The effects of ethanol extract from Lepidolaena clavigera (L. clavigera) on antifungal activity were investigated. The crude ethanol extract of L. clavigera inhibited the growth of the Gram positive bacterium Bacillus subtilis ATCC 19659, (1 mm inhibition zone at 150 ${\mu}g/disc$) and the dermatophytic fungus Trichophyon mentagrophyes ATCC 28185, (2 mm inhibition zone at 150 ${\mu}g/disc$), and cytotoxic to P388 murine leukaemia cells ATCC CCL 46 P388D1, (IC50 >12,500 ${\mu}g/mL\;at\;150\;{\mu}g/disc$) and cytotoxic to BSC monkey kidney cells (@ 5 mg/mL, 150 ${\mu}g/disc$; ++: 50% activity). We suppose that this crude ethanol extract of L. clavigera is the antifungal activity.
The effects of chloroform extract from Riccardia marginata on antifungal activity were investigated. The crude chloroform extract of R. marginata inhibited the growth of the Gram positive bacterium Bacillus subtilis ATCC 19659, (4 mm inhibition zone at $150\;{\mu}g/disc$) and the dermatophytic fungus Trichophyton mentagrophytes ATCC 28185, (6 mm inhibition zone at $150\;{\mu}g/disc$), and inactive to P388 murine leukaemia cells ATCC CCL 46 P388D1, ($IC_{50}\;>25,000\;{\mu}g/mL$ at $150\;{\mu}g/disc$). This crude chloroform extract of R. marginata showed strong antifungal activity against the dermatophytic fungus Trichophyton mentagrophytes.
Purpose: To evaluate the antimicrobial persistence of silver diamine fluoride (SDF) and silver fluoride (AgF) on Streptococcus mutans. Materials and Methods: An in vitro experiment was conducted to observe changes in the diameter of the inhibition zone of various materials, including AgF (Riva Star AquaTM step 1; SDI), potassium iodine (Riva star aquaTM step 2; SDI), Fluor protector® (FP, Ivoclar Vivadent), SDF (Riva starTM step 1; SDI), Ampicillin (Sigma-Aldrich), Amphotericin B (Nexstar) and negative control on S. mutans. Result: SDF, AgF and FP exhibited significant antimicrobial persistence over the 4 weeks period (P<0.05). At day 28, the diameter of inhibition zone was larger in SDF than in AgF. Conclusion: SDF and AgF have significant antibacterial durability against bacteria commonly associated with dental caries, with the antimicrobial effect lasting for at least 4 weeks. Further clinical studies are needed to validate these findings in vivo.
The anti-Helicobacter pylori activity, cytotoxicity, and anti-inflammatory activity of white ginseng extract (WGE) were investigated in vitro in this study. The antimicrobial effects of WGE toward H. pylori strains 52 J99, SSI, and 51 were tested using the disk diffusion method. Among these H. pylori strains, H. pylori 52 was the most sensitive, having the largest inhibition zone (19 mm), followed by J99, SSI, and 51. The zone of inhibition due to WGE increased significantly with increasing dosage. The cytotoxicity of WGE toward the human cancer cell lines A-549 (human lung carcinoma), HEC-1-B (human endometrial adenocarcinoma), HeLa (human uterin adenocarcinoma), and SW-156 (human kidney carcinoma) was measured using the 3-(4,5-dimethylthizol-2-yl)-2,5-diphenylate-tetrazolium bromide (MTT) assay. WGE exhibited an inhibitory effect on cell growth at 2.0 mg/mL for all tumor cell lines. An analysis of anti-inflammatory activity using the RAW 264.7 cell line showed that the inhibition of nitric oxide (NO) production increased as the WGE content increased. These results demonstrate the potential of WGE to be used as a health-promoting substance.
Objectives : This experimental study was performed to investigate the efficacy of Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution manufactured with freezing dryness method to use eye drop. Methods : After administering Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis, MIC(Minimum Inhibition Concentration) and the size of inhibition zone were measured. Anti-bacterial potency was also measured using the size of inhibition zone. Results : There was no response to MIC on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) after Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution was medicated. Conclusions : The present study suggests that Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution dosen't have anti-bacterial effects on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis. These study result recommends that more research on other herbal medicines of eye drop for Keratitis are required.
