Prevalence of Canine Influenza Infection in Pet Dogs and Canine Parvovirus Infection in Street Dogs of Bangladesh

  • A cross-sectional study was conducted to investigate the prevalence of canine influenza (CI) infection in pet dogs and canine parvovirus (CPV) infection in street dogs of different age and sex by collecting rectal and nasal swab samples from three districts, Dhaka, Mymensingh and Sirajgonj, in Bangladesh using a RapiGEN$^{(R)}$ Canine Influenza Virus Ag Test kit and RapiGEN$^{(R)}$ Canine Parvovirus Ag Test Kit. Out of 114 rectal swabs and 115 nasal swab samples, the overall prevalence of CI and CPV was found to be 11.30% and 32.45%, respectively. The prevalence of parvovirus infection was found to be significantly higher in puppies and dogs 6 months of age (50.0%) than those > 24 months of age (p = 0.005). The prevalence was also higher in males (34.42%) than females (30.18%). The prevalence of CI was higher (30.43%) in dogs up to 6 months of age (p = 0.011) than 6-12 month (7.93%) and 12-18 month (6.66%) old dogs. Moreover, the prevalence of CI was found to be higher in males (16.10%) than females (5.66%). The prevalence of CPV infection also varied significantly in different study areas (p = 0.0029), with 12.72%, 12.5% and 7.14% of dogs found to be CI positive in Dhaka, Mymensingh and Sirajgonj, respectively. Overall, the highest prevalence of CI was found in local breeds (6.08%) followed by German shepherds and Keeshonds (1.73%), and Bloodhounds and Terriers (0.86%). Additionally, there were more positive CI found in unvaccinated dogs (14.81%) than vaccinated (2.94%) dogs. Dogs with flu-like symptoms were more positive (19.23%) for CI relative to those without flu-like symptoms (4.76%) samples. Overall, the results of this study indicate that canine vaccination should be initiated to prevent the occurrence of diseases and that regular monitoring should be continued in Bangladesh.

인간 코로나 바이러스를 중심으로 2017-2019년 소아청소년 호흡기 바이러스 감염증의 역학 및 특성: 경기 북서부지역 단일기관의 후향적 연구 (Epidemiology and Characteristics of Pediatric Respiratory Virus Infection From 2017 to 2019 Focusing on Human Coronavirus: A Retrospective Study of a Single Center in Northwestern Gyeonggi-do)

  • 목적: Coronavirus disease 2019 (COVID-19) 발생 이후 인간 코로나바이러스를 포함한 호흡기 바이러스의 패턴에 변화가 있을 것으로 생각되어, COVID-19 발생 이전 경기도 명지병원에 급성 호흡기 감염증으로 입원한 소아청소년을 대상으로 바이러스 감염의 패턴을 보고하였다. 방법: 2017년 1월부터 2019년 12월까지 급성 호흡기 감염증으로 명지병원에 입원한 18세 이하 소아청소년을 대상으로 하였고 real-time reverse transcription polymerase chain reaction (RT-PCR) 결과를 바탕으로 후향적으로 의무기록을 고찰하였다. 결과: 총 3,557명 중에서 중복감염 포함하여 3,686건의 바이러스가 검출되었고 평균 양성률은 78.6%이었으며 여름에 비해 겨울에 PCR 양성률이 높았다. 파라인플루엔자바이러스, 메타뉴모바이러스, 보카바이러스는 4-6월에 주로 검출되고, 인간엔테로바이러스는 7-9월에, 호흡기세포융합바이러스와 인플루엔자바이러스는 겨울철에 주로 검출되었다. 진단별로 인두편도염에서는 아데노바이러스가 가장 많았고 폐렴에서는 호흡기세포융합바이러스가 가장 많이 검출되었다. 인간 코로나 바이러스는 겨울철에 주로 검출되었으며, 크루프 환자 중에서 3번째 높은 빈도로 나타났다. 결론: 본 연구결과가 이전에 보고되었던 소아청소년의 호흡기 바이러스 역학과 크게 다르지 않았다. 또한 COVID-19 발생 이후 호흡기 바이러스 패턴 비교 연구에 도움이 되고자 COVID-19 발생 이전 호흡기 바이러스 역학정보를 보고하는 바이다.

