제주 수산가공부산물 유래 기능성 소재 탐색 (Development of Bioactive Substances from Fishery Processing by-products in Jeju)

  • In this study, we investigated the bioactive substances of the Alcalase hydrolysate obtained from fishery processing by-products in Jeju by measuring bioactivities including radical scavenging acitivty, cytoprotective activity against 2,2-azobis-(2-amidino-propane) dihydrochloride (AAPH), and ACE inhibitory activity. This study is important because of utilization of unused fishery processing by-products in Jeju. The Alcalase hydrolysate was prepared through the hot water extraction and enzymatic hydrolysis, and then further separation of the Alcalase hydrolysate was performed by ultrafiltration using 10 kDa molecular weight cut-off membrane. The Alcalase hydrolysate showed the relatively higher DPPH and peroxyl radical scavenging activity ($IC_{50}$ value; 1.30 mg/ml and 0.888 mg/ml, respectively). Also, the Alcalase hydrolysate showed the ACE inhibitory activity with 1.87 mg/ml of $IC_{50}$ value. These biological activities are increased over 1.2 or 2.5 times through the ultrafiltration of the Alcalase hydrolysate. Therefore, the Alcalase hydrolysate obtained from fishery processing by-products in Jeju and the different molecular weight fractions should be given consideration for food and cosmetics ingredient. Furthermore, this research on the utility of fishery processing by-products might be a useful tool into the industry.

효소를 이용한 동물성 식품가공부산물의 식품 소재화 탐색 (Study on Utilization of Animal By-products from Food Processing by Enzyme Treatment)

  • 식품 가공의 부산물 중 동물성 부산물을 재활용하여 조미소재로의 개발 가능성을 탐색하였다. 부산물 원료로는 돼지뼈(PB)와 닭뼈(CB), 그리고 참치 혈합육(TDF)을 사용하였다. PB와 CB를 열수로 18시간 추출한 후 단백질과 지방 분해효소를 처리하여 가수분해물을 제조하였다. 각각의 열수 추출물에서 지방의 제거를 위하여 1차로 lipase활성이 있는 pancreatic enzyme을 처리하고 2차로 Flavourzyme을 사용하는 경우 가수분해도(DH)가 PB는 약 70%, CB는 약 80%까지 증가하였다. 지방이 적은 TDF는 단백질 분해효소인 Alcalase와 Flavourzyme으로 분해한 결과 고형분 회수율은 약 22%, 단백질 회수율은 9%이었으며, 효소분해액 중 유리 아미노산 함량은 고형분 중 27%까지 향상되었다. 세 종류 분해액에 가염한 후 관능적인 특성을 조사한 결과 전체적으로 TDF>PB>CB의 순으로 우수하여 참치 혈합육은 조미소재의 원료로 적합한 동물성 부산물로 판단되었다.

버섯 병재배 배지재료의 이화학성과 활용 (Physico-chemical Characteristics and Utilization of Raw Materials for Mushroom Substrates)

  • 본 시험은 버섯 병재배 농가에서 사용하고 있는 배지재료와 혼합배지의 수분함량, pH, 전탄소(T-C), 전질소(T-N), C/N율, 인산, 13종의 양이온 함량 등 이화학적 특성을 파악하여 기초자료로 활용하고자 수행하였다. 전국의 병재배 농가에서 임의로 선정한 35농가 중 팽나무버섯 13개소, 큰느타리버섯 14개소, 느타리버섯 8개소에서 배지재료 143점과 혼합배지 35점의 시료가 수집되었다. 배지재료의 성분함량은 동일한 명칭의 재료라도 2~5배로 차이가 많은 경우도 있었다. 그리고 농가에서 수집한 버섯재배용 혼합배지의 C/N율은 팽나무버섯과 큰 느타리버섯 배지가 $28{\pm}4$, 느타리버섯 병재배용 배지는 $21{\pm}2$의 범위이었으며 중금속 등 특정성분에 특이사항은 없었다. 그리고 배지재료별 성분함량표를 이용하여 버섯 재배용 혼합배지의 성분조성을 산출하기 위한 계산식을 제시하였다.