Objectives : This experimental study was performed to investigate the effect of Fel Ursi & Bovis Calculus pharmacopuncture solution(FUBCPS) manufactured by using alcohol/water extraction method for identify ing the use of it as eyedrops. Methods: FUBCPS was manufactured by using alcohol/water extraction method. Measure the size of inhibition zone and MIC(Minimum Inhibition Concentration) after administering FUBCPS on bacterial species. Staphylococcus aureus. Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum and Candida albicans, which cause keratitis. Administering cravit(Levonoxacin medicine) on bacterial species also performed to compare the anti-bacterial potency of this material, measured by using the size of inhibition zone Results : After administering FUBCPS on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans). there was no response to MIC and there was no anti -bacterial potency also. Conclusions : This study suggests that FUBCPS dose not have anti-bacterial effects on bacterial species which cause Keratitis. These study result recommends that we need to research more on herbal medicines of eye drop which have anti-bacterial effects on keratitis
To select active bacterial strains to control plant diseases, 57 bacterial strains were isolated from the rhizosphere of the plants growing in various areas such as coast, middle and top of Halla Mountain in Jeju Island. Anti-fungal effect of isolated bactrial strains was tested in vitro by incubating in potato dextrose agar with isolates of four fungal plant pathogens Rhizoctonia solani, Fusarium oxysporum, Colletotrichum gloeosporioides and C. orbiculare, respectively. Thirty-four bacterial strains inhibited the hyphal growth of the plant pathogens, from which 17 strains inhibited one of the tested fungi, 10 strains two fungi, six strains three and a strain TRL2-3 inhibited all of the tested fungi. Some bacterial strains could inhibit weakly the hyphal growth of the plant pathogens, whereas some did very strongly with apparent inhibition zone between the plant pathogens and bacterial strains indicating the unfavorable condition for hyphal growth. Although there was no apparent inhibition zone, some bacterial strains showed a strong suppression of hyphal growth of plant pathogens. Especially, the inhibition by TRL2-3 was remarkably strong in all cases of the tested plant pathogens in this study that could be a possible candidate for biological control of various plant diseases.
We isolated marine actinomycetes, strain D-5 which produces anti-methicillin resistant Staphylococcus aureus (anti-MRSA) compound. Streptomyces sp. D-5 relatively grew well in the 20~25℃, pH 8.0, and NaCl 3.0%. The ethyl acetate extract of D-5 culture was separated by C18 ODS open column and reverse phase HPLC to yield anti-MRSA compound. The molecular weight of this compound was determined to be 898 by a Liquid chromatograph-mass spectrometer (LC-MS). Compared with penicillin G, this compound showed significant anti-MRSA activity. It also exhibited an inhibition zone of 26 mm at a concentration of 64 ㎍/disk and an inhibition zone of 16 mm at a concentration of 16 ㎍/disk against the MRSA KCCM 40511. Furthermore, the co-treatment of HPLC peak 5 compound and vancomycin caused a more rapid decrease in MRSA cells than each compound alone. It showed 86.8% growth inhibition activity within 12 hours at a low concentration of 50 ㎍/mL during co-treatment, and 97.1% growth in-hibition activity within 48 hours against MRSA KCCM 40511. Taken together, our results suggest that Streptomyces sp. D-5 and its anti-MRSA compound could be employed as a potent agent in MRSA infection.
Helicobacter pylori에 대한 항균효과는 열수추출물에서는 Fomitopsis pinicola가 15 mm의 생육저해환으로 가장 높았으며 그 다음으로는 Codyceps militaris, Coriolus versicolor, Phellinus linteus, Fomitella traxinea가 비교적 높은 항균력을 나타내었다. 열수 추출물들의 pH가 낮았으나 항균력이 없는 것으로 보아서 버섯추출물의 수소이온 농도는 항균력에 영향을 미치지 않는 것으로 여겨졌다. Methanol 추출물에서도 Fomitopsis pinicola가 Helicobacter pylori에 대한 생육저해환의 크기가 44 mm로 가장 높은 항균력을 보였다. 이미 알려진 바와 같이 버섯의 주성분은 당과 단백질이며 주로 단백다당체를 이루고 있다. 이를 구성하는 성분 및 함량에 따라 다른 기작을 보이는 갓으로 추정되며 결합하고 있는 형태도 기작에 영향을 끼치는 것으로 사료된다. 본 실험을 통해 Fomitopsis pinicola methanol 추출물이 DEAE-Sepadex의 초기 분획에서 Helicobacter pylori에 대한 항균효과를 나타냈다. 이 분획은 당과 단백질의 함량이 높게 나타났다. Fomitopsis pinicola, Codyceps militaris는 Helicobacter pylori에 대한 항균력이 뛰어나 기능성식품에 이용될 수 있을 것으로 기대된다.

Minimal A agar medium에서는 tabtoxin 검색에 Agrobactcrium tumefaces가 가장 유용한 균주였으며 minimal glucose agar m$\varepsilon$dium 에서도 역시 마찬가시의 견과를 얻였다 Complex agar medium 에서는 사용된 모든 지시균주에 대해 증식저지환이 형성되지 않아, tabtoxin이 생성되지 않음알수 있었다. 배양온도에 따른 tabtox의 생성능은 $20^{\circ}C$ 빛 $30^{\circ}C$ 에서 최적이였으며 배양시간이 경과함에 따라 tabtoxin 생성량이 증가 하였다. Glutamine을 minimal glucose agar medium에 첨가하여 tabtoxin 에 대한 지시 균주의 반응은 첨 가한 glutamine 의 양이 증가할수록 tabtoxin에 의한 생육억제가 감소함을 알 수 있었다.
