025% seed dosage) with 0~300mg/L bicarbonate alkalinities. For synthetic solution(100mg/L initial P, 50mg/L calcium, 100mg/L bicarbonate alkalinity, 0.025% seed dosage), the phosphorus concentration was examined with $10{\sim}35^{\circ}C$. In addition, calcinated eggshell was injected to swine wastewater to test the applicability to actual wastewater.