넙치(Paralichthys olivaceus) 사료 내 어유(Fish oil) 대체원으로써의 diacylglycerol 이용성 평가 (Evaluation of Diacylglycerol as an Alternative to Dietary Fish Oil in Diets for Juvenile Olive Flounder Paralichthys olivaceus)

  • The objective of this study was to investigate the utilization of diacylglycerol (DAG) as a new dietary ingredient replacing fish oil in feed for juvenile olive flounder Paralichthys olivaceus. Fish oil based control diet (CON) was prepared and four other diets were formulated by replacing 50% of the fish oil in CON with one of five DAG: DAGL (1,3-lauryl glycerol) or DAGP (1,3-palmityl glycerol) in low or high concentrations (designated as DAGLL, DAGLH, DAGPL and DAGPH). Another diet was prepared replacing 100% of the fish oil in CON with a 1:1 mixture (DAGLP) of DAGL and DAGP. Olive flounder (13.4 g) were fed to apparent satiation, twice a day, for 12 weeks. Following the feeding trials, no significant differences were observed in growth performance, blood parameters and non-specific immune responses between CON and any of the DAG groups. Polyunsaturated fatty acid levels were not significantly affected by the inclusion of DAGs. Thus, DAGL or DAGP could be used to replace up to 50% of fish oil in fish feed without reducing growth performance, health or innate immunity. The replacement of up to 100% of dietary fish oil in olive flounder feed by DAGLP is also feasible.

Optimization and production of protein hydrolysate containing antioxidant activity from tuna cooking juice concentrate by response surface methodology

  • To optimize the hydrolysis conditions in the production of antioxidant hydrolysates from tuna cooking juice concentrate (TC) to maximize the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, TC containing 48.91% protein was hydrolyzed with Alcalase 2.4 L, and response surface methodology (RSM) was applied. The optimum hydrolysis conditions included a 2.2% (w/v) Alcalase concentration and 281 min hydrolysis time, resulting in the highest DPPH radical scavenging activity of 66.49% (0.98 µmol Trolox/mg protein). The analysis of variance for RSM showed that hydrolysis time was an important factor that significantly affected the process (p < 0.05). The effects of different drying methods (freeze drying, hot air drying, and vacuum drying) on the DPPH radical scavenging activity and amino acid (AA) profiles of TC hydrolysate (TCH) were evaluated. Vacuum-dried TCH (VD) exhibited an increase in DPPH radical scavenging activity of 81.28% (1.20 µmol Trolox/mg protein). The VD samples were further fractionated by ultrafiltration. The AA profiles and antioxidant activities in terms of the DPPH radical scavenging activity, 2,2'-azino-bis(3-ethylbenzthiazoline)-6-sulfonic acid (ABTS) radical scavenging activity, ferric reducing antioxidant power, and ferrous ion chelating activity were investigated. Glutamic acid, glycine, arginine, and cysteine were the major AAs found in the TCH fractions. The highest DPPH radical scavenging activity was found in the VD-1 fraction (< 5 kDa). The VD-3 fraction (> 10 kDa) exhibited the highest ABTS radical scavenging activity and ferric reducing antioxidant power. The ferrous ion chelating activity was the highest in VD-1 and VD-2 (5 to 10 kDa). In conclusion, this study provided the optimal conditions to obtain high antioxidant activities through TCH production, and these conditions could provide a basis for the future application of TCH as a functional food ingredient.