Enhancement of Antigen-specific Antibody and $CD8^+$ T Cell Responses by Codelivery of IL-12-encapsulated Microspheres in Protein and Peptide Vaccination

  • Background: Although IL-12 has been widely accepted to playa central role in the control of pathogen infection, the use of recombinant IL-12 (rIL-12) as a vaccine adjuvant has been known to be ineffective because of its rapid clearance in the body. Methods: To investigate the effect of sustained release of IL-12 in vivo in the peptide and protein vaccination models, rIL-12 was encapsulated into poly ($A_{DL}$-lactic-co-glycolic acid) (PLGA). Results: We found that codelivery of IL-12-encapsulated microspheres (IL-12EM) could dramatically increase not only antibody responses, but also antigen-specific $CD4^+\;and\;CD8^+$ T cell responses. Enhanced immune responses were shown to be correlated with protective immunity against influenza and respiratory syncytial virus (RSV) virus challenge. Interestingly, the enhancement of $CD8^+$ T cell response was not detectable when $CD4^+$ T cell knockout mice were subjected to vaccination, indicating that the enhancement of the $CD8^+$ T cell response by IL-12EM is dependent on $CD4^+$ T cell "help". Conclusion: Thus, IL-12EM could be applied as an adjuvant of protein and peptide vaccines to enhance protective immunity against virus infection.

Sequence analysis of the fusion protein gene of Newcastle disease virus isolated from breeder ducks in Korea

  • Newcastle disease (ND) is an infectious poultry disease that caused high mortality and reduced egg production. NDVs are regularly present in the domestic duck population. And ducks play a possible role in the maintenance and transmission of NDVs. While we were monitoring the Avian Influenza, NDVs were isolated from field samples by accident. So we analysed the biological and genetic characteristics of these viruses. Lentogenic NDVs were isolated from two farms among twenty breeder duck farms. The ages of ducks were 39 weeks old in the 'A' farm and 3~72 weeks old in the 'B' farm. And they were not inoculated with the NDVs vaccine. In the biological characteristics, the both viruses which separated from the farm 'A' and 'B' were thermostable. The amino acid sequence of a site from 112 to 119 in the fusion (F) protein was 'GKQGRLIG' which has monobasic motif in the samples of both farms. And this means the separated NDVs are lentogenic. Phylogenetic analysis was performed by entire nucleotide sequence of F protein. The virus strains from the A farm (MN095239) and the B farm (MN095240) belonged to class II genotype I. Using the analysis of whole F protein nucleic acid sequence, the MN095239 (GenBank) had homology with Ulster strain about 99.95% and the MN095239 (GenBank) had homology with KR/CK/KU_LBM255/09 strain about 99.89%. NDV surveillance is needed to investigate epidemiological relationship of domestic breeder duck isolates in Korea.

Insights of window-based mechanism approach to visualize composite biodata point in feature spaces

  • In this paper, we propose a window-based mechanism visualization approach as an alternative way to measure the seriousness of the difference among data-insights extracted from a composite biodata point. The approach is based on two components: undirected graph and Mosaab-metric space. The significant application of this approach is to visualize the segmented genome of a virus. We use Influenza and Ebola viruses as examples to demonstrate the robustness of this approach and to conduct comparisons. This approach can provide researchers with deep insights about information structures extracted from a segmented genome as a composite biodata point, and consequently, to capture the segmented genetic variations and diversity (variants) in composite data points.