Apparent digestibility coefficients of animal feed ingredients for olive flounder (Paralichthys olivaceus)

  • Apparent digestibility coefficients (ADCs) of dry matter, crude protein, crude lipid, nitrogen-free extract, energy and essential amino acids in animal-based feed ingredients were determined for olive flounder (Paralichthys olivaceus). A reference diet (RF) was formulated to contain 1.0% chromic oxide (Cr2O3) as an inert indicator. Nine test diets were formulated to contain RF and one of the feed ingredients (pollock meal [PM], jack mackerel meal [JMM], anchovy meal [AM], cod meal [CM], sardine meal [SM], sand eel meal [SEM], tuna meal [TM], meat meal [MM] and squid liver meal [SLM]) at a 7:3 ratio in each diet designated as PM, JMM, AM, CM, SM, SEM, TM, MM and SLM, respectively. Olive flounder, averaging 150 ± 8.0 g, were stocked at a density of 25 fish per tank in 400-L fiberglass tanks attached with fecal collection columns. Feces were collected from triplicate groups of fish one time a day for four weeks. Dry matter and crude protein ADCs of CM and SEM were significantly higher than the other tested ingredients. Lipid ADCs of JMM, CM and SEM were significantly higher than the other test ingredients. Energy ADCs of CM and SEM were significantly higher than that of the other tested ingredients. The availability of amino acids in CM was generally higher than the other animal protein sources. PM exhibited the lowest amino acid availability among the treatments. Interestingly, MM exhibited significantly higher nutrient digestibility than several marine-based ingredients. However, CM and SEM are seeming to be highly digestible and effective to use in olive flounder diet compared to the other tested ingredients. Overall, the results of this study provide information about the bioavailability of nutrients and energy in animal feedstuffs to apply when formulating cost-effective practical feeds for olive flounder.

Effects of biochar-based fertilizer on ammonia volatilization under controlled conditions

  • As the interest in sustainable and environmentally friendly agriculture continues to grow, there is a corresponding increase in organic fertilizers utilization. However, studies on ammonia (NH3) emissions, which are primarily generated in the agricultural sector, by organic fertilizers are lacking. Additionally, the reliance on imported ingredients in the production of organic fertilizers hinders the widespread adoption of organic fertilizers. This study aims to evaluate NH3 volatilization by incorporating rice husk biochar into organic fertilizers. The study also aims to assess whether domestically produced rice husk biochar can serve as a viable substitute for imported ingredients. Here, the dynamic chamber method was used under controlled conditions. Results show that inorganic fertilizers readily undergo hydrolysis, thereby rapidly generating significant amounts of NH3, particularly in the initial stages. In contrast, organic fertilizers decompose gradually, leading to relatively long-term NH3 emissions. The incorporation of rice husk biochar into organic fertilizers demonstrated diminished daily NH3 emissions compared to those from commercial organic fertilizers, resulting in decreased total NH3 volatilization. These findings show that the combination of rice husk biochar can reduce NH3 volatilization and serve as an alternative to imported ingredients for organic fertilizers. The results of this study can be utilized as fundamental information for the assessment of biochar as a potential ingredient for organic fertilizers.

피부노화 방지에 이용되는 천연물의 종류 및 추출연구 동향 (Trends in Extraction Research and Types of Natural Substances Used for Skin Aging Prevention)

  • 노인인구 증가와 피부미용에 관해 관심이 커지고 천연물에 대한 과학적 고찰의 전문화는 천연소재에 대한 활용 증가로 이어지고 있다. 본 논문은 천연원료를 피부노화 경감 소재로 활용한 문헌의 추출법 및 경감능을 기반으로 천연물의 종류, 추출물의 기능성 및 추출 기술을 조사하였다. 천연물 유래 기능성 소재의 피부노화 경감에 대한 직접적 영향을 평가하기 위해 콜라겐에 미치는 영향 중 Procollagen 합성능과 MMP-1 경감능을 기반으로 추출물의 기능성을 판단하였다. 각 천연물은 위의 평가법을 이용한 문헌 중 식물, 한약재 및 녹조류로 구분하여 각 원료로부터 기능성 소재의 확보를 위한 추출 기술과 주요 결과를 서술한다. 이에 따라 피부노화 완화를 위한 기능성 소재의 추출 기술과 연구 동향을 제공하여 천연물 활용 연구 분야에 신속한 접근을 제공한다.