Novel reassortant 2.3.4.4B H5N6 highly pathogenic avian influenza viruses circulating among wild, domestic birds in Xinjiang, Northwest China

  • Background: The H5 avian influenza viruses (AIVs) of clade 2.3.4.4 circulate in wild and domestic birds worldwide. In 2017, nine strains of H5N6 AIVs were isolated from aquatic poultry in Xinjiang, Northwest China. Objectives: This study aimed to analyze the origin, reassortment, and mutations of the AIV isolates. Methods: AIVs were isolated from oropharyngeal and cloacal swabs of poultry. Identification was accomplished by inoculating isolates into embryonated chicken eggs and performing hemagglutination tests and reverse transcription polymerase chain reaction (RT-PCR). The viral genomes were amplified with RT-PCR and then sequenced. The sequence alignment, phylogenetic, and molecular characteristic analyses were performed by using bioinformatic software. Results: Nine isolates originated from the same ancestor. The viral HA gene belonged to clade 2.3.4.4B, while the NA gene had a close phylogenetic relationship with the 2.3.4.4C H5N6 highly pathogenic avian influenza viruses (HPAIVs) isolated from shoveler ducks in Ningxia in 2015. The NP gene was grouped into an independent subcluster within the 2.3.4.4B H5N8 AIVs, and the remaining six genes all had close phylogenetic relationships with the 2.3.4.4B H5N8 HPAIVs isolated from the wild birds in China, Egypt, Uganda, Cameroon, and India in 2016-2017, Multiple basic amino acid residues associated with HPAIVs were located adjacent to the cleavage site of the HA protein. The nine isolates comprised reassortant 2.3.4.4B HPAIVs originating from 2.3.4.4B H5N8 and 2.3.4.4C H5N6 viruses in wild birds. Conclusions: These results suggest that the Northern Tianshan Mountain wetlands in Xinjiang may have a key role in AIVs disseminating from Central China to the Eurasian continent and East African.

소아에서 B형 인플루엔자 바이러스의 감염에서 oseltamivir 치료의 유용성: 2개 병원에서 조사된 환자들을 대상으로 (Clinical Characteristics of Influenza B Virus in Children and the Efficacy of Oseltamivir: Data from Two University Hospitals)

  • 목적: 본 연구는 인플루엔자 B 바이러스에 대한 oseltamivir의 효능을 알아보고자 치료군과 비치료군간의 임상적 특징의 차이를 비교하였다. 방법: 2012년 1월부터 2012년 7월까지 입원하거나 외래 진료실을 방문하여 인플루엔자 B 바이러스에 감염된 소아 환자를 대상으로 후향적으로 의무기록을 분석하였다. 결과: 인플루엔자 B 바이러스에 의한 폐렴, 급성 기관지염, 급성세기관지염, 크룹으로 진단된 72명의 평균 나이는 $3.6{\pm}2.8$세 였다. oseltamivir 치료군은 38명(53%), 비치료군은 34명(47%)이었고 치료 시기를 고려하지 않았을 때 두 군의 입원 기간($4.18{\pm}2.10$일 vs $4.79{\pm}1.49$일, P=.17), 총 발열 기간($5.32{\pm}2.07$일 vs $6.41{\pm}3.25$일, P=.09)을 포함한 임상적 양상에 유의한 차이가 없었다. 비치료군, 48시간 이내 치료군, 48시간 이후 치료군간의 비교에서 임상적 양상에 유의한 차이가 없었다. 3세 이하의 환자들을 대상으로 두 군간의 비교에서도 유의한 차이가 없었다. 결론: Oseltamivir 복용 전 치료군과 비치료군간의 임상적 특성에서 유의한 차이를 나타내는 항목이 없어 oseltamivir의 적응증을 도출하기 어려웠으며 치료 후 임상 경과의 유의한 차이도 없었다. 그러나 투약 시점에 따른 비교에서는 발열 시작 48시간 이내에 osetamivir를 투여한 군에서 발열 기간이 감소하는 경향을 보여주었으므로 향후 투약 시기를 고려하여 연령별 분석, 인플루엔자 B바이러스 종에 대한 분석을 포함하는 대단위 연구가 필요하다.

한탄바이러스 Nucleocapsid Protein 발현에 있어 S Genome 내 Noncoding Region의 역할 (The Role of Noncoding Region in Hantaan Viral S Genome for Expression of Nucleocapsid Protein)

  • The genome of Hantaan virus, the prototype of the hantavirus genus, is composed of three segmented, single stranded negative sense RNA genome. The 5' and 3' termini of the Hantaan virus RNA genome contain noncoding regions (NCRs) that are highly conserved and complementary to form panhandle structures. There are some reports that these NCRs seems to control gene expression and viral replication in influenza virus and vesicular stomatitis virus. In this study, we examined whether NCRs in Hantaan virus playa role in expression of the viral nucleocapsid protein (Np) and foreign (luciferase) gene. The 5' and/or 3' NCR-deleted mutants were constructed and analysed. The Np expression of 5' NCR-deleted clone was similar to that of the clone containing full S genome. In the case of 3' NCR-deleted clone, it showed 40% reduction. To investigate the role of NCR in foreign gene expression, the clones which are replaced ORF of Hantaan viral Np gene with that of luciferase gene were constructed. The results were similar to those of the experiments using Np gene. These results suggest that 3' NCR is more important than 5' NCR in protein expression. To find out a critical region of 3' NCR in protein expression, several clones with a deleted part of 3' NCR were constructed and analyzed. The deletion of the conserved region in 3' NCR showed $20{\sim}30%$ decrease in Np expression. However there were no change in luciferase activities between clones with or without non-conserved region of 3' NCR. These results suggest that the 3' NCR of Hantaan virus S genome, especially conserved region in 3' NCR, plays an important role in the expression of Hantaan viral Np and foreign genes.

Ascorbic acid의 항결핵균 작용에 관한 연구 (A Study on Antibacterial Effects of Ascorbic Acid against Mycobacterium tuberculosis)

  • Vitamin C is known as an integral element for the formation and maintenance of intercellular supporting structures. Ascorbic acid has been used for the treatment of certain poisonings and hypovitaminosis(scurvy) but also known as a powerful reducing agent, and can kill a variety of bacteria and detoxify bacterial exotoxins including various clostridial exotoxins in vitro. For viruses, vitamin C inactivates herpes virus, vaccinia virus and influenza virus and has been used for the prevention and treatment of the common cold. Thus ascorbic acid plays an important role in antimicrobial action. Scurvy also promotes the development of tubercles in experimentally infected guinea pig and the tuberculosis patients require more vitamin C than normal persons. However there is no reports that ascorbic acid could inhibit the growh of M. tuberculosis. In this paper, antibacterial effects of ascorbic acid against M. tuberculosis were studied. The results are as follows: 1. The single use of the ascorbic acid exhibited antibacterial effect in vitro against $5{\times}10^3/ml$ of M. tuberculosis $H_{37}$ Rv at the concentration of ascorbic acid 0.625mg/ml over 3 hours exposure and 0.05mg/ml over 9 hours exposure. 2. In vivo mice administered with ascorbic acid 50mg/day for 5, 10 and 15 days respectively were protected from M. tuberculosis $2LD_{50}$, $3LD_{50}$, $4LD_{50}$ and $5LD_{50}$ given intravenously.

Detection rate and clinical impact of respiratory viruses in children with Kawasaki disease

  • Purpose: The purpose of this prospective case-control study was to survey the detection rate of respiratory viruses in children with Kawasaki disease (KD) by using multiplex reverse transcriptase-polymerase chain reaction (RT-PCR), and to investigate the clinical implications of the prevalence of respiratory viruses during the acute phase of KD. Methods: RT-PCR assays were carried out to screen for the presence of respiratory syncytial virus A and B, adenovirus, rhinovirus, parainfluenza viruses 1 to 4, influenza virus A and B, metapneumovirus, bocavirus, coronavirus OC43/229E and NL63, and enterovirus in nasopharyngeal secretions of 55 KD patients and 78 control subjects. Results: Virus detection rates in KD patients and control subjects were 32.7% and 30.8%, respectively (P=0.811). However, there was no significant association between the presence of any of the 15 viruses and the incidence of KD. Comparisons between the 18 patients with positive RT-PCR results and the other 37 KD patients revealed no significant differences in terms of clinical findings (including the prevalence of incomplete presentation of the disease) and coronary artery diameter. Conclusion: A positive RT-PCR for currently epidemic respiratory viruses should not be used as an evidence against the diagnosis of KD. These viruses were not associated with the incomplete presentation of KD and coronary artery dilatation.