Evaluation of Amino Acid and Energy Utilization in Feedstuff for Swine and Poultry Diets

  • An accurate feed formulation is essential for optimizing feed efficiency and minimizing feed cost for swine and poultry production. Because energy and amino acid (AA) account for the major cost of swine and poultry diets, a precise determination of the availability of energy and AA in feedstuffs is essential for accurate diet formulations. Therefore, the methodology for determining the availability of energy and AA should be carefully selected. The total collection and index methods are 2 major procedures for estimating the availability of energy and AA in feedstuffs for swine and poultry diets. The total collection method is based on the laborious production of quantitative records of feed intake and output, whereas the index method can avoid the laborious work, but greatly relies on accurate chemical analysis of index compound. The direct method, in which the test feedstuff in a diet is the sole source of the component of interest, is widely used to determine the digestibility of nutritional components in feedstuffs. In some cases, however, it may be necessary to formulate a basal diet and a test diet in which a portion of the basal diet is replaced by the feed ingredient to be tested because of poor palatability and low level of the interested component in the test ingredients. For the digestibility of AA, due to the confounding effect on AA composition of protein in feces by microorganisms in the hind gut, ileal digestibility rather than fecal digestibility has been preferred as the reliable method for estimating AA digestibility. Depending on the contribution of ileal endogenous AA losses in the ileal digestibility calculation, ileal digestibility estimates can be expressed as apparent, standardized, and true ileal digestibility, and are usually determined using the ileal cannulation method for pigs and the slaughter method for poultry. Among these digestibility estimates, the standardized ileal AA digestibility that corrects apparent ileal digestibility for basal endogenous AA losses, provides appropriate information for the formulation of swine and poultry diets. The total quantity of energy in feedstuffs can be partitioned into different components including gross energy (GE), digestible energy (DE), metabolizable energy (ME), and net energy based on the consideration of sequential energy losses during digestion and metabolism from GE in feeds. For swine, the total collection method is suggested for determining DE and ME in feedstuffs whereas for poultry the classical ME assay and the precision-fed method are applicable. Further investigation for the utilization of ME may be conducted by measuring either heat production or energy retention using indirect calorimetry or comparative slaughter method, respectively. This review provides information on the methodology used to determine accurate estimates of AA and energy availability for formulating swine and poultry diets.

전나무 정유의 항진균 효과와 유효성분의 시너지효과 평가 (Evaluation on Anti-fungal Activity and Synergy Effects of Essential Oil and Their Constituents from Abies holophylla)

  • 본 연구는 전나무 정유의 Epidermophyton floccosum, Trichophyton mentagrophytes, Trichophyton rubrum과 같은 피부사상균에 대한 항진균 활성을 구명하고 피부염 치료제로서의 전나무 정유의 잠재력을 평가하고자 하였다. 전나무 정유와 분획물, 유효성분의 표준시약을 이용하여 한천희석법, paper disc 확산법, TEM을 이용한 세포 변화 관찰 등을 통해 항진균 활성을 평가하였고, 주성분은 GC-MS로 분석하였다. 유효성분들의 시너지효과는 체커보드법을 통해 평가하였다. 그 결과, 가장 높은 활성을 나타낸 물질은 ${\alpha}$-bisabolol을 함유하고 있는 분획 G4였으며 다른 분획 보다 뛰어나게 높은 활성을 나타냈다. 체커보드법을 통해 ${\alpha}$-bisabolol과 bornyl acetate를 병용시 뛰어난시너지 효과를 나타냈고 세포 변화 관찰을 통해 분획 G4에 노출된 E. floccosum과 T. rubrum 균사는 정상세포와 달리 세포막과 세포소기관이 파괴됨이 관찰되었다. 따라서 전나무 정유와 그 구성성분들은 높은 항진균 효과를 가지고 있기 때문에 피부염 치료제의 원료로 이용이 가능할 것이라 기대되며, 특히 ${\alpha}$-bisabolol의 우수한 항진균 활성에 주목할 필요가 있다고 사료된다.
